谷氨酰胺酶是催化谷氨酰胺分解为谷氨酸的氨基酸水解酶。它广泛存在于真核生物和原核生物中,在许多微生物和哺乳动物的氮代谢过程中起重要作用。但蓝藻中谷氨酰胺酶的酶学特征及生理功能尚不清楚,仅在一些蓝藻的基因组中发现有假定谷氨酰胺酶基因,这些基因编码的未知功能蛋白中有谷氨酰胺酶功能结构域,如集胞藻6803基因组中的slr2079基因。因此,本研究以模式蓝藻集胞藻6803 为研究对象,研究蓝藻谷氨酰胺酶的酶学特征及其生理功能。 为研究蓝藻谷氨酰胺酶的酶学特征,本研究克隆了集胞藻6803 slr2079基因,并在大肠杆菌中融合表达,经Ni-NTA亲合柱纯化后,通过对重组蛋白进行酶活测定及动力学分析,发现Slr2079蛋白是以谷氨酰胺为唯一催化底物的谷氨酰胺酶。 重组酶Slr2079的最适反应pH为9;最适反应温度为37C - 42C。该酶和绝大多数微生物源性的谷氨酰胺酶一样均为非磷酸依赖型。有趣的是该酶活性受Na+调节,而这种调节是通过提高对底物的亲和力来实现的。 为研究蓝藻谷氨酰胺酶在细胞内的生理功能,本研究通过基因插入失活,构建了缺失slr2079基因的集胞藻6803突变体,并对其进行生理、生化研究。在正常生长条件下,突变体和野生型蓝藻的生长未见差异,表明该基因不是集胞藻6803生长所必需的基因。但在700 mM NaCl胁迫条件下,突变体的生长速率比野生型快1.25倍。半定量RT-PCR结果显示,几个盐胁迫相关基因在突变体与野生型中的表达有所不同:与耐受盐胁迫的相关基因slr1608 (gdhB) 和slr1751 (prc)在突变体中表达提高,而盐敏感的基因sll0262 (desD) 和 slr0213 (guaA)在突变体中表达下降。由于重组的Slr2079具有谷氨酰胺酶活性,因此我们试图通过检测在蓝藻中参与氨同化作用的关键酶谷氨酸合成酶和谷氨酰胺合成酶在集胞藻6803中的表达情况来揭示Slr2079在集胞藻6803谷氨酰胺代谢中的生理功能。半定量RT-PCR结果显示,仅谷氨酸合成酶在突变体中表达提高,而谷氨酰胺合成酶表达未见明显变化。这些研究结果表明,在集胞藻6803中,Slr2079可能是通过调节与盐胁迫相关基因的表达来参与应对盐胁迫,而在氮代谢中起次要作用。

蓝藻是迄今地球上发现的最古老、分布最广和最具多样性的光合自养原核生物,其细胞结构简单,具有类似于植物的光合作用,是研究光合作用及其它代谢过程重要的模式生物。由于这类生物起源于远古前寒武纪,但至今依然繁多,在极端寒冷的南北极冰湖和近于沸腾温度的温泉,以及高盐、强碱的极端环境中均有存在,它们在漫长的进化过程中如何应对灾难性环境、针对随时可能遭遇的不同胁迫环境因子形成了怎样的分子适应机制,是近年来倍受关注但仍未诠释的问题之一。由于蓝藻与高等植物叶绿体在进化上密切相关,搞清楚这类生物适应不同胁迫环境因子的分子基础及其作用机制,对从进化的角度理解光合生物与环境相互作用、通过同源性发现作物抗逆育种新靶标,有重要的理论和实践意义。 逆境应答蛋白的表达是细胞对逆境胁迫的主要适应机制之一。在特定的逆境条件下,细胞通常会表达一组蛋白质,用于识别与传递环境胁迫信号、稳定细胞内环境、消除并修复逆境造成的损伤等。因此,逆境应答蛋白的系统鉴定和功能确认,是揭示逆境条件下细胞代谢网络及抗逆性分子机制的关键。单细胞模式蓝藻基因组序列的确定,极大地推动了蓝藻细胞蛋白质组成模式研究,也为系统发掘蓝藻逆境应答蛋白、理解和揭示分子适应机制提供了新的切入点。Synechocystis 6803是第一个完成基因组测序的放氧光合模式生物。由于其具有易培养、可转化、对环境条件变化反应快等优点,以该藻种为材料所展开的逆境应答特别是盐胁迫蛋白质组研究方面已经取得了重要的进展,而对高pH胁迫的蛋白质组研究还鲜有报道。因此,本论文以Synechocystis 6803为材料,从分离纯化的亚细胞组分入手,采用蛋白质组学研究手段,对蓝藻细胞应答高pH胁迫的蛋白质代谢网络进行探讨。利用蔗糖密度离心和水溶性两相分离法相结合的方法,分别获得了对照(pH7.5)和处理(pH11)细胞的质膜、外膜和类囊体膜,并分别构建了包括可溶性蛋白和膜组分的一维和二维蛋白质凝胶电泳图谱。分析结果表明,高pH胁迫下质膜和可溶性蛋白蛋白组分的变化较外膜和类囊体膜蛋白组分更为明显。在考马斯亮兰染色胶上共发现有近110个蛋白点上调或下调表达,其中有82个蛋白点来源于质膜。对质膜蛋白进行的差异荧光标记双向电泳(2-D DIGE)分析结果与考马斯亮兰染色结果基本一致。对质膜上的82个蛋白点进行胶内消化和MALDI-TOF和MALDI-TOF/TOF质谱鉴定,得到了39个不同基因产物,其中25个是上调蛋白,14个是下调蛋白。在这些发生变化的蛋白中,近1/3是ABC型转运蛋白,如3个磷转运蛋白(Sll0679,Sll0683,Sll0684)均在高pH胁迫下明显上调。其它高pH响应蛋白包括参与光合作用(PsaF,Sll0819;CpcA,Sll1578)、呼吸作用(CoxB,Sll0813)以及细胞分裂过程的蛋白(MinD,Sll0289)。还有LexA repressor (Sll1626)和Guanylyl cyclase(Cya2,Sll0646)等起调控作用的蛋白质。此外发现8个高pH胁迫响应蛋白为功能未知的新蛋白。生物信息学预测结果显示,在已鉴定的质膜响应蛋白中有17个蛋白具有信号肽。6个蛋白为具有跨膜域的膜蛋白,其中的3个膜蛋白是首次被证明定位于质膜上,且其表达与高pH胁迫有关。这些研究结果对从分子水平理解蓝藻细胞主动应对高pH胁迫、维护细胞内pH相对稳定机制有重要启示。

蓝藻是唯一可以进行有氧光合作用的原核生物,是水生食物链主要的初级生产者。氮素是蓝藻细胞必需的大量营养元素之一,揭示蓝藻如何应对环境中氮素的变化、维持自身碳氮平衡的分子机理,对深刻理解蓝藻与环境的相互作用、有效促进或控制蓝藻的生长与繁殖,有重要的理论和实践意义。已有的研究发现,蓝藻细胞的碳氮平衡主要是通过调控氨同化途径中的关键酶类实现。但先前的研究主要集中在固氮蓝藻谷氨酰胺合成酶(GS)-谷氨酸合成酶(GOGAT)循环的特性分析方面,而对催化谷氨酰胺水解生成谷氨酸和氨的主要酶之一谷氨酰胺酶的报道极少,其分子特性及生理学意义尚不明了。因此,本论文以模式固氮蓝藻鱼腥藻7120 和非固氮蓝藻集胞藻6803 为材料,采用分子生物学和生物化学方法,对蓝藻谷氨酰胺酶进行体外研究,并对其生物学功能进行了初步探讨,获得了如下主要结果:1)对体外重组蛋白的酶活性检测发现,两类蓝藻基因组编码的假定性谷氨酰胺酶,均具有谷氨酰胺酶催化活性,表明基因组注释是准确的;2)固氮蓝藻重组酶(All2934、All4774)与非固氮蓝藻重组酶(Slr2079)酶学特征差异显著,具有不同的最适pH、温度及底物亲和力;3)固氮蓝藻重组酶All2934 催化活性受磷酸盐的激活,而非固氮蓝藻重组酶Slr2079 在高Na+浓度下活性更高;4)RT-PCR 分析结果表明,在正常培养条件下,两类蓝藻的谷氨酰胺酶基因在细胞内均有表达;5)在缺氮培养条件下,固氮蓝藻谷氨酰胺酶基因all2934 的表达水平发生明显变化,而all4774 保持相对稳定,表明前者可能在这类细胞应对氮饥饿过程中起重要作用;6)在正常培养条件下,非固氮蓝藻谷氨酰胺酶基因的缺失突变体(Δslr2079)与野生型表型相似,但在盐胁迫条件下,突变体生长速率及光合放氧活性均高于野生型,表明该基因可能在提高非固氮蓝藻细胞高盐耐受力方面起负调控作用。上述重要发现,不仅初步揭示了光合自氧生物谷氨酰胺酶体外重组酶的分子特征,也为进一步研究谷氨酰胺酶在蓝藻细胞内的专一性功能奠定了重要基础。

Dew is an important water source for desert organisms in semiarid and arid regions. Both field and laboratory experiments were conducted to investigate the possible roles of dew in growth of biomass and photosynthetic activity within cyanobacterial crust. The cyanobacteria, Microcoleus vaginatus Gom. and Scytonema javanicum (Kutz.) Born et Flah., were begun with stock cultures and sequential mass cultivations, and then the field experiment was performed by inoculating the inocula onto shifting sand for forming cyanobacterial crust during late summer and autumn of 2007 in Hopq Desert, northwest China. Measurements of dew amount and Chlorophyll a content were carried out in order to evaluate the changes in crust biomass following dew. Also, we determined the activity of photosystem II(PSII) within the crust in the laboratory by simulating the desiccation/rehydration process due to dew. Results showed that the average daily dew amount as measured by the cloth-plate method (CPM) was 0.154 mm during fifty-three days and that the crust biomass fluctuated from initial inoculation of 4.3 mu g Chlorophyll a cm(-2) sand to 5.8-7.3 mu g Chlorophyll a cm(-2) crust when dew acted as the sole water source, and reached a peak value of approximately 8.2 mu g Chlorophyll a cm(-2) crust owing to rainfalls. It indicated that there was a highly significant correlation between dew amounts and crust moistures (r = 0.897 or r = 0.882, all P < 0.0001), but not a significant correlation between dew and the biomass (r = 0.246 or r = 0.257, all P > 0.05), and thus concluded that dew might only play a relatively limited role in regulating the crust biomass. Correspondingly, we found that rains significantly facilitated biomass increase of the cyanobacterial crust. Results from the simulative experiment upon rehydration showed that approximately 80% of PSII activity could be achieved within about 50 min after rehydration in the dark and at 5 degrees C, and only about 20% of the activity was light-temperature dependent. This might mean that dew was crucial for cyanobacterial crust to rapidly activate photosynthetic activity during desiccation and rehydration despite low temperatures and weak light before dawn. It also showed in this study that the cyanobacterial crusts could receive and retain more dew than sand, which depended on microclimatic characteristics and soil properties of the crusts. It may be necessary for us to fully understanding the influence of dew on regulating the growth and activity of cyanobacterial crust, and to soundly evaluate the crust's potential application in fighting desertification because of the available water due to dew. Iron deficiency can induce cyanobacteria to synthesize siderophore receptor proteins on the outer membrane to enhance the uptake of iron. In this study, an outer membrane of high purity was prepared from Anabaena sp. PCC 7120 based on aqueous polymer two-phase partitioning and discontinuous sucrose density ultra-centrifugation, and the induction of outer membrane proteins by iron deficiency was investigated using 2-D gel electrophoresis. At least. five outer membrane proteins were newly synthesized or significantly up-regulated in cells transferred to iron-deficient conditions, which were all identified to be siderophore receptor proteins according to MALDI-TOF-MS analyses. Bacterial luciferase reporter genes luxAB were employed to monitor the transcription of the encoding genes. The genes were induced by iron deficiency at the transcriptional level in different responsive modes. Luciferase activity expressed from an iron-regulated promoter may be used as a bioreporter for utilizable iron in natural water samples. The physiological differences for three bloom-forming cyanobacteria (Cylindrospermopsis raciborskii, Microcystis aeruginosa, and Aphanizomenon flos-aquae) were investigated. In comparison with M. aeruginosa and A. flos-aquae, C. raciborskii exhibited a significantly higher concentration of carotenoids, higher values in maximum photosynthesis rate (P-m), apparent photosynthetic efficieny (a), and maximum electron transport rate (ETRmax) during the growth period. In addition, higher extracellular alkaline phosphatase activities and lower light compensation point (I-c) were also detected in C raciborskii (p < 0.05, ANOVA). Therefore, it is suggested that the higher photosynthetic activities, more effective uptake and utilization to phosphate, and low light requirements might play important roles in the occurrence and invasive behavior of C. raciborskii. w Traditionally, nitrogen control is generally considered an important component of reducing lake eutrophication and cyanobacteria blooms. However, this viewpoint is refuted recently by researchers in China and North America. In the present paper, the traditional viewpoint of nitrogen control is pointed out to lack a scientific basis: the N/P hypothesis is just a subjective assumption; bottle bioassay experiments fail to simulate the natural process of nitrogen fixation. Our multi-year comparative research in more than 40 Yangtze lakes indicates that phosphorus is the key factor determining phytoplankton growth regardless of nitrogen concentrations and that total phytoplankton biomass is determined by total phosphorus and not by total nitrogen concentrations. These results imply that, in the field, nitrogen control will not decrease phytoplankton biomass. This finding is supported by a long-term whole-lake experiment from North America. These outcomes can be generalized in terms that a reduction in nitrogen loading may not decrease the biomass of total phytoplankton as it can stimulate blooms of nitrogen-fixing cyanobacteria. To mitigate eutrophication, it is not nitrogen but phosphorus that should be reduced, unless nitrogen concentrations are too high to induce direct toxic impacts on human beings or other organisms. Samples from stone surfaces were collected in pools within four unpolluted hillstreams (two shaded and two unshaded) in monsoonal Hong Kong (lat. 23 degrees N) to elucidate the extent of spatial (within and among streams) and temporal (seasonal) variations in algal biomass and assemblage composition. Sampling continued for over 12 months, incorporating the dry season when streams were at baseflow, and the wet season when spates were frequent. We anticipated that algal biomass would be lower in shaded streams and during the wet season, with associated seasonal differences in assemblage composition or relative abundance of different growth forms (e. g. erect versus prostrate). Benthic chlorophyll a (a proxy for algal biomass) varied among streams from an annual mean of 11.0-22.3 mg m(-2). Dry-season standing stocks were 18% higher than during the wet season when spate-induced disturbance reduced algal standing stocks. Algal biomass varied significantly at the stream scale, but not at the pool scale, and was lower in unshaded streams, where standing stocks may have been limited by high densities of algivorous balitorid loaches (mainly Pseudogastromyzon myersi). An overriding effect of grazers on algal biomass could also have reduced variations resulting from spate-induced disturbance. Significant differences in assemblage composition among streams, which were dominated by diatoms and cyanobacteria (totally 82 taxa) were not systematically related to shading conditions. Seasonal variations in algal assemblages were statistically significant but rather minor, and did not involve major shifts in composition or growth form caused by spate-induced disturbance. The abundance of filamentous cyanobacteria in all the streams may have been due to 'gardening' by balitorid loaches that removed erect or stalked diatoms and favoured cyanobacteria that persist through basal regeneration of filaments. This explanation requires validation through manipulative experiments.

Phyrobilisomes (PBS) are the major light-harvesting, protein-pigment complexes in cyanobacteria and red algae. PBS absorb and transfer light energy to photosystem (PS) II as well as PS I, and the distribution of light energy from PBS to the two photosystems is regulated by light conditions through a mechanism known as state transitions. In this study the quantum efficiency of excitation energy transfer from PBS to PS I in the cyanobacterium Synechococcus sp. PCC 7002 was determined, and the results showed that energy transfer from PBS to PS I is extremely efficient. The results further demonstrated that energy transfer from PBS to PS I occurred directly and that efficient energy transfer was dependent upon the allophycocyanin-B alpha subunit, ApcD. In the absence of ApcD, cells were unable to perform state transitions and were trapped in state 1. Action spectra showed that light energy transfer from PBS to PS I was severely impaired in the absence of ApcD. An apcD mutant grew more slowly than the wild type in light preferentially absorbed by phyrobiliproteins and was more sensitive to high light intensity. On the other hand, a mutant lacking ApcF, which is required for efficient energy transfer from PBS to PS II, showed greater resistance to high light treatment. Therefore, state transitions in cyanobacteria have two roles: (1) they regulate light energy distribution between the two photosystems; and (2) they help to protect cells from the effects of light energy excess at high light intensities. Using artificial systems to simulate natural lake environments with cyanobacterial blooms, we investigated plankton community succession by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) fingerprinting and morphological method. With this approach, we explored potential ecological effects of a newly developed cyanobacterial blooms removal method using chitosan-modified soils. Results of PCR-DGGE and morphological identification showed that plankton communities in the four test systems were nearly identical at the beginning of the experiment. After applying the newly developed and standard removal methods, there was a shift in community composition, but neither chemical conditions nor plankton succession were significantly affected by the cyanobacteria removal process. The planted Vallisneria natans successfully recovered after cyanobacteria removal, whereas that in the box without removal process did not. Additionally, canonical correspondence analysis indicated that other than for zooplankton abundance, total phosphorus was the most important environmental predictor of planktonic composition. The present study and others suggest that dealing with cyanobacteria removal using chitosan-modified soils can play an important role in controlling cyanobacterial blooms in eutrophicated freshwater systems.

This is the first to conduct Simultaneous determination of microcystin (MC) contaminations in multi-groups of vertebrates (fish, turtle, duck and water bird) from Lake Taihu with Microcystis blooms. MCs (-RR, -YR, -LR) in Microcystis scum was 328 mu g g(-1) DW. MCs reached 235 mu g g(-1) DW in intestinal contents of phytoplanktivorous silver carp, but never exceeded 0.1 mu g g(-1) DW in intestinal contents of other animals. The highest MC content in liver of fish was in Carassius auratus (150 ng g(-1) DW), followed by silver carp and Culter ilishaeformis, whereas the lowest was in common carp (3 ng g(-1) DW). In livers of turtle, duck and water bird, MC content ranged from 18 to 30 ng g(-1) DW. High MC level was found in the gonad, egg yolk and egg white of Nycticorax nycticorax and Anas platyrhynchos, suggesting the potential effect of MCs on water bird and duck embryos. High MC contents were identified for the first time in the spleens of N. nycticorax and A. platyrhynchos (6.850 and 9.462 ng g(-1) DW, respectively), indicating a different organotropism of MCs in birds. Lakes with deaths of turtles or water birds in the literatures had a considerably higher MC content in both cyanobacteria and wildlife than Lake Taihu, indicating that toxicity of cyanobacteria may determine accumulation level of MCs and consequently fates of aquatic wildlife. The effects of temperature and light on the growth and geosmin production of Lyngbya kuetzingii were determined. Of the three temperatures tested, 10, 25 and 35A degrees C, the maximal geosmin concentration and geosmin productivity were yielded at 10A degrees C, while the highest chl a production was observed at 25A degrees C. In the studies on light intensity, the maximal geosmin concentration and geosmin productivity were observed at 10 mu mol m(-2) s(-1), while the highest chl a production was at 20 mu mol m(-2) s(-1). It was suggested that more geosmin was synthesized with lower chl a demand. Meanwhile, the relative amounts of extra- and intracellular geosmin were investigated. Under optimum growth conditions (20 mu mol m(-2) s(-1), 25A degrees C; BG-11 medium), the amounts of extracellular geosmin increased as the growth progressed and reached the maximum in the stationary phase, while the intracellular geosmin reached its maximum value in the late exponential phase, and then began to decline. However, under the low temperature (10A degrees C) or light (10 mu mol m(-2) s(-1)) conditions, more intracellular geosmin was synthesized and mainly accumulated in the cells. The proportions of extracellular geosmin were high, to 33.33 and 32.27%, respectively, during the stationary phase at 35A degrees C and 20 mu mol m(-2) s(-1). It was indicated that low temperature or light could stimulate geosmin production and favor the accumulation of geosmin in cells, while more intracellular geosmin may be released into the medium at higher temperatures or optimum light intensity.

Microcystins are heptapeptide toxins produced by cyanobacteria. Microcystin-RR(MC-RR) is a common variant among the 80 variants identified so far. There have been many investigations documenting the toxic effects of microcystins on animals and higher plants, but little is known on the toxic effects of microcystins on algae, especially at molecular level. We studied the effects of MC-RR on gene expression profile of a few antioxidant enzymes and heat shock protein-70 (Hsp70) in Synechocystis sp. PCC6803. After two days post-exposure, a high dose toxin (5 mg/l, about 4.8 x 10(-3) mM) significantly increased expression levels of the genes gpx1, sodB, katG, acnB, gamma-TMTand dnaK2, while a relatively low dose toxin (1 mg/l, about 9.63 x 10(-4) mM) induced a moderate and slow increase of gene expression. Our results indicate that MC-RR could induce the oxidative stress in Synechocystis sp. PCC6803 and the increase in gene expression of antioxidant enzymes and Hsp70 might protect the organism from the oxidative damage. in addition, cell aggregation was observed during the early period of exposure, which might be a specific oxidative stress reaction to MC-RR. The planktivorous filter-feeding silver carp (Hypophthalmichthys molitrix) and bighead carp (Aristichthys nobilis) are the attractive candidates for bio-control of plankton communities to eliminate odorous populations of cyanobacteria. However, few studies focused on the health of such fishes in natural water body with vigorous toxic blooms. Blood parameters are useful and sensitive for diagnosis of diseases and monitoring of the physiological status of fish exposed to toxicants. To evaluate the impact of toxic cyanobacterial blooms on the planktivorous fish, 12 serum chemistry variables were investigated in silver carp and bighead carp for 9 months, in a large net cage in Meiliang Bay, a hypereutrophic region of Lake Taihu. The results confirmed adverse effects of cyanobacterial blooms on two phytoplanktivorous fish, which mainly characterized with potential toxicogenomic effects and metabolism disorders in liver, and kidney dysfunction. In addition, cholestasis was intensively implied by distinct elevation of all four related biomarkers (ALP, GGT, DBIL, TBIL) in bighead carp. The combination of LDH, AST activities and DBIL, URIC contents for silver carp, and the combination of ALT. ALP activities and TBIL, DBIL. URIC concentrations for bighead carps were found to most strongly indicate toxic effects from cyanobacterial blooms in such fishes by a multivariate discriminant analysis. (C) 2009 Elsevier B.V. A previously unknown cyanophage, PaV-LD (Planktothrix agardhii Virus isolated from Lake Donghu), which causes lysis of the bloom-forming filamentous cyanobacterium P. agardhii, was isolated from Lake Donghu, Wuhan, China. PaV-LD only lysed P. agardhii strains isolated from Lake Donghu and not those isolated from other lakes. The PaV-LD particle has an icosahedral, non-tailed structure, ca. 70 to 85 nm (mean +/- SD = 76 +/- 6 nm) in diameter. PaV-LD was stable at freezing temperature, but lost its infectivity at temperatures >50 degrees C. Lysis of host cells was delayed about 3 d after the PaV-LD treatment with chloroform, and the virus was inactivated by exposure to low pH (<= 4). The latent period and burst size of the PaV-LD were estimated to be 48 to 72 h and about 340 infectious units per cell, respectively. The regrowth cultures of surviving host filaments were not lysed by the PaV-LD suspension. To our knowledge, this is the first isolation and cultivation of a virus infectious to the filamentous bloom-forming cyanobacterium Planktothrix from a freshwater lake.
