The electrochemical behavior of Alizarin Red S(ARS) on GC electrode has been studied in acidic condition by spectroelectrochemistry with LOPTLC. It was found that there are three electrochemical reactions and followed by a chemical reaction of ARS in the potential range of 1.00——0.60V. The mechanism of electrode reactions has been studied and suggested based on the informations obtained from electrochemical and insitu spectroelectrochemical experiments.

Surface structure of the glassy carbon surface modified with cobalt tetraphenyl-porphyrin (CoTPP) by thermal-treatment has been studied by XPS, DTA and TG. During the thermal treatment a bond can be formed between the glassy carbon surface and TPP. Therefore the stability of electrode for the catalysis of dioxygen reduction is improved. Upon thermal treatment at 600 degrees C, FWHM of Co(2p(2/2)) is broadened, the reason is due to overlapping of peaks of multiple states, the spin orbit separation between Co (2p(1/2)) and Co (2p(3/2)) increases to 15.5-16.3eV, which indicated a change from low spin divalent states, the kinetic energy of Co L3VV Auger line and Auger parameter also increase. These changes of central cobalt ion provide a suitable redox potential for Co(III)/Co(II) which is related to the activity for catalysis of dioxygen reduction.

Using in vitro selection method to isolate nucleic acids, peptides and proteins has been studied intensively in recent years. In vitro mRNA display is a new and effective technique for peptides selection, and the rationale of this technique is that a synthetic mRNA with puromycin could covalently link with the protein that it encodes, thus an mRNA-protein fusion is formed. This approach has been used in identification of many functional peptides. The peptides binding with thymidylate synthase RNA were isolated using mRNA display technique from a large peptide library (>10(13) different sequences). The selection scheme was constructed, and the experimental conditions, including library synthesis, formation of RNA-peptide fusion and RNA immobilization were optimized. Eight cycles have been processed and the results confirmed that the selected peptides could bind with thymidylate synthase mRNA specifically. Compared the amino acid sequences of the selected peptides with those from the initial random library, the basic and aromatic residues in selected peptides were enriched significantly, suggesting these peptide regions may be important in the peptide-TS mRNA interaction. As a novel in vitro selection approach, mRNA display technique would be developed as a powerful tool for isolation of functional peptides and proteins that could interact with immobilized targets with high affinity and specificity.

Cu, Zn superoxide dismutases (SODs) are rnetalloenzymes that represent one important line of defence against reactive oxygen species (ROS). A cytoplasmic Cu. Zn SOD cDNA sequence was cloned from scallop Chlamys farreri by the homology-based cloning technique. The full-length cDNA of scallop cytoplasmic Cu, Zn SOD (designated CfSOD) was 1022 bp with a 459 bp open reading frame encoding a polypeptide of 153 amino acids. The predicted amino acid sequence of CfSOD shared high identity with cytoplasmic Cu. Zn SOD in molluscs, insects, mammals and other animals, such as cytoplasmic Cu, Zn SOD in oyster Crassostrea sostrea gigas (CAD42722), mosquito Aedes aegypti (ABF18094), and cow Bos taurus (XP_584414). A quantitative reverse transcriptase real-time PCR (qRT-PCR) assay was developed to assess the mRNA expression of CfSOD in different tissues and the temporal expression of CfSOD in scallop challenged with Listonella anguillarum, Micrococcus luteus and Candida lipolytica respectively. Higher-level mRNA expression of CfSOD was detected in the tissues of haemocytes, gill filaments and kidney. The expression of CfSOD dropped in the first 8-16 h and then recovered after challenge with L. anguillarum and M. litteus, but no change was induced by the C. lipolytica challenge. The results indicated that CfSOD was a constitutive and inducible acute-phase protein, and could play an important role in the immune responses against L. anguillarum and M. luteus infection. Authigenic carbonates were sampled in methane-enriched piston core sediments collected from gas venting sites on the western continental slope of the Ulleung Basin, East Sea of Korea. Multidisciplinary investigations on these carbonates, including the scanning electronic microscope (SEM) observations and mineralogical-geochemical compositions, were carried out to identify the carbon and oxygen sources and the forming mechanism of these carbonates. The authigenic carbonates from the study area correspond to semi-consolidated, compact concretions or nodules ranging from 2 to 9 cm in size. X-ray diffraction and electron microprobe analyses showed that most of the sampled carbonate concretions were composed of almost purely authigenic high-Mg calcite (10.7-14.3 mol% MgCO3). Characteristically, microbial structures such as filaments and rods, which were probably associated with the authigenic minerals, were abundantly observed within the carbonate matrix. The carbonates were strongly depleted in delta C-13 (-33.85 parts per thousand to -39.53 parts per thousand Peedee Belemnite (PDB)) and were enriched in delta O-18 (5.16-5.60 parts per thousand PDB), indicating that the primary source of carbon is mainly derived from the anaerobic oxidation of methane. Such methane probably originated from the destabilization of the underlying gas hydrates as strongly supporting from the enriched O-18 levels. Furthermore, the strongly depleted delta C-13 values (-60.7 parts per thousand to -61.6 parts per thousand PDB) of the sediment void gases demonstrate that the majority of the gas venting at the Ulleung Basin is microbial methane by CO2 reduction. This study provides another example for the formation mechanism of methane-derived authigenic carbonates associated with gas-hydrate decomposition in gas-seeping pockmark environments. 3DMove software, based on the three-dimension structural model of geologic interpretation, can forecast reservoir cracks from the point of view of formation of the structural geology, and analyze the characteristics of the cracks. 3DMove software dominates in forecasting cracks. We forecast the developments and directions of the cracks in Chengbei buried hill with the application of forecasting technique in 3DMove software, and obtain the chart about strain distributing on top in buried hill and the chart about relative density and orientation and the chart about the analysis of crack unsealing. In Chengbei 30 buried hill zone, north-west and north-east and approximately east-west cracks in Cenozoic are very rich and the main directions in every fault block are different. Forecasting results that are also verified by those of drilling approximately accord with the data from well logging, the case of which shows that the technique has the better ability in forecasting cracks, and takes more effects on exploration and exploitation of crack reservoir beds in ancient buried hill reservoirs.

Carbon isotopes of individual lipids in typical organisms from the Nansha sea area were measured by the GC-IRMS analytical technique. delta(13)C values of saturated fatty acids in different organisms examined are from -25.6parts per thousand to -29.7parts per thousand with the average values ranging from -26.4parts per thousand to -28.2parts per thousand and the variance range of 11.8parts per thousand, between different organisms is also observed. Unsaturated fatty acids have heavy carbon isotopic compositions and the mean differences of 2.9%.9-6.8parts per thousand compared to the same carbon number saturated fatty acids. delta(13)C values of n-alkanes range from -27.5%o to -29.7parts per thousand and their mean values, ranging from -28.6parts per thousand, to -28.9parts per thousand, are very close in different organisms. The mean difference in delta(13)C between the saturated fatty acids and n-alkanes is only 1.5parts per thousand, indicating that they have similar biosynthetic pathways. The carbon isotopic variations between the different carbon-number lipids are mostly within +/-2.0parts per thousand, reflecting that they experienced a biosynthetic process of the carbon chain elongation. At the same time, the carbon isotopic genetic relationships between the biological and sedimentary lipids are established by comparative studies of carbon isotopic compositions of individual lipids in organisms and sediments from the Nansha sea area, which provides scientific basis for carbon isotopic applied research of individual lipids.

The chloride extraction rules of iron artifacts were studied by immersion methods. Different chloride extraction results between the alkaline solution and a washing solution were obtained. The microstructure and the anti-corrosion performance of the samples before and after treatment, were, respectively studied by the scanning electron microscope (SEM) and the potentiodynamic scanning method. The results indicated that Cl- removed according to the diffusion law. The microstructure of the samples transformed after treatment. The rusts became more compact, and the porosity also increased. The chloride extraction effect in the washing solution was better than that in the NaOH solution.

In practice,many fracture reservoir was found,which has giant potential for exporation. For example,in limestone fracture reservoir,igneous rock fracture reservoir and shale fracture reservoir ,there are high yield oil wells found. The fracture reservoir has strong anisotropy and is very difficult to explore and produce.Since 1990’s,the techniques that use structure information and P-WAVE seismic attributes to detect fracture developed very rapidly,include stress and strain analysis,using amplitude,interval velocity,time-difference,azimuthal AVO analysis etc. Based on research and develop these advanced techniques of fracture detect,this paper selected two typical fracture reservoir as target area,according to the characters of research area,selected different techniques to pridect the fracture azimuth and density of target,and at last ,confirmed the favored area. This paper includes six parts:the first chapter mainly addresses the domestic and international research actuality about the fracture prediction and the evolement in ShengLi oil field,then according to the temporal exploration requirement,a research route was established; Based on the close relationship between structural fracture and the geotectonic movement and the procedure of rock distortion,the second chapter research the structural fracture predicting technique which is realized by computing the strain in every geotectonic movement ,which is by use of the forward and inversion of the growing history of structure; The third chapter discussed many kind of traditional techniques for fracture reservoir prediction,and point out their disadvantages.then research and develop the coherence volume computing technique which can distinguish from faults,the seismic wave absorbing technique,and other fracture predicting technique which is by use of seismic attributes ,such as azimuthal AVO FVO etc; The fourth chapter first establish the geological and petrophysical model by use of the existed log and drill well information, then research the variation of amplitude and seismic wave which is caused by fractures.based on it , the fracture predicting technique which is by use of variation of azimuthal impedance is researched;The fifth chapter is a case study,it selects shale fracture reservoir in LuoJia area as target,selects several kind of techniques to apply ,at last ,the fracture distribution of target reservoir and favored area were gotten;the sixth chapter is another case study,it selects limestone fracture reservoir in BoShen6 buried hill as target,selects several kind of techniques to apply,similarly favored area were gotten. Based on deeply research and development of the new techniques for fracture reservoir exploration, This paper selects two fracture reservoirs the most typical in ShengLi as targets to be applied ,good results show up a good application way ,which can be used for reference for future fracture exploration,and it can bring materially economic and social benefit.

With the development of seismic exploration, the target becomes more and more complex, which leads to a higher demand for the accuracy and efficiency in 3D exploration. Fourier finite-difference (FFD) method is one of the most valuable methods in complex structure exploration, which keeps the ability of finite-differenc method in dealing with laterally varing media and inherits the predominance of the phase-screen method in stablility and efficiency. In this thesis, the accuracy of the FFD operator is highly improved by using simulated annealing algorithm. This method takes the extrapolation step and band width into account, which is more suitable to various band width and discrete scale than the commonely-used optimized method based on velocity contrast alone. In this thesis, the FFD method is extended to viscoacoustic modeling. Based on one-way wave equation, the presented method is implemented in frequency domain; thus, it is more efficient than two-way methods, and is more convenient than time domain methods in handling attenuation and dispersion effects. The proposed method can handle large velocity contrast and has a high efficiency, which is helpful to further research on earth absorption and seismic resolution. Starting from the frequency dispersion of the acoustic VTI wave equation, this thesis extends the FFD migration method to the acoustic VTI media. Compared with the convetional FFD method, the presented method has a similar computational efficiency, and keeps the abilities of dealing with large velocity contrasts and steep dips. The numerical experiments based on the SEG salt model show that the presented method is a practical migration method for complex acoustical VTI media, because it can handle both large velocity contrasts and large anisotropy variations, and its accuracy is relatively high even in strong anisotropic media. In 3D case, the two-way splitting technique of FFD operator causes artificial azimuthal anisotropy. These artifacts become apparent with increasing dip angles and velocity contrasts, which prevent the application of the FFD method in 3D complex media. The current methods proposed to reduce the azimuthal anisotropy significantly increase the computational cost. In this thesis, the alternating-direction-implicit plus interpolation scheme is incorporated into the 3D FFD method to reduce the azimuthal anisotropy. By subtly utilizing the Fourier based scheme of the FFD method, the improved fast algorithm takes approximately no extra computation time. The resulting operator keeps both the accuracy and the efficiency of the FFD method, which is helpful to the inhancements of both the accuracy and the efficiency for prestack depth migration. The general comparison is presented between the FFD operator and the generalized-screen operator, which is valuable to choose the suitable method in practice. The percentage relative error curves and migration impulse responses show that the generalized-screen operator is much sensiutive to the velocity contrasts than the FFD operator. The FFD operator can handle various velocity contrasts, while the generalized-screen operator can only handle some range of the velocity contrasts. Both in large and weak velocity contrasts, the higher order term of the generalized-screen operator has little effect on improving accuracy. The FFD operator is more suitable to large velocity contrasts, while the generalized-screen operator is more suitable to middle velocity contrasts. Both the one-way implicit finite-difference migration and the two-way explicit finite-differenc modeling have been implemented, and then they are compared with the corresponding FFD methods respectively. This work gives a reference to the choosen of proper method. The FFD migration is illustrated to be more attractive in accuracy, efficiency and frequency dispertion than the widely-used implicit finite-difference migration. The FFD modeling can handle relatively coarse grids than the commonly-used explicit finite-differenc modeling, thus it is much faster in 3D modeling, especially for large-scale complex media.

The ionospheric parameter M(3000)F2 (the so-called transmission factor or the propagation factor) is important not only in practical applications such as frequency planning for radio-communication but also in ionospheric modeling. This parameter is strongly anti-correlated with the ionospheric F2-layer peak height hmF2,a parameter often used as a key anchor point in some widely used empirical models of the ionospheric electron density profile (e.g., in IRI and NeQuick models). Since hmF2 is not easy to obtain from measurements and M(3000)F2 can be routinely scaled from ionograms recorded by ionosonde/digisonde stations distributed globally and its data has been accumulated for a long history, usually the value of hmF2 is calculated from M(3000)F2 using the empirical formula connecting them. In practice, CCIR M(3000)F2 model is widely used to obtain M(3000)F2 value. However, recently some authors found that the CCIR M(3000)F2 model has remarkable discrepancies with the measured M(3000)F2, especially in low-latitude and equatorial regions. For this reason, the International Reference Ionosphere (IRI) research community proposes to improve or update the currently used CCIR M(3000)F2 model. Any efforts toward the improvement and updating of the current M(3000)F2 model or newly development of a global hmF2 model are encouraged. In this dissertation, an effort is made to construct the empirical models of M(3000)F2 and hmF2 based on the empirical orthogonal function (EOF) analysis combined with regression analysis method. The main results are as follows: 1. A single station model is constructed using monthly median hourly values of M(3000)F2 data observed at Wuhan Ionospheric Observatory during the years of 1957–1991 and compared with the IRI model. The result shows that EOF method is possible to use only a few orders of EOF components to represent most of the variance of the original data set. It is a powerful method for ionospheric modeling. 2. Using the values of M(3000)F2 observed by ionosondes distributed globally, data at grids uniformly distributed globally were obtained by using the Kriging interpolation method. Then the gridded data were decomposed into EOF components using two different coordinates: (1) geographical longitude and latitude; (2) modified dip (Modip) and local time. Based on the EOF decompositions of the gridded data under these two coordinates systems, two types of the global M(3000)F2 model are constructed. Statistical analysis showed that the two types of the constructed M(3000)F2 model have better agreement with the observational M(3000)F2 than the M(3000)F2 model currently used by IRI. The constructed models can represent the global variations of M(3000)F2 better. 3. The hmF2 data used to construct the hmF2 model were converted from the observed M(3000)F2 based on the empirical formula connecting them. We also constructed two types of the global hmF2 model using the similar method of modeling M(3000)F2. Statistical analysis showed that the prediction of our models is more accurate than the model of IRI. This demonstrated that using EOF analysis method to construct global model of hmF2 directly is feasible. The results in this thesis indicate that the modeling technique based on EOF expansion combined with regression analysis is very promising when used to construct the global models of M(3000)F2 and hmF2. It is worthwhile to investigate further and has the potential to be used to the global modeling of other ionospheric parameters.

The effect of thermal treatment on H-MCM-22 and H-ZSM-5 zeolites was investigated using the electron spin resonance technique. A six-line signal (denoted as A, g = 2.048, A = 22. 15 G) was detected on H-MCM-22 after He purging at high temperatures, whose intensities increased with the treating temperature. The same signal was also found on H-ZSM-5 zeolites with different crystal sizes. The paramagnetic center was identified as a V center, namely, a hole of an electron trapped on an oxygen atom bonding to a nearby aluminum atom. These signals appeared only on a dealuminated sample or a sample concomitantly with dealumination. The formation of the hole might involve an electron transferring from the lattice oxygen to a nonframework aluminum species, and the hyperfine splitting is caused by the interaction between the electron hole locating on the p orbit of oxygen and the framework aluminum bonding with the oxygen. The signal disappeared after the sample was exposed to air or oxygen at room temperature. However, the process was reversible. A new set of signals (denoted as B, g(1) = 2.008, g(2) = 2.003, g(3) = 1.9985) was observed after oxygen adsorption on the H-MCM-22 pretreated with He at 973 K or He purging at 973 K on the H-MCM-22 pretreated with oxygen at 813 K, which was attributed to the O- species.
