An antibody phage display library against White Spot Syndrome Virus (WSSV) was constructed. After four rounds of panning against WSSV, 192 out of 480 clones displayed WSSV binding activity. One of the positive clones, designated A1, had relatively higher activity specifically binding to WSSV A1-soluble, single-chain fragment variable (scFv) antibody has an affinity constant (K-aff) of 2.02 +/- 0.42 x 10(9) M-1. Dot blot assays showed that A1-soluble scFv could detect WSSV directly from shrimp hemolymph after 24-h feeding infection by WSSV. A1 scFv has potential for the development of a cheap, simple and sensitive diagnostic kit for WSSV in the field. (C) 2003 Elsevier Science B.V. All rights reserved.

A new gene with WD domains is cloned and characterized according to its differential transcription and expression between previtellogenic oocytes (phase I oocytes) and fully-grown oocytes (phase V oocytes) from natural gynogenetic silver crucian carp (Carassius auratus gibelio) by using the combinative methods of suppressive subtraction hybridization, SMART cDNA synthesis and RACE-PCR. The full-length cDNA is 1870 bp. Its 5 ' untranslated region is 210 bp, followed by an open reading frame of 990 bp, which has the typical vertebrate initiator codon of ANNATG. The open reading frame encodes a protein with 329 amino acids. It has 670 bp of 3 ' untranslated region and an AATAAA polyadenylation signal. Because it has 92% homology to STRAP (serine-threonine kinase receptor-associated protein), a recently reported gene, we named it FSTRAP (fish STRAP). Virtual Northern blotting indicated that the FSTRAP was transcribed in fully-grown oocytes (phase V oocytes), but not in previtellogenic oocytes (phase I oocytes). RT-PCR analysis showed that FSTRAP was transcribed in brain, heart, kidney, muscle, ovary, spleen and testis, but not in liver. And its mRNA could be detected in the oocytes from phase II to phase V. Western blotting also showed that FSTRAP protein could be detected in brain, heart, kidney, muscle, ovary, spleen and testis except liver. Results of Western blotting on various oocytes were also similar to the RT-PCR data. FSTRAP protein was not expressed in the previtellogenic oocytes. Its expression initiated from phase II oocytes after vitellogenesis, and was consistent with the mRNA transcription.

Neutron irradiated high resistivity (4-6 kOMEGA-cm) silicon detectors in the neutron fluence (PHI(n)) range of 5 X 10(11) n/cm2 to 1 X 10(14) n/cm2 have been studied using a laser deep level transient spectroscopy (L-DLTS). It has been found that the A-center (oxygen-vacancy, E(c) = 0.17 eV) concentration increases with neutron fluence, reaching a maximum at PHI(n) almost-equal-to 5 X 10(12) n/cm2 before decreasing with PHI(n). A broad peak has been found between 200 K and 300 K, which is the result of the overlap of three single levels: the V-V- (E(c) = 0.38 eV), the E-center (P-V, E(c) = 0.44 eV), and a level at E(c) = 0.56 eV that is probably V-V0. At low neutron fluences (PHI(n) < 5 X 10(12) n/cm2), this broad peak is dominated by V-V- and the E-centers. However, as the fluence increases (PHI(n) greater-than-or-equal-to 5 X 10(12) n/cm2), the peak becomes dominated by the level of E(c) = 0.56 eV.

A multistream reactor for high-throughput examining the surface acidity by NH3-TPD method by application of multistream mass spectrometer screening (MSMSS) technique has been developed. This method allows for examining the surface acidity of 10 catalyst samples in about 6 h, which is an improvement over the traditional process. The demonstration of the feasibility of high-throughput TPD can be significant in convincing the hardened traditionalists in the heterogeneous catalysis community that, combinatorial methods indeed should have an important place in scientific catalyst research and development. The developed method could also be used for almost all the temperature-programmed analysis theoretically with careful designed multistream reactors. (C) 2003 Elsevier B.V. All rights reserved.

The international and national organizations have constituted a series of methods to characterize the physical and chemical properties for substances. The applications to physical transition, chemical reaction, and characteristic parameters of substances are given in the article as examples.

In this article, a two-dimensional (2D) nanoplate and a 3D hierarchical structure of BiOCl were synthesized through a simple sonochemical route. Compared with previous preparation methods, the 2D nanoplates can be prepared at a relatively short time (about 30 min) with low energy used. Additionally, these 2D nanoplates can easily assemble into a 3D hierarchical structure with the surfactant reagents. The obtained products were well crystallized and subsequently characterized by a range of methods, such as X-ray powder diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), high-resolution transmission microscopy (HRTEM), selected area electron diffraction (SAED) and Raman spectroscopy.

In this work, glycyrrhetinic acid-modified chitosan (mGA-suc-CTS) used as liver targeted carrier for drug delivery, was prepared via hemisuccinate as a bridged group. The structure of the product was confirmed by IR and NMR methods and the degree of substitution (DS) of glycyrrhetinic acid groups was estimated via elemental analysis. Nanoparticles were formed by ionic gelation methold. The drug-loading and release behavior of the nanoparticles were investigated using BSA as the model drug. The results indicated that the carrier with a highest DS of 5.19% could be got and the DS was controlled by changing reaction temperature or feed ratio. BSA could be entrapped into the nanoparticles with the drug-loading ratio of 26.3% and the encapsulation efficiency of 81.5%. A sustained release over an 11-day period was observed in pH 7.4 in vitro.

Y0.9-xGdxEu0.1BO3 phosphors were synthesized by spray drying (SD) method, and the results were compared with those by conventional solid state (SS) and citrate gel (GC) methods. The PL intensity of phosphors increases with the increase of x value in Y0.9-xGdxEu0.1BO3 (prepared by SD) due to an energy migration process like Gd3+ - (Gd3+)(n) - Eu3+ occurred in the material. Compared with the latter two methods, the phosphor particles prepared by spray drying method have a better morphology, such as homogeneous size (about 1similar to3 mum) with spherical shape and smooth surface. Furthermore, the spray drying-derived phosphors have higher photoluminescence (PL) intensity than those by citrate gel method, but still a little lower than those by the solid state method.

Using low-temperature hydrothermal methods, nanoscale lanthanide phenylphosphonates species with different morphologies, namely, nanoparticles and nanorods, have been systematically synthesized. The possible growth mechanism of these nanorods was discussed. X-ray diffraction, transmission electron microscopy, electron diffraction, and photoluminescence spectra were used to characterize these materials. The photoluminescent properties of EU(O3PC6H5)(HO3PC6H5) and La0.91EU0.09(O3PC6H5)(HO3PC6H5) nanorods were discussed.

In this paper, we will report the preparation of a mixed-valence polyoxometalate compound (Bu4N)(4)[PMo12O40].2DMF.H2O (TBA = tetrabutylammonium; DMF = N,N-dimethyl formamide). The title compound has been photochemically synthesized and characterized by using elemental analysis, IR, solid diffusion reflectance electronic spectra, ESR spectra, XPS, CV and X-ray single-crystal analysis. The crystal lographic data are as follows: monoclinic, P2(1)/c, a = 14.124(3), b = 17.481(4), c = 22.744(5) Angstrom, beta = 101.66(3)degrees, V = 5500(2) Angstrom(3), C70H160Mo12N6O43P, M-r = 2956.29, Z = 2, D-c = 1.785 g/cm(3), F(000) = 2970 and mu(MoKalpha) = 1.412 mm(-1). The structure has been refined to R = 0.0638 and wR = 0.1975 by full-matrix least-squares methods. The title compound is composed of four tetrabutylammonium cations, one [(PMoMo11O40)-Mo-V](4-) heteropoly anion, two N,N-dimethyl formamide and one H2O molecule.

A new multilayer film fabricated based on electrostatic attraction in this laboratory was firstly characterized by the electrochemical impedance spectroscopy. The relationship between the charge-transfer resistance and double-layer capacitance with the number of layers was obtained through analyzing the impedance data. It demonstrated that the multilayer film showed a unique structure with the film growth. Compared to other electrochemical methods, the electrochemical impedance spectroscopy was proved to be a very sensitive and useful technique for characterizing the multilayer films.

Ultrathin multilayer films of a polybasic lanthanide heteropoly tungstate-molybdate complex and a cationic polymer of quaternized poly(4-vinylpyridine) partially complexed with osmium bis(2,2'-bipyridine) have been fabricated on a gold electrode precoated with a cysteamine self-assembled monolayer. The multilayer films have been characterized by optical spectroscopy, small-angle X-ray diffraction, and electrochemical methods (cyclic voltammetry and electrochemical impedance). Especially, the electrochemical impedance spectroscopy is developed to monitor the layer deposition processes. It provides important information such as double-layer capacitance and charge-transfer resistance. All obtained results reveal regular film growth with each layer adsorption. By mechanism-transformation (anionic --> cationic) poly(styrene-6-2-ethyl-2-oxazoline) diblock copolymer, PS-b-PEOx, was synthesized in two steps. The first step is the polymerization of styrene block capped with ethylene oxide and its tosylation; the second step is the cationic ring-opening polymerization of 2-ethyl-2-oxazoline. The products were thoroughly characterized by various methods, such as H-1-NMR, IR, DMA, TEM and SAXS. The results show that the copolymer obtained possesses high molecular weight and narrow molecular weight distribution.

The first step is the polymerization of butadiene block capped with ethylene oxide and its tosylation; the second step is the cationic ring-opening polymerization of 2-ethyl-2-oxazoline. The products were characterized by various methods, such as IR, (HNMR)-H-1, DMA, TEM and SAXS. The results show that the obtained copolymers possess high molecular weight and narrow molecular weighs distribution, and that the content of 1,4-structure was controllable.

The morphology of polyamidelOlO/polypropylene blends was found to significantly depend upon the concentration of the compatibilizer[polypropylene-grafted-acrylic acid (PP-g-AA)]. A significant reduction in phase size was observed because of the interaction that existed between the PP-g-AA and polyamide. These interactions have been confirmed by several methods. The tensile mechanical properties and impact behavior of the prepared blends were investigated and correlated with scanning electron microscope (SEM) analysis of the fracture surfaces. It was found that PP-g-AA as the compatibilizer has a profound effect upon the properties of the blends. This behavior is attributed to a series of chemical and physico-chemical interactions taking place between the two components.
