Practical testing of the feasibility of cyanobacterial inoculation to speed up the recovery of biological soil crusts in the field was conducted in this experiment. Results showed that cyanobacterial and algal cover climbed up to 48.5% and a total of 14 cyanobacterial and algal species were identified at the termination of inoculation experiment; biological crusts' thickness, compressive and chlorophyll a content increased with inoculation time among 3 years; moss species appeared in the second year; cyanobacterial inoculation increased organic carbon and total nitrogen of the soil; total salt, calcium carbonate and electrical conductivity in the soil also increased after inoculation. Diverse vascular plant communities composed of 10 and 9 species are established by cyanobacterial inoculation on the windward and leeward surface of the dunes, respectively, after 3 years. The Simpson index for the above two communities are 0.842 and 0.852, while the Shannon-Weiner index are 2.097 and 2.053, respectively. In conclusion, we suggest that cyanobacterial inoculation would be a suitable and effective technique to recover biological soil crusts, and may further restore the ecological system. In this report, recombinant interieukin-8 (rIL-8) was produced and its activity tested for the first time in fish. The rainbow trout rIL-8 was produced in Escherichia coli and purified using a 6xHis tag at the N-terminus. The rIL-8 induced a dose-dependent migration of head kidney leukocytes at concentrations from 0.1 to 10 ng/ml, with a peak response at 1 ng/ml. Trout rIL-8 also had a significant effect on superoxide production by head kidney cells, with maximal, activity at 0.1 and 1 ng/ml. When injected intraperitoneally into trout, rIL-8 had a clear effect on total leukocyte number in the peritoneal cavity, with increasing doses (up to 5 mu g) eliciting more cells. Of three leukocyte types distinguished, neutrophils were the dominant cell type, especially at higher rIL-8 concentrations. In contrast, the proportion of macrophages and lymphocytes decreased with rIL-8 administration, suggesting that they were not attracted at the same rate as neutrophils. (C) 2007 Elsevier Ltd. Micronutrients play a very important role in biological processes for wastewater treatment. Many industrial wastewaters lack in nutrients (macronutrients and micronutrients) required for microbial growth, and this is one of the main problems at many activated sludge plants treating industrial wastewater. The microbial community structure is one of the important factors controlling the pollutant-degrading capacity of biological wastewater treatment system. In this study, the concentrations of micronutrients of the textile wastewater discharged from a textile plant were determined, and the effects of micronutrients on treatment efficiency and microorganism community structure of the biological treatment system were studied. The results showed that the optimal concentrations of magnesium, molybdenum, zinc, thiamine and niacin in the textile wastewater were 5.0, 2.0, 1.0, 1.0 and 1.0mg/L, respectively. The COD removal rates when magnesium, molybdenum, zinc, thiamine and niacin were added individually to the wastewater in their optimal concentrations were 1.8, 1.4, 1.3, 1.6 and 2.2 times of that of the control, respectively. The improving effects of combinations of zinc and thiamine, zinc and niacin, thiamine and niacin were better than single micronutrient. The diversity of quinones (DQ) changed significantly after the micronutrient was added into the wastewater treatment system. This indicated that there was probably a feasibility of optimizing the biological treatment performances and microorganism community structure of textile wastewater treatment system through micronutrient supplement.

Micronutrients play an important role in biological processes for wastewater treatment. Many industrial wastewaters lack in nutrients required for microbial growth, and this is one of the problems at many activated sludge plants treating them. In this study, the effects of the micronutrient niacin on the COD removal rates of textile wastewater, together with the effect of Mixed Liquor Suspended Solids (MLSS) on niacin, were studied. Certain improvement effects were found on the removal rates of COD, when 0.5 similar to 2.0 mg/L niacin was added to the textile wastewater. The optimal concentration of niacin was 1.0 mg/L, which was continuously added during textile wastewater treatment, and removal rates were 1.31 times compared to those of the control system. The concentration of MLSS was probably one of the factors influencing treatment efficiency, and the biological performance of treatment system could be optimized through micronutrient niacin supplements.

White spot syndrome virus (WSSV) is one of the most significant viral pathogens causing high mortality and economic damage in shrimp aquaculture. Although intensive efforts were undertaken to detect and characterize WSSV infection in shrimp during the last decade, we still lack methods either to prevent or cure white spot disease. Most of the studies on neutralizing antibodies from sera have been performed using in vivo assays. For the first time, we report use of an in vitro screening method to obtain a neutralizing scFv antibody against WSSV from a previously constructed anti-WSSV single chain fragment variable region (scFv) antibody phage display library. From clones that were positive for WSSV by ELISA, 1 neutralizing scFv antibody was identified using an in vitro screening method based on shrimp primary lymphoid cell cultures. The availability of a neutralizing antibody against the virus should accelerate identification of infection-related genes and the host cell receptor, and may also enable new approaches to the prevention and cure of white spot disease.

Persistent bioaccumulative toxins (PBTs) are organic substances that are persistent, bioaccumulative and can cause severe toxic effects (e.g. potential oncogens, mutagenic, endocrine disrupters) to human health or environment which are the ones that need special attention. PBT chemicals could be released to the environment from several types of sources and are ubiquitous in environment. However, fast and efficiency monitoring and assessment methods to investigate PBTs in environment are still lacking. In this study, a cleaning-up procedure of analyzing PBTs in fuels combustion soot was developed and its performance was assessed through comparing the chromatograms of crude extracts with their cleaned extracts after the cleaning-up procedure. The results showed that polycyclic aromatic compounds (PACs) were the main components in fuel combustion soot and the clean-up procedure developed in this paper can be well used as the method of analyzing PBTs in fuels combustion soot.

Because of the obvious importance of P as a nutrient that often accelerates growth of phytoplankton (including toxic cyanobacteria) and therefore worsens water quality, much interest has been devoted to P exchange across the sediment-water interface. Generally, the release mode of P from the sediment differed greatly between shallow and deep lakes, and much of the effort has been focused on iron and oxygen, and also on the relevant environmental factors, for example, turbulence and decomposition, but a large part of the P variation in shallow lakes remains unexplained. This paper reviews experimental and field studies on the mechanisms of P release from the sediment in the shallow temperate (in Europe) and subtropical (in the middle and lower reaches of the Yangtze River in China) lakes, and it is suggested that pH rather than DO might be more important in driving the seasonal dynamics of internal P loading in these shallow lakes, i.e., intense photosynthesis of phytoplankton increases pH of the lake water and thus may increase pH of the surface sediment, leading to enhanced release of P (especially iron-bound P) from the sediment. Based on the selective pump of P (but not N) from the sediment by algal blooms, it is concluded that photosynthesis which is closely related to eutrophication level is the driving force for the seasonal variation of internal P loading in shallow lakes. This is a new finding. Additionally, the selective pump of P from the sediment by algal blooms not only explains satisfactorily why both TP and PO4-P in the hypereutrophic Lake Donghu declined significantly since the mid-1980s when heavy cyanobacterial blooms were eliminated by the nontraditional biomanipulation (massive stocking of the filter-feeding silver and bighead carps), but also explains why TP in European lakes decreased remarkably in the spring clear-water phase with less phytoplankton during the seasonal succession of aquatic communities or when phytoplankton biomass was decreased by traditional biomanipulation. Compared with deep lakes, wax and wane of phytoplankton due to alternations in the ecosystem structure is also able to exert significant influences on the P exchange at the sediment-water interface in shallow lakes. In other words, biological activities are also able to drive P release from sediments, and such a static P release process is especially more prominent in eutrophic shallow lakes with dense phytoplankton.

The changes of NH3-N, NO3-N, NO2-N and TN/TP were studied during growth and non-growth season in 33 subtropical shallow lakes in the middle and lower reaches of the Yangtze River. There were significant positive correlations among all nutrient concentrations, and the correlations were better in growth season than in non-growth season. When TP > 0.1 mgL(-1), NH3-N increased sharply in non-growth season with increasing TP, and NO3-N increased in growth season but decreased in non-growth season with TP. These might be attributed to lower dissolved oxygen and low temperature in non-growth season of the hypereutrophic lakes, since nitrification is more sensitive to dissolved oxygen and temperature than anti nitrification. When 0.1 mgL(-1)> TP > 0.035 mgL(-1), TN and all kinds of inorganic nitrogen were lower in growth season than in non-growth season, and phytoplankton might be the vital regulating factor. When TP < 0.035 mgL(-1), inorganic nitrogen concentrations were relatively low and NH3-N, NO2-N had significant correlations with phytoplankton, indicating that NH3-N and NO2-N might be limiting factors to phytoplankton. In addition, TN/TP went down with decline in TIP concentration, and TN and inorganic nitrogen concentrations were obviously lower in growth season than in non-growth season, suggesting that decreasing nitrogen (especially NH3-N and NO3-N) was an important reason for the decreasing TN/TP in growth season. The ranges of TN/TP were closely related to trophic level in both growth and non-growth seasons, and it is apparent that in the eutrophic and hypertrophic state the TN/TP ratio was obviously lower in growth season than in non-growth season. The changes of the TN/TP ratio were closely correlated with trophic levels, and both declines of TN in the water column and TP release from the sediment were important factors for the decline of the TN/TP ratio in growth season.

In order to identify genes encoding the outer membrane proteins (OMPs) of the myxobacter Flavobacterium columnare G(4), the expression library of the bacterium was screened by using rabbit antisera developed against its OMPs. Positive colonies of Escherichia coli M15 containing fragments encoding the bacterial OMPs were selected for cloning the relevant genes by genomic walking methods. Two genes encoding a membrane-associated zinc metalloprotease and prolyl oligopeptidase are reported in this paper. The membrane-associated zinc metalloprotease gene (map) is 1800 bp in length, coding for 449 amino acids (aa). Despite the presence of a conserved motif HEXXH for all metalloproteases, the special HEXXH similar to 32 aa similar to E motif of the F. columnare G(4) Map and its low level of identity with other reported zinc-containing metalloproteases may imply that the membrane-associated zinc metalloprotease of F. columnare G(4) represents a new family of zincins. The gene encoding prolyl oligopeptidase (Pop), a serine proteinase, is 2352 bp in length, coding for 649 aa. Sequence homology analysis revealed that the Pop is also novel as it has <50% identity with other reported prolyl oligopeptidase family proteins. The present study represents the first to employ anti-fish bacterial OMP sera to screen genes of membrane-associated proteases of fish pathogenic bacteria, and to provide necessary information for the examination of the role of the two genes in the infection and pathogenesis of F. columnare.

Gynogenetic silver crucian carp, Carassius auratus gibelio, is an intriguing model. system. In the present work, a systemic study has been initiated by introducing suppression subtractive hybridization technique into this model system to identify the differentially expressed genes in oocytes between gynogenetic silver crucian carp and its closely related gonochoristic color crucian carp. Five differential cDNA fragments were identified from the preliminary screening, and two of them are ZP3 homologues. Moreover, the full length ZP3 cDNAs were cloned from their oocyte cDNA libraries. The length of ZP3 cDNAs were 1378 bp for gyno-carp and 1367 bp for gono-carp, and they can be translated into proteins with 435 amino acids. Obvious differences are not only in the composition of amino acids, but also in the number of potential O-linked oligosaccharide sites. In addition, gyno-carp ZP3 amino acid sequence has an unexpected higher identity value with common carp (83.5%) than that with the closely related gono-carp (74.7%). The unique homology may be originated from the ancient hybridization. Northern blot analysis confirmed that expression of the ZP3 gene occurred exclusively in the oocytes. Because O-linked oligosaccharides on ZP3 have been demonstrated to play very important roles in fertilization, it is suggested that the extra O-linked glycosylation sites may be related to the unique sperm-egg recognition mechanism in gynogenesis.

A total of six stations in the Han River system were selected for establishing polyurethane foam units (PFUs) to collect protozoans, including phytomastigophorans, zoomastigophorans, amoebas and ciliates, in July 1993. In the bioassessment of microbial communities using the PFUs, the number of species decreased as pollution intensity increased. The diversity index values calculated at the main stations generally agreed with the pollution status of the stations. Anyang-Chon (Chon means stream) showed the lowest diversity value (1.89), and all stations, except Masok and Anyang-Chon, showed diversity index values ranging from 3.15 to 3.93. The highest heterotrophic index (HI) value was detected in Anyang-Chon followed by Masok-Chon. The number of species at the main stations reached a maximum on day 11 of being exposed to PFUs. The results of S-eq, G and T-90% all suggest that bioassessments using the PFU system were well matched with pollution status of the water. All microbial variables were significantly correlated with comprehensive chemical pollution indices, P-a and P-b, with correlation coefficients ranging from r=0.87 to r=0.96.
