工业革命以来,由于人口的快速增加和人类活动的强烈干扰(主要包括煤炭、石油等石化燃料的燃烧、化肥生产和使用)导致土地利用/覆被变化、大气CO2浓度升高、N沉降等一系列全球环境变化问题。有关陆地生态系统生物地球化学循环,尤其是陆地生态系统C、N循环及其耦合过程方面的研究成为全球变化科学研究领域的重要内容。 干旱/半干旱地区占地球陆地总面积的1/3。与湿润地区相比较,干旱/半干旱地区生态系统稳定性比较差,往往属于生态脆弱区。因此,全球变化对干旱/半干旱地区生态系统影响更加敏感。科尔沁沙地位于我国北方干旱/半干旱地区,是我国典型的农牧交错区和生态脆弱区。科尔沁沙地是世界上人口密度最高的干旱/半干旱地区之一,人类活动对其影响剧烈。然而,有关科尔沁沙地生态系统C、N元素生物地球化学循环过程对土地利用/覆被变化、N沉降等全球变化响应及其反馈机制的研究非常缺乏。因此,本文以科尔沁沙地退化沙质草地、农田、不同年龄樟子松和杨树人工林等生态系统为对象,开展了造林、模拟N沉降和凋落物管理对生态系统C、N元素循环过程影响的研究。 在科尔沁沙地东南缘,以退化沙质草地、樟子松(Pinus sylvestris var. mongolica)人工林(15、24和30年生)、杨树(Poplus xiaozhuanica)人工林(7、11和15年生)为对象,研究草地转变为林地对生态系统C、N储量影响;以退化草地、榆树疏林草地和32年生樟子松人工林为对象,比较草地造林对土壤C、N循环过程及其土壤微生物性状的影响;以农田和5、10、15年生杨树人工林为对象,研究退耕还林对生态系统C、N储量和循环过程影响;以35年生樟子松人工林为对象,模拟研究N沉降和凋落物管理对生态系统C、N循环过程影响。通过上述研究,得到以下主要结果: (1)草地生态系统总C储量为34.38 Mg ha-1,15、24和30年生樟子松人工林生态系统总C储量分别为43.56、60.45和66.59 Mg ha-1,7、11和15年生杨树人工林生态系统总C储量分别为34.54、48.26和78.77 Mg ha-1;与农田相比,退耕5年的杨树人工林生态系统总C库储量下降13%,而10年和15年杨树人工林分别增加了176%和5倍;随着人工林年龄的增加,地上植被生物量C库储量占生态系统总C库储量的比例逐渐增加,并主要分配在树干。草地生态系统总N库储量为2.54 Mg ha-1,15、24和30年生樟子松人工林生态系统总N库储量分别为1.96、2.10和2.19 Mg ha-1,7、11和15年生杨树人工林生态系统总N库储量分别为2.27、1.84和2.60 Mg ha-1;与农田相比,退耕5年的杨树人工林生态系统总N库储量下降32%,而10年和15年杨树人工林分别增加了47%和76%;农田和草地造林后生态系统N储量依然主要分配在土壤中。 (2)草地和农田造林后土壤C、N库储量的变化受多因子的影响,例如林龄、树种种类以及立地条件等。农田和草地造林初期,土壤C、N库储量表现出下降趋势,随着林龄的增加,土壤C、N储量逐渐恢复。草地营造樟子松人工林30年后,0–60 cm深度土壤C、N储量依然显著低于草地;与草地相比,15年生杨树人工林土壤C、N储量差异不显著。在立地条件较好的情况下,10年杨树人工林土壤C、N储量已显著高于农田;然而,在立地条件相对较差的情况下,15年杨树人工林土壤C、N储量仍然与农田相比差异不显著。 (3)土地利用变化能够强烈地改变土壤C、N循环过程。与草地或疏林草地相比,32年生樟子松人工林土壤C、N、P含量显著降低;土壤C、N矿化过程发生显著变化,并且受季节变化的影响;在不同季节,土壤微生物量碳含量、代谢熵(qCO2)、微生物熵(MBC/TOC)以及土壤酶活性等在不同土地利用条件下表现出规律不一致。同样,农田退耕杨树人工林能够显著影响土壤C、N矿化过程,土壤无机氮(铵态氮+硝态氮)含量,土壤微生物量碳含量以及土壤微生物活性。草地造林在一定程度上导致土壤质量下降。而农田造林有利于土壤质量改善,尤其在在立地条件较好情况下。 (4)N添加增加对沙地樟子松人工林地上和地下C、N元素含量影响不大;N添加1年后,仅林下植被C、N含量显著增加,高氮处理(N15)凋落物N含量显著增加。N添加抑制了沙地樟子松人工林凋落物的早期分解和N、P元素释放。5、6、8和9月份土壤无机N含量均随着N输入增加表现出一定程度的增加,然而,7月份N添加导致土壤无机N含量降低。N添加对土壤潜在N矿化速率影响不显著。7和8月份N添加影响土壤C矿化速率,而其它月份影响不显著。低氮处理(N5)有利于增加土壤微生物量碳含量,而高氮处理(N15)在一定程度上降低土壤微生物量碳含量。 (5)凋落物输入变化(凋落物添加和凋落物移出)在一定程度上改变了35年生沙地樟子松人工林生态系统C、N循环过程。凋落物移出(C0)增加了林下植被C含量,降低了树木叶片N含量。凋落物移出抑制了凋落物分解和P元素的释放,而增加了C元素的早期释放速率,对N元素释放过程影响不显著。凋落物输入变化对不同月份土壤无机N含量和土壤N矿化过程影响均不显著。仅在6月份凋落物移出显著抑制了土壤C矿化速率,其它月份差异均不显著。凋落物管理对土壤微生物量碳含量影响不显著。 以上研究结果表明,土地利用变化、N沉降和凋落物输入改变等能够影响半干旱地区沙地生态系统C、N储量和循环过程。尤其是土地利用变化强烈改变沙地生态系统C、N储量、分配格局和循环过程,并且受到多因子的影响。科尔沁沙地樟子松人工林生态系统C、N元素生物地球化学循环存在密切的耦合关系。今后有必要进一步结合3S技术、同位素技术、模型模拟以及分子生物学技术等,从微观-宏观不同尺度上,研究半干旱地区沙地生态系统C、N循环过程对全球变化的响应及其反馈机制。

制备了用作直接甲醇燃料电池的碳载Pt-Fe(Pt-Fe/C)阴极催化剂,X射线能量色散谱(EDX)、X射线衍射谱和电化学测量的结果表明,在Pt-Fe/C催化剂中,Fe以3种形式存在.质量分数大约为20%的Fe进入Pt的晶格,形成Pt-Fe合金,质量分数大约为80%的Fe没有进入Pt的晶格而以Fe和Fe2O3的形式单独存在.该催化剂经酸处理后,非合金化Fe和Fe2O3被溶解,而使Pt-Fe/C催化剂的电化学活性比表面积要比未经酸处理前的增加约30%左右,导致Pt-Fe/C催化剂对氧还原的电催化活性优于未经酸处理前的Pt-Fe/C催化剂.研究结果表明,Pt-Fe/C催化剂的电化学活性比表面积对氧还原的电催化活性起重要的作用,另外,只有与Pt形成合金的Fe能提高Pt对氧还原的电催化活性,而非合金化的Fe对Pt催化剂对氧还原的电催化活性基本没有影响.

A high pressure and high temperature method was used to efficiently extract on a large scale metallofullerenes M@C-2n (M=La,Ce) in a closed vessel under argon gas protection. With pyridine as the HPHT solvent, about 60-80% M@C-2n and 30-55% M@C-82 can be enriched, M@C-82 is dissolved selectively; With toluene as the HPHT solvent, M@C-2n can also be efficiently extracted, especially M@C-74, which is a new member of M@C-2n soluble species. A large and transparent Yb:FAP crystal with dimensions up to circle divide 30 mm x 85 mm has been grown by the Czochralski method. The preparation of the raw material has been investigated. X-ray power diffraction studies of Yb:FAP crystal confirm that the as-grown crystal is isostructural with the FAP crystal. The crystalline quality has been studied using X-ray rocking curve analysis. The segregation coefficient of Yb3+ in the Yb:FAP crystal has been also determined. Linear thermal expansion coefficients in [001] and [100] directions have been measured in the 30-800 degrees C temperature range. (c) 2005 Elsevier B.V. All rights reserved.

在中国北方大部分地区,水分始终是影响植物生长和分布的最主要限制性因子之一,植物在其生长期经常遭受水分胁迫。不仅如此,随着大气同温层中臭氧浓度的减少,过量的有害紫外辐射(主要是UV-B,280nm-320nm)将穿透大气层达到地球表面。随着全球变化的加剧,这些地区的植物将不可避免地受UV-B和水分胁迫的共同作用。 本实验是在北京东灵山暖温带森林生态系统中,选择常见灌丛土庄绣线菊(Spiraea pubescens),建立UV-B控制实验。连续三个生长季每天增补9.4kJ•m-2的辐射剂量,模拟臭氧衰减17%时近地表面UV-B辐射的增强。本实验的目的是观测在野外环境下,长时间人工增强UV-B辐射对土庄绣线菊水分生理、氮素利用以及形态特征的影响。具体对以下指标进行测定:叶片的气孔导度、碳同位素比率(δ13 C)、叶含水量、叶面积、水分利用效率(WUE)、叶全氮含量、叶氮素再吸收率。 实验结果表明,增强UV-B辐射显著减少了土庄绣线菊的叶面积(50.1%),提高了叶片全氮含量(102%),处理植株的氮素再吸收率比对照植株高出50.9%。同时,UV-B辐射还在一定程度上(尽管统计显示不显著)降低了气孔导度(16.1%)、胞间CO2浓度与大气CO2浓度之比(Ci/Ca) (4.0%)、提高了碳同位素比率(δ13 C)(20.5‰)、叶含水量(3.1%)及比叶重(SLW)(5.2%),从而导致水分利用效率(WUE)的增加(4.1%),植物的抗旱能力增强。值得注意的是,深层土壤(30-40cm)含水量变化会影响气孔导度、δ13 C、WUE对紫外辐射的响应程度:在土壤干旱的季节(6月和9月),气孔导度、δ13 C、WUE这些指标处理和对照的差异很小,但是当土壤水分充足时(7月和8月),处理和对照的差异就较为显著。另外,随着实验处理时间的延长UV-B的效应变得不显著。相关分析表明,UV-B辐射降低了土壤含水量(30-40cm)与土庄绣线菊叶含水量、δ13 C、Ci/Ca、气孔导度的相关系数,增强了WUE与土壤含水量的相关性,这也许是由于UV-B辐射增强了WUE对土壤水分变化的敏感性。本研究的结果表明UV-B辐射对土庄绣线菊的形态和生长有显著的影响,但对主要水分生理指标影响不显著。

A Gram-positive bacterium, designated strain CW 7(T), was isolated from forest soil in Anhui Province, south-east China. Cells were strictly aerobic, motile with peritrichous flagella and rod-shaped. The strain grew optimally at 30-37 degrees C and pH 7.0-8.0. The major fatty acids of strain CW 7(T) were anteiso-C-15:0, iso-C-15:0 and anteiso-C-17:0. The predominant menaquinone was MK-7. The cell-wall peptidoglycan contained meso-diaminopimelic acid. The G + C content of the genomic DNA was 42.3 mol%. Phylogenetic analysis indicated that strain CW 7(T) belonged to a monophyletic cluster within the genus Bacillus and showed 16S rRNA gene sequence similarities of less than 96.5% to recognized species of the genus Bacillus. The results of the polyphasic taxonomic study, including phenotypic, chemotaxonomic and phylogenetic analyses, showed that strain CW 7(T) represents a novel species of the genus Bacillus, for which the name Bacillus pallidus sp. nov. is proposed. The type strain is CW 7(T) (=KCTC 13200(T)=CCTCC AB 207188(T)=LMG 24451(T)).

A Gram-negative, rod-shaped, non-motile, non-spore-forming bacterium, designated strain HR2(T) was isolated from a soil sample from the Talklimaken Desert in Xinjiang Province, China. Strain HR2(T) grew optimally at pH 7.0-8.0 and 30-37 degrees C in the presence of 0-1% (w/v) NaCl. An analysis of 16S rRNA gene sequences revealed that strain HR2(T) fell within the radiation of the genus Pseudomonas, the highest level of similarity being found with respect to Pseudomonas luteola IAM 13000(T) (97.5%); the levels of sequence similarity with respect to other recognized Pseudomonas species were < 96.4%. DNA-DNA hybridization showed that the genetic relatedness between strain HR2(T) and P. luteola IAM 13000(T) was 53.2%. The G + C content of the genomic DNA of strain HR2(T) was 55.2 mol%. The major fatty acids were 18: 1, summed feature 3 and 16:0. The hydroxylated fatty acids 10:0 3-OH, 12:0 3-OH and 12:0 2-OH were also present. The data obtained in this polyphasic study indicated that this isolate represents a novel species of the genus Pseudomonas, for which the name Pseudomonas duriflava sp. nov. is proposed, The type strain is HR2(T) (=KCTC 221129(T) =CGMCC 1.6858(T)).

The taxonomic position of a novel Gram-negative strain, designated Sy1(T), isolated from a farm-soil sample obtained from Jiangsu Province, PR China, was characterized by using a polyphasic approach. The cells were non-motile, non-spore-forming rods. The organism grew optimally at 30-37 degrees C and at pH 6.0-8.0. Based on 16S rRNA gene sequence analysis, strain Sy1(T) is a member of the genus Sphingobacterium; Sphingobacterium multivorum JCM 21156(T) was the nearest relative (98.5% sequence similarity). The predominant fatty acids of strain Sy1T were isoC15:0 (32.90/o), C16:0 (10.9%) and summed feature 3 (iso-C-15:0 2-OH and/or C-16:1 omega 7c; 24.1%). The DNA G + C content was 38.5 mol%. The low level of DNA-DNA relatedness (2.2 %) to S. multivorum JCM 21156 T in combination with differential morphological and biochemical properties demonstrated that strain SY1(T) (=KCTC 22131(T)= CGMCC 1.6855(T)) should be classified as representing a novel species of the genus Sphingobacterium for which the name Sphingobacterium siyangense sp. nov. is proposed.

目的:评价新生豚鼠高胆红素血症时P50抑制值[即T/C值(T为实验,C为对照)]的动态变化对神经系统的毒性作用.方法:出生5~7天的新生豚鼠30只,随机分成5组,每组6只.其中,第一组为正常对照组(C),其余4组为实验组(T).5组新生豚鼠均在硫喷妥钠麻醉下行颅骨电极包埋术,待手术麻醉清醒后,分别测各组新生豚鼠的T/C值.检测完毕,分别向其中2组实验组动物腹腔注入胆红素溶液100 μg/g,4 h、8 h后观察;另2组实验组动物腹腔注入胆红素溶液200 μg/g,4 h、8 h后观察.正常对照组动物均向腹腔注入生理盐水0.5 ml.各组动物在观察完行为学变化和T/C值检测后,再迅速处死动物,取脑组织,在光镜、电镜下观察脑组织结构的变化.结果:实验豚鼠在注入胆红素溶液后,除Tlb组变化不明显外(P>0.05),其余各组豚鼠T/C值的变化差异均有统计学意义(P<0.01).不同实验组与正常对照组的T/C值的变化差异也均有统计学意义(P<0.01).结论:P50抑制(T/C)在高胆红素血症不同时段均有明显变化,可以较早期地(在胆红素聚集阶段)预测胆红素对神经系统的毒性作用,为临床预防和评价高胆红素血症对新生儿神经系统损伤提供一种新的方法.

研究了卵泡大小、卵泡液和卵丘细胞对牛卵泡卵母细胞体外成熟的影响。结果表明:直径2~6 mm 卵泡中的卵母细胞成熟 率(7211 %) 最高,与直径小于2 mm(5814 %) 、6~8 mm(5614 %) 及大于8 mm(3510 %) 卵泡中的卵母细胞组差异显著;成熟培养基 中添加10 %的新鲜牛卵泡液(bFF) 对牛卵母细胞的体外成熟有促进作用,但高浓度的bFF(20 %、30 %) 则抑制牛卵母细胞的体外成 熟;卵丘- 卵母细胞复合体的质量影响卵母细胞的体外成熟,A、B、C 三级卵母细胞成熟率分别为8611 %、6613 %、3516 % ,卵裂率分 别为4218 %、3119 %、1015 % ,差异均显著( P < 0105) 。

Our studies investigated the physico-chemical properties of alkaline phosphatase excreted by D. magna. This cladoceran mainly released alkaline phosphatase, though it also released a small amount of acid phosphatase. The alkaline phosphatase showed a broad pH optimum (8.05-10.0), and had a broad optimum temperature (30-35 degrees C) with a temperature coefficient (Q(10)) of 2.45. The K-m of the enzyme is 0.15 +/- 0.02 mM when p-nitrophenyl phosphate is used as a substrate, and the V-max is 0.43 +/- 0.01 mu M pNP mg(-1) DW h(-1). Even though alkaline phosphatase had been incubated in chloroform saturated with WC medium for 13 days, its activity was 54% that of the original. The enzyme was strongly inactivated by EDTA, and appeared to be zinc dependent. The alkaline phosphatase activity remained constant when D. magna was fed different quantities of Chlorella sp. The sensitivity of D. magna phosphatase activity to phosphate was time-dependent. During the first 16 hrs, the enzyme was insensitive to phosphate addition, after 24 hrs incubation the enzyme became sensitive to phosphate addition.
