The sensitivity of 61 cultures of bacteria isolated from fish towards chlortetracycline (CTC) at 5 ppm and 20 ppm levels has been determined on two solid media: sea water agar (SWA) and a distilled water based medium consisting of peptone, beef extract, glucose and NaCI (PBGA). The cultures employed consisted of (i) gram-negative rods of marine origin (Achromobacter, Pseudomonas, Vibrio and Flavobacterium) and (ii) gram positive organisms (Micrococci and Corynebacterium). Depending on the inhibition zone diameter, the order of CTC sensitivity was found to be PseudomonasCTC levels of 5 and 20 ppm. The SWA enhanced the development of resistance in majority of the cultures as compared to PBGA. PBG broth containing CTC at less than 10 ppm exerted a bacteriostatic effect only while that containing 10 ppm or more exerted a bactericidal effect on Achromobacter, Pseudomonas and Micrococci cultures. The significance of these results in fish preservation by CTC is discussed.

Quantitative and qualitative studies on the bacterial flora of fresh Indian mackerel (Rastrelliger kanagurta) have been made. The total native flora as well as 5 ppm CTC insensitive flora of the fish showed variations with season. About 90% of the fresh fish flora was sensitive to 5 ppm CTC. The natural flora of the fresh fish consisted of Vibrios, Pseudomonas, Achromobacter, Flavobacterium, Corynebacteria, Micrococci, Bacillus and yeasts. In the CTC insensitive flora, Vibrios predominated followed by yeasts. The selection of bacterial genera during storage of the fish in ice and in 5 ppm CTC incorporated ice has also been investigated. At the time of spoilage, Pseudomonas was found to be the dominant flora of the fish stored in both types of ice.

ENGLISH: We analyzed catches per unit of effort (CPUE) from the Japanese longline fishery for bigeye tuna (Thunnus obesus) in the central and eastern Pacific Ocean (EPO) with regression tree methods. Regression trees have not previously been used to estimate time series of abundance indices fronl CPUE data. The "optimally sized" tree had 139 parameters; year, month, latitude, and longitude interacted to affect bigeye CPUE. The trend in tree-based abundance indices for the EPO was similar to trends estimated from a generalized linear model and fronl an empirical model that combines oceanographic data with information on the distribution of fish relative to environmental conditions. The regression tree was more parsimonious and would be easier to implement than the other two nl0dels, but the tree provided no information about the nlechanisms that caused bigeye CPUEs to vary in time and space. Bigeye CPUEs increased sharply during the mid-1980's and were more variable at the northern and southern edges of the fishing grounds. Both of these results can be explained by changes in actual abundance and changes in catchability. Results from a regression tree that was fitted to a subset of the data indicated that, in the EPO, bigeye are about equally catchable with regular and deep longlines. This is not consistent with observations that bigeye are more abundant at depth and indicates that classification by gear type (regular or deep longline) may not provide a good measure of capture depth. Asimulated annealing algorithm was used to summarize the tree-based results by partitioning the fishing grounds into regions where trends in bigeye CPUE were similar. Simulated annealing can be useful for designing spatial strata in future sampling programs. SPANISH: Analizamos la captura por unidad de esfuerzo (CPUE) de la pesquería palangrera japonesa de atún patudo (Thunnus obesus) en el Océano Pacifico oriental (OPO) y central con métodos de árbol de regresión. Hasta ahora no se han usado árboles de regresión para estimar series de tiempo de índices de abundancia a partir de datos de CPUE. EI árbol de "tamaño optimo" tuvo 139 parámetros; ano, mes, latitud, y longitud interactuaron para afectar la CPUE de patudo. La tendencia en los índices de abundancia basados en árboles para el OPO fue similar a las tendencias estimadas con un modelo lineal generalizado y con un modelo empírico que combina datos oceanográficos con información sobre la distribución de los peces en relación con las condiciones ambientales. EI árbol de regresión fue mas parsimonioso y seria mas fácil de utilizar que los dos otros modelos, pero no proporciono información sobre los mecanismos que causaron que las CPUE de patudo valiaran en el tiempo y en el espacio. Las CPUE de patudo aumentaron notablemente a mediados de los anos 80 y fueron mas variables en los extremos norte y sur de la zona de pesca. Estos dos resultados pueden ser explicados por cambios en la abundancia real y cambios en la capturabilidad. Los resultados de un arbal de regresión ajustado a un subconjunto de los datos indican que, en el OPO, el patudo es igualmente capturable con palangres regulares y profundos. Esto no es consistente con observaciones de que el patudo abunda mas a profundidad e indica que clasificación por tipo de arte (palangre regular 0 profundo) podría no ser una buena medida de la profundidad de captura. Se uso un algoritmo de templado simulado para resumir los resultados basados en el árbol clasificando las zonas de pesca en zonas con tendencias similares en la CPUE de patudo. El templado simulado podría ser útil para diseñar estratos espaciales en programas futuros de muestreo. (PDF contains 45 pages.)

The direct measurement of in situ respiring bacteria using 5-cyano-2,3-ditolyl tetrazolium chloride (CTC) shows that, especially for Gram-negative bacteria, large numbers of viable but non-culturable (VBNC) bacteria are present in finished water from a conventional water treatment plant, and the regrowth of bacteria along distribution networks can be seen rapidly by using this very sensitive technique. The level of bacterial inactivation with chlorine is much less important than has been previously supposed (based on experiments with non-injured laboratory strains of bacteria and classical culture techniques). Threshold values of VBNC bacteria leaving water treatment plants or regrowing along distribution systems have to be determined for better control of coliform regrowth and health- risks associated with the consumption of drinking water.

Among the various antibiotics tried, tetracyclines particularly chlorotetracycline (CTC), chloramphenicol and chlorostrep were found to be fairly effective at 8 and 10 p.p.m. levels. The order of sensitivity to CTC among the six genera studied was found to be AchromobacterCTC at 10 p.p.m. level could partially inhibit this culture. The useful combinations of antibiotics for commercial application are discussed.

Effect of incorporating chlorotetracycline (CTC) in ice up to 5 ppm level on the keeping quality of prawns has been studied. A shelf life extension by nearly six days is obtained for the CTC-iced sample over the control. The headless prawns absorbed greater amounts of CTC than whole prawns during storage in CTC-ice. Traces of the antibiotic are found in the muscle of the CTC-iced prawns even after cooking for one hour. The cause of destruction of CTC when used for prawn preservation is discussed.

Effects of dips in 10 and 50 ppm Chlortetracycline solution on the storage-life of sardines in ice have been studied. 5 ppm-CTC-ice in conjunction with dip in 10 ppm CTC-solution has also been tried. CTC-treatment considerably reduced bacterial number and gave a better appearance to the fish. The high fat content in sardines resulted in rapid development of rancidity. CTC has been found to have no control on the development of rancidity. Hence, CTC treatment of sardines is found to be of limited advantage in extending the storage-life of the fish in ice.

The native flora of oil sardine and mackerel consisting of Pseudomonas spp; Moraxella spp., Acinetobacter spp. and Vibrio spp. underwent significant changes during ice storage. At the time of spoilage, Pseudomonas spp. were predominant. CTC treatment significantly reduced the Pseudomonas spp. in the initial stages of storage; but later Pseudomonas spp. reasserted and constituted the bulk of the spoilage flora. In prawn, the native flora was comprised of Pseudomonas spp., Acinetobacter spp., Moraxella spp. and Vibrio spp. At the time of spoilage a heterogeneous flora, consisting of Pseudomonas spp; Moraxella spp. and Acinetobacter spp. predominated. CTC treatment significantly changed the flora of prawns. During spoilage, Pseudomonas predominated in CTC treated prawns.
