树突状细胞(DC)是免疫系统中具有多种免疫功能的关键细胞,但在HIV/AIDS 患者 体内,虽有大量病毒及抗原存在,却不能有效地激发特异性免疫应答。最新的研究显示, HIV/AIDS 血源和性传染的途径虽有不同,但DC 均是早于T 细胞而最先遭受感染的“第一 靶细胞”。已知辅助蛋白(Nef、Rev、Tat、Vif、Vpr 和Vpu)是HIV-1 在宿主细胞内复制 和致病的根本因素。而哪种辅助蛋白、如何影响DC 功能,至今研究结果甚少,结论杂乱 不一。 本论文作为HIV-1 影响DC 功能研究的实验体系部分,旨在为系统比较研究6 个辅助 蛋白对DC 功能的调节和机理奠定基础。采用分子生物学和免疫学技术方法,把辅助蛋白 基因从DNA 和mRNA 水平导入目的细胞DC,在DC 前体及其分化、成熟的各个时相, 连续、动态地分析辅助蛋白对DC 特征性表面标志及免疫相关基因的表达水平、摄取和处 理抗原、激发和调控免疫应答等功能的影响,以期揭示HIV/AIDS 患者DC 功能明显异常 和失调的原因。 首先,分别将6 个辅助蛋白基因克隆到pEGFP-N2 和pCS2+表达载体,得到具有绿色 荧光蛋白融合基因的表达载体,将分别用于DNA 和mRNA 水平转染DC。再以K562 细胞 为模型建立mRNA 转染细胞的基本方法;以人外周血CD14 单核细胞为前体,建立了体外 “2+2”快速诱导DC 的方法。最后,利用Amaxa 转染系统,从DNA 水平研究辅助蛋白对 DC 前体(单核细胞)的功能影响。发现单核细胞转染辅助蛋白与GFP 融合基因5h 后,胞 内蛋白大量表达,且能维持表达48h;其中Nef、Tat、Vpu、Rev、Vif、Vpr 表达效率分别 为35.42%、34.42%、43.42%、 17.07%、13.65%、10.29%;单核细胞转染基因后,表型 CD14、HLA-DR、CD80、CD83、CD86、DC-SIGN 没有明显的表达变化;转染Nef、Vpu、 Rev 辅助蛋白后,单核细胞有10%凋亡;Vpr 能抑制单核细胞IL-10 的分泌,Nef 能促进分 泌IL-6。 总之,通过构建辅助蛋白的表达载体,优化DC 的体外培养过程以及mRNA 转染方法, 并成功将辅助蛋白导入DC 前体,鉴定其表型和功能变化。为研究辅助蛋白影响DC 的功 能和机制建立了稳定而可行的实验系统。

以化合物对HIV1诱导C8166细胞形成合胞体的抑制实验和化合物对HIV1感染细胞的保护实验作为初筛方法,筛选了来源于7科17种植物的47个样品,其中8个样品经测定是核糖体失活蛋白(RIPs),其余为粗提蛋白。进而测定了初筛有抗HIV活性的化合物对共培养、急性和慢性感染的HIV1p24抗原表达水平的影响,用间接荧光染色检测这些化合物对HIV抗原阳性细胞率的影响,以确证其抗HIV活性。天花粉蛋白(TCS)、南方栝楼蛋白峰Ⅴ、南方栝楼蛋白峰Ⅵ均显著地抑制合胞体的形成;巴Ⅱ、丝瓜子蛋白、油瓜根蛋白、木盍藤子蛋白有一定的抑制作用。老鼠拖瓜蛋白、大叶木鳖子根蛋白、西双版纳根Ⅱ等粗提蛋白有显著的抑制作用。RIPs不能保护HIV1感染细胞的死亡。TCS显著地抑制了HIV1急性感染中p24抗原的表达,减少了HIV抗原阳性细胞数,但它们均不影响共培养细胞融合、HIV1慢性感染中p24抗原表达水平。结果表明,除TCS等已知RIPs外,还有一些新的RIPs具有不同程度的抗HIV活性。

Five models for human interleukin-7 (HIL-7), HIL-9, HIL-13, HIL-15 and HIL-17 have been generated by SYBYL software package. The primary models were optimized using molecular dynamics and molecular mechanics methods. The final models were optimized using a steepest descent algorithm and a subsequent conjugate gradient method. The complexes with these interleukins and the common gamma chain of interleukin-2 receptor (IL-2R) were constructed and subjected to energy minimization. We found residues, such as Gln127 and Tyr103, of the common gamma chain of IL-2R are very important. Other residues, e.g. Lys70, Asn128 and Glu162, are also significant. Four hydrophobic grooves and two hydrophilic sites converge at the active site triad of the gamma chain. The binding sites of these interleukins interaction with the common gamma chain exist in the first helical and/or the fourth helical domains. Therefore, we conclude that these interleukins binds to the common gamma chain of IL-2R by the first and the fourth helix domain. Especially at the binding sites of some residues (lysine, arginine, asparagine, glutamic acid and aspartic acid), with a discontinuous region of the common gamma chain of IL-2R, termed the interleukins binding sites (103-210). 选用实验室克隆的17个鳙鱼微卫星分子标记分析四川泸州和江西鄱阳湖的两个种群鳙鱼的遗传多样性及种质特性,计算和统计了杂合度、多态信息含量(PIC)、有效等位基因数、等位基因频率、遗传距离、遗传相似系数、Hardy-Weinberg平衡偏离指数等方面内容。结果表明:选择使用17个微卫星标记,其中有4个为单态标记,13个为多态标记。江西和四川鳙鱼群体每个微卫星位点的平均等位基因数分别为3.325及3.882,平均有效等位基因数分别为3.531及2.676,多态位点百分率分别为82.4及70.5,17个微卫星标记

用RACE PCR获得了鳜白介素-1β(IL-1β)的全长cDNA.鳜IL-1β的cDNA全长为1298 nt(核苷酸),其5′非编码区包含UTR 93 nt;3′非编码区包含452 nt;其开放阅读框内包含753 nt,翻译成251个氨基酸.将鳜IL-1β克隆到原核表达载体pET-32a上,在大肠杆菌Rosetta- gami(DE3)内以包涵体形式得以高效表达.

用刀豆蛋白A(ConA)刺激诱导草鱼和中华鳖脾细胞 ,收集细胞培养上清液 ,用小鼠胸腺细胞增殖试验和对小鼠L92 9细胞系杀伤试验检测上清液中白细胞介素 2 (简称IL 2 )活性。结果表明 :草鱼、中华鳖脾细胞培养上清液中有IL 2样活性物质 ,这种物质使小鼠的胸腺细胞3H TdR掺入量明显增加 ,在相同效靶比的条件下对小鼠L92 9细胞系杀伤率也显著增强 ,这种IL 2活性均能被抗人rIL 2血清所抑制。中华鳖脾细胞培养上清液 (含IL 2 )对中华鳖胸腺细胞也有较明显的促增殖作用并能消除兔抗中华鳖胸

结果显示 :当年草鱼种和中华鳖脾细胞培养上清液中能检测出IL 2活性 ,而且 1龄以上草鱼、中华鳖的IL 2活性高于当年孵化的草鱼和中华鳖。草鱼、中华鳖脾细胞在 2 5℃培养温度条件下 ,其上清液中IL 2活性最高 ,35℃次之 ,1 5℃最低。因此 ,草鱼、中华鳖IL 2活性在一定范围内是随着年龄增加而增强和依赖温度的。通过小鼠胸腺细胞增殖和对小鼠L92 9细胞杀伤率的实验表明 37℃比 2 5℃检测温度下所测的IL 2活性高 ,而中华鳖胸腺细胞增殖实验却显示 2 5℃检测温度下IL 2活性高于 37℃

Present in the excrement of humans and animals, 17 beta-estradiol (E-2) has been detected in the aquatic environment in a range from several nanograms to several hundred nanograms per liter. In this study, the sensitivities of rare minnows during different life stages to E-2 at environmentally relevant (5, 25, and 100 ng l(-1)) and high (1000 ng l(-1)) concentrations were compared using vitellogenin (VTG) and gonad development as biomarkers under semistatic conditions. After 21 days of exposure, VTG concentrations in whole-body homogenates were analyzed; the results indicated that the lowest observed effective concentration for VTG induction was 25 ng l(-1) E-2 in the adult stage, but 100 ng l(-1) E-2 in the larval and juvenile stages. After exposure in the early life stage, the larval and juvenile fish were transferred to clean water until gonad maturation. No significant difference in VTG induction was found between the exposure and control groups in the adults. However, a markedly increased proportion of females and appearance of hermaphrodism were observed in the juvenile-stage group exposed to 25 ng l(-1) E-2. These results showed that VTG induction in the adult stage is more sensitive than in larval and juvenile stages following exposure to E-2. The juvenile stage may be the critical period of gonad development. Sex ratio could be a sensitive biomarker indicating exposure to xenoestrogens in early-life-stage subchronic exposure tests. The results of this study provide useful information for selecting sensitive biomarkers properly in aquatic toxicology testing.

In this study, an IL-8 homologue has been cloned and identified from a reptile, Chinese soft-shelled turtle for the first time. The full-length cDNA of turtle IL-8 was 1188 bp and contained a 312 bp open reading frame (ORF) coding for a protein of 104 amino acids. The chemokine CXC domain, which contained Glu-Leu-Arg (ELR) motif and four cysteine residues, was well conserved in turtle IL-8. The 4924 bp genomic DNA of turtle IL-8 contained four exons and three introns. Phylogenetic analysis showed that the amino acid sequence of turtle IL-8 clustered together with birds. RT-PCR analysis showed that turtle IL-8 mRNA was constitutively expressed liver, spleen, kidney, heart, blood and intestine tissues of control turtles. Real-time quantitative PCR analysis further indicated that the turtle IL-8 mRNA expression was apparent in various tissues at 8 h and up-regulated significantly during 8 h-7 d after Aeromonas hydrophila infection. The present studies will help us to understand the evolution of IL-8 molecule and the inflammatory response mechanism in reptiles. (C) 2009 Elsevier Ltd. All rights reserved.

This study investigates the ozonation of 17 alpha-ethinylestradiol (EE2) in aqueous solution. The affecting factors on the degradation of EE2 were studied and described in details, such as initial EE2 concentration, initial pH value and ozone concentration. In addition, some parameters such as pH. electrical conductivity, mineralization efficiency and degradation products were monitored during the process. The mineralization efficiency of EE2 could reach 53.9%. During the ozonation process the rapid decrease of pH and the sharp increase of electrical conductivity indicated the fort-nation of acidic by-products, small fragments and ions which were confirmed by high performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GUMS) analysis. Results showed that there were intermediate products of smaller molecule with higher polarity produced during the course of EE2 degradation. Then a possible reaction pathway for EE2 degradation involving all intermediates detected is proposed. During the ozonation process EE2 was first oxidized into hydroxyl-semiquinone isomers which were subsequently degraded into low molecular weight compounds such as oxalic acid, malonate, glutarate, and so on. Furthermore. these organic acids are easily oxidized by ozone into carbon dioxide (CO2). This work shows that ozonation process is promising for the removal of EE2. The results can provide some useful information for the potential treatment of EE2 by ozonation in aqueous solution. (c) 2005 Elsevier B.V. All rights reserved.

This paper reports that lnAs/In0.53Ga0.47As/AlAs resonant tunnelling diodes have been grown on InP substrates by molecular beam epitaxy. Peak to valley current ratio of these devices is 17 at 300K. A peak current density of 3kA/cm(2) has been obtained for diodes with AlAs barriers of ten monolayers, and an In0.53Ga0.47As well of eight monolayers with four monolayers of InAs insert layer. The effects of growth interruption for smoothing potential barrier interfaces have been investigated by high resolution transmission electron microscope.
