We report the morphology and phase behaviors of blend thin films containing two poly styrene-b-poly (methyl methacrylate) (PS-b-PMMA) diblock copolymers with different blending compositions induced by a selective solvent for the PMMA block, which were studied by transmission electron microscopy (TEM). The neat asymmetric PS-b-PMMA diblock copolymers employed in this study, respectively coded as a(1) and a(2), have similar molecular weights but different volume fractions of PS block (f(PS) = 0.273 and 0.722). Another symmetric PS-b-PMMA diblock copolymer, coded as s, which has a PS block length similar to that of a(1), was also used. For the asymmetric a(1)/a(2) blend thin films, circular multilayered structures were formed. For the asymmetric a(1)/symmetric s blend thin films, inverted phases with PMMA as the dispersed domains were observed, when the weight fraction of s was less than 50%. The origins of the morphology formation in the blend thin films via solvent treatment are discussed. Combined with the theoretical prediction by Birshtein et al. (Polymer 1992, 33, 2750), we interpret the formation of these special microstructures as due to the packing frustration induced by the difference in block lengths and the preferential interactions between the solvent and PMMA block.

利用浸渍法制备含Ce-La稀土助剂改性的Pt-Rh三效催化剂,采用多种表征手段和高温抗硫水热老化实验,考察Ce-La稀土对Al2O3载体的热稳定性和催化剂活性的影响。结果表明:稀土La2O3的添加主要改善载体Al2O3的高温高比表面性能;CeO2的引入明显提高了CO和NO的催化转化活性,但对HC的转化几乎无影响;CeO2的添加也促进了催化剂的抗硫水热老化性能,并显著拓宽催化剂的三效窗口。主要归因于CeO2的添加促进了催化剂上CO、HC和NO间的相互作用以及CeO2所特有的储氧功能.

Isothermal and non-isothermal crystallization kinetics of three metallocene-catalysed short-chain-branched polyethylene (SCBPE) fractions with different degree of branching were investigated by using differential scanning calorimetry (DSC). Narrow molecular weight fractions (M-w = 20,000 and M-w/M-n < 1.15) are used and the degree of branching (CH3 per 1000C) are 1.6, 10.4, 40 respectively. The regime I - II transition temperature are 119.8C, 115.9 degreesC, 113.3 degreesC with the decreasing of degree of branching. Increasing the branch content decreases the rate of secondary nucleation, i,relative to the rate of surface spreading and so increases the range of supercooling over which regime I exists. The rate of bulk crystallization for both isothermal and non-isothermal crystallization decreases with the increasing of degree of branching. Both Ozawa Equation and Kissinger Equation are invalid for non-isothermal crystallization kinetics of SCBPE fractions,that means the effects of the branched chain on crystallization process are more complex than expected.

本工作用DSC方法对三种不同支化度的茂金属短链支化聚乙烯的等温、非等温结晶行为进行了研究 .样品为乙烯和己烯 1的共聚物 ,短支链主要为正丁基 ,分子量Mw =2 0 ,0 0 0 ,Mw/Mn<1 15 ,支化度 (每10 0 0C中CH3 数目 )分别为 1 6、10 4、40 .实验结果表明 ,茂金属短链支化聚乙烯结晶方式Ⅰ Ⅱ转变温度随支化度增加而降低 ,分别为 119 8℃、115 9℃、113 3℃ ;同时支链的存在降低了二次成核速率 ,增大了方式Ⅰ的结晶范围 ,总的结晶速度随支化度增大而减小

A blend of polyethersulfone (PES) and polycarbonate (PC) with a ratio of 40/60 was studied by scanning electron microscopy (SEM), dynamic mechanical analysis, and transmission electron microscopy (TEM). It was found that the PES-PC blend is a partially miscible, two-phase system, and an interfacial layer exists between the phases of PES and PC. Specific interaction resulting from the n-complex between PES and PC provides the driving force for formation of the interfacial layer. In addition, phase inversion behavior was also observed for the 40/60 composition.

Layered double hydroxides (LDHs) pillared with heteropolyoxometalate have been synthesized via both restructuring of hydrotalcite-like compounds and direct anion exchange under microwave field. LDHs and their derivatives are both found to be efficient for alkylation of isobutane with butene with a higher butene conversion and selectivity towards C-8(0).

C-type lectin is a family of Ca2+ dependent carbohydrate-recognition proteins which play crucial roles in the innate immunity of invertebrates by mediating the recognition of host cells to pathogens and clearing microinvaders as a pattern recognition protein (PRP). The cDNA of Zhikong scallop Chlamys farreri C-type lectin (designated CFLec-1) was cloned by expressed sequence tag (EST) and RACE techniques. The full-length cDNA of CFLec-1 was 1785 bp, consisting of a 5'-terminal untranslated region (UTR) of 66 bp and an unusually long 3' UTR of 1040 bp with seven polyadenylation signal sequences AATAAA and a poly(A) tail. The CFLec-1 cDNA encoded a polypeptide of 221 amino acids with a putative signal peptide of 15 amino acid residues and a mature protein of 206 amino acids. Analysis of the protein domain features indicated a typical long-form carbohydrate-recognition domain (CRD) of 130 residues in the CFLec-1 deduced amino acid sequence. The expression pattern of CFLec-1 transcripts in healthy and bacterial challenged scallops was studied by semi-quantitative RT-PCR. mRNA transcripts of CFLec-1 could be mainly detected in the tissues of haemocytes, gill, gonad and mantle of unchallenged scallops, whereas the expression of CFLec-1 transcripts was increased in all the tested tissues after heat-killed Vibrio anguillarum challenge. The temporal expression of CFLec-1 mRNA in haemolymph challenged by Micrococcus luteus and V anguillarum was both up-regulated and reached the maximum level at 8 and 16 It post stimulation, respectively, and then dropped back to the original level. In order to investigate its immune functions, CFLec- I was recombined and expressed in Escherichia coli BL21(DE3)-pLysS as a fusion protein with thioredoxin. The recombinant CFLec-1 agglutinated bacteria E. coli JM109 in vitro, and the agglutination was Ca2+ dependent which could be inhibited by EDTA. But it did not agglutinate M. luteus, Candida lipolytica and animal erythrocytes including rabbit, rat, mouse, chicken, human group A, human group B, human group O. Meanwhile, the recombinant CFLec-1 could inhibit the growth of both E. coli JM 109 and M. luteus, but no inhibition activity against V anguillarum. These result indicated that CFLec-1 was a constitutive and inducible PRP which was involved in the reorganization and clearance of invaders in scallop. (c) 2006 Elsevier Ltd. All rights reserved.

本文通过在胶州湾及邻近海区的野外调查,其中包括海底表层沉积物采集、海水样采集、浊度测量、ADCP走航式测量,获得大量数据;结合历史资料,用如下方法研究了胶州湾及邻近海区的沉积动力学:(1)以地貌单元完整性和特征距离一致性为依据,将研究区划分为若干小区;采用Gao-Collins粒径趋势分析模型提取海底沉积物净搬运方向的信息,并寻找天然示踪沉积物方面的证据;(2)通过现代声学理论分析,获得了用ADCP进行走航式悬沙浓度测量的方法;(3)用~(210)Pb方法和沉积物平衡法计算百年尺度的沉积速率;(4)通过对原始水槽试验数据的重新分析,获得了用于沉积物搬运率计算的经典公式之一-Bagnold型方程的系数校正公式。胶州湾及邻近海区海底沉积物种类繁多,而且呈现斑状分布特征。沉积物粒径趋势结果显示,胶州湾内的大型垄脊和口门延伸至外海的深槽尾部是两个主要的沉积物汇聚区,这不仅与地貌及水动力相吻合,还与根据重矿物扩散趋势一致;与颗石藻化石空间分布型式变有一定的相关性。对于相同的沉积物样品来说,基于不同粒度分析方法所获得的粒径趋势矢量具有一致性。因此,粒径趋势分析模型不仅可用于采样间距不一致、沉积物类型复杂的深水海湾;也可用于传统方法获得的历史粒度资料的处理。胶州湾沙滩供沙的主要机制是通过风暴潮期间强烈的动力作用,使沙滩沉积物以悬移质、底移质形式向深水区扩散或搬运。胶州湾悬沙浓度的时空变化主要与水动力状况、陆地径流和天气状况有关。野外实测悬沙浓度和用卫星图像遥感算法获得的结果均表明,胶州湾西北部潮滩外缘海域悬沙浓度较高,海湾东部和邻近海区悬沙浓度较低。悬沙浓度的年变化、季节变化均较为显著。春季、冬季风浪大,悬沙浓度较高;夏季风暴潮通常导致悬沙浓度急剧升高。理论分析获得了海水中悬沙浓度与ADCP输出的回声强度之间的函数关系式;在胶州湾口门进行走航式测量,同步采水样进行现场校正来率定相应的ADCP声学信号,获得了计算悬沙浓度的半经验公式;进而得到走航断面准同步的悬沙浓度剖面。结果发现,胶州湾口门断面流速沿水深呈垂向均匀分布的特征较显著;悬沙浓度具有一定的水平成层分布特征,水流流速的增大有利于悬沙浓度的提高。在普通大潮潮周期内,胶州湾悬移质输向外海。多种研究方法表明,胶州湾沉积速率较小,其量级为10~0mm yr~(-1);这主要与胶州湾沉积物来源较少有关。

结核分枝杆菌(M.tuberculosis),俗称结核杆菌,是引起结核病的病原菌。可侵犯全身各器官,但以肺结核为最多见。结核病至今仍为重要的传染病。估计世界人口中1/3感染结核分枝杆菌。据WHO报道,每年约有800万新病例发生,至少有300万人死于该病。我国建国前死亡率达200-300人/10万,居各种疾病死亡原因之首,建国后,随着人民生活水平提高,卫生状态改善,特别是开展了群防群治,儿童普遍接种卡介苗,结核病的发病率和死亡率大为降低。由于耐药结核的出现已及HIV共感染等因素影响,目前发病率又有上升趋势。对于结核病的防治而言,其首要问题是结核病的诊断,而结核病的实验室检查已成为临床医生诊断结核病、判断疗效和评估预后不可缺少的手段。结核病的临床诊断主要根据病史、痰培养、涂片抗酸染色、胸片等结果,其中痰结核检查是诊断肺结核的重要依据之一,检测方法常规采用细菌学检查。痰中细菌学检查主要采用痰涂片和培养方法,涂片虽简单易行,但阳性率低,培养虽为金标准,但周期太长,均难以满足临床需要。细菌对部分患者可造成误诊或漏诊。近年来随着现代科学技术的不断进展,分子生物学技术在结核病诊断方面取得了显著的成绩,实时动态荧光定量(FQ—PCR)技术又是在原有的PCR技术上一次质的飞跃。 尽管许多文献已经报道了结核耐药的分子机理,但耐药的分子机理并没有充分了解。本研究利用全基因组鸟枪法测序和高通量测序技术,测定了两株结核分枝杆菌菌株TFJA和TFJB全基因组序列,分别为药物敏感株和耐多药株。在序列初步拼接后,获得了由189个DNA contigs(DNA序列连接群)组成的基因组框架图。借助10kb左右插入片段克隆的正反向末端序列信息以及PCR扩增技术,确定了全部DNA contigs在基因组上的物理位置,最后补上了所有“间隙(gap)” ,得到了全基因组序列。三种限制性酶切物理图谱证明了序列拼接的正确性。两株结核菌基因组大小为4.4Mbp左右,GC含量都在65.6%左右,其中TFJB株比TFJA株大7kbp,分别具有3591、3648个基因,83.7%的基因位于先导链上。我们比较了两株细菌的949SNPs位点,发现94个在TFJB中不同nsSNPs,这些SNPs位点的碱基在其它的结核分枝杆菌基因组中是相同的。我们去掉了那些代谢途径与毒力代谢无关的基因,最后选择了6个基因的18个SNP,检测这些SNP在277株耐药或者药物敏感结核分枝杆菌中的保守性以及其耐药相关性。用PCR扩增含有SNP的序列,用统计学检验这些SNP是否耐药相关,结果发现,TFJB0109_CDS04405基因的407位点SNP(g->a)和TFJB0109_CDS00149基因上的SNP(720a->c,722插入t,727t->g,729g->t,以及735g->t)可能与SM耐药相关,其P-value分别是0.04128、0.02160和0.01394(见表3)。 结核是一种慢性传染病,其病原体为TB。在全基因组序列分析研究的同时,我们筛选到一套灵敏度较高、特异性较好的REAL TIME-PCR检测引物,并用它对105份不同发病时间的病人标本进行了检测。初步结果表明,应用本方法,样本检出率为98%,这一结果比涂片及分离培养更灵敏。

The effect of water temperature on gut mass and digestive enzyme activity in sea cucumber Apostichopus japonicus, including relative gut mass (RGM), amylase, lipase, pepsin and trypsin activities were studied at temperatures of 7, 14, 21, and 28A degrees C over a period of 40 days. Results show that RGM significantly decreased after 40 days at 21A degrees C and markedly decreased over the whole experiment period at 28A degrees C; however, no significant effect of duration was observed at 7 or 14A degrees C. At 14A degrees C, trypsin activity significantly decreased over 10 and 20 days, then increased; amylase and trypsin activity significantly decreased after 40 days at 28A degrees C. However, no significant effect of duration was found on amylase, pepsin or trypsin activities in the other temperature treatment groups. At 28A degrees C, lipase activity peaked in 20 days and then markedly decreased to a minimum at the end of the experiment. On the other hand, pepsin activity at 28A degrees C continuously increased over the whole experimental period. Principle component analysis showed that sea cucumbers on day 40 in the 21A degrees C group and in the previous 20 days in the 28A degrees C group were in the prophase of aestivation. At 28A degrees C, sea cucumbers aestivated at 30-40 days after the start of the experiment. It is concluded that the effect of temperature on the digestion of A. japonicus is comparatively weak within a specific range of water temperatures and aestivation behavior is accompanied by significant changes in RGM and digestive enzyme activities.

在食物中含10%和20%蛋白质的条件下,采用食物平衡法测定了单宁酸对根田鼠食物摄入量和蛋白质消化率的作用。食物蛋白质含量为10%时,第1-5天,单宁酸对根田鼠食物摄入量具有显著的抑制作用,自第6天,其作用不明显,以3%和6%单宁酸处理的根田鼠,其食物蛋白质消化率较对照组分别降低22%和47.67%;在食物蛋白质含量为20%的条件下,单宁酸对根田鼠的食物摄入量和蛋白质消化率无显著作用。上述结果验证了植物次生化合物能抑制植食性小哺乳动物食摄入量及蛋白质消化率的假设。
