A simple and sensitive flow injection method is presented for the determination of histidine based on its enhancement of electrogenerated chemiluminescence (ECL) of luminol. After optimization of the experimental parameters, the working range for histidine was in 1.0 x 10(-6) to 1.0 x 10(-3) mol/L with a detection limit (S/N = 3) of 0.56 mumol/L. The relative standard deviation was 1.6% for 11 measurements of 5 x 10(-5) mol/L histidine solution. The proposed method has been successfully applied to the determination of histidine in real pharmaceutical preparation.

Poly (ethylene oxide) (PEO) and poly (trimethopropane trimethacrylate) (PTMPTMA) interpenetrate networks have been synthesized. The confined crystallization behavior of PEO in the PTMTYTMA networks has been investigated by a differential scanning calorimeter and scanning electron microscope. The degree of PEO crystallinity in PEO/PTMPTMA interpenetrate networks reduces with the increase of PTMPTMA. PEO is in an amorphous state when the concentration of PEO is lower than 50% in the interpenetrate networks system. The melting points of crystalline PEO in the networks are lower than that of pure PEG, and the melting point of PEO in the networks is higher and increases with the increase of PEO in the interpenetrate networks. Wide-angle X-ray diffraction results show that the PEO crystallite size perpendicular to the (120) plane is not affected as much as PEO in silica networks. (C) 2001 Elsevier Science Ltd. Noncompatibilized and compatibilized ABS-nylon1010 blends were prepared by melt mixing. Polystyrene and glycidyl methacrylate (SG) copolymer was used as a compatibilizer to enhance the interfacial adhesion and to control the morphology. This SG copolymer contains reactive glycidyl groups that are able to react with PA1010 end groups (-NH2 or -COOH) under melt conditions to form SG-g-Nylon copolymer. Effects of the compatibilizer SG on the rheological, thermal, and morphological properties were investigated by capillary rheometer, DSC, and SEM techniques. The compatibilized ABS-PA1010 blend has higher viscosity, lower crystallinity, and smaller phase domain compared to the corresponding noncompatibilized blend. (C) 1999 John Wiley & Sons, Inc.

A novel engineering thermoplastic, phenolphthalein poly (ether-ether-sulfone) (PES-C) was blended with a commercial thermotropic liquid crystalline polymer(TLCP), Vectra A950, up to 30 weight percent of TLCP. A rheometrics dynamic spectrometer (RDS-I) and a CEAST capillary rheometer, a rheoscope 1000 were employed to investigate the melt rheology and extrusion behaviour at both the low and high shearing rates. The morphologies of the blends under different shearing were observed with a scanning electron microscope(SEM) and correlated to the observed rheology. The principal normal stress differences measured with cone-and-plate geometry give a temperature-independent correlation for both blend and PES-C when they are plotted against shear stress. But the extrudate swell of the blends showed a strong temperature dependence at each shear stress. The concentration dependence of extrudate swell shows a contrary behaviour to that of the inorganic filled system. A reasonable hypothesis based on the relaxation and disorientation of TLCP during flowing in the capillary and exiting was given to explain it. The melt fracture was checked after extrusion from capillary and was discussed.

The rheological properties of the novel engineering thermoplastic phenophthalein poly(ether ether ketone) (PEK-C) have been investigated using both a rotational and a capillary rheometer. The dependence of the viscosity on the shear rate and temperature was obtained. The activation energy was evaluated both from the Arrhenius and the Williams-Landel-Ferry (WLF) equation. An estimate for the proper E(eta) (dependent only on the chemical structure of the polymer) has been found from the WLF equation at temperatures about T-g + 200 degrees C. Measurements of the die swell have been performed. The first normal stress differences were evaluated from the die swell results and compared with the values obtained from the rotational rheometer at low shear rates.

高效毛细管电泳(High performance capillary electrophoresis缩写HPCE)是一种高效、快速分离复杂混合物的强有力工具.毛细管电泳电化学检测(Capillary electrophoresiselectrochemistry缩写CEEC)在HPCE中是一个极有前途的研究领域.安培检测由于灵敏度高,死体积小,成本低,而受到极大关注.把化学修饰电极(Chemically modified electrode缩写CME)用于CEEC的报道很少.本文介绍了一种新颖的方法将铂修饰到碳纤维电极上,并用于CEEC中检测肼.肼是一类比较难氧化的化合物,在普通固体电极上过电位很高,不适于电化学检测.我们做的修饰电极,对肼不仅有很好的催化活性,而且有很好的稳定性.

The solution of non-volatile solutes can be concentrated to saturation by membrane distillation. If the solute is easy to crystalize, the membrane distillation-crystallization phenomenon will appear during the membrane distillation of saturated solutions. It is possible that crystalline products are separated from concentrated solutions by a membrane process. In this work the PVDF capillary membrane, which was improved on hydrophobicity by using LiCl instead of a water-soluble polymer as an additive, has been used for treating the waste water of taurine. The crystalline product has been obtained from the waste water by the membrane distillation-crystallization technique. The results have shown good prospects for a membrane distillation application for treatment of industrial waste water.

带电粒子与绝缘体表面相互作用的研究揭示了很多新的物理现象和物理规律,目前该研究领域已拓展到原子物理,固体物理,等离子体物理等领域,特别是促进了纳米材料,超小尺寸半导体芯片制作,固体结构分析等应用领域的发展。本论文介绍了低能高流强电子束与绝缘体相互作用实验平台的建设,包括强流LaB6电子枪,64通道一维位置灵敏法拉第筒,电流电压转换放大器,基于Lab VIEW的数据获取软件系统等。研究了低能高流强电子束与锥形绝缘毛细管之间的相互作用。测量了出射电子的角分布与能谱,研究了入射电子能量、流强对电子传输效率的影响,分析了电子与高电荷态粒子同毛细管相互作用的区别。用低能高流强电子束与等内径弯曲绝缘管相互作用,测量了出射电子的角分布,研究了入射能量与流强对导向效应的影响。研究表明,与高点和态离子相比,电子与绝缘管相互作用有许多新的特性,电子传输效率较高电荷态粒子小1-2个数量级,且能量损失严重。量子反射和多次小角度散射是电子导向的主要原因

A pattern recognition protein (PRP), lipopolysaccharide and beta-1,3-glucan binding protein (LGBP) cDNA was cloned from the haemocyte of Chinese shrimp Fenneropenaeus chinensis by the techniques of homology cloning and RACE. Analysis of nucleotide sequence revealed that the full-length cDNA of 1,275 bp has an open reading frame of 1,098 bp encoding a protein of 366 amino acids including a 17 amino acid signal peptide. Sequence comparison of the deduced amino acid sequence of F. chinensis LGBP showed a high identity of 94%, 90%, 87%, 72% and 63% with Penaeus monodon BGBP, Litopenaeus stylirostris LGBP, Marsupenaeu japonicus BGBP, Homarus gammarus BGBP and Pacifastacus leniusculus LGBP, respectively. The calculated molecular mass of the mature protein is 39,857 Da with a deduced pI of 4.39. Two putative integrin binding motifs, RGD (Arg-Gly-Asp) and a potential recognition motif for beta-1,3-linkage of polysaccharides were observed in LGBP sequence. RT-PCR analysis showed that LGBP gene expresses in haemocyte and hepatopancreas only, but not in other tissues. Capillary electrophoresis RT-PCR method was used to quantify the variation of mRNA transcription level during artificial infection with heat-killed Vibrio anguillarum and Staphylococcus aureusin. A significant enhancement of LGBP transcription was appeared at 6 h post-injection in response to bacterial infection. These results have provided useful information to understand the function of LGBP in shrimp.

A new antimicrobial protein gene of the anti-lipopolysaccharide factor family (tentatively named as ALFFc) has been cloned from hemocytes of the Chinese shrimp Fenneropenaeus chinensis by rapid amplification of 3' and 5' complementary DNA ends with polymerase chain reaction. The full-length complementary DNA of ALFFc consists of 600 bp with a 369-bp open reading frame, encoding 123 amino acids. The deduced peptide contains a putative signal peptide of 25 amino acids and mature peptide of 98 amino acids. The molecular mass of the deduced mature peptide is 13799.16 Da. It is highly cationic, with a theoretical pI of 10.3. The deduced amino acid sequence of ALFFc showed 56% homology with sequences of Tachypleus tridentatus and L. polyhemus. The tissue expression profile of this gene was studied by Northern blot, and ALFFc transcripts were mainly detected in hemocytes, gill, and intestine. RNA in situ hybridization showed that ALFFc was constitutively expressed in hemocytes. Capillary electrophoresis reverse transcriptase PCR was used to quantify the variation of messenger RNA transcription level during the artificial infection process with Vibrio anguillarum. Significant enhancement of ALFFc transcription appeared during the first 24 hours in response to Vibrio infection. These results provide useful information for understanding the function of ALFFc in shrimp.

Lectin is regarded as a potential molecule involved in immune recognition and phagocytosis through opsonization in crustacean. Knowledge on lectin at molecular level would help us to understand its regulation mechanism in crustacean immune system. A novel C-type lectin gene (Fclectin) was cloned from hemocytes of Chinese shrimp Fenneropenaeus chinensis by 3' and 5' rapid amplification of cDNA ends (RACE) PCR. The full-length cDNA consists of 1482 bp with an 861 bp open reading frame, encoding 287 amino acids. The deduced amino acid sequence contains a putative signal peptide of 19 amino acids. It also contains two carbohydrate recognition domains/C-type lectin-like domains (CRD1 and CRD2), which share 78% identity with each other. CRD1 and CRD2 showed 34% and 30% identity with that of mannose-binding lectin from Japanese lamprey (Lethenteron japonicum), respectively. Both CRD1 and CRD2 of Fclectin have I I amino acids residues, which are relatively invariant in animals' C-type lectin CRDs. Five residues at Ca2+ binding site I are conserved in Fclectin. The potential Ca2+/carbohydrate-binding (site 2) motif QPD, E, NP (Gln-Pro-Asp, Glu, Asn-Pro) presented in the two CRDs of Fclectin may support its ability to bind galactose-type sugars. It could be deduced that Fclectin is a member of C-type lectin superfamily. Transcripts of Fclectin were found only in hemocytes by Northern blotting and RNA in situ hybridization. The variation of mRNA transcription level in hemocytes during artificial infection with bacteria and white spot syndrome virus (WSSV) was quantitated by capillary electrophoresis after RT-PCR. An exploration of mRNA expression variation after LPS stimulation was carried out in primarily cultured hemocytes in vitro. Expression profiles of Fclectin gene were greatly modified after bacteria, LPS or WSSV challenge. The above-stated data can provide us clues to understand the probable role of C-type lectin in innate immunity of shrimp and would be helpful to shrimp disease control. (c) 2006 Elsevier Ltd. The possibility of the brine shrimp Artemia to produce dormant embryo (cysts) in diapause is a key feature in its life history. In the present study, we obtained a proteomic reference map for the diapause embryo of Artemia sinica using two-dimensional gel electrophoresis with a pH range of 4-7 and a molecular weight range of 10-100 kDa. Approximately 233 proteins were detected, and 60 of them were analyzed by capillary liquid chromatography tandem mass spectrometry (LC-MS/MS). Of these, 39 spots representing 33 unique proteins were identified, which are categorized into functional groups, including cell defense, cell structure, metabolism, protein synthesis, proteolysis, and other processes. This reference map will contribute toward understanding the state of the diapause embryo and lay the basis and serve as a useful tool for further profound studies in the proteomics of Artemia at different developmental stages and physiological conditions.

A new member of antimicrobial protein genes of the Crustin family was cloned from haemocytes of the Chinese shrimp Fenneropenaeus chinensis by 3' and 5' RACE. The full-length cDNA of Crustin-like gene contains a 390 bp open reading frame, encoding 130 amino acids. The deduced peptide contains a putative signal peptide of 17 amino acids and mature peptide of 113 amino acids. The molecular mass of the deduced mature peptide is 12.3 ku. It is highly cationic with a theoretical isoelectric point of 8.5. The deduced amino acids sequence of this Crustin showed high homology with those of Penaeus (Litopenaeas) setferus. Northern blotting showed that the cloned Crustin gene was mainly expressed in haemocytes, gill, intestine, and RNA in situ hybridization indicated that the Crustin gene was constitutively expressed exclusively in haemocytes of these tissues. Capillary electrophoresis RT-PCR analysis showed that Crustin was up-regulated dramatically from 12 to 48 h after a brief decrease of mRNA during first 6 h in response to microbe infection. The level of Crustin mRNA began to restore at 72 h post-challenge. This indicated that Crustin gene might play an important role when shrimps are infected by bacterial pathogen.

The fatty acids composition in different parts of full-grown Rhopilema esculentum jellyfish from Yellow Sea was investigated. The lipids, extracted from the umbrella and oral arms and gonads of R. eculentum jellyfish, respectively were analysed by combined capillary gas chromatography/mass spectrometry. The results show that there are more than thirty kinds of fatty acids in jellyfish, and the fatty acid compositions of three parts of R. esculentum are almost the same. In the three parts, percentages of polyunsaturated fatty acids (PUFA) are high, and range from 36.23% to 38.74%. Docosahexaenoic acid (DHA), eicosatetraenoic acid (AA) and eicosapentaenoic acid (EPA) are three major PUFA.

Microsatellites were screened in a backcross family of the Pacific oyster, Crassostrea gigas. Fifteen microsatellite loci were distinguishable and polymorphic with 6 types of allele-combinations. Null alleles were detected in 46.7% of loci, accounting for 11.7% of the total alleles. Four loci did not segregate in Mendelian Ratios. Three linkage groups were identified among 7 of the 15 segregating loci. Fluorescence-based automated capillary electrophoresis (ABI 310 Genetic Analyzer) that used to detect the microsatellite loci, has been proved a fast, precise, and reliable method in microsatellite genotyping.
