ARGONAUTE4 (AGO4) and RNA polymerase IV (Pol IV) are required for DNA methylation guided by 24 nucleotide small interfering RNAs (siRNAs) in Arabidopsis thaliana. Here we show that AGO4 localizes to nucleolus-associated bodies along with the Pol IV subunit NRPD1b; the small nuclear RNA (snRNA) binding protein SmD3; and two markers of Cajal bodies, trimethylguanosine-capped snRNAs and the U2 snRNA binding protein U2B''. AGO4 interacts with the C-terminal domain of NRPD1b, and AGO4 protein stability depends on upstream factors that synthesize siRNAs. AGO4 is also found, along with the DNA methyltransferase DRM2, throughout the nucleus at presumed DNA methylation target sites. Cajal bodies are conserved sites for the maturation of ribonucleoprotein complexes. Our results suggest a function for Cajal bodies as a center for the assembly of an AGO4/NRPD1b/siRNA complex, facilitating its function in RNA-directed gene silencing at target loci.

DNA methylation directed by 24-nucleotide small RNAs involves the small RNA-binding protein ARGONAUTE4 (AGO4), and it was previously shown that AGO4 localizes to nucleolus-adjacent Cajal bodies, sites of snRNP complex maturation. Here we demonstrate that AGO4 also localizes to a second class of nuclear bodies, called AB-bodies, which are found immediately adjacent to condensed 45S ribosomal DNA (rDNA) sequences. AB-bodies also contain other proteins involved in RNA-directed DNA methylation including NRPD1b (a subunit of the RNA Polymerase IV complex, RNA PolIV), NRPD2 (a second subunit of this complex), and the DNA methyltransferase DRM2. These two classes of AGO4 bodies are structurally independent--disruption of one class does not affect the other--suggesting a dynamic regulation of AGO4 within two distinct nuclear compartments in Arabidopsis. Abolishing Cajal body formation in a coilin mutant reduced overall AGO4 protein levels, and coilin dicer-like3 double mutants showed a small decrease in DNA methylation beyond that seen in dicer-like3 single mutants, suggesting that Cajal bodies are required for a fully functioning DNA methylation system in Arabidopsis.

Data fusion can be defined as the process of combining data or information for estimating the state of an entity. Data fusion is a multidisciplinary field that has several benefits, such as enhancing the confidence, improving reliability, and reducing ambiguity of measurements for estimating the state of entities in engineering systems. It can also enhance completeness of fused data that may be required for estimating the state of engineering systems. Data fusion has been applied to different fields, such as robotics, automation, and intelligent systems. This paper reviews some examples of recent applications of data fusion in civil engineering and presents some of the potential benefits of using data fusion in civil engineering.

One of the greatest obstacles facing the nuclear industry is that of sustainability, both in terms of the finite reserves of uranium ore and the production of highly radiotoxic spent fuel which presents proliferation and environmental hazards. Alternative nuclear technologies have been suggested as a means of delivering enhanced sustainability with proposals including fast reactors, the use of thorium fuel and tiered fuel cycles. The debate as to which is the most appropriate technology continues, with each fuel system and reactor type delivering specific advantages and disadvantages which can be difficult to compare fairly. This paper demonstrates a framework of performance metrics which, coupled with a first-order lumped reactor model to determine nuclide population balances, can be used to quantify the aforementioned pros and cons for a range of different fuel and reactor combinations. The framework includes metrics such as fuel efficiency, spent fuel toxicity and proliferation resistance, and relative cycle performance is analysed through parallel coordinate plots, yielding a quantitative comparison of disparate cycles. © 2011 Elsevier Ltd. High-resolution γ-ray spectroscopy is essential to fully exploit the unique, high-quality beams available at the next generation of radioactive ion beam facilities such as the TRIUMF isotope separator and accelerator (ISAC). The 8π spectrometer, which consists of 20 Compton-suppressed HPGe detectors, has recently been reconfigured for a vigorous research programme in weak interaction and nuclear structure physics. With the addition of a variety of ancillary detectors it has become the world's most powerful device dedicated to β-decay studies. This paper provides a brief overview of the apparatus and highlights from recent experiments. © 2005 IOP Publishing Ltd.

Lifetimes for decays linking near-yrast states in 107Cd have been measured using the recoil distance method (RDM). The nucleus of interest was populated via the 98Mo(12C,3n)107Cd fusion-evaporation reaction at an incident beam energy of 60 MeV. From the measured lifetimes, transition probabilities have been deduced and compared with the theoretical B(E2) values for limiting cases of harmonic vibrational and axially deformed rotational systems. Our initial results suggest a rotor-like behaviour for the structure based on the unnatural-parity, h11/2 orbital in 107Cd, providing further evidence for the role of this 'shape-polarizing' orbital in stabilizing the nuclear deformation in the A ∼ 100 transitional region. © 2005 IOP Publishing Ltd.

The potential for countries that currently have a nominal nuclear energy infrastructure to adopt thorium–uranium-fuelled nuclear energy systems, using a once-through ‘open’ nuclear fuel cycle, has been suggested by the International Atomic Energy Agency. This review paper highlights generation II, III and III+ nuclear energy technologies that could potentially adopt an open thorium–uranium fuel cycle and qualitatively highlights the main differences between the open thorium–uranium and open uranium fuel cycles.

Restoring a scene distorted by atmospheric turbulence is a challenging problem in video surveillance. The effect, caused by random, spatially varying, perturbations, makes a model-based solution difficult and in most cases, impractical. In this paper, we propose a novel method for mitigating the effects of atmospheric distortion on observed images, particularly airborne turbulence which can severely degrade a region of interest (ROI). In order to extract accurate detail about objects behind the distorting layer, a simple and efficient frame selection method is proposed to select informative ROIs only from good-quality frames. The ROIs in each frame are then registered to further reduce offsets and distortions. We solve the space-varying distortion problem using region-level fusion based on the dual tree complex wavelet transform. Finally, contrast enhancement is applied. We further propose a learning-based metric specifically for image quality assessment in the presence of atmospheric distortion. This is capable of estimating quality in both full-and no-reference scenarios. The proposed method is shown to significantly outperform existing methods, providing enhanced situational awareness in a range of surveillance scenarios. © 1992-2012 IEEE.
