研究了氯化钐对冷束缚应激大鼠血清激素的影响及其抗应激作用。用放射免疫法测定了血清GH、FSH、LH、TSH、T_3、T_4和皮质醇浓度,并用光镜和透射电镜观察了胃粘膜和脑垂体促肾上腺皮质激素细胞及肾上腺皮质束状带细胞超微结构的变化。结果表明,在应激反应中,氯化钐具有明显的提高血清GH和TSH值(p<0.01),显著降低血清FSH、T_4和皮质醇值(p<0.01),抑制促肾上腺皮质激素细胞分泌和保护胃粘膜的作用。

Selenium binding proteins (SeBP) represent a family of proteins that are believed to be involved in controlling the oxidation/reduction in many physiological processes. The cDNA of Zhikong Scallop Chlamys farreri selenium binding protein (zSeBP) was cloned by expressed sequence tag (EST) and RACE techniques. The high similarity of zSeBP deduced amino acid sequence with the SeBP in other organisms, such as bird, fish, frog, mosquito, fruit fly, mammalian, and even nematode and microorganism indicated that zSeBP should be a member of SeBP family. The temporal expression of zSeBP in the hemocytes was measured by semi-quantitative RT-PCR after scallops were stimulated by either oxidative stress or microbial challenge. The expression of zSeBP was up-regulated progressively after stimulation, and then dropped gradually to the original level. Meanwhile, malondialdehyde (MDA) measured by the colorimetric method in the microbial challenged scallops increased immediately after scallops was challenged by microbes, and was significantly higher than that in the control scallops. Results indicated that the microbial infection could incense the disorder of oxidation/reduction and may result in high MDA production. The negative correlation between the expression level of zSeBP and the MDA content suggested that zSeBP could play an important role in mediating the anti-oxidation mechanisms and immune response in marine invertebrates. The objective of the study is to investigate the suitability of using Pulse-coherent Acoustic Doppler Profiler (PCADP) to estimate suspended sediment concentration (SSC). The acoustic backscatter intensity was corrected for spreading and absorption loss, then calibrated with OBS and finally converted to SSC. The results show that there is a good correlation between SSC and backscatter intensity with R value of 0.74. The mean relative error is 22.4%. Then the time span of little particle size variation was also analyzed to exclude the influence of size variation. The correlation coefficient increased to 0.81 and the error decreased to 18.9%. Our results suggest that the PCADP can meet the requirement of other professional instruments to estimate SSC with the errors between 20% and 50%, and can satisfy the need of dynamics study of suspended particles.

应用稳定碳、氮同位素比值法和脂肪酸标志分析法,较为系统地研究了贝藻混养系统中滤食性贝类的食物来源,评估了大型藻类对混养系统及滤食性贝类的物质贡献。主要研究结果如下: 1.综述了典型生态系统中大型藻类和滤食性贝类各自的生态学地位和作用,大型藻类与滤食性贝类不仅在水体营养盐方面存在互利关系,二者在物质循环与收支方面同样具有耦合性,大型藻类提供的颗粒态有机质可以为滤食性贝类提供饵料来源。 2.总结了稳定同位素比值法和脂肪酸标志法在海洋生态系统食物来源及食物网分析中的应用,并建立了两种方法的具体操作规程。 3.分析了栉孔扇贝Chlamys farreri和海带Laminaria japonica混养系统中海带碎屑形成及释放不同阶段的生态学特征,评估了碎屑对扇贝的饵料贡献。海带在6周内释放了自身约27%的碳;碎屑形成及释放过程中C:N比值显著下降,同时伴随着旺盛的细菌降解,碎屑中也发现有大量硅藻类和原生动物存在。稳定同位素分析证实海带碎屑是混养期间扇贝的主要食物来源。 4.查明了春季胶州湾潮间带自然分布的长牡蛎Crassostrea gigas、紫贻贝Mytilus galloprovincialis和湾内浅海筏式养殖栉孔扇贝的可能食物来源。湾内栉孔扇贝饵料组成中浮游硅藻类为最主要部分,同时混杂有陆源有机质和细菌类物质;潮间带自然生长的牡蛎和贻贝饵料组成中,浮游植物占86.2-89.0%,种类组成中除硅藻外还包括一定比例的金藻和甲藻类;潮间带繁盛的孔石莼Ulva pertusa藻床为两种贝类提供了8.7-11.0%的补充食物来源。 5.揭示了桑沟湾贝藻混养海区春、夏季栉孔扇贝饵料来源组成情况及其季节变化,评估了海带养殖区碎屑碳量季节变化及海带来源碳对扇贝组织碳的贡献。结果表明,湾内贝藻混养区碎屑碳量为75.52-265.19 μg l-1,其在水体总颗粒态有机碳中的比例为25.6-73.8%。海带来源碎屑碳对栉孔扇贝组织碳的贡献比例为14.1-42.8%,且与水体碎屑碳比例的季节变化存在极显著相关性(F=0.992, P=0.004)。5月份湾外海带养殖区水体碎屑碳量为110.12-144.71 μg l-1,显著高于湾内无海带区(75.52 μg l-1),湾外养殖的扇贝组织中海带来源碳比例为22.0-24.1%,显著高于湾内单养区扇贝(9.6%)。估算结果表明,桑沟湾每年收获的6967吨(总湿重)栉孔扇贝中,海带提供了约57.1吨碳,换算为海带干物质为219.6吨。脂肪酸标志分析结果表明,2月份至8月份硅藻类在扇贝饵料组成中比例逐渐下降,而细菌类比例逐渐升高。整个采样期间,EPA/DHA比例较低,说明扇贝饵料组成中可能包括高DHA含量的组分。

The Bohai Sea was the site of the Chinese national GLOBEC programme. During the June 1997 cruises of R/V Science No.1, observations and experiments on zooplankton feeding were conducted. At five 48 h time-series stations the following observations and measurements on zooplankton were carried out: (1) diurnal vertical migration, by collecting samples at different layers every 3 h with a closing net; (2) diurnal feeding rhythms, by gut pigment analysis; and (3) ingestion rate, by both gut pigment analysis and the dilution method. A classification by body size was used to deal with the diversity of species and developmental stages of zooplankton assemblages. Samples were separated into three size groups: small (200-500 mu m), medium (500-1000 mu m) and large (> 1000 mu m). The results showed that the copepods (Calanus sinicus, Paracalanus parvus, Acartia bifilosa and Centropages mcmurrichi) performed clear diurnal vertical migrations. However, their behaviour was different at different stations. The variation in gut pigment content over the 24 h cycle showed strong diurnal feeding rhythms, particularly for the large size group. Gut pigment contents reached their daily maximum during the time from dusk to midnight (18:00-24:00). The peak value was about 10 times the minimum observed in the daytime. The in situ daily grazing rate, based on gut pigment contents and evacuation experiments, was 4.00-12.65 ng chla ind(-1) day(-1) for the small size group, 5.99-66.58 ng chla ind(-1) day(-1) for the medium size group and 31.31-237.13 ng chla ind(-1) day(-1) for the large size group. The copepods consumed only a small part (2.90-13.52%) of the phytoplankton biomass hut about 77% of the daily production. The grazing mortality rate of phytoplankton by microzooplankton (<200 mu m) measured by the dilution method ranged from 0.43 to 0.69 day(-1) The calculated daily consumption of phytoplankton biomass was 35-50%, and 85-319% of the potential production.

以青海省达日县高寒草甸原生高寒嵩草(Kobresia)草甸封育系统为对照,研究了土地退化对植被生产力的影响,检验了不同人工重建措施(两个人工种植处理:混播(HB)、翻耕单播(DBF)和1个退化草地封育自然恢复处理(NR)及1个退化草地自然状态(SDL))对植被生产力的相对影响程度。结果表明,原生植被封育处理(YF)地上总生物量为265.1g•m~(-2),混播(HB)和翻耕单播(DBF)处理中地上总生物量分别为原生植被封育处理的116%和68%。退化草地封育自然恢复处理(NR)和重度退化自然状态下地上总生物量分别为原生植被封育的76%和53%。YF处理根系生物量远大于其它处理。原生植被封育系统中植被地上部分碳储量为110.14g•m~(-2),地下根系(0~30cm)碳储量为2957g•m~(-2),植被总碳储量为3067.14g•m~(-2);重度退化草地系统中植被地上部分碳储量为57.07g•m~(-2),地下根系(0~30cm)碳储量为357g•m~(-2),植被总碳储量为414.07g•m~(-2)。由此可见,高寒草甸严重退化后,通过植物组织流失的碳达到2653.35g•m~(-2),即86.5%的碳损失;原生植被封育系统植被总氮储量为56.85g•m~(-2),而重度退化草地植被总氮储量为18.02g•m~(-2),高寒草甸严重退化使植物组织68.30%氮损失。与重度退化地相比,由于恢复重建措施增加了植物的生物量输入和群落组成,除翻耕单播处理外,其它恢复重建措施均能恢复系统植被的碳氮储量。这些恢复重建措施将会逐步改善土壤的物理和化学特性,最终使这些生态系统逐步由碳源向碳汇方向的转变成为可能。

茶卡-共和盆地及其毗邻地区位于青海省的东部,约处于北纬34°45'~37°00',东经98°45'~101°30'.海拔2 800~5 305 m,面积约38 300 km2,属于高原大陆性气候类型.本区共有野生种子植物59科、277属、854种.分别占青海省总科数的65.56%、总属数的54.53%、总种数的37.37%.区系特征如下:1、种类相对较贫乏,木本更贫乏.2、就属的层面而言,本区属于以北温带成分,特别是欧亚大陆温、寒地带典型成分为优势的,兼具旱生温性、耐寒温性和高寒类型的温带区系性质.3、许多种、属和热带类型均以本区为边缘分布区,因而区系的边缘性质明显.4、为青藏高原高寒区系、黄土高原温性(草原)植物区系和亚洲东部荒漠区系在我国的交汇过渡区,区系的过渡性质明显.5、是青藏高原植物亚区唐古特地区的一部分.6、是青海植物区系或"唐古特植物地区"划分植物亚地区的关键区域.

对青藏高原海北站区的自然土壤和扰动土壤进行高分辨率采样,测定土壤根系、有机碳及其~(14)C含量;用~(14)C示踪技术探讨土地利用变化对高寒草甸土壤有机质更新的影响.研究表明,土地利用变化对高寒草甸土壤碳循环影响显著. 耕作活动导致扰动土壤有机碳储量比自然土壤增加29.35%;扰动土壤剖面10~50 cm深土壤有机质的14C含量相对富集;自然土壤大多数有机碳储存在土壤表层,更新时间<50 a,同一深度扰动土壤有机碳储量显著少,更新时间长(171~294 a);自然土壤10 cm以下有机碳主要为更新时间>1 000 a的稳定碳所控制,扰动土壤的相应值出现在40 cm以下;自然土壤有机质更新产生的CO2通量为114 gC•m~(-2)•a~(-1),扰动土壤为48.7 gC•m~(-2)•a~(-1).

利用5-9月植物生长期间所观测的不同植被盖度条件下土壤温、湿度资料,分析了矮嵩草草甸植被盖度对土壤温、湿度的影响.结果表明:有植被覆盖区随季节进程,植被盖度增加,土壤温度升高,湿度下降;8:00-19:00土壤温度随植被盖度的增加而增加,14:00土壤温度随植被盖度增加而降低;不论是早晚还是中午,土壤湿度都随植被盖度的增加而增加.

通过对高寒草甸10 种雀形目鸟类繁殖生产力研究表明: 高寒草甸鸟类的繁殖生产力与鸟类栖息地所处的地理环境、营巢类型和繁殖方式密切相关, 其中繁殖季节长度和窝卵数是影响高寒草甸鸟类繁殖生产力的两个最重要的因子( P < 0.05) 。鸟类平均繁殖生产力为2.80(只/ 对·年)。

本文应用限制性内切酶分析藏鼠兔和高原鼠兔的线粒体DNA限制性片段长度多态性(mtDNARFLP),并结合形态、生境、地史和化石资料综合探讨两物种的异同。结果表明,形态上差异显著的两个物种,在分子水平上也同样具有明显的区别。用16种酶分别消化各样品,共检出30种限制性类型,两物种间存在丰富的多态性;它们的mtDNA分子均为17.9kb(千碱基对)左右;平均遗传距离为0.0455,分歧时间约为距今二百三十万年前的上新世晚期,相当于我国哺乳动物时代的榆社期晚期或泥河湾期早期。

Based on the 16S mitochondrial partial gene sequences of 29 genera, containing 26 from Oedipodidae and one each from Tanaoceridae, Pyrgomorphidae and Tetrigidae (as outgroups), the homologus sequences were compared and phylogenetic analyses were performed. A phylogenetic tree was inferred by neighbor-joining (NJ). The results of sequences compared show that: (i) in a total of 574 bp of Oedipodidae, the number of substituted nucleotides was 265 bp and the average percentages of T, C, A and G were 38.3%, 11.4%, 31.8% and 18.5%, respectively, and the content of A+T (70.1%) was distinctly richer than that of C+G (29.9%); and (ii) the average nucleotide divergence of 16S rDNA sequences among genera of Oedipodidae were 9.0%, among families of Acridoidea were 17.0%, and between superfamilies (Tetrigoidea and Acridoidea) were 23.9%, respectively. The phylogenetic tree indicated: (i) the Oedipodidae was a monophyletic group, which suggested that the taxonomic status of this family was confirmed; (ii) the genus Heteropternis separated from the other Oedipodids first and had another unique sound-producing structure in morphology, which is the type-genus of subfamily Heteropterninae; and (iii) the relative intergeneric relationship within the same continent was closer than that of different continents, and between the Eurasian genera and the African genera, was closer than that between Eurasians and Americans.

Prolonged floral longevity and bumblebees as dominate pollinators in alpine ecosystem have been suggested to overcome pollination limitation of alpine plants arising from the decrease of pollinator activity with increasing altitude. However, this conclusion has never been examined in the Qinghai-Tibetan Plateau (QTP), the highest and largest plateau in the world. In this study, we intended to test year-to-year correlations between floral longevity, visiting frequency and pollen limitation of this species between two populations (at 3200 m and 4000 m) of Gentiana straminea in this plateau. Pollinator exclusion elongated both male and female phases greatly at both sites, and durations of both male and female phases in natural condition varied greatly over three years. The visiting frequency of bumblebees varied greatly at the higher altitude, but seemed to be stable at the lower altitude. Seed production was pollination limited in both populations in most studied years. The floral durations, pollinator frequency and pollination limitation showed no significant and consistent variations with the increase of altitude. The previous hypothesis that the prolonged floral longevity of alpine plants can compensate for low levels of pollinator visitation therefore could not be confirmed, and our results further suggested that in the QTP platform, the altitude shows no consistent effect on the reproductive performance of this species, despite that the fluctuation of visit frequency intensified at the higher altitude.

A novel, simple and accurate fingerprint method was developed using high-performance liquid chromatography-photodiode array detection (HPLC-DAD) for the quality control of Qianghuo, a Tibetan folk and Chinese herbal medicine used as a diaphoretic, an antifebrile and an anodyne. For the first time, the feasibility and advantages of employing chromatographic fingerprint were investigated and demonstrated for the evaluation of Qianghuo by systematically comparing chromatograms of aqueous extracts with the professional analytical software recommended by State Food and Drug Administration (SFDA). Our results revealed that the chromatographic fingerprint combing similarity evaluation could efficiently identify and distinguish raw herbs of Qianghno from different sources and different species. The effects on Notopterygium forbesii Boiss (Apiaceae) chromatographic fingerprints resulted from collecting locations, harvesting time were also examined. (c) 2006 Elsevier lrelanc Ltd.
