A multi-disciplinary team based at Heriot-Watt University and other Universities has been set up to tackle the design and manufacturing of lab-on-a-chip for industries as one of the demonstrators of the EPSRC Grand Challenge project "3D-Mintegration". The team focuses on the analysis of foetal genetic material extracted from maternal blood as a smart alternative to invasive prenatal testing such as amniocentesis. The first module of the microsystem envisaged achieves a separation of blood cells from plasma. This system permits the testing of different manufacturing techniques.

Placing a gene of interest under the control of an inducible promoter greatly aids the purification, localization and functional analysis of proteins but usually requires the sub-cloning of the gene of interest into an appropriate expression vector. Here, we describe an alternative approach employing in vitro transposition of Tn Omega P(BAD) to place the highly regulable, arabinose inducible P(BAD) promoter upstream of the gene to be expressed. The method is rapid, simple and facilitates the optimization of expression by producing constructs with variable distances between the P(BAD) promoter and the gene. To illustrate the use of this approach, we describe the construction of a strain of Escherichia coli in which growth at low temperatures on solid media is dependent on threshold levels of arabinose. Other uses of the transposable promoter are also discussed.

State-of-the-art large vocabulary continuous speech recognition (LVCSR) systems often combine outputs from multiple subsystems developed at different sites. Cross system adaptation can be used as an alternative to direct hypothesis level combination schemes such as ROVER. In normal cross adaptation it is assumed that useful diversity among systems exists only at acoustic level. However, complimentary features among complex LVCSR systems also manifest themselves in other layers of modelling hierarchy, e.g., subword and word level. It is thus interesting to also cross adapt language models (LM) to capture them. In this paper cross adaptation of multi-level LMs modelling both syllable and word sequences was investigated to improve LVCSR system combination. Significant error rate gains up to 6.7% rel. were obtained over ROVER and acoustic model only cross adaptation when combining 13 Chinese LVCSR subsystems used in the 2010 DARPA GALE evaluation. © 2010 ISCA.

The paper describes the architecture of VODIS, a voice operated database inquiry system, and presents some experiments which investigate the effects on performance of varying the level of a priori syntactic constraints. The VODIS system includes a novel mechanism for incorporating context-free grammatical constraints directly into the word recognition algorithm. This allows the degree of a priori constraint to be smoothly varied and provides for the controlled generation of multiple alternatives. The results show that when the spoken input deviates from the predefined task grammar, a combination of weak a priori syntax rules in conjunction with full a posteriori parsing on a lattice of alternative word matches provides the most robust recognition performance. © 1991.

Vision trackers have been proposed as a promising alternative for tracking at large-scale, congested construction sites. They provide the location of a large number of entities in a camera view across frames. However, vision trackers provide only two-dimensional (2D) pixel coordinates, which are not adequate for construction applications. This paper proposes and validates a method that overcomes this limitation by employing stereo cameras and converting 2D pixel coordinates to three-dimensional (3D) metric coordinates. The proposed method consists of four steps: camera calibration, camera pose estimation, 2D tracking, and triangulation. Given that the method employs fixed, calibrated stereo cameras with a long baseline, appropriate algorithms are selected for each step. Once the first two steps reveal camera system parameters, the third step determines 2D pixel coordinates of entities in subsequent frames. The 2D coordinates are triangulated on the basis of the camera system parameters to obtain 3D coordinates. The methodology presented in this paper has been implemented and tested with data collected from a construction site. The results demonstrate the suitability of this method for on-site tracking purposes.

Cross system adaptation can be used as an alternative to direct hypothesis level combination schemes such as ROVER. The standard approach involves only cross adapting acoustic models. To fully exploit the complimentary features among sub-systems, language model (LM) cross adaptation techniques can be used. Previous research on multi-level n-gram LM cross adaptation is extended to further include the cross adaptation of neural network LMs in this paper. Using this improved LM cross adaptation framework, significant error rate gains of 4.0%-7.1% relative were obtained over acoustic model only cross adaptation when combining a range of Chinese LVCSR sub-systems used in the 2010 and 2011 DARPA GALE evaluations. Space heating accounts for a large portion of the world's carbon dioxide emissions. Ground Source Heat Pumps (GSHPs) are a technology which can reduce carbon emissions from heating and cooling. GSHP system performance is however highly sensitive to deviation from design values of the actual annual energy extraction/rejection rates from/to the ground. In order to prevent failure and/or performance deterioration of GSHP systems it is possible to incorporate a safety factor in the design of the GSHP by over-sizing the ground heat exchanger (GHE). A methodology to evaluate the financial risk involved in over-sizing the GHE is proposed is this paper. A probability based approach is used to evaluate the economic feasibility of a hypothetical full-size GSHP system as compared to four alternative Heating Ventilation and Air Conditioning (HVAC) system configurations. The model of the GSHP system is developed in the TRNSYS energy simulation platform and calibrated with data from an actual hybrid GSHP system installed in the Department of Earth Science, University of Oxford, UK. Results of the analysis show that potential savings from a full-size GSHP system largely depend on projected HVAC system efficiencies and gas and electricity prices. Results of the risk analysis also suggest that a full-size GSHP with auxiliary back up is potentially the most economical system configuration. © 2012 Elsevier Ltd.

Cross system adaptation, in which model adaptation is performed using the outputs from another sub-system, can be used as an alternative to hypothesis level combination schemes such as ROVER. Normally cross adaptation is only performed on the acoustic models. However, there are many other levels in LVCSR systems' modelling hierarchy where complimentary features may be exploited, for example, the sub-word and the word level, to further improve cross adaptation based system combination. It is thus interesting to also cross adapt language models (LMs) to capture these additional useful features. In this paper cross adaptation is applied to three forms of language models, a multi-level LM that models both syllable and word sequences, a word level neural network LM, and the linear combination of the two. Significant error rate reductions of 4.0-7.1% relative were obtained over ROVER and acoustic model only cross adaptation when combining a range of Chinese LVCSR sub-systems used in the 2010 and 2011 DARPA GALE evaluations. © 2012 Elsevier Ltd. We quantify the conditions that might trigger wide spread adoption of alternative fuel vehicles (AFVs) to support energy policy. Empirical review shows that early adopters are heterogeneous motivated by financial benefits, environmental appeal, new technology, and vehicle reliability. A probabilistic Monte Carlo simulation model is used to assess consumer heterogeneity for early and mass market adopters. For early adopters full battery electric vehicles (BEVs) are competitive but unable to surpass diesels or hybrids due to purchase price premium and lack of charging availability. For mass adoption, simulations indicate that if the purchase price premium of a BEV closes to within 20% of an in-class internal combustion engine (ICE) vehicle, combined with a 60% increase in refuelling availability relative to the incumbent system, BEVs become competitive. But this depends on a mass market that values the fuel economy and CO2 reduction benefits associated with BEVs. We also find that the largest influence on early adoption is financial benefit rather than pro-environmental behaviour suggesting that AFVs should be marketed by appealing to economic benefits combined with pro-environmental behaviour to motivate adoption. Monte Carlo simulations combined with scenarios can give insight into diffusion dynamics for other energy demand-side technologies. © 2012 Elsevier Inc.

Spoken content in languages of emerging importance needs to be searchable to provide access to the underlying information. In this paper, we investigate the problem of extending data fusion methodologies from Information Retrieval for Spoken Term Detection on low-resource languages in the framework of the IARPA Babel program. We describe a number of alternative methods improving keyword search performance. We apply these methods to Cantonese, a language that presents some new issues in terms of reduced resources and shorter query lengths. First, we show score normalization methodology that improves in average by 20% keyword search performance. Second, we show that properly combining the outputs of diverse ASR systems performs 14% better than the best normalized ASR system. © 2013 IEEE.
