Effects of Microcystis blooms on the crustacean plankton were studied using enclosure experiments during July-September, 2000. Eight enclosures were set in the hypereutrophic Donghu Lake. Different nutrient concentrations through additional nutrient and sediment in enclosures were expected to result in different abundance of Micropystis. From July to early August, the phytoplankton community was dominated by Chlorophyta, Cryptophyta, Bacillariophyta and Cyanophyta other than Microcystis aeruginosa. M. aeruginosa showed a rapid increase during early August in all enclosures and predominated. Crustacean plankton was dominated by the herbivorous Moina micrura, Diaphanosoma brachyurum and Ceriodaphnia cornuta, and the predaceous Mesocyclops sp. and Thermocyclops taihokuensis. During the pre-bloom period, the dynamics of M. micrura population appeared to be mainly affected by the predaceous cyclopoids. With the development of Microcystis blooms, such interaction between M. micrura and cyclopoids seemed weakened, especially when the Microcystis biomass was high. But there was no apparent influence on the interaction between Leptodora kindti and its zooplanktonic prey. The density of two cyclopoids decreased with the enhancement of Microcystis. The density decline of M. micrura was caused by both predation and inhibition by Micropystis. The low food availability of other edible phytoplankton during the blooms led to low densities of both C. cornuta and D. brachyurum by late August. It appears that dense Microcystis blooms exert strong negative effects on the herbivorous cladocerans and the predaceous cyclopoids.

Phytoplankton assemblages in the subtrophical oligotrophic Lake Fuxian, the second deepest lake in China, were investigated monthly from September 2002 to August 2003. A total of 113 species belonging to seven phyla were identified, among them, a filamentous green alga, Mougeotia sp., dominated almost throughout the study period and comprised most of the total phytoplankton biomass. Mougeotia sp. has made a substantial development during the past decades: it was absent in 1957, only occasionally present in 1983, increased substantially in 1993, and became predominant in 2002-2003. It is likely that natural invasion of the Taihu Lake noodlefish (Neosalanx taihuensis) has led to a change of dominant herbivorous zooplankton from small to large calanoid, which has increased grazing pressure on small edible algae, and thus has indirectly favored the development of the inedible filamentous Mougeotia sp.

Tissue distributions and seasonal dynamics of the hepatotoxic microcystins-LR and -RR in a freshwater snail (Bellamya aeruginosa) were studied monthly in a large shallow, eutrophic lake of the subtropical China during June-November, 2003. Microcystins (MCs) were quantitatively determined by High-Performance Liquid Chromatography (HPLC) with a qualitative analysis by a Finnigan LC-MS system. On the average of the study period, hepatopancreas was the highest in MC contents (mean 4.14 and range 1.06-7.42 mug g(-1) DW), followed by digestive tracts (mean 1.69 and range 0.8-4.54 mug g(-1) DW) and gonad (mean 0.715 and range 0-2.62 mug g(-1) DW), whereas foot was the least (mean 0.01 and range 0-0.06 mug g(-1) DW). There was a positive correlation in MC contents between digestive tracts and hepatopancreas. A constantly higher MC content in hepatopancreas than in digestive tracts indicates a substantial bioaccumulation of MCs in the hepatopancreas of the snail. The average ratio of MC-LR/MC-RR showed a steady increase from digestive tracts (0.44) to hepatopancreas (0.63) and to gonad (0.96), suggesting that MC-LR was more resistant to degradation in the snail. Since most MCs were present in the hepatopancreas, digestive tracts and gonad with only a very small amount in the edible foot, the risk to human health may not be significant if these toxic parts are removed prior to snail consumption. Microcystins are naturally occurring hepatotoxic cyclic heptapeptides produced by some toxic freshwater cyanobacterial species. In this study, crude extract of toxic cyanobacterial blooms from Dianchi Lake in southwestern China was used to determine the effects of microcystins on rape (Brassica napus L.) and rice (Oryza sativa L.). Experiments were carried out on a range of doses of the extract (equivalent to 0, 0.024, 0.12, 0.6 and 3 mug MC-LR/ml). Investigations showed that exposure to microcystins inhibited the growth and development of both rice and rape seedlings, however, microcystins had more powerful inhibition effect on rape than rice in germination percentage of seeds and seedling height. Microcystins significantly inhibited the elongation of primary roots of rape and rice seedlings. Determination of the activities of peroxidase and superoxide dismutase demonstrated that microcystin stress was manifested as an oxidative stress. Using ELISA, microcystins were examined from the extract of exposed rape and rice seedlings, indicating that consumption of edible plants exposed to microcystins via irrigation route may have health risks. Significantly different levels of recovered microcystins between exposed rice and rape seedlings Suggested that there might be different tolerant mechanisms toward microcystins. We studied in the laboratory the population growth rates of four cladocerans fed both with decomposed Microcystis aeruginosa and with a mixture of fresh colonial M. aeruginosa and Scenedesmus obliquus. The neonates of Diqphanosoma brachyurum and Daphnia carinata were able to develop into adults when they were fed with <64mum decomposed M. aeruginosa, while those of Moina micrura could not use decomposed M. aeruginosa. The population growth rate of the largest species, D. carinata, was less affected by the presence of fresh colonial M. aeruginosa than the other three species. D. carinata obtained the highest growth rate at a biomass level of 10 mg L-1 fresh colonial M. aeruginosa, indicating that, to some extent, it can use colonial M. aeruginosa at a size range of 64-112mum. The population growth rate of M. micrura was negatively correlated with fresh colonial M. aeruginosa within a range of 10-100 mg L-1. The population growth rates of D. brachyurum and Ceriodaphnia cornuta were remarkably decreased by fresh colonial M. aeruginosa, although no significant difference was found within the M. aeruginosa biomass range of 10-100 mg L-1 for either cladoceran. At a biomass level of 50 mg L-1 M. aeruginosa, the population growth rates of the four cladocerans positively correlated with S. obliquus biomass within a range of 0.1-5.0 mg L-1. Our results indicate that the zooplankton community under bloom condition is shaped by the quantity of both M. aeruginosa and other edible algae.

The community structure of zooplankton was studied in a eutrophic, fishless Japanese pond. The ecosystem was dominated by a dinoflagellate, Ceratium hirundinella, two filter-feeding cladocerans, Daphnia rosea and Ceriodaphnia reticulata, and an invertebrate predator, the dipteran Chaoborus flavicans. The midsummer zooplankton community showed a large change in species composition (the Daphnia population crashed) when a heavy Ceratium bloom occurred. It is shown that (i) the rapid density decline of D.rosea in mid-May was mainly caused by a shortage of edible phytoplankton, which was facilitated by the rapid increase in C.hirundinella abundance; (ii) the low density of D.rosea in June-July was considered to be mainly caused by the blooming of Ceratium hirundinella (which may inhibit the feeding process of D.rosea), while predation by C.flavicans larvae, the changing temperature, the interspecific competition and the scarcity of edible algae were not judged to be important; (iii) the high summer biomass of the planktonic C.flavicans larvae was maintained by the bloom of C.hirundinella, because >90% of the crop contents of C.flavicans larvae were C.hirundinella during this period. The present study indicates that the large-sized cells or colonies of phytoplankton are not only inedible by most cladocerans, but the selective effect of the blooming of these algae can also influence the composition and dominance of the zooplankton community, especially for the filter-feeding Cladocera, in a similar way as the selective predation by planktivorous fish. The large-sized phytoplankton can also be an important alternative food for ominivorous invertebrate predators such as Chaoborus larvae, and thus may affect the interactions between these predators and their zooplanktonic prey. In this way, such phytoplankton may play a very important role in regulating the dynamics of the aquatic food web, and become a driving force in shaping the community structure of zooplankton.

采用样方法研究了首曲湿地功能区"黑土滩"退化草甸改良后3 a的群落特征变化,分析了围栏封育和补播"高寒1号"生态组合草种对退化草甸的恢复改良效果.结果显示:仅封育3 a后,"黑土滩"退化草甸群落的盖度、高度、地上生物量和可食牧草比例均显著提高,丰富度指数由0.55增加到0.75,多样性指数由0.07增加到了0.25;封育后补播"高寒1号"生态草种相对于封育前,使得退化草甸的盖度增加了56.00%,高度增加了11.74 cm,地上生物量增加了222.24 g/m~2,可食牧草比例增加了55.98%,物种数由5种/m~2增加到了15种/m~2,丰富度指数由0.55增加到了3.29,多样性指数由0.07增加到了1.85,均匀度指数由0.06增加到了0.27.相对围栏封育而言,封育后补播是一种更有效的"黑土滩"退化草甸改良恢复措施.围栏封育和补播配套实施可以显著改善"黑土滩"退化草地的群落貌相、草地生产力和组分结构状况.

随着工农业的快速发展,土壤重金属尤其Cd和Pb污染日益严重。筛选和培育具有重金属低积累特性的农作物排异品种被认为是当前应对土壤重金属污染最为合理和有效的途径之一。本文通过盆栽试验、大田试验和砂培试验,研究了大白菜品种对Cd和Pb的吸收和积累的品种差异、对Cd、Pb胁迫的响应以及大白菜安全生产的调控技术,得出以下结论: 1) 盆栽梯度试验中,80种大白菜地上部对Cd的吸收存在显著差异(p < 0.05)。在3种Cd处理下(1.0, 2.5和5.0 mg/kg),80种大白菜Cd含量浓度范围分别为(mg/kg) 0.22–2.46, 0.90–14.10和2.03–18.01, 其平均值分别为 0.79, 3.76 和6.79 mg/kg DW。大白菜对Cd胁迫具有较强的耐性。大田试验中,15种大白菜的富集系数和转运系数与盆栽梯度试验的结果基本一致。排异植物的筛选和鉴定标准包括:(1)该植物的地上部和根部的Cd含量都很低或者可食部位低于有关标准;(2) 富集系数(BF) < 1.0;(3) 转运系数(TF) < 1.0;(4)该植物具有较高的Cd耐性,在较高的Cd污染下能够正常生长且生物量无显著下降。采用此标准,结合盆栽梯度试验和大田试验结果,北京新3号、绿星70和丰源新3号可鉴定为Cd排异品种。秋傲和赛新5号具有排异Cd特性,但其对Cd的耐性较差。 2) 盆栽梯度试验中,在Pb投加浓度为500和1500 mg/kg处理下,30种大白菜地上部对Pb的吸收存在显著差异 (p < 0.05),其Pb浓度的范围分别为:0.52–8.68 和1.86–16.20 mg/kg, 其平均值分别为3.01 和6.87 mg/kg DW。并且,随着Pb浓度的增加,白菜地上部Pb含量有随之增加的趋势。大白菜对Pb具有较强的耐性。低浓度的Pb处理对大白菜的生物具有一定的促进作用。结合盆栽试验和大田试验的结果,秋傲、世博秋抗和福星80可鉴定为Pb排异大白菜品种。 3) 砂培试验中,在Cd和Pb胁迫下,大白菜地上部的丙二醛(MDA)含量增加,随着Cd处理浓度的增加,超氧化物歧化酶(SOD)活性呈现先下降后上升接着下降的趋势,酸菜王的SOD活性要高于北京新3号SOD的活性。随着Pb处理浓度的增加,福星80地上部的SOD活性随之增加,而绿星大棵菜地上部的SOD活性先下降后增加。在不同梯度的Cd处理下,大白菜地上部的可溶性蛋白(SP)含量未见显著降低,甚至有所增加,而在不同梯度的Pb处理下,大白菜地上部的SP含量有所降低。 4) 施用改良剂可升高土壤的pH值和降低土壤中的有效态Cd,从而对大白菜的生长具有促进作用。施用改良剂可显著降低大白菜对Cd和Pb的吸收和累积。

Melatonin is a highly conserved molecule that not only exists in animals, but also is present in bacteria, unicellular organisms and in plants. Since melatonin is an antioxidant, in plants melatonin was speculated to protect them from intrinsic and environmental oxidative stress. More importantly, melatonin in edible plants inevitably enters animals and human through feed and food. In this study, more than 100 Chinese medicinal herbs were analyzed using the methods of solid phase extraction and HPLC-FD on-line with MS to determine whether melatonin is present in these commonly used herbs. Melatonin was detected in majority of these plants. Sixty-four of them contain melatonin in excess of 10 ng per gram dry mass. Melatonin levels in several herbs are in excess of 1000 ng/g. It is well known that normal average physiological plasma levels of melatonin are only 10-60 pg/mL. These high level-melatonin containing plants are traditionally used to treat diseases which presumably involve free radical damage. The current study provides new information concerning one potentially effective constituent present in a large number of medicinal herbs. The results suggest that these herbs should be reevaluated in reference to their nutritional and medicinal value. (C) 2003 Elsevier Science Inc. All rights reserved.

Chromatography-amperometric detection of nitrite with a polypyrrole modified glassy carbon electrode doped with tungstodiphosphate anion (Dawson-type P2W18O626-/PPy/GC electrode) based on its electrocatalytic reduction of nitrite is described. The cyclic and hydrodynamic voltammetry of nitrite at the P2W18O626-/PPy/GC electrode was studied. The factors affecting the detection of nitrite and the analytical performance of the modified electrode in flowing stream were investigated. The results show that the modified electrode has a good sensitivity (the limit of detection is 1 mu mol dm(-3)) and a satisfactory reproducibility (RSD = 3.78%, N = 21). The modified electrode was used in the chromatographic detection of nitrite spiked in the liquid from a tin of mushrooms and the mineralized spring water. It was found that the modified electrode exhibited good selectivity for nitrite.

A transformation model for Laminaria japonica was established from 1993 to 1998, on the basis of which the transgenic kelp with heterologous gene encoding hepatitis B surface antigen (HBsAg) was obtained by using the micro-particle bombardment transformation method. Results of quantitative ELISA showed that HBsAg in transgenic kelp was 0.529 mug/mg soluble proteins on average and the highest value was 2.497 mug/mg, implying that recombinant HBsAg had natural epitope. Further support for the integration of HBsAg gene into kelp genome was obtained by PCR-Southern and total DNA hybridization. Prospect of kelp bio-reactor producing high value materials such as edible HBV vaccine was discussed as well.
