The changes of NH3-N, NO3-N, NO2-N and TN/TP were studied during growth and non-growth season in 33 subtropical shallow lakes in the middle and lower reaches of the Yangtze River. There were significant positive correlations among all nutrient concentrations, and the correlations were better in growth season than in non-growth season. When TP > 0.1 mgL(-1), NH3-N increased sharply in non-growth season with increasing TP, and NO3-N increased in growth season but decreased in non-growth season with TP. These might be attributed to lower dissolved oxygen and low temperature in non-growth season of the hypereutrophic lakes, since nitrification is more sensitive to dissolved oxygen and temperature than anti nitrification. When 0.1 mgL(-1)> TP > 0.035 mgL(-1), TN and all kinds of inorganic nitrogen were lower in growth season than in non-growth season, and phytoplankton might be the vital regulating factor. When TP < 0.035 mgL(-1), inorganic nitrogen concentrations were relatively low and NH3-N, NO2-N had significant correlations with phytoplankton, indicating that NH3-N and NO2-N might be limiting factors to phytoplankton. In addition, TN/TP went down with decline in TIP concentration, and TN and inorganic nitrogen concentrations were obviously lower in growth season than in non-growth season, suggesting that decreasing nitrogen (especially NH3-N and NO3-N) was an important reason for the decreasing TN/TP in growth season. The ranges of TN/TP were closely related to trophic level in both growth and non-growth seasons, and it is apparent that in the eutrophic and hypertrophic state the TN/TP ratio was obviously lower in growth season than in non-growth season. The changes of the TN/TP ratio were closely correlated with trophic levels, and both declines of TN in the water column and TP release from the sediment were important factors for the decline of the TN/TP ratio in growth season.

According to outdated paradigms humic substances (HS) are considered to be refractory or inert that do not directly interact with aquatic organisms. However, they are taken up and induce biotransformation activities and may act as hormone-like substances. In the present study, we tested whether HS can interfere with endocrine regulation in the amphibian Xenopus laevis. In order to exclude contamination with phyto-hormones, which may occur in environmental isolates, the artificial HS 1500 was applied. The in vivo results showed that HS 1500 causes significant estrogenic effects on X. laevis during its larval development and results of semi-quantitative RT-PCR revealed a marked increase of the estrogenic biomarker estrogen receptor mRNA (ER-mRNA). Furthermore, preliminary RT-PCR results showed that the thyroid-stimulating hormone (TSH beta-mRNA) is enhanced after exposure to HS1500, indicating a weak adverse effect on T3/T4 availability. Hence, HS may have estrogenic and anti-thyroidal effects on aquatic animals, and therefore may influence the structure of aquatic communities and they may be considered environmental signaling chemicals. Submersed macrophytes in Yangtze lakes have experienced large-scale declines due to the increasing human activities during past decades. To seek the key factor that affects their growth, monthly investigations of submersed macrophytes were conducted in 20 regions of four Yangtze lakes during December, 2001-March, 2003. Analyses based on annual values show that the ratio of Secchi depth to mean depth is the key factor (50% of macrophyte biomass variability among these lakes is statistically explained). Further analyses also demonstrate that the months from March to June are not only the actively growing season for most macrophytes, but the key time the factor acts. Five key-time models yielding higher predictive power (r(2) reaches 0.75,0.76,0.77,0.69 and 0.81) are generated. A comparison between key-time models and traditional synchronic ones indicates that key-time models have higher predictive power. Analyses of transparency thresholds during macrophyte growing season and the limitations of the models are presented. The models and other results may benefit the work concerning submersed macrophyte recovery in Yangtze lakes. (c) 2005 Elsevier B.V. All rights reserved.

Phytoplanktivorous bighead carp were injected i.p. with extracted microcystins (mainly MC-RR and -LR) at two doses, 200 and 500 MC-LReq. mu g kg(-1) bw, and the changes in extractable MCs in liver and in the ultrastructure of hepatocytes were studied at 1, 3, 12, 24 and 48 h after injection. Quantitative and qualitative determinations of MCs in the liver were conducted by HPLC and LC-MS, respectively. MC concentration in the liver reached the maxima at 12 It (2.89 mu g MCs g(-1) dry weight at the lower dose) or at 3 h (5.43 mu g MCs g(-1) dry weight at the higher dose) post-injection, followed by sharp declines afterwards, whereas the ultrastructural changes of hepatocytes in both dose groups suggest progressive increases in severity toward the directions of apoptosis and necrosis from I to 24 h, respectively. There were two new findings in fish: widening of intercellular spaces was among the early ultrastructural changes induced by MCs and ultrastructural recovery of hepatocytes was evident at 48 h post-injection in both dose groups. Both the present and previous studies suggest that with in vivo or in vitro exposure to microcystins, hepatocyte damage in fish tends to proceed toward the direction of apoptosis at lower MC concentrations but toward the direction of necrosis at high MC concentrations. The temporal dynamics of MCs in the liver suggest that bighead carp may have a mechanism to degrade or bind MC-LR actively after it enters the blood system. (c) 2005 Elsevier Ltd. The Southeast Asia and Western Pacific regions contain half of the world's children and are among the most rapidly industrializing regions of the globe. Environmental threats to children's health are widespread and are multiplying as nations in the area undergo industrial development and pass through the epidemiologic transition. These environmental hazards range from traditional threats such as bacterial contamination of drinking water and wood smoke in poorly ventilated dwellings to more recently introduced chemical threats such as asbestos construction materials; arsenic in groundwater; methyl isocyanate in Bhopal, India; untreated manufacturing wastes released to landfills; chlorinated hydrocarbon and organophosphorous pesticides; and atmospheric lead emissions from the combustion of leaded gasoline. To address these problems, pediatricians, environmental health scientists, and public health workers throughout Southeast Asia and the Western Pacific have begun to build local and national research and prevention programs in children's environmental health. Successes have been achieved as a result of these efforts: A cost-effective system for producing safe drinking water at the village level has been devised in India; many nations have launched aggressive antismoking campaigns; and Thailand, the Philippines, India, and Pakistan have all begun to reduce their use of lead in gasoline, with resultant declines in children's blood lead levels. The International Conference on Environmental Threats to the Health of Children, held in Bangkok, Thailand, in March 2002, brought together more than 300 representatives from 35 countries and organizations to increase awareness on environmental health hazards affecting children in these regions and throughout the world. The conference, a direct result of the Environmental Threats to the Health of Children meeting held in Manila in April 2000, provided participants with the latest scientific data on children's vulnerability to environmental hazards and models for future policy and public health discussions on ways to improve children's health. The Bangkok Statement, a pledge resulting from the conference proceedings, is an important first step in creating a global alliance committed to developing active and innovative national and international networks to promote and protect children's environmental health.

Wu-Han Xiao, Jian-Guo Wang and Lian-Xiang Li (2002) Taxonomic studies of parasitic nyctotherans from Chinese Anura amphibians III. Wichtermania. Zoological Studies 41(1): 69-76. This paper describes 6 new species belonging to the genus Wichtermania from Anura amphibians distributed in southern China. The 6 species are Wichtermania multigranularis sp. nov., W. oviformis sp. nov., W vesiformis sp. nov., W. reticulatis sp. nov., W granuliformis sp. nov., and W obliquoides sp. nov. The characteristics for diagnosis of the 6 new species are described in this paper.

Lian-Xiang U, Jian-Guo Wang and Wu-Han Xiao (2002) Taxonomic studies of parasitic nyctotherans from Chinese Anura amphibians IV. Spirocytopharynxa gen. nov. and Macrocytopharynxa gen. nov. Zoological Studies 41(1): 77-84. This paper describes 2 new genera and 5 new species of nyctotherans from Anura amphibians distributed in southern China. Based on the composition of the upper and bottom flaps, the number and position of sutural lines, the length of the oral groove, and the morphology and position of the end of the cytopharynx, the adoral zone of the membranelle (AZM), and the macronucleus, two new genera were established. The 3 new species, Spirocytopharynxa sinensis, S. guangxiensis, and S. quadranus, belong to the new genus Spirocytopharynxa, Another 2 species, Macrocytopharynxa (Nyctotheroidae Nie, 1932) pyriformis n. comb. and M. lingchuanensis n. sp., belong to the new genus Macrocytopharynxa. The characteristics for diagnosis of the 2 new genera and 5 new species are described in this paper.

This paper describes the long-term dynamics of phosphorus concentrations in both the lake water and the sediment in a subtropical Chinese lake, Lake Donghu. The total phosphorus (TP) concentration in the lake water experienced an upward trend from the 1950s, and peaked in 1983/1984, but declined obviously afterwards. From the 1950s to the 1990s, TP content in the upper 10 cm sediment of the lake increased steadily from 0.307 to 1.68 mg Pg DW-1 at Station I and from 0.151 to 0.89 mg Pg DW-1 at Station II, respectively. The TP increase in the lake water before mid-1980s was mainly attributed to the massive input of sewage P. The outbreak of cyanobacterial blooms coincided with the peaks of TP and Orthophosphate (PO4-P) in the water in mid-1980s, and the maximum TP of the water reached as high as 1.349 mg/1 at Station I and 0.757 mg/l at Station II (in 1984), respectively. The declines of TP and PO4-P in the water after mid-1980s was coincident with the disappearance of cyanobacterial bloom. (C) 2001 Elsevier Science Ltd. All rights reserved.

The development of an implantable five channel microelectrode array is presented for neural signal recordings. The detailed fabrication process is outlined with four masked used. The SEM images show that the probe shank is 1.2mm long, 100 mu m wide and 30 mu m thick with the recording sites spaced 200 mu m apart for good signal isolation. The plot of the single recording site impedance versus frequency is shown by test in vitro and the ompedence declines with the increasing frequency. Experiment in vivo using this probe is under way.

通过对两栖类动物大蹼铃蟾(B朋b了na勿日对朋)皮肤分泌物及匀浆物的分离,纯化,从其碱性组分中得到了全新的三叶因子多肤(BIn--TFFZ)和膜联蛋白H相关蛋白(BAIIRP),它们分别具有诱导或抑制人血小板活化的全新功能。以血小板膜糖蛋白GPVI受体激动剂stejnulxin为对照,研究了Bm一TFFZ活化人血小板的信号传导途径。哺乳动物的三叶因子(TFF)蛋白的主要功能在于通过修复粘膜损伤,维持粘膜层的完整。它们在肿瘤抑制及癌症侵润方面也有一定程度的作用。我们从两栖类动物大蹼铃蟾皮肤分泌物分离,纯化而得到了一个全新的三叶因子多肤(Bm一TFFZ)。它是单亚基蛋白,表观分子量为13000,并具有全新的诱导人血小板活化的功能,我们研究了其诱导人血小板活化的活性的量效关系。它对血小板的激活涉及血小板整合素a:Ibp3的活化,Bm一TFFZ可诱导经阿斯匹林及三磷酸腺昔"双磷酸酶(Apyrase)处理的血小板的聚集,表明B,TFFZ的活性不依赖于血小板的ADP及血栓嗯烷A2(TXAZ)的自泌正反馈。F工TC标记的B爪一TFFZ蛋白能与血小板膜结合。通过专一性药理学抑制剂的研究,我们初步排除了Bm一TFFZ蛋白作用于血小板膜已知的G一蛋白偶联受体(如ADP,TXAZ及血小板活化因子受体)的可能性。以血小板GPVI激动剂stejnulxin为对照,我们仔细研究了Bm一TFFZ蛋白刺激血小板后的信号传导分子事件,发现其酪氨酸磷酸化图谱明显不同于血小板GPVI激动剂5tejnu1X如,尤其是磷脂酶CYZ并不磷酸化。因此,我们认为Bm一TFFZ蛋白激活血小板的信号传导通路是活化磷脂酶C,并刺激胞内钙离子的释放,活化血小板伪:。p3,进而导致血小板的聚集。从Bm--TFFZ蛋白的cDNA克隆序列推知它由104个氨基酸组成,含有两个三叶因子结构域,与人TFFZ和非洲爪蟾xPZ序列中相同的氨基酸分别占28%及34%。为了确定两栖类的三叶因子蛋白具有诱导人血小板活化的功能,我们仔细地研究了整个分离纯化过程中所有组份的血小板聚集活性并比较了它们的相对活性差异。Bm一TFFZ是第一个来自两栖类动物的血小板激动剂,也是我们第一个报道的具有诱导血小板聚集活性的三叶因子多肤。·我们还通过阴离子交换,凝胶过滤和阳离子交换层析,从大蹼铃蟾皮肤匀浆物中纯化了一个表观分子量为33kD。的单链蛋白。N一末端序列比较分析显示,该·蛋白与来自非洲爪蟾、红色原鸡和人膜联蛋白H的N一末端序列相同的氨基酸分别占70%、64%和56%。该蛋白具有以钙依赖的方式抑制专一性血小板膜糖蛋白VI(GPVI)受体激动剂-Stejnu1Xin诱导洗涤人血小板聚集的生物学功能,最大抑制率达48%。结合其N一末端序列搜索结果及其活性的钙依赖性,推测该蛋白是与膜联蛋白H相关的一类蛋白质。

A two-dimensional (2D) multi-channel silicon-based microelectrode array is developed for recording neural signals. Three photolithographic masks are utilized in the fabrication process. SEM images show that the microprobe is 1. 2mm long,100μm wide,and 30μm thick, with recording sites spaced 200μm apart for good signal isolation. For the individual recording sites, the characteristics of impedance versus frequency are shown by in vitro testing. The impedance declines from 14MΩ to 1.9kv as the frequency changes from 0 to 10MHz. A compatible PCB (print circuit board) aids in the less troublesome implantation and stabilization of the microprobe.

本研究针对川西北高山草甸缺乏科学管理,过度放牧导致草场退化,并由此引发的一系列生态环境问题,选取红原县瓦切乡1996 年草地承包后形成的四个放牧强度草场,即不放牧、轻度(1.2 头牦牛hm-1)、中度(2.0 头牦牛hm-1)和重度放牧(2.9 头牦牛hm-1),作为研究对象,研究了不同放牧强度对草地植物-土壤系统中碳、氮这两个最基本物质的分布格局和循环过程的影响,并探讨了放牧干扰下高山草甸生态系统的管理。 1.放牧对草地植物群落物种组成,尤其是优势种,产生了明显的影响。不放牧、轻度、中度和重度放牧草地群落物种数分别为22,23,26,20 种,群落盖度分别是不放牧96.2%>中度93.6%>轻度89.7%>重度73.6%。随放牧强度的增加, 原植物群落中的优势种垂穗鹅冠草( Roegneria nutans )、发草(Deschampsia caespitosa)和垂穗披碱草(Elymus nutans)等禾草逐渐被莎草科的川嵩草(Kobresia setchwanensis)和高山嵩草(Kobresia pygmaea)所取代成为优势种。同时,随放牧强度的增加,高原毛茛(Ranunculus brotherusii)、狼毒(Stellera chamaejasme)、鹅绒委陵菜(Potentilla anserina)和车前(Plantagodepressa)等杂类草的数量也随之增加。 2.生长季6~9 月份,草地植物地上和地下生物量(0~30cm)都是从6 月份开始增长,8 月份达到最高值,9 月份开始下降。每个月份,通常地上生物量以不放牧为最高,重度放牧总是显著小于不放牧;地下生物量随放牧强度的增加表现为增加的趋势,通常重度和中度放牧显著高于不放牧和轻度放牧草地。不放牧、轻度、中度和重度放牧草地6~9 月份4 个月的植物总生物量平均值分别是1543、1622、2295 和2449 g m-2,但随放牧强度的增加越来越来多的生物量被分配到了地下部分,地下生物量占总生物量比例的大小顺序分别是重度88%>中度82%>轻度76%>不放牧69%。生物量这种变化主要是由于放牧使得群落优势种发生改变而引起的,其分配比例的变化体现了草地植物对放牧干扰的适应策略。 3.植物碳氮贮量的季节变化类似与生物量的变化。每个月份,不同放牧强度间植物地上碳氮的贮量有所不同,一般重度放牧会显著减少植物地上碳氮贮量。植物根系(0~30cm)碳氮贮量随放牧强度的增加表现为增加的趋势,通常重度和中度放牧显著高于不放牧和轻度放牧草地。不放牧、轻度、中度和重度放牧草地6~9 月份4 个月的植物总碳平均值分别是547、586、847 和909 g m-2,根系碳贮量占植物总碳的比例大小顺序分别是重度88%>中度82%>轻度76%>不放牧69%;放牧、轻度、中度和重度放牧草地6~9 月份4 个月的植物总氮平均值分别是17、17、23 和26 g m-2,根系氮贮量占植物总氮的比例大小顺序分别是重度79%>轻度71%>中度70%>不放牧65%。 4. 土壤有机碳贮量(0~30cm)的季节变化表现为7 月份略有下降,8 月开始增加,9 月份达到的最大值。土壤氮贮量的季节变化表现为随季节的推移逐渐增加的趋势。增加的放牧强度不同程度的增加土壤有机碳氮的贮量。不放牧、轻度、中度和重度放牧6~9 月份4 个月的土壤有机碳贮量的平均值分别是9.72、10.36、10.62 和11.74 kg m-2,土壤氮贮量分别为1.45、1.56、1.66 和1.83 kg m-2。土壤中有机碳(氮)的贮量都占到了植物-土壤系统有机碳(氮)的90%以上,但不同放牧强度之间的差异不明显。 5. 土壤氮的总硝化和反硝化,温室气体N2O 和CO2 的释放率的季节变化表现为从6 月份开始增加,7 月份达到最大值,8 月份开始下降,9 月份降为最小值。增加的放牧强度趋向于增加土壤氮的总硝化和反硝化作用,温室气体N2O和CO2 的释放率,通常情况下,中度放牧和重度放牧显著地加强了这些过程。 6.垂穗鹅冠草(Roegneria nutans)和川嵩草(Kobresia setchwanensis)凋落物在不同放牧强度下经过1 年的分解,两种凋落物的失重率及其碳氮的损失率3都随放牧增加表现为增加的趋势。在同一放牧强度下,川嵩草凋落物的失重率和碳氮的损失率都高于垂穗鹅冠草凋落物。 7. 尽管重度放牧显著增加了土壤碳氮的贮量,但同时也显著降低了植被群落盖度,降低了植物地上生物量,因此,久而久之会减少植物向土壤中的碳氮归还率;与不放牧和轻度放牧相比,重度放牧又显著增加了土壤CO2 和NO2 的排放量,这是草地生态系统碳氮损失的重要途径。由此可见,对于这些地处青藏高原的非常脆弱的高山草甸生态系统,长期重度放牧不仅导致植物生产力降低,而且将导致草地生态系统退化,甚至造成土壤中碳氮含量减少。 Long-term overgrazing has resulted in considerable deterioration in alpine meadowof the northwest Sichan Province. In order to explore management strategies for thesustainability of these alpine meadows, we selected four grasslands with differentgrazing intensity (no grazing-NG: 0, light grazing-LG: 1.2, moderate grazing-MG: 2.0,and heavy grazing-HG: 2.9 yaks ha-1) to evaluate carbon, nitrogen pools and cyclingprocesses within the plant-soil system in Waqie Village, Hongyuan County, Sichuan Province. 1. Grazing obviously changed the plant species composition, especially ondominant plant species. Total number of species is 22, 23, 26, and 20 for NG, LG, MGand HG, respectively. Vegetation coverage under different grazing intensity ranked inthe order of 96.2% for HG>93.6% for MG>89.7% for LG>73.6% for NG. Thedominator of HG community shifted from grasses-Roegneria nutans andDeschampsia caespitosa dominated in the NG and LG sites into sedges-Kobresiapygmaea and K. setchwanensis. At the same time, with the increase of grazingintensity, the numbers of forbs, such as Ranunculus brotherusii, Stellera chamaejasme,Potentilla anserine and Plantago depressa, increased with grazing intensity. 2. Over the growing season, aboveground and belowground biomass showed a 5single peak pattern with the highest biomass in August. For each month, abovegroundbiomass usually was the highest in the NG site and lowest in the HG site.Belowground biomass showed a trend of increase as grazing intensity increased and itwas significantly higher in the HG and MG site than in the NG and LG sites. Totalplant biomass averaged over the growing season is 1543, 1622, 2295 and 2449 g m-2for NG, LG, MG and HG, respectively. The proportion of biomass to total plantbiomass for NG, LG, MG and HG is 88%, 82%, 76% and 69%, respectively. Higherallocation ratio for is an adaptive response of plant to grazing. 3. Carbon and nitrogen storage in plant components followed the similar seasonalpatterns as their biomass under different grazing intensities. Generally, heavy grazingsignificantly decreases aboveground biomass carbon and nitrogen compared to nograzing. Carbon and nitrogen storage in root tended to increase as grazing increasedand they are significantly higher in the HG and MG sites compared to the LG and NGsite. Total Carbon storage in plant system averaged over the growing season is 547,586, 847 and 909 g m-2 for NG, LG, MG and HG, respectively, while 17, 17, 23 and 26g m-2 for nitrogen. The proportion of carbon storage in root to total plant carbon forNG, LG, MG and HG is 88%, 82%, 76%, 69%, respectively, while 65%, 71%, 70%and 79% for nitrogen. 4. Carbon storage in soil (0-30cm) decreased slightly in July, then increased inAugust and peaked in September. Nitrogen storage in soil tended to increase withseason and grazing intensity. Total Carbon storage in soil averaged over the growingseason is 9.72, 10.36, 10.62 and11.74 kg m-2 for NG, LG, MG and HG, respectively,while 1.45, 1.56, 1.66 and 1.83 for nitrogen. The proportion of carbon (nitrogen)storage in soil to plant-soil system carbon (nitrogen) storage for NG, LG, MG and HGis more than 90%, which is not markedly different among different grazing intensities. 5. Gross nitrification, denitrification, CO2 and N2O flux rates in soil increasedfrom June to July and then declined until September, all of which tended to increasewith the increase of grazing intensity. Generally, heavy and moderate grazing intensitysignificantly enhanced these process compared to no and light grazing intensity. 6. After decomposing in situ for a year, relative weight, carbon and nitrogen loss in the litter of Roegneria nutans and Kobresia setchwanensis tended to increase asgrazing intensity increased. Under the same grazing intensity, relative weight, carbonand nitrogen loss in the litter of Kobresia setchwanensis were higher than these in thelitter of Roegneria nutans. 7. Although heavy grazing intensity resulted in higher levels of carbon andnitrogen in plant and soil, it decreased vegetation coverage and aboveground biomass,which are undesirable for livestock production and sustainable grassland development.What is more, heavy grazing could also introduce potential carbon and nitrogen lossvia increasing CO2 and N2O emission into the atmosphere. Grazing at moderateintensity resulted in a plant community dominated by forage grasses with highaboveground biomass productivity and N content. The alpine meadow ecosystems inTibetan Plateau are very fragile and evolve under increasing grazing intensity by largeherbivores; therefore, deterioration of the plant-soil system, and possible declines insoil C and N, are potential without proper management in the future.

青藏高原东南缘由于特殊的生态地理条件,有着丰富的森林资源,这些资源是长江上游涵养水源、保持水土的生态屏障,是生物多样性的资源宝库。但随着过量的森林采伐,使该区曾经丰富的生物多样性资源遭受了前所未有的破坏,天然林的质量严重下降,生态系统退化,功能减弱。与此同时,许多物种的种群规模正在锐减,物种的遗传多样性也严重丧失。川西云杉是西部地区分布最广的云杉树种之一,在较高海拔的地区有着重要的生态学功能,是一种适应性很强的乡土树种。本项目采用简单序列重复标记(SSR)和特定序列位点(STS)研究不同生境条件下川西云杉群体的遗传变异及其时空分布格局,考察遗传变异与复杂的山地生态环境间的潜在联系,系统地揭示川西云杉天然群体与环境系统相互作用的生态适应与分子进化机制。研究成果能有效地为该树种遗传资源的科学保护与合理利用提供理论依据和科学指导,可为中国西南部亚高山天然林的可持续经营及退化生态系统的恢复与重建提供依据。主要研究结果如下: 1 STS和SSR两种分子标记的研究结果表明:川西云杉群体拥有中等水平的遗传多样性(基于SSR标记,平均He = 0.640;基于STS标记,平均He = 0.553)。造成这种中等水平的遗传多样性,可能是由于历史原因,川西云杉天然林被大量采伐,导致了遗传多样性的丧失。两种方法都检测出群体BT有着最高水平的遗传多样性。 2 两种标记的结果都一致说明:检测的10个川西云杉群体间遗传分化比较高,其存在群体间的遗传变异比例要明显地高于广泛分布的挪威云杉、横贯大陆的黑云杉以及兼有连续分布和不连续分布的西加云杉等,但要低于分布范围狭窄且呈不连续分布的粗枝云杉。青藏高原东南部的片段化生境可能是导致高水平遗传分化的主要原因。 3 基于两种标记的UPGMA 聚类和PCA分析结果,以及基于SSR标记的FCA分析结果都显示:群体BY遗传上明显区别于其它群体,其可能原因是青藏高原东南缘山脉阻隔而导致的生殖隔离的结果。 4 根据软件Bottleneck 1.2.02的检测以及不依赖哈迪一温伯格平衡的M比率检测结果:川西云杉种群很有可能经历了近期的遗传瓶颈效应。在本研究中遗传瓶颈效应并没有显著影响到物种的遗传多样性。然而,在这些片断化群体未来的子代群体中很可能出现遗传瓶颈导致的遗传多样性下降的效应。 Due to the extremely complex topography and climatic conditions, the southeast of the Qinghai-Tibet Plateau is region abundant in forest resource, which is benefit to the upper reaches of the Yangtze River water conservation, and protecting natural environment and biodiversity resources. However, by the reason of a plenty of trees were cut in these yeas, the biodiversity resource was destroy with degraded ecosystem and imperfect funcation. Picea balfouriana is one of the regionally distributed conifer species in the southeast of the Qinghai-Tibet Plateau and considered as a constructive species within its distribution area. It is an optimal species for the production of biomass. Moreover, it is well adapted to stressful environments at high altitude, especially to cold and drought conditions which are generally harsh for other trees. In our study, two types of molecular markers (SSR and STS) were used to estimate the genetic diversity and genetic structure of P. balfouriana populations originating from the southeast of the Qinghai-Tibet Plateau with varying climatic and geographical conditions. The results will not only provide a deep insight into its genetic diversity and population genetic structure but also valuable information for further management and breeding programs in P. balfouriana. In summary, the results obtained in this study revealed that: 1 A moderate degree of genetic variation is present in P. balfouriana in the southeast of the Qinghai-Tibet Plateau and it may caused by many trees were cut in the past. Population BT owns the highest level of genetic diversity by both two types of markers. 2 Considerable population differentiation exists among the ten P. balfouriana populations based on both SSR and STS markers, possibly caused by habitat fragmentation and heterogeneous environments. 3 The UPGMA clustering and PCA analyses based on SSR and STS marker, and FCA analyses based on SSR marker congruously separated population BY from other populations, which is likely due to the presence of mountain barriers. 4 The results of bottleneck analysis indicated that P. balfouriana populations that have undergone recent declines. In our research, the bottleneck effect don’t have a significant impact on the genetic diversity of species, however, the level of genetic diversity of P. balfouriana offspring populations growing in the fragmented habitats will decline in the future.

揭示水体中繁殖的两栖动物在异质性景观中的空间扩散特点,探讨景观面积丧失和破碎化对于两栖动物的影响,为两栖动物的保护提供理论依据。本文以四川西北部若尔盖湿地自然保护区的高原林蛙(Rana kukunoris)为研究对象,通过运用地理信息系统及建立景观模型等方法,在分析若尔盖湿地自然保护区范围内现有景观格局的基础上,建立了高原林蛙的景观扩散模型,并模拟了“沼泽→草甸”的湿地逆演化过程下高原林蛙的空间分布与景观连接的变化特点。主要结果是: 1.若尔盖湿地自然保护区呈典型的沼泽—草甸式斑块—基质景观格局。草甸面积占整个景观面积的79.42%,景观蔓延度指数(CONTAG)为79.00远离最小值0而更趋向于最大值100,面积和景观蔓延度指数表明草甸是整个景观中面积占绝对优势且景观连接好的类型,构成了景观的基质,对景观的动态格局演变起主导作用。沼泽面积仅占整个景观面积的18.08%,但却是整个景观中斑块数目最多的单元,占所有斑块数的82.9%。因此沼泽斑块与草甸基质之间的动态结构对高原林蛙的扩散起着决定性的作用。 2.空间扩散模型表明,其它类型的景观不但扩展了高原林蛙的活动范围,而且也为高原林蛙在不同沼泽斑块间的连接提供了通道。高原林蛙的空间扩散区域使得彼此间成斑块化隔离状分布的沼泽形成了潜在景观功能连接,促进了不同斑块间物种的交流。小型沼泽作为垫脚石(stepping-stone),使得整个景观中的相隔距离较远的大型斑块联结为一个功能整体,促进了高原林蛙在整个景观中的相互动态联系。 3.模拟“沼泽→草甸”的湿地逆演化过程表明,大量小型沼泽湿地的消失将会 对在沼泽中繁殖并扩散到其它景观类型中去的高原林蛙造成潜在影响。逆演化过程不仅使沼泽斑块的分布范围,沼泽源斑块的面积和空间扩散面积减少,而且对景观连接也有很大影响。小型沼泽的消失,将使得景观斑块的功能连接变小,使得依靠小型沼泽作为跳板的动物在沼泽斑块之间的移动将变得更加困难。 本文是对生境丧失与破碎化影响下两栖动物的行为反应的一种尝试。影响模型的因素很多,包括动物对各种类型景观的偏好程度,地理数据的精度,及模型的可靠程度都是制约模型准确度的因素。 The spatial diffusion of water—breeding amphibian through heterogeneous landscape and the effects of landscape losing and fragmentation to amphibian were the core theory of the landscape ecology of amphibian. Geographical information system (GIS) and landscape model were used to model the diffused area of Rana kukunoris in Zoige Wetland Natural Reserve. Model was also used to analysis the spatial distribution variation of R. kukunoris and the change of landscape connectivity when simulated the retrogressive succession of landscape. The main results are below: 1. There was peatland—meadow pattern which was typical patch—matrix landscape pattern in Zoige Natural Reserve. The meadow area occupied 79.42% of the entire landscape area, contagion index (CONTAG) was 79.00 which was far away the minimum value (0) but tend to the maximum value (100). Both of these showed that meadow was the largest part and the most continue units. It was shown that meadow was matrix of the landscape, which evolved the leading role to the landscape dynamic pattern. Though their area only occupies 18.08% of entire landscape area, peatlands were according to 82.9% of the total patches. Dynamic of the pattern between peatlands and meadows decided the spatial diffusion of R. kukunoris. 2.The model indicated that the other types of landscape not only expanded diffusion of R. kukunoris, but also have provided the potential channels for frog's connections among different peatlands. The spatial diffusion zone of R. kukunoris forced isolated patch peatlands to be potential landscape functional connectivity. The small peatlands, as stepping-stone, made the large peatlands connect as a functional one and promoted the integrated and dynamic connectivity of R. kukunoris in the whole landscape. 3. The simulation of “peatlands→meadows”retrogressive succession process indicated that the decrease of small peatlands will have potential effect to R. kukunoris because they must bred in peatlands and diffuse to other type of the landscape. Retrogressive succession process not only made the decrease of distribution of peatlands, patches number of peatlands and diffused area of R. kukunoris, but also reduced the connectivity among source patches. As stepping-stone, the disappearance of small peatlands will made the migration of R. kukunoris among patches more difficult. The model was an experiment of the amphibian behavior reaction to habitat losing and fragmentation. There were many factors that could influence the accuracy of model, such as the preference of animals to each type of landscape, the geographical data precision, reliable degree of model.
