The technique of mtDNA restriction fragments length polymorphism (RFLP) was used to survey the population structure of D. albomicans. Remarkable mtDNA polymorphism has been observed in D. albomicans populations. A total of 34 nucleomorphs were detected from 82 isofemale lines assayed by only 8 restriction enzymes. The cause and the effect of this phenomenon were discussed. As a result, it is suggested that a mechanism which maintains mtDNA diversity exists in this fly, and that the high intra-populational polymorphism could numerically conceal the extent of differentiation between populations. In addition, on the base of restriction maps, it was found that the mtDNA molecule of D. albomicans might be impacted by the selection pressure during its evolution process both on the nucleotide composition and on the functional regions.

 目的 观察丙泊酚对大鼠海马CA1 区锥体神经元产生的长时程抑制(L TD) 的影响,并 分析其可能机制。方法 断头法分离wistar 大鼠(13~19 d) 海马半脑,用切片机切出400μm 厚度的 海马脑片。实验分三组:脂肪乳剂组( I 组) ,丙泊酚组(P 组) ,SR95531 + 丙泊酚组( GP 组) 。I 组和P 组以90μL 脂肪乳剂或丙泊酚(相当于100μmol/ L) 预孵脑片60 min ,然后给予低频刺激(L FS) ,记录 L TD 的表达情况; GP 组先在循环液中加入10μmol/ L SR95531 预孵脑片30 min ,再加入100μmol/ L 丙泊酚继续孵育60 min ,继而给予L FS ,记录L TD 的表达情况。结果 I 组给予L FS 后,产生L TD , L FS 后10~40 min 的兴奋性突触后电流( EPSC) 值为基础值的57185 %;P 组给予L FS 后10~40 min 的EPSC 值为基础值的40182 % ,明显低于I 组( P < 0105) ; GP 组给予L FS 后10~40 min 的EPSC 值为基础值的56151 % ,与I 组比较差异无显著意义( P > 0105) ,与P 组比较差异有显著意义( P < 0105) 。结论 100μmol/ L 丙泊酚使大鼠海马CA1 区锥体神经元L TD 表达增强,这种作用与其增强 GABAA 受体功能有关;当阻断GABAA 受体后,这种易化作用消失。

本文根据历年收集的标本和生态资料记述了云南倒刺鲃的基本生物学特性。云南倒刺鲃喜居湖泊沿岸浅水区的中下层;成体以水草为主食,幼体则以丝状藻类为主食;生长速度明显快于相近种;繁殖期为每年的5—8月,喜在砂砾底质并具流水处产卵;体长274.0—400.0mm的个体,其绝对繁殖力为8800—13632粒。调查资料还表明,云南倒刺鲃在异龙湖已绝灭,在阳宗海和星云湖也已濒临灭绝,仅在抚仙湖尚存有一定的数量。

2007年9~11月和2008年4~5月,对云南省丽江玉龙雪山自然保护区的鸟类资源进行了调查,共记录鸟类184种,结合中国科学院昆明动物研究所鸟类标本室收藏的该地区标本和文献记录的种类,保护区内共记录鸟类330种(另13亚种),隶属47科(另4亚科),18目,占云南省所录鸟类种数848种的38.92%,全国鸟类种数1329种的24.83%。其中国家一级保护物种1种,二级保护物种29种。黑眉长尾山雀Aegithalos iouschistos和大嘴乌鸦Corvus macrorhynchos为保护区内的优势种类。保护区记录的鸟类有留鸟230种和亚种,占所录鸟类种和亚种数的67.05%。区系成分以东洋界物种为主,有182种,占繁殖鸟类总种数的66.42%。亚区级区系分析表明保护区的鸟类以西南山地亚区的种类为主。结合该区鸟类生境分布的特点,提出云南松高山松林、针阔混交林和寒温性暗针叶林的保护是保护工作的重点。

报道了美姑脊蛇Achalinus meiguensis线粒体基因组全序列.美姑脊蛇线粒体全序列长17239bp,由22个tRNA,2个rRNA和13个蛋白质基因及2个非编码的控制区或D-loop组成,存在着基因重排现象.对已报道蛇类线粒体基因组全序列进行比对分析后,发现一些蛇类线粒体基因组进化规律:双控制区现象在爬行动物进化历史中独立地发生,有不同的演化历史;tRNA假基因是在真蛇下目(Caenophidia)中进化形成的;TΨC臂的相对较短(一般少于5bp)和缺失"DHU"臂造成蛇类tRNA较短.通过M

对鲢、鳙单倍体和二倍体单个囊胚胚胎进行培养,结果表明,单倍体和二倍体细胞对培养条件的要求相差不大,其原代培养细胞的生长特点表现为新生细胞主要沿着组织块边缘重叠生长,向外扩展生长的速度非常缓慢,二倍体的鲢和鳙胚胎细胞培养物成活率相对高于单倍体,其中鲢二倍体胚胎细胞传代细胞占细胞培养数的18.06%。本结果对深入开展鲢、鳙核移植及胚胎干细胞研究具有积极意义。

Microorganisms play an important role in removing pollutants from constructed wetlands. We investigated the microbial characteristics in a novel integrated vertical-flow constructed wetland (IVCW), which has been in operation in Wuhan, China since 1998. We used phospholipid fatty acid (PLFA) and amoA gene to analyze the structure and diversity of the microbial community within the IVCW. PLFA results suggested that the amount of bacterial PLFA was significantly higher than that of fungal PLFA, but the total microbial biomass represented by PLFA index was low in the system. Microbial spatial distribution showed significantly higher bacterial (both G(+) and G(-)) and fungal biomass in the surface than in the subsurface layers. The ratios of monounsaturated to branched PLFA demonstrated that an anaerobic layer sandwiched by two aerobic layers existed in the IVCW, consistent with the redox potential results. Analysis of the amoA revealed the presence of Nitrosomonas-like sequences in the surface substrate of the downflow chamber and apparent diversities of ammonia-oxidizing bacteria in the system. These results suggest that microorganisms, despite their relatively low biomass, have inhabited the IVCW, and the results will offer some valuable information on microbe to system designers and managers.

Synechocystis sp. PCC 6803 exposed to chill (5 degrees C)-light (100 mu mol photons m(-2) s(-1)) stress loses its ability to reinitiate growth. From a random insertion mutant library of Synechocystis sp. PCC 6803, a sll1242 mutant showing increased sensitivity to chill plus light was isolated. Mutant reconstruction and complementation with the wild-type gene confirmed the role of sll1242 in maintaining chill-light tolerance. At 15 degrees C, the autotrophic and mixotrophic growth of the mutant were both inhibited, paralleled by decreased photosynthetic activity. The expression of sll1242 was upregulated in Synechocystis sp. PCC 6803 after transfer from 30 to 15 degrees C at a photosynthetic photon flux density of 30 mu mol photons m(-2) S-1. sll1242, named ccr (cyanobacterial cold resistance gene)- 1, may be required for cold acclimation of cyanobacteria in light.

A novel fish chemokine receptor gene, chemokine (C-X-C motif) receptor 3 (CXCR3)-like was isolated from the grass carp Ctenopharyngodon idella , with its full-length genomic sequence. The cDNA of grass carp CXCR3-like (gcCXCR3-like) consists of 1261 bp with a 49bp 5'-UTR and a 189 bp 3'-UTR. An open reading frame of 1023 bp encodes a 341-amino acid peptide, with seven transmembrane helices. The deduced amino acid sequence showed the same sequence identities (37.8%) with its counterparts in goat and human. The gcCXCR3-like gene consists of two exons, with one intervening intron, spaced over approximately 2 kb of genomic sequence. Phylogenetic analyses clearly demonstrated that the gcCXCR3-like resembles the CXCR3s of other vertebrates. Real-time PCR analysis showed that gcCXCR3-like was expressed in all tested organs except heart and the expression level of gcCXCR3-like was highest in brain. Flow cytometric analyses showed the positive rate of labelled leukocytes from the healthy grass carp was 17.3%, and the labelled leukocytes were divided into three types by cell sorting. Immunohistochemical localization revealed that gcCXCR3-like expressed in whole brain regions including cerebel, diencephalon, medulla oblongata, optic lobe, and rhinencephalon, and that the labelled leukocytes are actually populations of monocyte and/or phagocyte, lymphocyte and the granulocyte. Based on the band anticrossing model, the effects of the strain-compensated layer and the strain-mediated layer on the band structure, gain and differential gain of GaInNAs/GaAs quantum well lasers have been investigated. The results show that the GaNAs barrier has a disadvantage in increasing the density of states in the conduction band. Meanwhile, the multilayer quantum wells need higher transparency carrier density than the GaInNAs/GaAs single quantum well with the same wavelength. However, they help to suppress the degradation of the differential gain. The calculation also shows that from the viewpoint of band structure, the strain-compensated structure and the strain-mediated structure have similar features.

We present a novel contactless and nondestructive method called the surface electron beam induced voltage (SEBIV) method for characterizing semiconductor materials and devices. The SEBIV method is based on the detection of the surface potential induced by electron beams of scanning electron microscopy (SEM). The core part of the SEBIV detection set-up is a circular metal detector placed above the sample surface. The capacitance between the circular detector and whole surface of the sample is estimated to be about 0.64 pf It is large enough for the detection of the induced surface potential. The irradiation mode of electron beam (e-beam) influences the signal generation. When the e-beam irradiates on the surface of semiconductors continuously, a differential signal is obtained. The real distribution of surface potentials can be obtained when a pulsed e-beam with a fixed frequency is used for irradiation and a lock-in amplifier is employed for detection. The polarity of induced potential depends on the structure of potential barriers and surface states of samples. The contrast of SEBIV images in SEM changes with irradiation time and e-beam intensity.
