Surface replacement reaction of thiol-derivatized, single-stranded oligonucleotide (HS-ssDNA) by mercaptohexanol (MCH) is investigated in order to reduce surface density of the HS-ssDNA adsorbed to Au(111) surface. Cyclic voltammograms (CVs) and scanning tunneling microscopy (STM) are employed to assess the composition and state of these mixed monolayers. It is found that each CV of mixed self-assembled monolayers (SAMs) only shows a single reductive desorption peak, which suggests that the resulted, mixed SAMs do not form discernable phase-separated domains. The peak potential gradually shifts to negative direction and the peak area increases step by step over the whole replacement process. By analyzing these peak areas, it is concluded that two MCH molecules will replace one HS-ssDNA molecule and relative coverage can also be estimated as a function of exposing time. The possible mechanism of the replacement reaction is also proposed. The DNA surface density exponentially reduces with the exposing time increasing, in other words, the replacement reaction is very fast in the first several hours and then gradually slows down. Moreover, the morphological change in the process is also followed by STM.

The probability distribution of the four-phase invariants in the case of single isomorphous replacement has been developed to estimate some individual phases. An example of its application to obtain the phases having special values of 0, pi or +/-pi /2 is given for a known protein structure in space group P2(1)2(1)2(1). The phasing procedure includes the determination of starting phases and an iterative calculation. The initial values of starting phases, which are required by the formula, can be obtained from the estimate of one-phase seminvariants and by specifying the origin and enantiomorph. In addition, the calculations lead to two sets of possible phases for each type of reflection by assigning arbitrarily an initial phase value. The present method provides a possibility for the multisolution technique to increase greatly the number of known phases while keeping the number of the trials quite small.

A method for estimating the one-phase structure seminvariants (OPSSs) having values of 0 or pi has been proposed on the basis of the probabilistic theory of the three-phase structure invariants for a pair of isomorphous structures [Hauptman (1982). Acta Cryst. A38, 289-294]. The test calculations using error-free diffraction data of protein cytochrome c(550) and its PtCl42- derivative show that reliable estimates of a number of the OPSSs can be obtained. The reliability of the estimation increases with the increase of the differences between diffraction intensities of the native protein and its heavy-atom derivative. A means to estimate the parameters of the distribution from the diffraction ratio is suggested.

RNA interference (RNAi) is an evolutionarily conserved mechanism by which double-stranded RNA (dsRNA) initiates post-transcriptional silencing of homologous genes. Here we report the amplification and characterisation of a full length cDNA from black tiger shrimp (Penaeus monodon) that encodes the bidentate RNAase III Dicer, a key component of the RNAi pathway. The full length of the shrimp Dicer (Pm Dcr1) cDNA is 7629 bp in length, including a 51 untranslated region (UTR) of 130 bp, a 3' UTR of 77 bp, and an open reading frame of 7422 bp encoding a polypeptide of 2473 amino acids with an estimated molecular mass of 277.895 kDa and a predicted isoelectric point of 4.86. Analysis of the deduced amino acid sequence indicated that the mature peptide contains all the seven recognised functional domains and is most similar to the mosquito (Aedes aegypti) Dicer-1 sequence with a similarity of 34.6%. Quantitative RT-PCR analysis showed that Pm Dcr1 mRNA is most highly expressed in haemolymph and lymphoid organ tissues (P 0.05). However, there was no correlation between Pm Dcr1 mRNA levels in lymphoid organ and the viral genetic loads in shrimp naturally infected with gill-associated virus (GAV) and Mourilyan virus (P > 0.05). Treatment with synthetic dsRNA corresponding to Pm Dcr1 sequence resulted in knock-down of Pm Dcr1 mRNA expression in both uninfected shrimp and shrimp infected experimentally with GAV. Knock-down of Pm Dcr1 expression resulted in more rapid mortalities and higher viral loads. These data demonstrated that Dicer is involved in antiviral defence in shrimp. (c) 2007 Elsevier Ltd. Research related to carbon geochemistry and biogeochemistry in the East China Sea is reviewed in this paper. The East China Sea is an annual net sink for atmospheric CO, and a large net source of dissolved inorganic carbon to the ocean. The sea absorbs CO, from the atmosphere in spring and summer and releases it in autumn and winter. The East China Sea is a CO, sink in summer because Changjiang River freshwater flows into it. The net average sea-air interface carbon flux of the East China Sea is estimated to be about 4.3 X 10(6) t/y. Vertical carbon transport is mainly in the form of particulate organic carbon in spring; more than 98% of total carbon is transported in this form in surface water, and the number exceeds 68% in water near the bottom. In the southern East China Sea, the average particulate organic carbon inventory was about one-tenth that of the dissolved organic carbon. Research indicates that the southern Okinawa Trough is an important site for particulate organic carbon export from the shelf. The annual cross-shelf exports are estimated to be 414 and 106 Gmol/y for dissolved organic carbon and particulate organic carbon, respectively. Near-bottom transport could be the key process for shelf-to-deep sea export of biogenic and lithogenic particles.

Using heterogeneous vegetation in alpine grassland through grazing is a necessary component of deintensification of livestock systems and conservation of natural environments. However, better understanding of the dynamics of animal feeding behaviour would improve pasture and livestock grazing managements, particularly in the early part of the spring season when forage is scarce. The changes in behaviour may improve the use of poor pastures. Then, enhancing management practices may conserve pasture and improve animal productivity. Grazing behaviour over 24 In periods by yaks in different physiological states (lactating, dry and replacement heifers) was recorded in the early, dry and later, germinating period of the spring season. Under conditions of inadequate forage, the physiological state of yaks was not the primary factor affecting their grazing and ruminating behaviour. Forage and sward state affected yaks' grazing and ruminating behaviour to a greater extent. Generally, yaks had higher intake and spent more time grazing and ruminating during the later part of the spring season, following germination of forage, than during the earlier dry part of the season. However, the live weight of yaks was less during pasture germination than during the early dry part of the season because the herbage mass is low, and the yaks have to expend much energy to seek feed at this particular time. Expansive soil is a kind of typical unsaturated soil with characteristics of high swelling-shrinking deformation, cracks and over consolidation. It is very harmful to civil engineering, As a new processing method deal with expansive soil, Chemistry treatment has widespread applied in developed countries such as Europe and America, and also gained remarkable result. Based on the embankment filling soil improving testing projects in Meng-Xin freeway, this paper proposed a new processing method to expansive soil embankment wrapped with PAS-treated soil, experimental study of expansive soil chemical improved by PAS is been carried out. The water content change is the external factor which causes expansive soil to have swelling-shrinkage deformation. this reflected that the soil body swelling-shrinkage characteristic mainly depends on its mineral ingredient and the soil-water mutual function. This paper takes expansive soil as one kind of ordinary high plastic clay from angle of clay-water mutual function explained the expansive soil swelling-shrinkage deformation mechanism on microscopic. And take this swelling-shrinkage mechanism as the master line, Cooperates with the China Academy of Chemistry, we developed the new method PAS treatment, trough ionic exchange, joint, package and flocculation, the stronger static electricity function weakened the level through adsorption and the stronger static electricity function, PAS can weakened the negative charge repulsion between levels, causes the electric potential to reduce, diffusion layer thickness to be thinner, and improves the water affinity performance of expansive soil effectively. Moreover the space network architecture compromised with PAS and soil enhanced the joint strength between the clay particles , enable the soil body to have comparatively high strength and the distortion rate. pointed proposed the PAS modified principle. Combine with the construction of experimented road, this paper sums up and presents the construction craft and technology requirement of PAS treatment to expansive soil embankment. Through many experimental studied the basic physical property, the intensity characteristic and water stability changes of expansive soil and PAS-treated soil. The results of study indicate that adding lime into the expansive soil can reduce the content of clay gain obviously, reduce the plasticity notably, increase the strength greatly, control the property of swelling and shrinking effectively, and can meliorate the stability of sucking water clearly. Simultaneity PAS don’t change the cultivate capacity of the soil, the modified slope of the embankment can adopt plant fixed slope method as ecology protection. Finally the processing effect of use different treatment has analyzed through numerical simulation, summarized the PAS chemical wrapping treatment process in the actual project application, and appraised its processing effect and the project efficiency. The research indicated that PAS chemical treatment is one effective method to improve expansive soil. Compare with long-distance replacement, especially in the high plastic expansive soil massive distribution area, PAS treatment has the very greatly economical superiority to be promoted. The study in the paper not only afforded technique method to Meng-Xin expressway construction but also important for improvement of the expressway construction theory in swelling soil areas. Key words: PAS; expansive soil; swelling-shrinkage deformation mechanism; wrapping embankment; chemical modified treatment.

Reservoir prediction techniques from prestack seismic are among the most important ones for exploration of lithologic hydrocarbon reservoir. In this paper, we set the turbidite fan sandstone reservoir in Liao-Zhong depress as our researching target, and aims to solve the apllication difficulties on pre-stack inversion in the area, where the drilling data is scarce and the reservoir is lateral varied. Meanwhile, AVO analysis and pre-stack inversion for gas-bearing detection is systematically researched. The seismic reflection characters of gas-bearing sandstone in turbidite fan with different fluid content are defined, after analyzing results from AVO seismic simulation and porous fluid replacement of real log data, and under the guides of the seismic characters from classical gas-bearing sandstone reservoir and numerical simulation for complicate gas-bearing sandstone. It is confirmed that detecting gas-bearing sandstone in turbidite fan via AVO technologies is feasible. In terms of AVO analysis, two AVO characters, fluid detection factor and product of intercept and gradient, can effectively identify top and bottom boundaries and lateral range of tuibidite gas sand by comparing real drilling data. Cross-plotting of near and far angle stack data could avoid the correlation existing in P-G analysis. After comparing the acoustic impedance inversions with routine stacked data and AVO intercept, impedance derived from AVO intercept attribute could reduce the acoustic impedance estimating error which is caused by AVO. On the aspect of elastic impedance inversion, the AVO information in the pre-stack gathers is properly reserved by creating partial angle stack data. By the far angle elastic impedance alone, the gas sand, with abnormally low range of values, can be identified from the background rocks. The boundary of gas sand can also be clearly determined by cross-plotting of near and far angle elastic impedances. The accuracy of far angle elastic impedance is very sensitive to the parameter K, and by taking the statistical average of Vp/Vs on the targeted section in key wells, the accuracy of low frequency trends is gurranteed; the intensive absorsion within the area of the gas sand, which tends to push the spectral of seismic data to the lower end, will cause errors on the inversion result of elastic impedance. The solution is to confine the inversion on the interested area by improving the wavelet. On the aspect of prestack AVA simultaneous inversion, the constraint of local rock-physical trends between velocities of P-wave、S-wave and density successfully removes the instability of inversion, thus improves the precision of the resulting elastic parameters. Plenty of data on rock properties are derived via AVO analysis and prestack seismic data inversion. Based on them, the fluid anomaly is analysized and lithological interpretation are conducted. The distribution of gas sand can be consistently determined via various of ways, such as cross-plotting of P and G attributes, near and far partial angle stack data, near and far angle elastic impedances, λρ and Vp/Vs, etc. The shear modulo and density are also reliable enough to be used for lithological interpretation. We successfully applied the AVO analysis and pre-stack inversion techniques to gas detecting for turbidite fan sand reservoir in Liao-Zhong depression.
