Electrochemically induced three conformational transitions of calf thymus DNA from B-10.4 to Z(10.2)-DNA and from B-10.2 to B-10.4 and to C-DNA in 10 mM phosphate buffer solution (pH 7.21) at glassy carbon electrode are found and studied by in situ circular dichroism (CD) thin layer spectroelectrochemistry with singular value decomposition least square (SVDLS) analysis. It indicates that the so-called B-10.2 form and the C-form of DNA may be composed of B-10.4 and left-A DNA and of B-10.4 and right-A DNA, respectively. The irreversible electrochemical reduction of adenine and cytosine groups in the DNA molecule is studied by UV-Vis spectroelectrochemistry. Some electrochemical parameters alphan = 0.17, E-0' = -0.70 V (vs. Ag/AgCl), and the standard heterogeneous electron transfer rate constant, k(0) = 1.8 x 10(-5) cm s(-1) are obtained by double logarithmic analysis and non-linear regression. The anodic voltammetric behavior of medecamycin (MD) in the presence of various electrolytes was studied by linearsweep voltammetry, differential-pulse voltammetry and cyclic voltammetry at a glassy carbon electrode. In phosphate buffer solutions (pH = 9.4), MD is oxidized irreversibly. The peak potential is at about +0.75 V (vs.Ag/AgCl). The height of the peak is linearly increased with the concentration of MD over the range of 5 x 10(-5) similar to 1 x 10(-1) g/L. The method has been used for the direct determination of MD in tablets. The relative standard deviation (n = 10) is 1.8%. The recoveries of MD in urine samples are in the range of 95% similar to 115%.

Capillary electrophoresis with amperometric detection is evaluated for the simultaneous determination of 2-aminothiazole (A), 2-amino-benzothiazole (AB), 2-mercaptobenzothiazole (AM). The cyclic voltammogram, hydrodynamic voltammogram, effect of pH, concentration of buffer and separation voltage on the separation and the detection were studied. The conditions were optimized as follows: 50 mM phosphate buffet; pH 6.0, 2s at 17.5 kV sample injection, separation at 17.5 kV, 1.2 V as detection potential. The method provided low detection limit as 0.5 mu M, 0.05 mu M and 0.01 mu M, wide linear range 2-200 mu M, 10-200 mu M and 0.025-100 mu M for A, AB, and AM, respectively. The variations in peak current and migration time for 15 consecutive injections of a standard containing 5 mu M each compound were 3.7, 2.1, and 3.9%, and 1.2, 0.8, and 1.2%, for A, AB and AM, respectively. This method was employed to analyze river water.

A capillary electrophoresis-amperometric detection system was developed for the determination of propranolol (PRO) at a 33 mu m carbon fiber microdisk electrode (CFE). The cyclic voltammogram, the hydrodynamic voltammograms and the effect of pH were studied. Under the optimum conditions: separation Voltage 15 kV; injection 3 s at 15 kV; 10 mM pH 7.5 phosphate buffer, 1.15 V (vs. Ag/AgCl) detection potential, the detection limit (LOD) for PRO was 0.05 mu M (S/N = 3). The response for PRO was linear over two orders of magnitude with a linear correlation coefficient of 0.994. The feasibility of this method was demonstrated by the detection of PRO in urine sample.

Four phenothiazines, promethazine, dioxypromethazine, chlorpromazine, and trifluoperazine have been separated by capillary electrophoresis using N, N, -dimethylformamide (DMF) as separation medium with UV absorbance detection. High voltage and concentrated buffer were used with small current and low electroosmosis. Good resolution and high column efficiency were obtained. Separation selectivity in DMI; was different from that in water because of the different solvation interactions. The influence of buffer composition on separation selectivities and electroosmosis were also studied.

A stable film was prepared by casting dipalmitoylphosphatidylcholine (DPPC) and rutin onto the surface of a glassy carbon (GC) electrode. The electrochemistry behavior of rutin in the DPPC film was investigated. The modified electrode coated with rutin shows a quasi-reversible reduction-oxidation peak on the cyclic voltammogram in phosphate buffer (pH 7.4). This model of biological membrane was not only used to provide biological environment but also to investigate the oxidation of ascorbic acid by rutin. The DPPC-rutin modified electrode behaves as electrocatalytic oxidation to ascorbic acid. The oxidation peak current of ascorbic acid increases drastically and the peak potential of 4 x 10(-4) mol L-1 ascorbic acid shifts negatively about 100 mV compared with that obtained at a bare glassy carbon electrode. The catalytic current increased linearly with the ascorbic acid concentration in the range of 2 x 10(-4) mol L-1 and 1.4 x 10(-3) mol L-1 at a scan rate of 50 mV s(-1).

A novel amperometric glucose biosensor was constructed by electrochemical formation of a polypyrrole (PPy) membrane in the presence of glucose oxidase (GOD) on the surface of a horseradish peroxidase (HRP) modified ferrocenecarboxylic acid (FCA) mediated sol-gel derived ceramic carbon electrode. The amperometric detection of glucose was carried out at +0.16 V (vs. SCE) in 0.1 mol/L phosphate buffer solution (pH 6.9) with a linear response range between 8.0x10(-5) and 1.3x10(-3) mol/L of glucose. The biosensor showed a good suppression of interference and a negligible deviation in the amperometric detection.

Electroreduction of vitamin B-2 (VB2) was studied by in situ circular dichroism (CD) spectroelectrochemistry (SEC) with a long optical path length thin layer cell (LOPLTLC). The results showed that the electroreduction of VB2 in phosphate buffer solution (PBS) (PH 6.8) was a two-electron electrochemical process with weak adsorption of the reactant at the glassy carbon (GC) electrode surface. The CD spectra change of VB2 in the reduction process was explained with the theory of electronic states. We also treated the CD spectra with a singular value decomposition least square (SVDLS) method, and have found not only the number of components and their spectra, but also the fraction distribution of each component in the electroreduction process of VB2.

A ferrocene-dimyristoyl phosphatidylcholine (DMPC) film electrode was prepared by casting the solution of ferrocene and DMPC in chloroform onto a glassy carbon electrode surface. Ferrocene retained in the biological membrane gave a couple of irreversible peaks of cyclic voltammogram. The electrode exhibited good electrocatalytic activity for the oxidation of ascorbic acid (H(2)A) in phosphate buffer (pH 6.64) with an anodic peak potential of +340 mV (vs. Ag/AgCl). The anodic current was directly proportional to the square root of the scan rate below 150 mV s(-1). The influence of the pH value was investigated and it was observed that pH 6.64 was the suitable value to the anodic peak potential and current. The thickness of the film and the interference of uric acid were also studied. The electrode can be used to determine H(2)A in the presence of equimolar uric acid. The catalytic peak current increased linearly with the concentration of H(2)A in the range of 1 X 10(-4)-5 X 10(-3) mol L-1.

Combination of affinity extraction procedures with mass spectrometric analyses is termed affinity-directed mass spectrometry, a technique that has gained broad interest in immunology and is extended here with several improvements from methods used in previous studies. A monoclonal antibody was immobilized on a nitrocellulose (NC) membrane, allowing the corresponding antigen to be selectively captured from a complex solution for analysis by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS). This method was also used to rapidly determine the approximate binding region responsible for the antibody/antigen interaction. The tryptic fragments of antigen protein in buffer were applied to the antibody immobilized on NC film and allowed to interact. The NC film was then washed to remove salts and other unbound components, and subjected to analysis by MALDI-TOFMS. Using interferon-alpha (2a) and anti-interferon-alpha (2a) monoclonal antibody IgG as a model system, we successfully extracted the antigen protein and determined the approximate binding region for the antigen/antibody interaction (i.e., the tryptic fragment responsible). A simple method was proposed for the separation of allopurinol (AP) and its active metabolite oxypurinol (OP) by capillary electrophoresis with end-column amperometric detection. A running buffer composed of 15 rum Na2HPO4/NaH2PO4 at a pH 9.55, electrokinetic injection 7 s at 5 kV, separation voltage at 15 kV and detection potential at 1.20 V were investigated to be the optimal condition for the separation. The method exhibited low detection Emit (S/N = 3) as 1 x 10(-8) mol/l for AP and OP, wide linearity range of 2 x 10(-7) to 1 x 10(-4) mol/l, 1 x 10(-7) to 1 x 10(-4) and high efficiency of 1.2 x 10(5) and 1.8 x 10(5) N/m for AP and OP, respectively. The potential application examined for the method was the determination of the spiked urine sample, which was proved to be sensitive and efficient. (C) 2001 Elsevier Science B.V. The elucidation of key influence factors for electrostatic adsorption is very important to control protein nonspecific adsorption on modified surfaces. In this study, real-time surface plasmon resonance technique is used to characterize the electrostatic adsorption of two proteins (mouse IgG and protein A) on carboxymethyldextran-modified surface. The results show that protein solution pH and ionic strength are key influence factors for efficient electrostatic adsorption. The influence of protein, solution pH on the amount of electrostatic adsorption depends on the type of the charge and the charge density of both protein and modified matrix on the surface. The electrostatic adsorption process involves a competition between the positively charged protein and other positively charged species in the buffer solution. A decrease of ionic strength leads to an increasing electrostatic adsorption. The kinetic adsorption constants of protein A at different pH values were also calculated and compared.

The infrared spectra of BaLnB(9)O(16):Re, along with the VUV excitation spectra, have been measured. The spectra were tentatively interpreted in terms of the data on absorptions of the borate groups and band structure. It was observed that there are absorption due to BO3 and BO4 groups, indicating that there are BO3 and BO4 groups in BaLnB(9)O(16). It is found that absorption of the borate groups is located in the range from 120 to 170 mn. This result reveals that there is an energy transfer from host to the rare earth ions. It also observed that the energy of charge transfer band, the host absorption, the total crystal field splitting of d-levels of Tb3+ increase with the decrease in the Ln(3+) radius. The p-toluene sulfonic acid (MA) in phenol matrix was separated and determined by capillary electrophoresis with ultraviolet detector. the effect of the concentration and pH of the buffer on separation was investigated. Cinnamic acid has been chosen as the internal standard from four compounds, the calibration curves of PTSA in 50 mg/L phenol matrix were obtained with and without the internal standard. The linear range was from 1.25 to 12.5 mg/L and the correlation coefficient was 0.9999 for both curves. The limit of detection of PISA was 0.75 mg/L at 3 times of SIN. Finally, the concentration of PTSA in four synthesized samples was determined with method of standard additions, and the effect of matrix was discussed. The values of MA in these samples were 1.01, 0.94, 1.56 and 0.00 mg/L respectively.

We have made a cheap microsystem of capillary electrophoresis with a new method, integrating the electrodes, injection channel, separation channel, buffer reservoirs and detection cell on a polymethylmethacrylate (PMMA) chip. Using an integrated micro carbon fiber disk electrode as the working electrode in three electrodes system, 1 x 10(-4) mol/L dopamine(DA) could be detected with end-column amperometric detection. The reproducibility was good. Peak current was 6.73 nA,theoretical plate number was 71300/m and height equivalent of one theoretical plate height was 14.0 mum for 1 x 10(-4) mol/L DA. The limit of detection was 3.6 x 10(-8) mol/L and the linear range was extended from 5 x 10(-7) mol/L to 1 x 10(-4) mol/L for DA. 1 x 10(-4) mol/L catechol (CA) and 5 x 10(-5) mol/L DA were also separated completely with R-s = 10.1.
