Serine protease inhibitors, critical regulators of endogenous proteases, are found in all multicellular organisms and play crucial roles in host physiological and immunological effector mechanisms. The first mollusk serine proteinase inhibitor (designated AISPI) cDNA was obtained from the bay scallop Argopecten irradians by randomly sequencing a whole tissue cDNA library and rapid amplification of cDNA ends (RACE). The full-length cDNA of the scallop serine protease inhibitor was 1020 bp, consisting of a 5'-terminal untranslated region (UTR) of 39 bp, a 3'-terminal UTR of 147 bp with a canonical polyadenylation signal sequence AATAAA and a poly(A) tail, and an open reading frame of 834 bp. The AISPI cDNA encoded a polypeptide of 278 amino acids with a putative signal peptide of 22 amino acids and a mature protein of 256 amino acids. The deduced amino-acid sequence of AISPI contained six tandem and homologous domains similar to that of Kazal-type serine protease inhibitors, including the conserved sequence C-X(7)-C-X(6)-Y-X(3)-C-X(2,3)-C and six cysteine residues responsible for the formation of disulfide bridges, indicating that the AISPI protein from bay scallop should be a member of the Kazal-type serine protease inhibitor family. The temporal expression of AISPI was measured by semi-quantitative RT-PCR after injury or bacterial challenge. After the adductor muscle was wounded or injected with Vibrio anguillarum, the expression of AISPI mRNA in hemolymph was up-regulated and reached the maximum level at 8 and 16 h, respectively, and then progressively dropped back to the original level. The results indicated that AISPI could play an important role in injury healing and immune response in mollusks as it could be induced by injury and bacterial challenge. (c) 2005 Elsevier Ltd. A gene-clone-library-based molecular approach was used to study the nirS-encoding bacteria-environment relationship in the sediments of the eutrophic Jiaozhou Bay. Diverse nirS sequences were recovered and most of them were related to the marine cluster I group, ubiquitous in estuarine, coastal, and marine environments. Some NirS sequences were unique to the Jiaozhou Bay, such as the marine subcluster VIIg sequences. Most of the Jiaozhou Bay NirS sequences had their closest matches originally detected in estuarine and marine sediments, especially from the Chesapeake Bay, indicating similarity of the denitrifying bacterial communities in similar coastal environments in spite of geographical distance. Multivariate statistical analyses indicated that the spatial distribution of the nirS-encoding bacterial assemblages is highly correlated with environmental factors, such as sediment silt content, NH4+ concentration, and OrgC/OrgN. The nirS-encoding bacterial assemblages in the most hypernutrified stations could be easily distinguished from that of the least eutrophic station. For the first time, the sedimentological condition was found to influence the structure and distribution of the sediment denitrifying bacterial community.

The technique of balancing cross-sections, an important method for studying the tectonic history of sedimentary basins, has many applications. It enables one to compile charts for petroleum exploration and development, and growth sections of ancient structures can be restored so that the structural growth history can be studied. In order to study tectonic evolution in the Zhuanghai area of the Bohai-Bay basin, we selected two seismic profiles and compiled two structural growth sections. Based on the two balanced cross-sections, the evolution can be divided into four phases: the Triassic-Middle Jurassic phase, Late Jurassic - Cretaceous phase, Palaeogene extension phase, and Late Palaeogene-to-present phase. The whole area was uplifted during the Triassic-Middle Jurassic phase because of intense extrusion stress related to the Indo-China movement. During the Late Jurassic and Early Cretaceous, intense extension occurred in east China, and the whole area rifted, leading to the deposition of a thick sedimentary sequence. In the Late Cretaceous, the area suffered uplift and compression associated with the sinistral strike slip of the Tanlu fault. In the Palaeogene, a rifting basin developed in the area. Finally, it became stable and was placed in its present position by dextral strike-slip motion. In addition, some problems associated with compiling balanced cross-sections are discussed.

Offshore active faults, especially those in the deep sea, are very difficult to study because of the water and sedimentary cover. To characterize the nature and geometry of offshore active faults, a combination of methods must be employed. Generally, seismic profiling is used to map these faults, but often only fault-related folds rather than fracture planes are imaged. Multi-beam swath bathymetry provides information on the structure and growth history of a fault because movements of an active fault are reflected in the bottom morphology. Submersible and deep-tow surveys allow direct observations of deformations on the seafloor (including fracture zones and microstructures). In the deep sea, linearly aligned cold seep communities provide indirect evidence for active faults and the spatial migration of their activities. The Western Sagami Bay fault (WSBF) in the western Sagami Bay off central Japan is an active fault that has been studied in detail using the above methods. The bottom morphology, fractured breccias directly observed and photographed, seismic profiles, as well as distribution and migration of cold seep communities provide evidence for the nature and geometry of the fault. Focal mechanism solutions of selected earthquakes in the western Sagami Bay during the period from 1900 to 1995 show that the maximum compression trends NW-SE and the minimum stress axis strikes NE-SW, a stress pattern indicating a left-lateral strike-slip fault.

Studies of abundance, diversity and distribution of antibiotic-resistant bacteria and their resistance determinants are necessary for effective prevention and control of antibiotic resistance and its dissemination, critically important for public health and environment management. In order to gain an understanding of the persistence of resistance in the absence of a specific antibiotic selective pressure, microbiological surveys were carried out to investigate chloramphenicol-resistant bacteria and the chloramphenicol acetyltransferase resistance genes in Jiaozhou Bay after chloramphenicol was banned since 1999 in China. About 0.15-6.70% cultivable bacteria were chloramphenicol resistant, and the highest abundances occurred mainly in the areas near river mouths or sewage processing plants. For the dominant resistant isolates, 14 genera and 25 species were identified, mostly being indigenous estuarine or marine bacteria. Antibiotic-resistant potential human or marine animal pathogens, such as Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis and Shewanella algae, were also identified. For the molecular resistance determinants, the cat I and cat III genes could be detected in some of the resistant strains, and they might have the same origins as those from clinical strains as determined via gene sequence analysis. Further investigation about the biological, environmental and anthropogenic mechanisms and their interactions that may contribute to the persistence of antibiotic-resistance in coastal marine waters in the absence of specific antibiotic selective pressure is necessary for tackling this complicated environmental issue.

Environmental microbiology investigation was performed to determine the molecular diversity of beta-lactamase genes among ampicillin-resistant bacteria from Jiaozhou Bay. beta-lactamase genes were detected in 93.8% of the bacterial isolates identified as Enterobacteriaceae. The most frequently detected gene was bla(TEM), followed by bla(SHV), bla(OAX-1), bla(MOX) and bla(CMY). Most of the isolates (68.8%) were positive for the intI1 integrase gene, and two isolates were also found for the intI2 gene. The dfr and aadA gene cassettes were predominant. Anthropogenic contamination from onshore sewage processing plants might contribute predominantly to the beta-lactamase gene reservoir in the studied coastal waters. Environmental antibiotic-resistant bacteria and resistance genes may serve as bioindicators of coastal environmental quality or biotracers of the potential contamination sources. This is the first report of the prevalence and characterization of beta-lactamase genes and integrons in coastal Enterobacteriaceae from China.

The stratigraphic architecture, structure and Cenozoic tectonic evolution of the Tan-Lu fault zone in Laizhou Bay, eastern China, are analyzed based on interpretations of 31 new 2D seismic lines across Laizhou Bay. Cenozoic strata in the study area are divided into two layers separated by a prominent and widespread unconformity. The upper sedimentary layer is made up of Neogene and Quaternary fluvial and marine sediments, while the lower layer consists of Paleogene lacustrine and fluvial facies. In terms of tectonics, the sediments beneath the unconformity can be divided into four main structural units: the west depression, central uplift, east depression and Ludong uplift. The two branches of the middle Tan-Lu fault zone differ in their geometry and offset: the east branch fault is a steeply dipping S-shaped strike-slip fault that cuts acoustic basement at depths greater than 8 km, whereas the west branch fault is a relatively shallow normal fault. The Tan-Lu fault zone is the key fault in the study area, having controlled its Cenozoic evolution. Based on balanced cross-sections constructed along transverse seismic line 99.8 and longitudinal seismic line 699.0, the Cenozoic evolution of the middle Tan-Lu fault zone is divided into three stages: Paleocene-Eocene transtension, Oligocene-Early Miocene transpression and Middle Miocene to present-day stable subsidence. The reasons for the contrasting tectonic features of the two branch faults and the timing of the change from transtension to transpression are discussed. The Qikou Depression is the largest hydrocarbon bearing depression in the western part of the Bohai bay basin, dominated by fan delta and lacustrine strata with volcanic and volcaniclastic rocks. In this study, the formation pressures and hydrochemistry of the formation water in the Qikou depression are investigated. It is found that a significant overpressure occurs in the Dongying (Ed) Formation and the first member (Est), the second member (Es2), the third member (Es3) of the Shahejie Formation. The pressure coefficients commonly range from 1.2 to 1.6 with the highest pressure coefficient being 1.7. The analysis of hydrochemistry data shows that the whole depression is dominated by NaHCO3 water type. The concentration of total dissolved solid (TDS) ranges from 2.13 to 53.16 g/L and shows a distinct vertical variation of salinity and ion ratios. High salinity water (TDS> 10 g/L) occurs below a depth of 2500 m, which coincides with the presence of the overpressured system. However, the increasing trend of TDS is diminished below 3500 m because the generation of organic acids in Qikou Depression is inhibited in the presence of overpressure. The analysis of the relationship among different ions indicates that the present-day characteristics of the formation water result from the albitization of feldspar and the dissolution of sodium-rich silicate minerals and halite in the different hydrochemical and pressure systems. The Jiyang superdepression is one of the richest hydrocarbon accumulations in the Bohai Bay basin, eastern China. Comprehensive seismic methods have been used in buried hill exploration in Jiyang to describe these fractured reservoirs better. Accurate seismic stratigraphic demarcation and variable-velocity mapping were applied to reveal the inner structure of the buried hills and determine the nature of the structural traps more precisely. Based on the analysis of rock properties and the characteristics of well-developed buried hill reservoirs, we have successfully linked the geology and seismic response by applying seismic forward technology. Log-constrained inversion, absorption coefficient analysis and tectonic forward-inversion with FMI loggings were applied to analyse and evaluate the buried hill reservoirs and gave satisfying results. The reservoir prediction was successful, which confirmed that the comprehensive utilization of these methods can be helpful in the exploration of buried hill reservoirs.

A three-dimensional ocean circulation model, called Princeton Ocean Model (POM), is employed to simulate tides and tidal currents in Liaodong Bay. The nested grid technique is adopted to improve the computation precision. Computed harmonic constants of M-1, M-2 tides at five tidal gauge stations and surface elevations at two oil platforms are compared with those observed, and show good agreements with them. Based on the calculated results, the co-amplitude and co-phase tag lines of nil and M-2 tidal constituents, the residual current field of M-2 constituent, tidal form, tidal Current ellipse and the moving style of tidal current are given. It is found that diurnal tidal constituents have no amphidromic point whereas semi-diurnal constituents have one in the region of interest. Meanwhile, some meaningful results are concluded and presented, which are conducive to a thorough knowledge of the characteristics of tides and tidal currents in the Liaodong Bay.

[1] The evolution of freshwater plumes and the associated salinity fronts in the northern Bay of Bengal ( henceforth the bay) is studied using rotated empirical orthogonal function (REOF) analysis and extended associate pattern analysis (EAPA). The results show that sea surface salinity distribution is featured by eastern-bay and western-bay plumes in the northern bay during different seasons. The western-bay plume begins in early July, peaks in late August, and then turns into a bay-shaped plume with the two plumes in either side of the bay, which peaks in late October. The southward extension of the western-bay plume can be explained by the southwestward geostrophic flow associated with the cyclonic gyre in the northern bay, which counters the northeastward Ekman drift driven by wind stress. The offshore expansion of the western-bay plume is induced by the offshore Ekman drift which also produces a salinity front near the east coast of India. The bay-shaped plume appears when the cyclonic gyre shifts westward and a weak anticyclonic gyre occupies the northeastern bay. As the season advances, the western part of the bay-shaped plume decays while the eastern part persists until the following June, which is believed to be associated with the anticyclonic gyre in the northern bay. The evolution of the plumes except the eastern part of the bay-shaped plume in fall can be partly explained by the seasonal variation of mass transport associated with the Sverdrup balance. The fact that the western-bay (eastern-bay) plume appears when surface freshwater flux in the northeastern bay increases ( decreases) dramatically suggests that the plumes are not produced directly by surface freshwater flux. River discharge seems to be the freshwater source for the plumes and has little to do with the evolution of the plumes.

A comprehensive field study has been undertaken to investigate sediment resuspension dynamics in the Moreton Bay, a large semi-enclosed bay situated in South East Queensland, Australia. An instrumented tripod, which housed three current meters, three OBS sensors and one underwater video camera, was used to collect the field data on tides, currents, waves and suspended sediment concentrations at four sites (Sites 1, 2, 4, and 5) in the bay. Site I was located at the main entrance, Site 2 at the central bay in deep water, and Sites 4 and 5 at two small bays in shallow water. The bed sediment was fine sand (d(50) = 0. 2 mm) at Site 1, and cohesive sediment at the other three sites. Based on the collected field data, it is found that the dominant driving forces for sediment resuspension are a combination of ocean swell and tidal currents at Site 1, tidal currents at Site 2, and wind-waves at Sites 4 and 5. The critical bed shear stress for cohesive sediment resuspension is determined as 0. 079 Pa in unidirectional flow at Site 2, and 0. 076 Pa in wave-induced oscillatory flow at Site 5.

We examined the growth, survival and immune response of the scallop, Chlamys farreri, during a 1-year period in deep water of Haizhou Bay. Scallops were cultured using two methods: (1) in lantern nets at a 5 m depth and (2) in a bottom culture system (sleeves) on the seabed at about a 25 m depth. Shell heights, meat dry weight and immune activities in the haemolymph (superoxide dismutase and myeloperoxidase) were measured bimonthly or quarterly from July 2007 to June 2008. Survival was measured at the end of the study and environmental parameters in the experimental layers were monitored during the experiment. The growth and immune activities of scallops were lower when the water temperature was high, which was consistent with the main mortality occurring in summer. The growth and immunity of scallops were higher in the suspended culture than in the bottom culture during the experiment, with the exception of shell growth during the last study period. Survival of scallops in the suspended culture (54.6 +/- 12.3%) was significantly lower than that in the bottom culture (86.8 +/- 3.5%) at the end of this study. We conclude from our results that the high mortality of C. farreri can be prevented by culturing them in a bottom culture system before November of the first year, and then transferring them to a suspended culture to improve scallop production.

The species composition and abundance of microzooplankton at 10 marine and five coastal stations (Hongdao, Daguhe, Haibohe, Huangdao and Hangxiao) in the Jiaozhou Bay (Qingdao, China) were studied in 2001. The microzooplankton community was found to be dominated by Tintinnopsis beroidea, Tintinnopsis urnula, Tintinnopsis brevicollis and Cvdonellopsis sp. The average abundance of microzooplankton was highly variable among stations. Specifically, the abundance of microzooplankton was higher at inshore stations and lower in the center of the bay (St. 5), bay mouth (St. 9) and outside the bay (St. 10). The highest average annual densities (346 ind./L) was observed at St. 3, while the lowest (55 ind./L) was at St. 10. Two abundance peaks were recorded in May (324 ind./L) and February (300 ind./L). The distribution of microzooplankton in three sampling layers at the 10 stations was relatively homogenous and the abundance decreased slightly as the water depth increased. At coastal stations, the highest average annual density was recorded at Hongdao Station (677 ind./L), followed by Daguhe Station (616 ind./L), Haibohe Station (400 ind./L), Huangdao Station (275 ind./L) and Hangxiao Station (73 ind./L). Furthermore, a 24-h sampling analysis conducted at Hangxiao Station revealed that the microzooplankton assemblages were characterized by a bimodal diel vertical migration pattern, with the highest densities occurring at dusk (154 ind./L), followed by dawn (146 ind./L), noon (93 ind./L) and midnight (77 ind./L). The density of microzooplankton in the Jiaozhou Bay was in the middle range of the densities of temperate coastal waters worldwide.
