A hemorrhagic proteinase, jerdohagin, was purified from Trimeresurus jerdonii venom by gel filtration and ion-exchange chromatographies. It was a single chain polypeptide with an apparent molecular weight of 96 kDa as estimated by SDS-PAGE under the non-reducing and reducing conditions. Internal peptide sequencing indicated that it consisted of metalloproteinase, disintegrin-like and cysteine-rich domains and belonged to the class III snake venom metalloproteinases (class P-III SVMPs). Like other typical metalloproteinases, hemorrhagic activities of jerdohagin were completely inhibited by EDTA, but not by PMSF. Jerdohagin preferentially degraded a-chain of human fibrinogen. Interestingly, jerdohagin did not activate human prothrombin, whereas it cleaved human prothrombin and fragment F1 of activated human prothrombin. (C) 2004 Elsevier Ltd. All rights reserved.

Bat flight poses intriguing questions about how flight independently developed in mammals. Flight is among the most energy-consuming activities. Thus, we deduced that changes in energy metabolism must be a primary factor in the origin of flight in bats. The respiratory chain of the mitochondrial produces 95% of the adenosine triphosphate (ATP) needed for locomotion. Because the respiratory chain has a dual genetic foundation, with genes encoded by both the mitochondrial and nuclear genomes, we examined both genomes to gain insights into the evolution of flight within mammals. Evidence for positive selection was detected in 23.08% of the mitochondrial-encoded and 4.90% of nuclear-encoded oxidative phosphorylation (OXPHOS) genes, but in only 2.25% of the nuclear-encoded nonrespiratory genes that function in mitochondria or 1.005% of other nuclear genes in bats. To address the caveat that the two available bat genomes are of only draft quality, we resequenced 77 OXPHOS genes from four species of bats. The analysis of the resequenced gene data are in agreement with our conclusion that a significantly higher proportion of genes involved in energy metabolism, compared with background genes, show evidence of adaptive evolution specific on the common ancestral bat lineage. Both mitochondrial and nuclear-encoded OXPHOS genes display evidence of adaptive evolution along the common ancestral branch of bats, supporting our hypothesis that genes involved in energy metabolism were targets of natural selection and allowed adaptation to the huge change in energy demand that were required during the origin of flight.

Five models for human interleukin-7 (HIL-7), HIL-9, HIL-13, HIL-15 and HIL-17 have been generated by SYBYL software package. The primary models were optimized using molecular dynamics and molecular mechanics methods. The final models were optimized using a steepest descent algorithm and a subsequent conjugate gradient method. The complexes with these interleukins and the common gamma chain of interleukin-2 receptor (IL-2R) were constructed and subjected to energy minimization. We found residues, such as Gln127 and Tyr103, of the common gamma chain of IL-2R are very important. Other residues, e.g. Lys70, Asn128 and Glu162, are also significant. Four hydrophobic grooves and two hydrophilic sites converge at the active site triad of the gamma chain. The binding sites of these interleukins interaction with the common gamma chain exist in the first helical and/or the fourth helical domains. Therefore, we conclude that these interleukins binds to the common gamma chain of IL-2R by the first and the fourth helix domain. Especially at the binding sites of some residues (lysine, arginine, asparagine, glutamic acid and aspartic acid), with a discontinuous region of the common gamma chain of IL-2R, termed the interleukins binding sites (103-210). The beta-delayed neutron and gamma energy spectra taken from the decay of neutron-rich nucleus N-21 were measured by using the beta - gamma and beta - n coincidence detection method. Thirteen new neutron groups ranging from 0.28MeV to 4.98 MeV and with a total branching ratio of 88.7 +/- 4.2% were observed and presented. One gamma transition with an energy of 1222 keV emitted from the excited state of O-21, and four gamma transitions with energies of 1674, 2397, 2780, and 3175 keV emitted from the excited states of O-20 were identified in the 3 decay chain of N-21. The beta decay half-life for N-21 is determined to be 82.9 +/- 1.9 ms. The uncertainty of half-life is much smaller than the previous result.

A novel wholly aromatic diamine, 2,2 '-bis(3-sulfobenzoyl)benzidine (2,2 '-BSBB), was successfully prepared by the reaction of 2,2 '-dibenzoylbenzidine (2,2 '-DBB) with fuming sulfuric acid. Copolymerization of 1,4,5,8-naphathlenetetracarboxylic dianhydride with 2,2 '-BSBB and 2,2 '-DBB generated a series of rigid-rod sulfonated polyimides. The synthesized copolymers with the -SO3H group on the side chain of polymers possessed high molecular weights revealed by their high viscosity and the formation of tough and flexible membranes. The copolymer membranes exhibited excellent oxidative stability and mechanical properties due to their fully aromatic structure extending through the backbone and pendent groups. They displayed clear anisotropic membrane swelling in water with negligibly small dimensional changes in the plane direction of the membrane. The proton conductivities of copolymer membranes increased with increasing IEC and temperature, reaching value above 1.25 x 10(-1) S/cm at 20 degrees C, which is higher than that of Nafion (R) 117 at the same measurement condition. They displayed reasonably high proton conductivity due to the higher acidity of benzoyl sulfonic acid group, the larger interchain spacing, which is available for water to occupy, taking their lower water uptake (WU) into account. Consequently, these materials proved to be promising as proton exchange membrane.

The monolayer of the mixture of octadecanoic acid and octadecylamine with molar ratio 1: 1 has been investigated at the air-water interface. It was found that the monolayer shows a rather stable state at the surface pressure of 30 mN/m and this monolayer can be transferred onto a CaF2 plate by Langmuir-Blodgett (LB) technique. The infrared spectra of LB films indicated that octadecyl ammonium octadecanoate is formed by an intermolecular proton exchange between adjacent carboxylic and aminic groups (COO- and NH3+). In three-layer LB film, the CH2 scissoring mode of the long hydrocarbon chains of octadecyl ammonium octadecanoate shows a broad band feature at about 1468 cm(-1) while this vibrational mode of three-layer LB film of the mixture (1: 1) of deuterated stearic acid and octadecylamine (octadecylammonium octadecanoate-d35, C18H37NH3+C17D35COO-) only shows a narrow band. The broad feature of the CH2 scissoring mode in octadecylammonium octadecanoate probably originates from the coupling between the chain of stearic acid and that of octadecylamine while this kind of coupling could be completely removed in octadecylammonium octadecanoate-d35.

Three fully amorphous comb-branch polymers based on poly(styrene-co-maleic anhydride) as a backbone and poly(ethylene glycol) methyl ether of different molecular weights as side chains were synthesized. SiO2 nanoparticles of various contents and the salt LiCF3SO3 were added to these comb-branch polymers to obtain nanocomposite polymer electrolytes. The thermal and transport properties of the samples have been characterized. The maximum conductivity of 2.8x10(-4) S cm(-1) is obtained at 28 degreesC. In the system the longer side chain of the comb-branch polymer electrolyte increases in ionic conductivity after the addition of nanoparticles. To account for the role of the ceramic fillers in the nanocomposite polymer electrolyte, a model based on a fully amorphous comb-branch polymer matrix in enhancing transport properties of Li+ ions is proposed.

A novel synthesis of asymmetric bis(chlorophthalimide)s (3,4-BCPIs) has been established. The polymerizations of them produced higher molecular weight (0.38-0.51 dL/g) polyimides containing biphenyl units than those of isomeric polymers derived from symmetric bis(chlorophthalimide)s (4,4'-BCPIs) and 3,3'-BCPIs. The distribution of the formed biphenyl units of head to tail, head to head, and tail to tail in the chain of the polymers was about 58.0:21.0:21.0, determined by C-13 NMR spectra of the polymers. The composition of model compounds, determined by HPLC, was well consistent with the 13C NMR spectrum result. Comparing with polymers derived from 4,4'-BCPIs and 3,3'-BCPIs, the polymers derived from 3,4-BCPIs showed better solubilities in N,N-dimethylacetamide (DMAc), N,N-dimethyl-formamide (DMF), and N-methylpyrrolinone (NMP). Flexible films could be cast from the polymer solution with the inherent viscosities of above 0.35 dL/g. The polymer derived from asymmetric bis(chlorophthimide)s gave the highest T-g among the isomeric polymers.

The conformation of microperoxidase-11 (MP-11) adsorbed on roughened silver electrodes was studied using surface-enhanced Fourier transform Raman spectroscopy. The results demonstrate that MP-11 was initially adsorbed via its polypeptide chain with a alpha-helix conformation, as indicated by the enhancement of the characteristic bands related to the amides I and III. The weak resonance effect of the porphyrin macrocycle in the near IR region contributes to the spectrum of the heme group. The presence of imidazole as the sixth ligand to the heme iron influences the conformation of the polypeptide chain of MP-11 on the electrode surface. Evaporation of solvent water results in an opened conformation of the adsorbed MP-11. which allows the heme group to contact the electrode surface directly.

The compatibility and morphology of HIPS/PC and HIPS-g-GMA/PC blends were studied. The compatibility and morphology of HIPS/PC blends were characterized by DSC and SEM, respectively. The result of DSC shows that T-g of PS doesn't change with the blend composition, and T-g of PC decreases with the increase in weight fraction of HIPS, which indicates that the PC/HIPS blend is a partially miscible system. Results of SEM indicate that the decrease in T-g of PC results from PS interpenetrating into the phase of PC, and no change in T-g of PS results from PC not interpenetrating into the phase of PS. The copolymer of HIPS-g-GMA was prepared by reactive grafting method. The IR spectrum shows that GMA is grafted on the chain of HIPS. The compatibility and morphology of HIPS-gGMA (35)/PC (65) were studied by DSC and SEM. PC (65)/HEPS-g-GMA (35) blend exhibits reduced size of disperse phase, enhanced interface adhesion and lower T-g of PC phase as compared with the PC(65)/HIPS(35) blend. It implies that HIPS-g-GMA is an effective compatibilizer of the HIPS/PC blend.

Rabies virus was used as the antigen to immunize laying chickens. Anti-rabies virus immunoglobulin Y(IgY) was isolated from yolks of the eggs laid by these chickens using a two-step salt precipitation and one-step gel filtration protocol. The purified IgY was reduced with dithiothreitol, and heavy chains (HC) and light chains (LC) were obtained. In addition, the purified IgY was digested with pepsin and the fragment with specific antigen binding properties (Fab) was produced. Using matrix-assisted laser desorption/ionization mass spectrometry (MALDI-TOFMS), the average molecular weights of IgY, HC, LC, and Fab were determined as 167 250, 65 105, 18 660, and 45,359 Da, respectively. IgY has two structural differences compared with mammalian IgGs. First, the molecular weight of the heavy chain of IgY is larger than that of its mammalian counterpart, while the molecular weight of the light chain of IgY is smaller. Second, upon pepsin digestion, anti-rabies virus IgY is degraded into Feb, in contrast to mammalian IgG, which has been reported to be degraded into F(ab')(2) under the same conditions. A high-resolution C-13 n.m.r. spectrum of soluble polyaniline in DMF-d7 solution was recorded. The assignment for the various resonance peaks in the spectrum was tentatively performed and the chain structure of polyaniline was analysed. It has been shown that the main chain of pristine state polyaniline is composed of alternating benzoid-quinoid and successive benzoid-quinoid sequences with the former being present in greater concentration. The sequence distribution is random. In addition to the benzoid-type and quinoid-type structures, there is a small amount of other structural units in the main chain.

In chain molecules of 1, 2-PBD, there are two kinds of gauche arrangements, which is the cause of making the spectrum of the secondary carbon in main chain of the polymer split. In such a complex system, the gauche arrangements of the secondary carbon and the tertiary carbon occupy an important position. Hence, the contribution of the tertiary carbon to the chemical shifts of the secondary carbon has a decisive effect on the sequence structure distribution. The South China Sea (SCS) is one of the most active areas of internal waves. We undertook a program of physical oceanography in the northern South China Sea from June to July of 2009, and conducted a 1-day observation from 15:40 of June 24 to 16:40 of June 25 using a chain of instruments, including temperature sensors, pressure sensors and temperature-pressure meters at a site (117.5A degrees E, 21A degrees N) northeast of the Dongsha Islands. We measured fluctuating tidal and subtidal properties with the thermistor-chain and a ship-mounted Acoustic Doppler Current Profiler, and observed a large-amplitude nonlinear internal wave passing the site followed by a number of small ones. To further investigate this phenomenon, we collected the tidal constituents from the TPXO7.1 dataset to evaluate the tidal characteristics at and around the recording site, from which we knew that the amplitude of the nonlinear internal wave was about 120 m and the period about 20 min. The horizontal and vertical velocities induced by the soliton were approximately 2 m/s and 0.5 m/s, respectively. This soliton occurred 2-3 days after a spring tide.
