为了揭示黄土高原南部地区不同质地类型土壤剖面坚实度的变化及其与土壤含水率的定量关系,以黄墡土、土娄土、裸露在地表的粘化层耕作剖面为研究对象,定位观测其0~45 cm土壤坚实度与含水率的变化。结果表明,黄墡土、土娄土、裸露在地表粘化层耕作剖面的犁底层平均坚实度均大于耕层,犁底层平均坚实度较耕层分别高194.8%,87.3%,10.4%;剖面土壤质地越粘其平均坚实度越大;土壤坚实度与含水率呈负相关关系;土壤坚实度变化速率为0时,以上3种土壤剖面临界含水率分别为0.1712,0.1757,0.1835;质地不同的土壤剖面坚实度时空变化特征有差异,其中黄墡土剖面0~20 cm土层土壤坚实度为350~500 kPa,受土壤含水率变化的影响较小;20~30 cm土层土壤的坚实度为500~1400 kPa,不易受外界环境影响;30 cm以下土层土壤坚实度为700~1600 kPa,受土壤含水率变化影响较大。土娄土剖面0~40 cm土层土壤坚实度为600~1200 kPa,受含水率变化影响较大;40 cm以下土层土壤坚实度稳定在1 800 kPa左右。粘化层剖面0~15 cm土层土壤坚实度在2000 kPa左右,受环境影响较...

细根周转与土壤养分密切相关,但由于根系研究方法的差异以及研究对象的不同,土壤养分对细根周转影响的研究存在不一致的结论。本文以水曲柳(Fraxinus mandshurica)人工林为对象,应用3种方法研究施氮肥对细根生产和周转的影响。结果表明:施肥降低了活细根现存生物量,但施氮肥样地细根年生产量平均值(93.105g·m-2·a-1)与对照样地(93.505g·m-2·a-1)没有差异,不同方法得出施氮肥样地细根平均周转率(0.917次·a-1)大于对照样地(0.710次·a-1);不同土层内细根的生产量显著不同,表层生产量最大,土层越深细根生产量越低,但细根周转率一般随土壤加深而加快;不同的研究方法得出细根的年生产量和周转率差异较大,分室模型法最高,其次是内生长土芯法,极差法和积分法最低。

A probabilistic soil moisture dynamic model is used to estimate the soil moisture probability distribution and plant water stress of irrigated cropland in the North China Plain. Soil moisture and meteorological data during the period of 1998 to 2003 were obtained from an irrigated cropland ecosystem with winter wheat and maize in the North China Plain to test the probabilistic soil moisture dynamic model. Results showed that the model was able to capture the soil moisture dynamics and estimate long-term water balance reasonably well when little soil water deficit existed. The prediction of mean plant water stress during winter wheat and maize growing season quantified the suitability of the wheat-maize rotation to the soil and climate environmental conditions in North China Plain under the impact of irrigation. Under the impact of precipitation fluctuations, there is no significant bimodality of the average soil moisture probability density function.

本研究围绕萤火虫的荧光素酶克隆表达和发光行为两方面来进行。首先对萤火虫的发光体系和发光行为进行了概述,然后对萤火虫荧光素酶cDNA进行了克隆表达分析,探讨了日行性和夜行性萤火虫荧光素酶的差异。最后通过萤火虫的饲养和发光行为观察研究,对卵发光的意义进行了探讨。本研究克隆了三种云南萤火虫的荧光素酶基因,并成功表达出两种有活性的荧光素酶。其中日行性萤火虫云南窗萤(Pyrocoelia pygidialis)的荧光素酶基因完成了首次克隆,并在大肠杆菌中成功表达出有功能的荧光素酶。测序结果表明:P. pygidialis荧光素酶的cDNA序列有1647个碱基,编码548个氨基酸残基。系统发育分析表明云南窗萤与同属三种发光很亮的萤火虫不形成单系,在一定程度上印证了P. pygidialis以及同属微弱发光的日行性种类与持续发出很亮荧光的夜行性种类构成窗萤属两个分支的观点;并且从发光器官的形态差异初步推断这两个分支之间是独立进化的。荧光素酶的结构模拟分析表明,云南窗萤荧光素酶的N端和C端结构域之间的裂沟处存在着五个多肽环,这是已知荧光素酶推测的活性位点。日行性的云南窗萤和三种夜行性萤火虫的荧光素酶有13个不同氨基酸,位于模拟的分子结构表面。这些氨基酸的差异,有利于进一步分析日行性和夜行性萤火虫荧光素酶结构的不同。对栉角雪萤(Diaphanes pectinealis)的荧光素酶cDNA也完成了克隆表达。结果表明其cDNA序列有1647个碱基,编码548个氨基酸残基,比从基因组中获得基因序列多3个碱基。体外表达产物在底物存在时发出黄绿色光。对雌光萤科Rhagophthalmus sp.的荧光素酶基因进行了克隆和氨基酸序列比对。得出其荧光素酶基因与来自同一科的荧光素酶基因有最高的一致性为90.7%,和鞘翅目另外三个科的物种的一致性在65%以下,显示出雌光萤科独特的分类地位。通过对采自昆明郊区的云南窗萤和一种扁萤属萤火虫的饲养和发光行为的观察,得出萤火虫的卵和蛹阶段持续发微弱的光的结论,且首次发现了卵发光在发育过程中的变化。本文对卵发光行为进行了描述,并在此基础上对萤火虫卵和蛹两阶段发光的原因和意义进行了探讨。

Among the functional nucleic acids studied, adenine-rich nucleic acids have attracted attention due to their critical roles in many biological processes and self-assembly-based nanomaterials, especially deoxyribonucleic acids (abbreviated as poly(dA)). Therefore the ligands binding to poly(dA) might serve as potential therapeutic agents. Coralyne, a kind of planar alkaloid, has been firstly found that it could bind strongly to poly(dA). This work herein reports an approach for visual sensing of the coralyne-poly(dA) interaction. This method was based on the coralyne inducing poly(dA) into the homo-adenine DNA duplex and the difference in electrostatic affinity between single-stranded DNA and double-stranded DNA with gold nanoparticles (GNPs). Furthermore, we applied the recognition process of the interaction between coralyne and poly(dA) into specific coralyne detection with the assistance of certain software (such as Photoshop). A linear response from 0 to 728 nM was obtained for coralyne, and a detection limit of 91 nM was achieved.

We report the morphology and phase behaviors of blend thin films containing two poly styrene-b-poly (methyl methacrylate) (PS-b-PMMA) diblock copolymers with different blending compositions induced by a selective solvent for the PMMA block, which were studied by transmission electron microscopy (TEM). The neat asymmetric PS-b-PMMA diblock copolymers employed in this study, respectively coded as a(1) and a(2), have similar molecular weights but different volume fractions of PS block (f(PS) = 0.273 and 0.722). Another symmetric PS-b-PMMA diblock copolymer, coded as s, which has a PS block length similar to that of a(1), was also used. For the asymmetric a(1)/a(2) blend thin films, circular multilayered structures were formed. For the asymmetric a(1)/symmetric s blend thin films, inverted phases with PMMA as the dispersed domains were observed, when the weight fraction of s was less than 50%. The origins of the morphology formation in the blend thin films via solvent treatment are discussed. Combined with the theoretical prediction by Birshtein et al. (Polymer 1992, 33, 2750), we interpret the formation of these special microstructures as due to the packing frustration induced by the difference in block lengths and the preferential interactions between the solvent and PMMA block.

A poly(o-toluidine) (POT)/2,5-dimercapto-1,3,4-thiadiazole (DMcT) composite was prepared. When POT and DMcT are mixed in a proper solvent, POT in a medium-oxidation state is reduced, and DMcT in turn is oxidized to its soluble dimer when the molar ratio of DMcT to POT is higher than 0.5. Therefore, the composite was soluble in organic solvents such as tetrahydrofuran (THF), dimethylformamide (DMF), and N-methyl-2-pyrrolidone (NMP) and exhibited very high electroactivity, two orders of magnitude higher than that of pure POT and three orders of magnitude higher than that of pure DMcT. Molecular-level contact between POT and DMcT is the reason for the improved catalytic effect of POT on DMcT, compared to that of polyaniline on DMcT. (C) 1999 The Electrochemical Society. Artemia has evolved a unique developmental pattern of encysted embryos to cope with various environmental threats. Cell divisions totally cease during the preemergence developmental stage from gastrula to prenauplius. The molecular mechanism of this, however, remains unknown. Our study focuses on the involvement of p90 ribosomal S6 kinase (RSK), a family of serine/threonine kinase-mediating signal transduction downstream of mitogen-activated protein kinase cascades, in the termination of cell cycle arrest during the post-embryonic development of Artemia-encysted gastrula. With immunochemistry, morphology, and cell cycle analysis, the identified Artemia RSK was established to be specifically activated during the post-embryonic and early larval developmental stages when arrested cells of encysted embryos resumed mitoses. In vivo knockdown of RSK activity by RNA interference, kinase inhibition, and antibody neutralization consistently induced defective larvae with distinct gaps between the exoskeleton and internal tissues. In these abnormal individuals, mitoses were detected to be largely inhibited in the affected regions. These results display the requirement of RSK activity during Artemia development and suggest its role in termination of cell cycle (G(2)/M phase) arrest and promotion of mitogenesis. Our findings may, thus, provide insights into the regulation of cell division during Artemia post-embryonic development and reveal further aspects of RSK functions.

The population genetic structure of the crimson snapper Lutjanus erythropterus in East Asia was examined with a 427-bp hypervariable portion of the mtDNA control region. A total of 262 samples were collected and 75 haplotypes were obtained. Neutrality tests (Tajima's and Fu's) suggested that Lutjanus erythropterus in East Asia had experienced a bottleneck followed by population expansion since the late Pleistocene. Despite the low phylogeographic structures in mtDNA haplotypes, a hierarchical examination of populations in 11 localities from four geographical regions using analysis of molecular variance (AMOVA) indicated significant genetic differentiation among regions (Phi(CT) = 0.08564, p < 0.01). Limited gene flow between the eastern region (including a locality in the western Pacific Ocean and two localities in the East Sea) and three geographic regions of the South China Sea largely contributed to the genetic subdivision. However, comparisons among three geographic regions of the South China Sea showed little to no genetic difference. Populations of Lutjanus erythropterus in East Asia are inferred to be divided into two major groups: an eastern group, including populations of the western Pacific Ocean and the East Sea, and a South China Sea group, consisting of populations from northern Malaysia to South China. The results suggest that fishery management should reflect the genetic differentiation and diversity in East Asia. (c) 2006 International Council for the Exploration of the Sea. Seed rearing is an important part in large scale clam culture industry. Since the nutritional history affects early development in bivalve, the condition of larval nutrition plays a key role in successful seed rearing. So far, the molecular mechanism of nutrient uptake in bivalve larvae is unclear. As one of the important proteolytic enzymes, cathepsin B of several organisms has been reported to be involved in digestion. We intended to analyze whether cathepsin B is involved in larval nutrient metabolism in the economic bivalve, clam Meretrix meretrix. The full length of M. meretrix cathepsin B (MmeCB) cDNA was cloned, which is 1647 bp with an open reading frame of 1014 bp. The deduced amino acid sequence encoded a preproenzyme of 337 residues with Cys-114, His-282 and Asn-302 composing cathepsin B activity center. The temporal and spatial expressions of MmeCB mRNA were examined from trochophore to post larva stages by whole mount in situ hybridization. In trochophore stage, no detectable signal was found. In the later three stages, MmeCB mRNA was detected in the digestive gland, suggesting a possible role of MmeCB in digestion. Moreover, MmeCB mRNA was also observed in the epidermal cells in D-veligers. Cathepsin B specific inhibitor (CA074 methyl ester) was applied to block the activity of cathepsin B in unfed larvae. The average shell lengths of treated larvae were smaller than that in control groups. The results of mRNA epidermal distribution and inhibitor treatment in D-veligers indicated that MmeCB may be also associated with other pathway of nutrient metabolism in larval epidermis. The overall results in this paper revealed that MmeCB might play a role in larval nutrient metabolism. (C) 2008 Elsevier B.V. All rights reserved.
