由于人为因素导致的全球范围内的第六次物种大灭绝已经成为不争的事实,但人类还不清楚这种现象对生态系统功能的影响。在回答生物多样性与生态系统功能关系的问题上,补偿作用是一个争论的热点。为了阐明生物多样性对生态系统功能的影响,于2005年夏开始,在内蒙古温带典型草原开展了一个研究生物多样性与生态系统功能的物种去除试验。本研究是该项目的一部分。 元素循环是生态系统的重要功能之一,而氮素是限制草地生态系统生产力的主要因素,氮矿化是氮循环的关键步骤,因此,本研究重点讨论植物功能群对土壤氮矿化作用的影响,提出3点假设:1. 不同植物功能群对土壤氮矿化速率影响不同;2. 植物功能群去除前后氮矿化速率不同;3.植物功能群之间存在补偿效应。为了证明这些假设,于2006年9月、2007年6月和2007年8月份分别进行了室内培养(温度25℃,湿度60%田间最大持水量)用于测量氮矿化速率,同时于2007年6月和2007年8月份进行野外培养用于测量野外条件下的氮矿化速率,并在去除处理2年后得到以下主要结果: 1. 植物功能群去除数与土壤氮矿化速率呈单峰曲线关系(P<0.05),去除少量植物功能群氮矿化速率上升,去除更多的植物功能群后氮矿化速率下降; 2. 植物功能群去除数与土壤硝态氮含量呈线性正相关关系(P<0.0001),植物功能群的丧失加剧了土壤NO3--N的流失; 3. 多年生非禾草(PF)比其他植物功能群显著降低了氮矿化速率(P<0.05); 4. 短期内(去除处理1年内)在凋落物回填的情况下,去除0个植物功能群与去除全部植物功能群的氮矿化速率无显著差异(P>0.05); 5. 内蒙古温带典型草原在近3年内(2005、2006、2007)土壤碳库、氮库变化较小。

大气CO2 浓度和降水量增加有可能大幅提高中国北方部分草地生态系统净初级生产力,进而导致向土壤中输送的有机物相应增加。本研究以位于内蒙古自治区东乌珠穆沁旗内的半干旱草地生态系统为研究对象,通过向10−20 cm 土层添加不同质量和数量的植物凋枯物碎屑模拟有机物输入增加和喷灌模拟降水量提升,同时测定土壤微生物群落动态和植物生长指标,探讨在增加有机物输入和土壤水分的情况下土壤生物过程的变化及其对土壤碳排放和贮存的反馈作用。 研究结果表明,有机物添加可促进植物地上部分及根系的生长,并显著提高土壤中可溶性有机碳(能量)和氮(养分)的含量。土壤能量和养分水平的提高促进了土壤微生物的生长:在底物可利用性水平较高时,r−对策微生物(指具有生长迅速、C/N 值较低的微生物群组)在群落中占优势地位;随着底物水平的降低,土壤中K-对策微生物(指具有生长缓慢、C/N 值较高的微生物群组)在群落中逐渐占据优势地位。土壤微生物群落组成的改变进一步导致了微生物功能群代谢活性和特征的变化,具体表现为提高了有机物添加处理中土壤细菌群落的代谢潜能,并使细菌在群落水平上的生理剖面特征明显区别于未添加有机物的处理。 研究样地内土壤微生物主要受到底物中的能量(碳)限制,土壤活性有机质库作为可利用性较高的能量和养分的重要来源,对土壤微生物活性和土壤碳周转起着比水分因子更加重要的作用。土壤水分主要影响植物生长和根系活性,并增加了土壤微生物对底物响应的复杂性,但它对地下生物过程的作用程度以底物中能量和养分水平为前提。 利用稳定性13C 同位素示踪技术测量后发现,添加C4-植物凋枯物会加速C3 底物中碳的分解速度。结合有机物添加后土壤有机质库的变化,可以推测植物凋枯物(即能量物质)输入增加会导致土壤原生有机碳的正向激发效应。在此过程中,土壤微生物群落组成及功能群代谢活性的变化起着至关重要的作用。 不同光合途径(C3 和C4)的植物和同一植物不同器官组织(地上部分和根系)的凋枯物添加对地下生物过程,特别是土壤微生物群落代谢功能的影响是不同的。在添加C3-草本凋枯物的处理中,土壤细菌群落主要利用的碳源为氨基酸类化合物;而在添加C4-植物凋枯物的处理中,土壤细菌群落主要利用的碳源为羧酸类化合物。 本研究在野外自然条件下证明了在能量缺乏的中国北方草地生态系统中,土壤有机物输入增加不但不会提高土壤有机碳库的大小,而且可能导致土壤原生有机碳的激发效应。在利用土壤呼吸与环境因子(如温度)的关系进行模拟预测土壤碳排放时,需要考虑不同生态系统底物中的能量和养分水平,以及土壤微生物和植物根系等地下生物过程对底物水平的适应性。

mtDNA genotypes of six domestic horses (three adult short horses whose heights are under 1 m and three common domestic horses) from a small region of 15 km(2) in Malipo county of Yunnan province of China were investigated by the technique of restriction fragment length polymorphism (RFLP) with restriction endonucleases which recognize 6-bp sequences. An average of fragments for an individual was obtained. Unlike other domestic animals, this population of horses exhibits high mtDNA genetic diversity. Each of the six horses has a specific mtDNA genotype showing a pattern of multiple maternal origins, as suggested by fossil and literature records. We think the population of horses is an amazing seed-resource pool of horses and hence deserves to be paid more attention from the view of conservation genetics. However it is also remarkable that we did not find any typical mtDNA genetic markers which would discriminate between short horses and common domestic horses.

In this study, a detailed analysis of both previously published and new data was performed to determine whether complete, or almost complete, mtDNA sequences can resolve the long-debated issue of which Asian mtDNAs were founder sequences for the Native American mtDNA pool. Unfortunately, we now know that coding region data and their analysis are not without problems. To obtain and report reasonably correct sequences does not seem to be a trivial task, and to discriminate between Asian-and Native American mtDNA ancestries may be more complex than previously believed. It is essential to take into account the effects of mutational hot spots in both the control and coding regions, so that the number of apparent Native American mtDNA founder sequences is not erroneously inflated. As we report here, a careful analysis of all available data indicates that there is very little evidence that more than five founder mtDNA sequences entered Beringia before the Last Glacial Maximum and left their traces in the current Native American mtDNA pool.

The decipherment of the meager information provided by short fragments of ancient mitochondrial DNA (mtDNA) is notoriously difficult but is regarded as a most promising way toward reconstructing the past from the genetic perspective. By haplogroup-specific hypervariable segment (HVS) motif search and matching or near-matching with available modem data sets, most of the ancient mtDNAs can be tentatively assigned to haplogroups, which are often subcontinent specific. Further typing for mtDNA haplogroup-diagnostic coding region polymorphisms, however, is indispensable for establishing the geographic/genetic affinities of ancient samples with less ambiguity. In the present study, we sequenced a fragment (similar to 982 bp) of the mtDNA control region in 76 Han individuals from Taian, Shandong, China, and we combined these data with previously reported samples from Zibo and Qingdao, Shandong. The reanalysis of two previously published ancient mtDNA population data sets from Linzi (same province) then indicates that the ancient populations had features in common with the modem populations from south China rather than any specific affinity to the European mtDNA pool. Our results highlight that ancient mtDNA data obtained under different sampling schemes and subject to potential contamination can easily create the impression of drastic spatiotemporal changes in the genetic structure of a regional population during the past few thousand years if inappropriate methods of data analysis are employed.

A new blind loach of Triplophysa Rendahl 1933 was collected from a subterranean pool in a cave at Wulong County, Chongqing City, China, in November 2002. The new species, named Triplophysa rosa sp. nov., can be distinguished from its congeners by the following unique characters: eyes vestigial; 9 branched dorsal-fin rays; 12 branched pectoral-fin rays; 7 branched pelvic-fin rays; 6 branched anal-fin rays; 7 + 7 branched caudal-fin rays; distal margin of dorsal-fin concave; tip of pelvic-fin surpasses vertical level of anus; caudal-fin deeply forked; whole body scaleless and colourless. P>Sex controls have been performed in some farmed fish species because of significant growth differences between females and males. In yellow catfish (Pelteobagrus fulvidraco), adult males are three times larger than female adults. In this study, six Y- and X-linked amplified fragment length polymorphism fragments were screened by sex-genotype pool bulked segregant analysis and individual screening. Interestingly, sequence analysis identified two pairs of allelic genes, Pf33 and Pf62. Furthermore, the cloned flanking sequences revealed several Y- and X-specific polymorphisms, and four Y-linked or X-linked sequence characterized amplified region (SCAR) primer pairs were designed and converted into Y- and X-linked SCAR markers. Consequently, these markers were successfully used to identify genetic sex and YY super-males, and applied to all-male population production. Thus, we developed a novel and simple technique to help commercial production of YY super-males and all-male populations in the yellow catfish.

Gibel carp ( Carassius auratus gibelio) is a uniquely gynogenetic species with a minor ratio of males in natural habitats, but its male origin and sex determination mechanisms have been unknown. In this study, a male-biased mutant family was discovered from the gynogenetic gibel carp, and a male-specific SCAR marker was identified from the mutant family. Normal spermatogenesis was observed in the male testes by immuno. fluorescence histochemistry. Nearly identical AFLP profiles were observed between males and females, but a male-specific 86 bp AFLP fragment was screened by sex-pool bulked segregant analysis and individual screening. Based on the male-specific AFLP fragment, a total of 579 bp sequences were cloned by genome walking. Subsequently, a male-specific SCAR marker was designed, and the male-specific DNA fragment was confirmed to be steadily transmitted to the next generation and consistently detected only in males. The population genetic structure of fish parasitic nematode, Camallanus cotti, collected from the Yangtze River, Pearl River and Minjiang River in China was investigated. From these parasites, the similar to 730 bp of the first internal transcribed spacer of ribosomal DNA (ITS1 rDNA) and the 428 bp of mitochondrial cytochrome c oxidase subunit I (COI) gene were sequenced. For the ITS1 rDNA data set, highly significant Fst values and low rates of migration were detected between the Pearl River group and both the Yangtze River (Fst = 0.70, P < 0.00001; Nm = 0.21) and Minjiang River (Fst = 0.73, P < 0.00001; Nm = 0.18) groups, while low Fst value (Fst = 0.018, P > 0.05) and high rate of migration (Nm = 28.42) were found between the Minjiang and the Yangtze rivers. When different host/locality populations (subpopulations) within each river were considered, subpopulations between the Yangtze River and Minjiang River had low Fst values (<= 0.12) and high Nm values (>3.72), while Pearl River subpopulations were significantly different from the Yangtze River and Minjiang River subpopulations (Fst >= 0.59; Nm < 1). The COI gene data set revealed a similar genetic structure. Both phylogenetic analyses and a statistical parsimony network grouped the Pearl River haplotypes into one phylogroup, while the Yangtze River and Minjiang River haplotypes formed a second group. These results suggested that the Yangtze River and Minjiang River subpopulations constituted a single reproductive pool that was distinct from the Pearl River subpopulations. In addition, the present study did not find host-related genetic differentiation occurring in the same drainage. (C) 2009 Published by Elsevier B.V.

Samples from stone surfaces were collected in pools within four unpolluted hillstreams (two shaded and two unshaded) in monsoonal Hong Kong (lat. 23 degrees N) to elucidate the extent of spatial (within and among streams) and temporal (seasonal) variations in algal biomass and assemblage composition. Sampling continued for over 12 months, incorporating the dry season when streams were at baseflow, and the wet season when spates were frequent. We anticipated that algal biomass would be lower in shaded streams and during the wet season, with associated seasonal differences in assemblage composition or relative abundance of different growth forms (e. g. erect versus prostrate). Benthic chlorophyll a (a proxy for algal biomass) varied among streams from an annual mean of 11.0-22.3 mg m(-2). Dry-season standing stocks were 18% higher than during the wet season when spate-induced disturbance reduced algal standing stocks. Algal biomass varied significantly at the stream scale, but not at the pool scale, and was lower in unshaded streams, where standing stocks may have been limited by high densities of algivorous balitorid loaches (mainly Pseudogastromyzon myersi). An overriding effect of grazers on algal biomass could also have reduced variations resulting from spate-induced disturbance. Significant differences in assemblage composition among streams, which were dominated by diatoms and cyanobacteria (totally 82 taxa) were not systematically related to shading conditions. Seasonal variations in algal assemblages were statistically significant but rather minor, and did not involve major shifts in composition or growth form caused by spate-induced disturbance. The abundance of filamentous cyanobacteria in all the streams may have been due to 'gardening' by balitorid loaches that removed erect or stalked diatoms and favoured cyanobacteria that persist through basal regeneration of filaments. This explanation requires validation through manipulative experiments.

To conserve and utilize the genetic pool of gynogenetic gibel carp (Carassius auratus gibelio), the Fangzheng and Qihe stock hatcheries have been established in China. However, little information is available on the amount of genetic variation within and between these populations. In this study, clonal diversity in 101 fish from these two stock hatcheries and 35 fish from two other hatcheries in Wuhan and Pengze respectively was analysed for variation in serum transferrin. Thirteen clones were found in Fangzheng and Qihe, of which 12 were novel. Six clones were specific to Fangzheng and three specific to Qihe, whereas four were shared among the Fangzheng and Qihe fish. To obtain more knowledge on genetic diversity and genealogical relationships within gibel carp, the complete mitochondrial DNA (mtDNA) control region (similar to 920 bp) was sequenced in 64 individuals representing all 14 clones identified in the four hatcheries. Differences in the mtDNA sequences varied remarkably among hatcheries, with the Fangzheng and Qihe lines demonstrating high diversity and Wuhan and Pengze showing no variation. The Fangzheng and Qihe lines might represent two distinct matrilineal sources. One of the Qihe samples carried the haplotype shared by a most widely cultivated Fangzheng clone, indicating that a Fangzheng clone escaped from cultivated ponds and moved into the Qihe hatchery. Four Fangzheng samples clustered within the lineage formed mainly by Qihe samples, most likely reflecting historical gene flow from Qihe to Fangzheng. It is suggested that clones in Wuhan originated from Fangzheng, consistent with their introduction history, supporting the hypothesis that gibel carp in Pengze were domesticated from individuals in the Fangzheng hatchery.

Many experimental studies have documented the impact of microcystins (MC) on fish based on either intraperitoneal injection, or oral gavaging via the diet, but few experiments were conducted by MC exposure through natural food uptake in lakes. In this study, the phytoplanktivorous silver carp were stocked in a large pen set in Meiliang Bay of Taihu Lake where toxic Microcystis blooms occurred in the warm seasons. Fish samples were collected monthly and MC concentrations in liver and kidney of the fish were determined by LC-MS. The maximum MC concentrations in liver and kidney were present in July when damages in ultrastructures of the liver and kidney were revealed by electron microscope. In comparison with previous studies on common carp, silver carp showed less damage and presence of lysosome proliferation in liver and kidney. Silver carp might eliminate or lessen cell damage caused by MC through lysosome activation. Recovery in the ultrastructures of liver and kidney after Microcystis blooms was companied with a significant decrease or even disappearance of MC. Catalase and glutathione S-transferase in liver and kidney of silver carp during Microcystis blooms were significantly higher than before and after Microcystis blooms. The high glutathione pool in liver and kidney of silver carp suggests their high resistance to MC exposure. The efficient antioxidant defence may be an important mechanism of phytoplanktivorous fish like silver carp to counteract toxic Microcystis blooms. (C) 2007 Published by Elsevier Ltd.

Pollution resulting from increased human activities is threatening Lake Donghu, its effects being characterized by serious eutrophication. A steady increase of phosphorus loading is the most important factor of the lake eutrophication. Pollution external control projects are being implemented and will be accomplished before the year 2010. In order to predict the restoration rate by the lake's self-purification after the projects of external control, a model of predicting the removal rate of total phosphorus (TP) from lake water is developed, and a brief method of estimating the release and export rate of sediment phosphorus is suggested. Results show that, on the premise of external loading fully controlled. The restoration needs about 55 years or more. Obviously, the great P pool in the sediment will be a limiting factor of preventing the improvement of water quality after the external loading is under control. Based on the estimation we conclude that after the external control projects before 2010, in order to restore the lake in a few years, although highly cost, the first step must be the sediment dredging to remove internal loading. The second step is diverting water of River Changjiang into the lake to accelerate the improvement of lake water. Otherwise, removal of pollutant sources will become meaningless. The species-specific production of extracellular phosphatases in phytoplankton of a subtropical polymictic take was investigated from March to May 2004. Phosphatase activity was detected directly at the site of enzyme action using the enzyme-labelled fluorescence (ELF) technique. Size fractionation of bulk phosphatase activity (PA), concentrations of soluble reactive phosphorus (SRP), chlorophyll a, and phytoplankton composition were determined in parallel. Phosphatase-positive cells were present in every phytoplankton sample; labelled cells were detected in 33 algal taxa, including many chlorophytes, dinoflagellates and some diatoms, but never among cyanobacteria. We recorded an unusual dinoflagellate bloom (Peridiniopsis sp.), of which similar to 25% of the cells were phosphatase-positive. Several populations were partly phosphatase-positive whenever present, while some other species never showed any activity. The production of extracellular phosphatases was not primarily regulated by ambient P concentrations; algae produced these enzymes even if SRP concentrations were high. Moreover, heterotrophic nanoflagellates most probably contributed to the pool of particle-bound PA in some samples.
