采用样方法对云雾山干草原区本氏针茅草地群落进行长期定位监测,对未封育样地和封育5,10,15,20和25年样地群落的平均高度、盖度、多度和地上生物量以及植物多样性进行实地调查分析,以研究围栏封育对草地植物群落特征动态变化的影响。结果表明,围封10年样地群落的高度、多度以及地上生物量最大,而群落的盖度则随封育年限的增加呈显著增加趋势,在封育25年达到最大;围栏封育的不同年限也显著影响了群落的物种多样性,在围封10年样地群落具有最高的多样性指数,围封15年样地具有最高的丰富度指数,而均匀度指数则是在未封育群落中表现为最高值,最低值是在物种丰富度最高时出现。由此可见,围栏封育在一定时间范围内可以显著改善群落的特征,增加群落的生产力,对于本研究地本氏针茅群落的最佳封育期限为10~15年,但围封时间过长在草地退化方面有一定程度的影响。

The optimum growth condition of GaInNAs/GaAs quantum wells (QWs) by plasma-assisted molecular beam epitaxy was investigated. High-resolution X-ray diffraction and photoluminescence (PL) measurements showed that ion damage drastically degraded the quality of GaNAs and GaInNAs QWs and that ion removal magnets can effectively remove the excess ion damage. Remarkable improvement of PL intensity and obvious appearance of pendellosung fringes were observed by removing the N ions produced in the plasma cell. When the growth rate increased from 0.73 to 1.2 ML/s, the optimum growth temperature was raised from 460 degreesC to 480 degreesC and PL peak intensity increased two times. Although the N composition decreased with increasing growth rate, degradation of optical properties of GaInNAs QWs was observed when the growth rate was over 0.92 ML/s. Due to low-temperature growth of GaInNAs QWs, a distinctive reflection high-energy electron diffraction pattern was observed only when the GaAs barrier was grown under lower As-4 pressure. The samples with GaAs barriers grown under lower As-4 pressure (V/III ratio about 24) exhibited seven times increase in PL peak intensity compared with those grown under higher As-4 pressure (V/III ratio about 50). We report all optical clock recovery based on a monolithic integrated four-section amplified feedback semiconductor laser (AFL), with the different sections integrated based on the quantum well intermixing (QWI) technique. The beat frequency of an AFL is continuously tunable in the range of 19.8-26.3 GHz with an extinction ratio above 8 dB, and the 3-dB linewidth is close to 3 MHz. All-optical clock recovery for 20 Gb/s was demonstrated experimentally using the AFL, with a time jitter of 123.9 fs. Degraded signal clock recovery was also successfully demonstrated using both the dispersion and polarization mode dispersion (PMD) degraded signals separately.

半干旱草原是陆地生态系统的主要类型,对社会经济的可持续发展至关重要,已有研究表明农田扩张是半干旱草原面积缩减的主要原因之一。村庄作为基层单元,既是大尺度要素作用的承受者又是土地利用变化的直接执行者,其社会、经济、文化和自然资源特征对土地利用方式往往起决定作用,然而,尚未见从村庄层面上探讨中国半干旱草原牧区农田扩张机制的研究报道。村庄是一个内部有序的等级体系,其中所存在的数据自相关既能造成分析结果的不准确性又能用于土地利用变化预测。因此,本论文以位于科尔沁草原50多年经历了大面积草地开垦的乌兰敖都村为研究对象,以多层线性模型、空间插值技术和贝叶斯推理为分析手段,通过引入田块、单户和村庄3个尺度, 综合考虑自然和社会经济要素,揭示了半干旱草原牧区村庄的农田扩张机制,提出了处理土地利用变化研究中的数据自相关的方法,为半干旱草原的有效保护和管理提供了科学依据。 本研究表明: 1)草地开垦主要发生在甸子地,所有住户都倾向于在优质土地上开荒种粮,而且劳动力数量多、文化程度高和收入更大程度上依赖于农牧业生产的住户更易开荒种粮,但当牲畜数量较多时,住户开荒种地的倾向性降低,这与农牧交错带典型偏农区的研究结果相反。所开土地的90%用于种植玉米和水稻,其中玉米耕种面积主要受价格因素影响,而水稻的种植面积主要受土地条件影响。 2)离居民点近的草地比离居民点远的草地更易被开垦为农田,打草场比放牧场更易被开垦为农田。 3)多层线性模型将驱动因子置于不同尺度,通过引入随机方差成分避免统计谬误,因此可用于处理驱动力研究中的数据自相关。空间自相关不仅可用于预测土地利用变化趋势,也为深入了解土地利用变化过程提供了新视角。 4)在中小学中加强草地保护教育、在牧民中开展草地保护宣传、转移剩余劳动力、制定强有力措施保护居民点周围的草场是控制开荒种粮的有效措施。

西南地区在我国的经济发展和生态环境建设中占重要地位,但也是我国生态环境最脆弱的地区之一,生态系统退化,生态功能减弱,严重制约着西南林业的可持续经营与发展。本项目采用DNA 分子标记SSR 研究不同生境条件下粗枝云杉群体的遗传变异及其时空分布格局,考察遗传变异与复杂的山地生态环境间的潜在联系,系统地揭示粗枝云杉天然群体与环境系统相互作用的生态适应与分子进化机制。粗枝云杉适应性强,生长迅速,在植树造林和工业用材方面占有重要地位,研究成果可为中国西南部亚高山天然林的可持续经营及退化生态系统的恢复与重建提供理论依据和科学指导。主要研究结果如下: 1. SSR 位点变异丰富,等位基因频率的分布格局多样。7 个SSR 标记全是多态位点,每位点的等位基因数变化范围为13~24,平均为19.9 个。SSR 位点的等位基因片段长度范围变化较大。73.1%的等位基因变异遵循逐步突变模型(SSM)而发生1 个重复基元的变化,22.3%和4.6%的变异分别按两阶段突变模型(TMP)发生1 个重复基元以上的变化和在SSR 位点侧翼区发生1 个碱基变化的插入-删除事件。 2. 粗枝云杉拥有中等偏高水平的遗传多样性和相对大的群体间遗传分化。通过分析代表10 个群体的250 个个体在7 个SSR位点的变化,调查了源自中国西南山区的粗枝云杉的微卫星变异。相当高的遗传多样性和强烈的群体分化发生在粗枝云杉中, 其群体平均Nei's 期望杂合度为0.707 , 群体间遗传距离为0.121~0.224(FST)和0.100~0.537(RST)。然而,群体间遗传距离与地理距离之间无相关性,从而排除了简单的距离分离模式并暗示迁移不是影响粗枝云杉遗传变异格局的主要因素。事实上,使用私有等位基因估算的基因流数量非常低,仅等于0.753。等位基因置换检验(Allele permutation tests)揭示逐步突变及遗传漂变都对群体间分化有贡献。另外,在多数位点检测到显著的群体间遗传差异,这个结果说明自然选择,假设通过环境压力,是引起粗枝云杉微地理分化的主要因素之一。根据SSR基因型,250 个粗枝云杉个体的70%被正确地归类入其各自的来源群体,结果表明微卫星(SSR)对区分来自中国不同生态地理位点的粗枝云杉基因型是有效的。 3. 在SSR、RAPD 和AFLP 位点,显著的群体间遗传结构被发现的,但三种标记间遗传分化程度和群体遗传关系有差异。利用来自10 个群体的247 个个体,我们报告了关于样本粗枝云杉群体间遗传关系的总体看法。根据各自对评价遗传关系的信息能力和适用性,SSR、RAPD 和AFLP 标记被选用,三种技术非常有效地区别这些基因型。使用的SSR、RAPD 和AFLP 标记分别估计平均Dice 相似性系数。Mantel 检验产生显著但相对低的共表型适合度(RAPD = 0.63£AFLP = 0.60和SSR = 0.75)。比较三种标记系统,RAPD 和AFLP 共表型指数相对高地相关(r =0.59),而RAPD 和SSR 及SSR 和AFLP 之间的相关系数分别是0.53 和0.35。所有系统树,包括不同标记资料结合获得的系统树,反映了多数群体依据它们的地理条件而成某种特定关系。结果暗示单个或结合标记系统能用来深入洞察粗枝云杉遗传研究,并且不同标记系统合并资料能提供更可靠的信息。 Southwestern region plays an important role in economic developmentand ecological construction in China. Yet, it is also one of the weak regionsof ecological environment in China with degraded ecosystem and imperfectfunction, which restricts the sustaining management and development ofsouthwestern forestry. The genetic variation and spatial distribution patternof P. asperata populations originating from different habitats wereinvestigated using SSR molecular markers in this study. The correlationsbetween genetic variation and ecological and environmental conditionswere detected, and the interaction between P. asperata populations andenvironmental system and the mechanism of ecological adaption -molecular evolution were revealed. Given the significant ecological andeconomic roles of the fast-growing and wide-adaptive species in reforestation and production of pulp wood and timber, the study couldprovide a strong theoretical evidence and scientific direction for thesustaining management of subalpine natural forest, and the afforestationand rehabilitation of degraded ecosystem. The results are as follows: 1. The genetic variation at SSR loci was abundant and the distributionof allelic frequencies was uneven. All seven loci were polymorphic, and thenumber of alleles per locus varied from 13 to 24 with a mean valueequaling 19.9. The allele sizes at SSR loci were found to vary widely.73.1% of allelic variation followed stepwise mutation model (SSM) whichresults increase or decrease by one repeat type, and 22.3% and 4.6% wereresulted from two-phase mutation model (TMP) with allele size varying bymore than one repeat type and from insertion-deletion events in theflanking regions at SSR loci with a single basepair changing, respectively. 2. P. asperata possessed a moderate to high level of genetic diversityand considerable genetic differentiation. Microsatellite variation of P.asperata. originating from the mountains of southwestern China wasinvestigated by analyzing variation at seven SSR loci in 250 individualsrepresenting ten populations. A fair degree of genetic diversity and strongpopulation subdivision occurred with the mean gene diversity (H) of 0.707,and genetic distances among populations varying between 0.121 and 0.224(FST) and between 0.100 and 0.537 (RST). However, inter-populationgenetic distances showed no correlation with geographic distances between the population sites. This ruled out a simple isolation by distance modeland suggested that migration does not have a great impact. In fact, theamount of gene flow, detected using private alleles, was very low, equalingonly 0.753. Allele permutation tests revealed that stepwise-like mutations,coupled with genetic drift, could contribute to population differentiation.Moreover, significant genetic differences between populations weredetected at most loci. The results indicate that natural selection, presumablythrough environmental stress, may be one of the main factors causingmicro-geographical differentiation in the genetic structure of P. asperata.Based on SSR genotypes, 70% of the 250 individuals were correctlyclassified into their sites of origin. This suggests that microsatellites (SSRs) are effective in distinguishing genotypes of P. asperata originating fromdiverse eco-geographical sites in China. 3. Using a set of 247 individuals from ten P. asperata populations wereport an overview on the genetic relationship among the sampled P.asperata populations. RAPD, AFLP and SSR were used in terms of theirinformativeness and applicability for evaluate relationship and all threetechniques discriminated the genotypes very effectively. Mean Dicesimilarities coefficient were estimated using RAPD, AFLP and SSR,respectively. The Mantel test resulted in a significant but relatively low fit(RAPD = 0.63, AFLP = 0.60 and SSR = 0.75) of cophenetic values.Comparing the three marker systems to each other, RAPD and AFLP cophenetic indices were highly correlated (r = 0.59), while correlationcoefficient between RAPD and SSR was r = 0.53 and between SSR andAFLP was r = 0.35. For all markers a relatively high similarity indendrogram topologies was obtained although some differences wereobserved. All the dendrograms, including that obtained by the combineduse of all the marker data, reflect some relationships for most of thepopulations according to their geographic conditions. The results indicatethat single or combined marker system could be used to insight into geneticstudy in P. asperata and the combined data of different marker systems canprovide more reliable information.

青藏高原东南缘由于特殊的生态地理条件,有着丰富的森林资源,这些资源是长江上游涵养水源、保持水土的生态屏障,是生物多样性的资源宝库。但随着过量的森林采伐,使该区曾经丰富的生物多样性资源遭受了前所未有的破坏,天然林的质量严重下降,生态系统退化,功能减弱。与此同时,许多物种的种群规模正在锐减,物种的遗传多样性也严重丧失。川西云杉是西部地区分布最广的云杉树种之一,在较高海拔的地区有着重要的生态学功能,是一种适应性很强的乡土树种。本项目采用简单序列重复标记(SSR)和特定序列位点(STS)研究不同生境条件下川西云杉群体的遗传变异及其时空分布格局,考察遗传变异与复杂的山地生态环境间的潜在联系,系统地揭示川西云杉天然群体与环境系统相互作用的生态适应与分子进化机制。研究成果能有效地为该树种遗传资源的科学保护与合理利用提供理论依据和科学指导,可为中国西南部亚高山天然林的可持续经营及退化生态系统的恢复与重建提供依据。主要研究结果如下: 1 STS和SSR两种分子标记的研究结果表明:川西云杉群体拥有中等水平的遗传多样性(基于SSR标记,平均He = 0.640;基于STS标记,平均He = 0.553)。造成这种中等水平的遗传多样性,可能是由于历史原因,川西云杉天然林被大量采伐,导致了遗传多样性的丧失。两种方法都检测出群体BT有着最高水平的遗传多样性。 2 两种标记的结果都一致说明:检测的10个川西云杉群体间遗传分化比较高,其存在群体间的遗传变异比例要明显地高于广泛分布的挪威云杉、横贯大陆的黑云杉以及兼有连续分布和不连续分布的西加云杉等,但要低于分布范围狭窄且呈不连续分布的粗枝云杉。青藏高原东南部的片段化生境可能是导致高水平遗传分化的主要原因。 3 基于两种标记的UPGMA 聚类和PCA分析结果,以及基于SSR标记的FCA分析结果都显示:群体BY遗传上明显区别于其它群体,其可能原因是青藏高原东南缘山脉阻隔而导致的生殖隔离的结果。 4 根据软件Bottleneck 1.2.02的检测以及不依赖哈迪一温伯格平衡的M比率检测结果:川西云杉种群很有可能经历了近期的遗传瓶颈效应。在本研究中遗传瓶颈效应并没有显著影响到物种的遗传多样性。然而,在这些片断化群体未来的子代群体中很可能出现遗传瓶颈导致的遗传多样性下降的效应。 Due to the extremely complex topography and climatic conditions, the southeast of the Qinghai-Tibet Plateau is region abundant in forest resource, which is benefit to the upper reaches of the Yangtze River water conservation, and protecting natural environment and biodiversity resources. However, by the reason of a plenty of trees were cut in these yeas, the biodiversity resource was destroy with degraded ecosystem and imperfect funcation. Picea balfouriana is one of the regionally distributed conifer species in the southeast of the Qinghai-Tibet Plateau and considered as a constructive species within its distribution area. It is an optimal species for the production of biomass. Moreover, it is well adapted to stressful environments at high altitude, especially to cold and drought conditions which are generally harsh for other trees. In our study, two types of molecular markers (SSR and STS) were used to estimate the genetic diversity and genetic structure of P. balfouriana populations originating from the southeast of the Qinghai-Tibet Plateau with varying climatic and geographical conditions. The results will not only provide a deep insight into its genetic diversity and population genetic structure but also valuable information for further management and breeding programs in P. balfouriana. In summary, the results obtained in this study revealed that: 1 A moderate degree of genetic variation is present in P. balfouriana in the southeast of the Qinghai-Tibet Plateau and it may caused by many trees were cut in the past. Population BT owns the highest level of genetic diversity by both two types of markers. 2 Considerable population differentiation exists among the ten P. balfouriana populations based on both SSR and STS markers, possibly caused by habitat fragmentation and heterogeneous environments. 3 The UPGMA clustering and PCA analyses based on SSR and STS marker, and FCA analyses based on SSR marker congruously separated population BY from other populations, which is likely due to the presence of mountain barriers. 4 The results of bottleneck analysis indicated that P. balfouriana populations that have undergone recent declines. In our research, the bottleneck effect don’t have a significant impact on the genetic diversity of species, however, the level of genetic diversity of P. balfouriana offspring populations growing in the fragmented habitats will decline in the future.

日益增强的旅游活动干扰已成为九寨沟世界自然遗产的有效保护与持续管理不可回避的问题,也已成为当前区域生态保护与经济发展的焦点问题之一。评估乡土植物种的基本特征、质量、种子休眠程度以及萌发能力;筛选适宜的植被恢复乡土物种并选择有效植被恢复技术措施是开展植被恢复试验的重要前提和保障。本文以九寨沟世界自然遗产为例,通过对景区乡土植物种子基本特征的测定以及采用乡土物种作为植被恢复技术试验与恢复技术筛选的试验物种,在景区旅游干扰区的三种生境类型:原始针叶林、混交林和边坡,根据不同的影响因素设计试验,从而探索适宜的恢复措施和植被恢复物种。初步结论如下: 1. 本文研究的28种乡土植物中,17种测定的物种活力较高(>72%),均具有作为恢复物种的潜力。但28种种子中75%的发芽率为0%。大部分种子存在一定程度的休眠,包括甘肃山楂、称花藤、疏花槭、陇东海棠、球花荚迷、巴东小檗、蔷薇、宝兴栒子、光泽栒子、光枝柳叶忍冬、甘青铁线莲、毛果铁线莲、宽叶旌节花、糙叶五加和独活。因而种子存在深度休眠是乡土物种自然更新和植被恢复的主要障碍,需要根据种子的形态生理特征,寻求打破乡土植物种子休眠的有效方法,提高种子发芽率。 2.低温层积只显著提高了蔷薇、毛果铁线莲、宽叶旌节花和假升麻3种种子的发芽率,也相对较早地提高了陇东海棠、鲜黄小檗和独活的萌发。对于其它物种的种子,低温层积没能有效打破休眠提高发芽率。综合分析结果表明九寨沟28种林下或林缘乡土植物中窄叶鲜卑花、毛果铁线莲、宽叶旌节花、假升麻、陇东海棠、鲜黄小檗和独活等12种可直播或通过低温层积处理后直播用于旅游干扰退化地段的植被恢复中,而其它的种类尚需进一步寻求打破休眠的措施以提高种子发芽能力。 3.在原始林生境下,七筋菇、巴东小檗、耳叶风毛菊、长叶毛花忍冬及独活5种乡土植物种子的出苗能力不高,因此目前急需提出有关提高种子出苗的方法措施。在相同的处理条件下,不同物种出苗数存在着较大的差异,相比而言,巴东小檗、耳叶风毛菊和长叶毛花忍冬种子出苗与幼苗存活较高,对于其环境适应能力较强,因此它们更适宜采用播种的方式应用于九寨沟原始林践踏地段地表的植被恢复实践;而七筋菇和独活在室内和自然气候条件下萌发都很困难,在植被恢复过程中,可考虑采用其它播种方式。而在边坡和混交林生境试验结果表明,供试物种无萌发和出苗现象,这可能跟外界环境条件、干扰和种子命运有关。 4. 本研究中腐殖质、松土和苔藓覆盖处理措施均未能提高物种出苗,且对样方内植被结构以及物种丰富度也无显著影响,其中的影响因素也是多方面的,因此还需进一步研究探索。但腐殖质对幼苗存活能力方面有一定的积极作用。 The increasing tourism disturbance is an unavoidable issue to effective conservation and sustaind tourist management of Jiuzhaigou Nature Reserve. It has become one of the focal problems of regional ecological protect and economic development. Assessing the traits, quality, seed dormancy and germination of native seed and choosing the suitable species and ways for vegetation restoration were the important precondition and guarantee before vegetation restoration. In this paper, as an example to Jiuzhaigou world nature heritage, through the estimation to the basic characteristics of native seeds and the use of native species for the test of vegetation restoration and the choosing of vegetation restoration techniques, in three different habitats being disturbed, including the primitive forest, mixed forest and side slope. According to different factors, designing experiments in order to explore the appropriate measures and species of vegetation restoration . Our results are as follows: 1. There are 77% seeds of the 28 species with high viability(>72%), which had the potential possibility of being restoration species. The germination of 75% of the seed was 0%.Most of the seed was dormant, So dormancy of seed was the barrier for natural regeneration and vegetation restoration, Further research is needed on investigating the other more effective methods to release seed dormancy of these native species in the future. 2. Cold stratification did not release seed dormancy of most species. In our study, there are only 3 species including Clematis peterae var. trichocarpa, Stachyurus chinensis and Aruncus sylvester (P<0.05) with germination percentage increased by cold stratification. By comprehensive analysis, the result shows: In 28 undergrowth native seeds, Sibiraea angustata, Clematis peterae var. trichocarpa, Stachyurus chinensis, Aruncus Sylvester, Malus kansuensis, Berberis diaphana,and Heracleum hemsleyanum could be used to vegetation restoration by sowing directly or sowing after cold stratification in degenerate gland being disturbed by turism, and othes seed of 28 were needed to search other methods to release seed dormancy for advancing the ability of germination. 3.In the habitat of primitive forest, the ability of emergence of the native species Clintonia udensis, Berberis henryana, Saussurea neofranchetii, Lonicera trichosantha var.xerocalyx and Heracleum hemsleyanum was low, so it is urgently needed to search about the methods of advancing emergence. In the sametreatment, the number of emergence of different species was very different. Relatively speaking, the emergence and seedling survival of the species Berberis henryana, Saussurea neofranchetii and Lonicera trichosantha var.xerocalyx was higher, and the adaptability of the species was better, so they were more suitable for vegetation restoration on trampling-induced degraded patches of primitive forest in Jiuzhaigou; But the germination of Clintonia udensis and Heracleum hemsleyanum was both difficult in laboratory and natural conditions, so it could be considered to apply other sowing methods. However, the result showed that: applied species didn’t germinate or emergent in the habitat of side slope and mixed forest, which could be related to external environment conditions, interference and the fate of the seed. 4.In this study, humus, scatification and bryophyte cover measures didn’t advance the emergence of the species or the structure of vegetation and richness in plot significantly, the impact factors of which were many, therefore further research is needed . However, humus had a certain positive effect on the ability of seedling survival.

木质纤维素原料种类多、分布广、数量巨大,通过燃料乙醇生产技术、厌氧沼气发酵技术将其转化成乙醇、沼气等二次能源,一定程度上可以缓解化石能源的不断消耗所带来的能源危机,也解决了农林废弃物引起的环境污染问题。其中以木质纤维素原料生产燃料乙醇,还可以避免以淀粉类和糖类原料生产燃料乙醇时带来的“与人争粮”等一系列问题。因此具有重要的经济效益、环境效益和社会效益。 然而,木质纤维素原料结构致密,木质素包裹在纤维素、半纤维素外围,导致其很难被降解利用,必须进行适当的预处理,去除木质素,打破原有的致密结构,利于原料的后续利用。因此,预处理成为木质纤维素原料能源化利用的关键。而目前预处理环节的费用过于昂贵,于是寻找一种高效、低成本的预处理方法是当今研究的热点。 本论文采用组合白腐真菌对木质纤维素原料进行生物预处理研究,与其他物理化学法相比,该法有着专一性较强、反应温和、不造成环境污染、成本低等优势。白腐真菌主要通过分泌木质素降解酶对木质素进行降解,从而破坏原料的致密结构,提高后续利用效率。所以木质素降解酶酶活的高低是影响原料预处理效果的一个关键因素。于是本论文首先通过将白腐真菌进行组合的方式提高木质素降解酶(漆酶,Lac)酶活;接着对组合菌的菌株相互作用机理进行研究,阐明组合菌Lac 酶活提高的原因,为菌株组合提高Lac 酶活这种方法的应用提供理论依据,同时也为后续组合白腐真菌预处理木质纤维素原料提供指导;进一步采用固态发酵和木质素降解酶两种方式对木质纤维素原料进行预处理研究,最大化去除木质素成分,破坏原料的致密结构;最终对预处理后原料的酶解糖化进行初步研究,为原料后续的能源化应用奠定基础。具体研究结果如下: (1) 以实验室保存的三株主要分泌Lac 的白腐真菌为出发菌株,筛选得到一组Lac 酶活明显提高的组合菌55+m-6,其中菌株55 为Trametes trogii sp.,m-6 为Trametes versicolor sp.,组合后Lac 酶活较单菌株分别提高24.13倍和4.07 倍。组合菌的最适产酶条件为pH 6.5、C/N 16:1、Tween 80 添加量为0.01%,在该条件下组合菌的Lac 酶活峰值比未优化时提高4.11倍。 (2) 对组合菌55+m-6 菌株间相互作用机理进行研究,发现菌株之间不存在抑制作用;平板培养时,菌丝交界处Lac 酶活最高并分泌棕色色素;液体培养时,菌株m-6 对组合后Lac 酶活的提高起着更为重要的作用:菌株m-6的菌块、过滤灭菌胞外物以及高温灭菌胞外物均能明显刺激菌株55 的Lac产生;菌株55、m-6 进行组合后,同工酶种类未发生增减,但有三种Lac同工酶浓度有所提高;对菌株胞外物进行薄层层析和质谱分析,结果表明组合前后菌株胞外物中各物质在浓度上存在较大的变化。推测组合菌Lac酶活的明显提高,主要是由于菌株m-6 胞外物中的一些物质能刺激菌株55 分泌大量Lac 进行代谢,且这些刺激物质并非菌株m-6 特有,菌株55自身也可以代谢生成,但是适当的浓度才能刺激Lac 的大量分泌。 (3) 将组合菌55+m-6 用于固态发酵预处理木质纤维素原料,发现其对玉米秆的降解程度最大,在粉碎度40 目、含水率65%的最优处理条件下,处理至第15d,秸秆失重率为41.24%,其中木质素、纤维素、半纤维素均有降解,且Lac 和纤维素酶(CMC)酶活以及还原糖量均达到峰值。 (4) 对玉米秆进行木质素降解酶预处理,发现Lac/1-羟基苯并三唑(HBT)系统对玉米秆木质素的降解效果最好,在最优处理条件时,即HBT 用量0.2%、处理时间1d、Lac 用量50U/g,木质素降解率可达12.60%。预处理后玉米秆的致密结构被破坏,比表面积增大,利于后续酶与纤维素、半纤维素成分的结合。 (5) 对预处理后的玉米秆进行酶解糖化,其中组合菌固态发酵预处理后玉米秆的糖化率比对照高4.33 倍;Lac/HBT 系统预处理后玉米秆的糖化率比对照高2.99%,糖化液中主要含有木糖、葡萄糖两种单糖。 There are many kinds and large quantities of lignocellulosic biomass widely distributed on the earth. They can be converted into secondary energy such as fuel ethanol, biogas, et al., which can relieve the energy crisis caused by consumption of fossil energy resources and solve the problem of environmental pollution caused by agriculture and forestry waste. Meanwhile, the production of fuel ethanol from lignocellulosic biomass can ensure food supply to human kind instead of starch- and sugar-containing raw materials. So the energy conversion of lignocellulosic biomass contributes considerable economic, environment and social benefits. However, lignocellulosic biomass has the compact structure, in which lignin surrounds cellulose and hemicellulose, so it must be pretreated before energy usage and pretreatment is one of the most critical steps in the energy conversion of lignocellulosic biomass. At present, the cost of pretreatment is too expensive, so looking for an efficient and low-cost pre-treatment method is one of recent research hot spots. In this research, combined white rot fungi pretreatment method was used, which had some advantages in low cost, high specificity, mild reacting conditions and friendly environmental effects compared with the other physical and chemical methods. White rot fungi secrete lignin degrading enzymes to degrade the content of lignin and damage the contact structure of lignocellulosic biomass, so the activity of the lignin degrading enzymes is the key factor to the degradation effect of raw materials. Firstly, the combined fungi with high laccase activity were screened; secondly, the interaction mechanism between strains was studied, and the cause of higher laccase activity after strains combination was also preliminary clarified; under the guidance of the mechanism, lignocellulosic biomass was pretreated by the combined fungi; lastly, the enzymatic hydrolysis of pretreated lignocellulosic biomass was also preliminary studied; all of the researches could lay the foundation for the energy application of lignocellulosic biomass. The specific research results were as follows: (1) The combined fungi 55+m-6 with significant higher laccase activity were screened from the three white rot fungi stored in our lab which mainly secreted laccase. Strain 55 and strain m-6 were Trametes trogii sp. and Trametes versicolor sp., respectively. The laccase activity of combined fungi was 24.13 and 4.07-fold than strain 55 and strain m-6, respectively. The optimized condition for laccase production of the combined fungi in liquid medium was pH 6.5, C/N 16:1 and Tween 80 0.01%. In this optimized condition, the laccase activity of combined fungi was 4.11-fold higher comparing with which in non-optimized medium. (2) The interaction mechanism between strain 55 and strain m-6 was further studied, and no inhibition effect was observed. Brown pigment was secreted on the junction of the two strains on the plate, where the highest laccase activity was detected. Strain m-6 was much important to boost laccase activity of combined fungi in liquid medium, and strain 55 was stimulated by fungal plug, filter sterilized extracellular substances and high temperature sterilized extracellular substances of strain m-6 to produce laccase. The types of laccase isozymes did not change after combining strain 55 and strain m-6, but the concentrations of three types increased. Mass Spectrometry and TLC analysis of extracellular substances of each strain showed that concentration of some substances considerably changed after strains were combined. It was supposed that the cause of higher laccase activity of combined fungi was mainly due to some extracellular substances of strain m-6 with the appropriate concentration which stimulated laccase secretion of strain 55 and generated not only by strain m-6 but also by strain 55. (3) Combined fungi 55+m-6 were used to lignocellulosic biomass pretreatment with the type of solid-state fermentation. The highest degree of degradation of corn straw was obtained, including the rate of weight loss was 41.24% and the lignin, cellulose and hemicellulose were degraded partially under the optimized condition of 40 mesh, 65% water content on 15th day. Laccase, CMCase activities and content of reducing sugar reached the maximum value on that day. (4) Lignin degrading enzymes from combined fungi 55+m-6 were used for corn straw pretreatment. The most remarkable degradation of lignin in corn straw with Lac/1-hydroxybenzotriazole (HBT) system was observed, and the 12.60% lignin degradation was obtained under the optimized condition of 0.2% HBT, 50 U/g laccase for 1 d. After pretreated by Lac/HBT, the tight structure of corn straw was demolished and specific surface area increased, which had advantages for accessible of enzyme to cellulose and hemicellulose. (5) The corn straws pretreated by combined fungi 55+m-6 with the type of solid-state fermentation and Lac/HBT were used for enzymatic hydrolysis, and the saccharification rates of each pretreatment type were 4.33 times and 2.99% higher than CK, respectively. The enzymatic hydrolysis liquid of corn straw pretreated by Lac/HBT mainly contained xylose and glucose.

Adoption of a sintered stainless steel fiber felt was evaluated as gas diffusion backing in air-breathing direct methanol fuel cell (DMFC). By using a sintered stainless steel fiber felt as an anodic gas diffusion backing, the peak power density of an air-breathing DMFC is 24 mW cm(-2), which is better than that of common carbon paper. A 30-h-life test indicates that the degraded performance of the air-breathing DMFC is primarily due to the water flooding of the cathode. Twelve unit cells with each has 6 cm(2) of active area are connected in series to supply the power to a mobile phone assisted by a constant voltage diode. The maximum power density of 26 mW cm(-2) was achieved in the stack, which is higher than that in single cell. The results show that the sintered stainless steel felt is a promising solution to gas diffusion backing in the air-breathing DMFC, especially in the anodic side because of its high electronical conductivity and hydrophilicity. Natural humic water was treated with ultraviolet (UV) light and UV + hydrogen peroxide . The effects on the dissolved organic carbon content (DOC), the UV-absorbance at 254 nm (UV-abs.), the molecular size distribution, pH, and mutagenic activity were monitored, and the identity and concentrations of the most abundant gas chromatographable organic degradation products were determined. The DOC content and the UV-abs. of the water decreased substantially during treatment with. The decreases were dependent on the time of irradiation (UV dose) as well as on the H2O2 dose applied. The humus macromolecules were degraded to smaller fragments during irradiation. At higher UV doses, however, part of the dissolved organic matter (DOM) was found to precipitate, probably as a result of polymerization. Oxalic acid, acetic acid, malonic acid, and n-butanoic acid were the most abundant degradation products detected. These acids were found to account for up to 20% and 80% of the DOM in UV- and waters, respectively. No mutagenic activity was generated by the UV irradiation or the treatment. It is further concluded that the substantial mutagenic activity formed during chlorination of humic waters cannot be decreased by using UV irradiation as a pretreatment step.

Exactly measuring the degradation degree of ecosystems is the basis and precondition of restoring ecosystems. At present,degradation degree is usually analyzed qualitatively.In this paper,quantitative classification method was used to study the degradation degree of ecosystem in hilly region of western Liaoning,which was measured by the degradation degree of habitat.The result shows that the habitat in the northern slope of mountains has been degraded to certain stage between shrubbery and pioneer tree forest,which provides theoretical basis for vegetation restoration practice. Moreover,the result is beneficial for studying the degradation degree of ecosystems in other regions or of other types.

针对黄土高原半干旱区灌草植被退化严重的关键问题 ,进行了长期的定位研究 ,提出了灌草植被封育、改良、立体配置等快速恢复与重建的技术体系。试验结果表明 :主要草地群落本氏针茅、百里香适宜封育期为 3~ 5a ,产草量可提高 5 1~ 7 5倍。大针茅适宜封育期为 5a ,产草量可提高 1 5~ 5 0倍 ;改良草地最佳组合荒山穴播为本氏针茅 +杂类草、达乌里胡枝子 +本氏针茅群落 ,产草量提高 6 1~ 6 4倍。撂荒地开沟种植为本氏针茅 +冷蒿、本氏针茅 +红豆草、本氏针茅 +达乌里胡枝子群落 ,产草量提高 4 6~ 4 8倍。荒山隔带耕翻种植为芨芨草、本氏针茅 +紫花苜蓿、本氏针茅 +老芒麦、达乌里胡枝子 +本氏针茅群落 ,产草量提高4 5~ 6 5倍 ;灌草立体配置结合工程整地措施 ,建立了以柠条 +芨芨草、沙棘 +草木樨和山桃 +芨芨草为主的集流灌草配置模式 ,现已形成可更新的稳定的灌草群落类型。该体系的组装配套与试验、示范、推广为黄土高原农牧交错区灌草植被的快速恢复提供了重要的科学依据。

应用离心机法测定土壤水分特征,研究黄土高原典型草原带退耕地植被恢复演替过程中土壤持水性能的变化特点及趋势。结果表明:土壤含水量与土壤水吸力之间符合幂函数θ=aSb,参数a的变化随着植被的演替呈增加趋势;由于植被的影响,在同一吸力范围内的土壤含水量不同,植被为顶级长芒草(Stipabungeana)群落时,土壤在各吸力段的含水量最高,坡耕地最低;其它群落土壤含水量随着水吸力的增加变化趋于一致;退耕地植被在演替过程中通过提高土壤有机质含量改善结构,降低容重并增加毛管孔隙度,对土壤的储水和持水性能产生作用。
