The successful application of techniques to enhance detection of age marks in biological specimens is of vital importance in fisheries research. This manual documents age determination techniques used by staff at the Woods Hole Laboratory, National Marine Fisheries Service. General information on procedures for preparing anatomical structures is described, together with criteria used to interpret growth patterns and assign ages. Annotated photographs of age structures are provided to illustrate criteria. Detailed procedures are given for the following species: Atlantic herring (Clupea harengus), haddock (Melanogrammus aeglefinus), Atlantic cod (Gadus morhua), pollock (Pollachius virens), silver hake (Merluccius bilinearis), red hake (Urophycis chuss), black sea bass (Centropristis striata), weakfish (Cynoscion regalis), Atlantic mackerel (Scomber scombrus), butterfish (Peprilus triacanthus), redfish (Sebastes fasciatus), summer flounder (Paralichthys dentatus), winter flounder (Pseudopleuronectes americanus), witch flounder (Glyptocephalus cynoglossus), American plaice (Hippoglossoides platessoides), yellowtail flounder (Limanda ferruginea), surf clam (Spisula solidissima), and ocean quahog (Arctica islandica). After 20 annual meetings it is worth to have a look back and to see how it has started. There has been very little collaboration on research projects between member institutes under the auspices of WEFTA, co-operation in more neutral areas of common interest was developed at an early stage. The area which has proved very fruitful is methodology. It was agreed that probably the best way to make progress was to arrange meetings at each laboratory in turn where experienced, practising scientists could describe in detail how they carried out analyses. In this way, difficulties could be demonstrated or uncovered, and the accuracy, precision, efficiency and cost of the methods used in different laboratories could be compared.

Representatives from the family of Lemnaceae (duckweed) make ideal experimental material for research into a succession of phytophysiological processes with regard to growth rate and vegetative reproduction. They are also easy to maintain in sterile cultures. Lemnaceae belong to the higher flowering plants (flowers are rarely produced), however they are distinguished by a much simplified morphological and anatomical structure. As water plants they possess the advantage, that they can be cultivated in synthetic media under laboratory conditions controlled by the application of both a known light intensity and temperature. This paper describes experimental research of growth of Lemnaceae in different conditions. Some of the variables were mineral media, illumination and aeration.

In recent collaborative biological sampling exercises organised by the Nottingham Regional Laboratory of the Severn-Trent Water Authority, the effect of handnet sampling variation on the quality and usefulness of the data obtained has been questioned, especially when this data is transcribed into one or more of the commonly used biological methods of water quality assessment. This study investigates if this effect is constant at sites with similar typography but differing water quality states when the sampling method is standardized and carried out by a single operator. An argument is made for the use of a lowest common denominator approach to give a more consistent result and obviate the effect of sampling variation on these biological assessment methods.

Environmental managers strive to preserve natural resources for future generations but have limited decision-making tools to define ecosystem health. Many programs offer relevant broad-scale, environmental policy information on regional ecosystem health. These programs provide evidence of environmental condition and change, but lack connections between local impacts and direct effects on living resources. To address this need, the National Oceanic and Atmospheric Administration/National Ocean Service (NOAA/NOS) Cooperative Oxford Laboratory (COL), in cooperation with federal, state, and academic partners, implemented an integrated biotic ecosystem assessment on a sub-watershed 14-digit Hydrologic Unit Code (HUD) scale in Chesapeake Bay. The goals of this effort were to 1) establish a suite of bioindicators that are sensitive to ecosystem change, 2) establish the effects of varying land-use patterns on water quality and the subsequent health of living resources, 3) communicate these findings to local decision-makers, and 4) evaluate the success of management decisions in these systems. To establish indicators, three sub-watersheds were chosen based on statistical analysis of land-use patterns to represent a gradient from developed to agricultural. The Magothy (developed), Corsica (agricultural), and Rhode (reference) Rivers were identified. A random stratified design was developed based on depth (2m contour) and river mile. Sampling approaches were coordinated within this structure to allow for robust system comparisons. The sampling approach was hierarchal, with metrics chosen to represent a range from community to cellular level responses across multiple organisms. This approach allowed for the identification of sub-lethal stressors, and assessment of their impact on the organism and subsequently the population. Fish, crabs, clams, oysters, benthic organisms, and bacteria were targeted, as each occupies a separate ecological niche and may respond dissimilarly to environmental stressors. Particular attention was focused on the use of pathobiology as a tool for assessing environmental condition. By integrating the biotic component with water quality, sediment indices, and land- use information, this holistic evaluation of ecosystem health will provide management entities with information needed to inform local decision-making processes and establish benchmarks for future restoration efforts.

This document contains analytical methods that detail the procedures for determining major and trace element concentrations in bivalve tissue and sediment samples collected as part of the National Status and Trends Program (NS&T) for the years 2000-2006. Previously published NOAA Technical Memoranda NOS ORCA 71 and 130 (Lauenstein and Cantillo, 1993; Lauenstein and Cantillo, 1998) detail trace element analyses for the years 1984-1992 and 1993-1996, respectively, and include ancillary, histopathology, and contaminant (organic and trace element) analytical methods. The methods presented in this document for trace element analysis were utilized by the NS&T Mussel Watch and Bioeffects Projects. The Mussel Watch Project has been monitoring contaminants in bivalves and sediment for over 20 years, and is the longest active contaminant monitoring program operating in U.S. costal waters. Approximately 280 Mussel Watch sites are monitored on biennial and decadal timescales using bivalve tissue and sediment, respectively. The Bioeffects Project applies the sediment quality approach, which uses sediment contamination measurements, toxicity tests and benthic macroinfauna quantification to characterize pollution in selected estuaries and coastal embayments. Contaminant assessment is a core function of both projects. Although only one contract laboratory was used by the NS&T Program during the specified time period, several analytical methods and instruments were employed. The specific analytical method, including instrumentation and detection limit, is noted for each measurement taken and can be found at http://NSandT.noaa.gov. The major and trace elements measured by the NS&T Program include: Al, Si, Cr, Mn, Fe, Ni, Cu, Zn, As, Se, Sn, Sb, Ag, Cd, Hg, Tl and Pb.

Three direct plating methods and two most probable number (MPN) procedures were compared for the enumeration of Clostridium perfringens in seafoods the sulfitecycloserine (SC) agar, sulfite-polymyxin-sulfadiazine (SPS) agar, tryptone-sulfite- neomycin (TSN) agar, LS medium MPN procedure and iron milk MPN procedure. Isolates were confirmed as C. perfringens. The two MPN procedures compared very well with the three plating media tested with stock culture of C. perfringens from our laboratory collection and the reference strain NCIB 6125. But in fish samples, the two liquid media were found to be more sensitive and hence the MPN procedure using LS medium for the detection of C. perfringens in seafoods is suggested.
