In order to avoid the hygroscopicity of LiCl specimem, the method of directly chlorinating Li_2CO_3 with NH_4Cl was successfully introducing into the thermal analysis of the system containing LiCl. The three fusibility diagram of LiCl-KCl, LiClNaCl, LiCl-LiF were determined using the method. The results are in agreement wish the values reported in the literature, and phase diagram of LiCl-KCl-LiF ternary system was constructed based on these results. An improved method of PCR in which the small segment of conchocelis is amplified directly without DNA extraction was used to amplify a RUBISCO intergenic spacer DNA fragment from nine species of red algal genus Porphyra (Bangiales, Rhodophyta), including Porphyra yezoensis (Jiangsu, China), P. haitanensis (Fujian, China), P. oligospermatangia (Qingdao, China), P. katadai (Qingdao, China), P. tenera (Qingdao, China), P. suborboculata (Fujian, China), P. pseudolinearis (Kogendo, Korea), P. linearis (Devon, England), and P. fallax (Seattle, USA). Standard PCR and the method developed here were both conducted using primers specific for the RUBISCO spacer region, after which the two PCR products were sequenced. The sequencing data of the amplicons obtained using both methods were identical, suggesting that the improved PCR method was functional. These findings indicate that the method developed here may be useful for the rapid identification of species of Porphyra in a germplasm bank. In addition, a phylogenetic tree was constructed using the RUBISCO spacer and partial rbcS sequence, and the results were in concordant with possible alternative phylogenies based on traditional morphological taxonomic characteristics, indicating that the RUBISCO spacer is a useful region for phylogenetic studies.

The main chlorophyll a/b light-harvesting complex (LHC 11) has been isolated directly from thylakoid membranes of marine green alga (Bryopsis corticulans Setch.) by two consecutive runs of anion exchange and gel-filtration chromatography. LHC 11 proteins in the membrane extracts treated with 3% n-Octyl-b-D-glucopyranoside (OG) obtained specific binding ability on Q Sepharose column, and thus were isolated from the thylakoid membranes in a highly selective fraction. The monomeric, trimeric and oligomeric subcomplexes of LHC 11 have been obtained by fractionation of the LHC 11 mixes with sucrose density gradient ultracentrifugation. The SDS-PAGE analysis of peptide composition and absorption spectrum showed that LHC 11 monomers, trimers and oligomers prepared through this work were intact and in high purity. Our report is the first to show that it is possible to purify LHC If directly from thylakoid membranes without extensively biochemical purification.

It is known that global climate changed from the early Tertiary “Green House” to the Quaternary “Ice House” of cyclic glacial-interglacial climatic changes. Since the middle Pleistocene, the climate cycles changed from 40 kyr to 100 kyr, and the amplitudes of climatic fluctuations increased significantly. Therefore, it is important to study the climate changes since the middle Pleistocene. The loess-paleosol sequence in China is considered as one of the most continuous continental records of the last 2.58 Ma. Paleoclimatic and environmental changes have been widely extracted through various climatic parameters. However, the history of paleovegetation on the Loess Plateau still remains unclear. Did an extensive broadleaf forest ever exist on the Loess Plateau? Pollen preserved in the loess and paleosol provides a direct record for vegetation and paleoenvironmental change on the Plateau. However, because it is difficult to extract sufficient pollen grains from loess, the pollen record since the middle Pleistocene especially in the central part of the Chinese Loess Plateau has not been well studied. So we preliminarily focus on the palynological records of the loess-paleosol sequence spanning the last 630 kyr at Luochuan and aim to understand the evolution of vegetation and climate change on the Chinese Loess Plateau. The main results and conclusions are as follows: 1. The palynological results show that the grassland has been a dominant vegetation in the Luochuan area since 630 kyr, even during the intervals of relatively warm and wet climatic conditions. 2. The pollen concentration of Luochuan section sharply decreases from the bottom of S1 to downward depth. This decrease can be attributed to depositional environment rather than climate change. In loess, not only oxidation, but also the PH of deposits and bacteria or fungi have been able to degrade sporopollenin. 3. The paleoclimatic condition during S4 stage, characterized with warmer condition during the early stage, was warmer and wetter than that during S5 in Luochuan area. Paleoclimate was warmer and wetter during the early stage of S5 and became colder and drier later. The special pedogenic features of S5-I can be attributed to a prolonged pedogenic duration rather than a warm-wet climate. 4. Evidence from pollen assemblage suggests that the Holocene vegetation has been affected by human impacts, especially after the Yangshao Culture. 5. The present steppe environment on the loess plateau is mainly due to natural conditions. Temperature, seasonal precipitation and soil structure are three important factors which control the vegetation type. 6. The vegetation on the loess plateau is characterized with zonal or azonal distribution. So local conditions should be taken into account when recover natural vegetation. Finally, the restoration and reconstruction of ecosystem on the loess plateau area should be focused on planting grassland rather than forests.

Direct methanol fuel cells (DMFCs) consisting of multi-layer electrodes provide higher performance than those with the traditional electrode. The new electrode structure includes a hydrophilic thin film and a traditional catalyst layer. A decal transfer method was used to apply the thin film to the Nafion(R) membrane. Results show that the performance of a cell with the hydrophilic thin film is obviously enhanced. A cell with the optimal thin film electrode structure operating at I M CH3OH, 2 atm oxygen and 90degreesC yields a current density of 100 mA/cm(2) at 0.53 V cell voltage. The peak power density is 120 mW/cm(2). The performance stability of a cell in a short-term life operation was also increased when the hydrophilic thin film was employed. Ti-substituted mesoporous SBA-15 (Ti-SBA-15) materials have been synthesized by using a new approach in which the hydrolysis of the silicon precursor (tetramethoxysilane, TMOS) is accelerated by fluoride. These materials were characterized by powder X-ray diffraction patterns (XRD), X-ray fluorescence spectroscopy (Y-RF), N-2 sorption isotherms, diffuse-reflectance UV-visible (UV-vis) and UV-Raman spectroscopy, Si-29 MAS NMR, and the catalytic epoxidation reaction of styrene. Experiments show that Ti-SBA-15 samples of high quality can be obtained under the following conditions: F/Si greater than or equal to 0.03 (molar ratio), pH less than or equal to 1.0, aging temperature less than or equal to 80 degreesC, and Ti/Si less than or equal to 0.01. It was found that the hydrolysis rate of TMOS was remarkably accelerated by fluoride, which was suggested to play the main role in the formation of Ti-SBA-15 materials of high quality. There is no stoichiometric incorporation of Ti, and the Ti contents that are obtained are quite low in the case of the approach that is proposed. The calcined Ti-SBA-15 materials show highly catalytic activity in the epoxidation of styrene.

Oxygen adsorption and desorption on a Pd(100) surface with a mesoscopic defect were studied by photoemission electron microscopy (PEEM). The defect surface, with an area of approximately 200 x 60 mu m(2), behaved differently from the perfect Pd(100) surface towards the adsorption of oxygen. When saturated, both surface oxygen and subsurface oxygen coexisted on the defect surface, whereas only surface oxygen was present on the Pd(100) surface. Upon heating, subsurface oxygen diffused back to the surface and desorbed with surface oxygen at the same time. The difference in oxygen adsorption ability between the defect surface and the perfect Pd(100) surface can be attributed to different structures of these two surfaces. (C) 1999 Elsevier Science B.V.
