Several local groups have come together for this project to addresses water quality concerns in the Gabilan Watershed – also known as the Reclamation Ditch Watershed (Fig. 1.1). These are Moss Landing Marine Laboratories (MLML), the Resource Conservation District of Monterey County (RCDMC), Central Coast Watershed Studies (CCoWS), Return of the Natives (RON), Community Alliance with Family Farmers (CAFF), and Coastal Conservation and Research (CC&R). The primary goal is to reduce non-point source pollution – particularly suspended sediment, nutrients, and pesticides – and thereby improve near-shore coastal waters of Moss Landing Harbor and the Monterey Bay. Research cruises were conducted in August-October 2007 to complete the third annual remotely operated vehicle (ROV)-based assessments of nearshore rocky bottom finfish at ten sites in the northern Channel Islands. Annual surveys at the Channel Islands have been conducted since 2004 at four sites and were expanded to ten sites in 2005 to monitor potential marine protected area (MPA)effects on baseline fish density. Six of the ten sites are in MPAs and four in nearby fished reference areas. In 2007 the amount of soft-only substrate on the 141 track lines surveyed was again estimated in real-time in order to target rocky bottom habitat. These real-time estimates of hard and mixed substrate for all ten sites averaged 57%, 1% more than the post-processed average of 56%. Surveys generated 69.9 km of usable video for use in finfish density calculations, with target rocky bottom habitat accounting for 56% (39.1 km) for all sites combined. The amount of rocky habitat sampled by site averaged 3.8 km and ranged from 3.3 km sampled at South Point, a State Marine Reserve (SMR) off Santa Rosa Island, to 4.7 km at Anacapa Island SMR. A sampling goal of 75 transects at all 10 sites was met using real-time habitat estimates combined with precautionary over-sampling by 10%. A total of seventy kilometers of sampling is projected to produce at least seventy-five 100 m2 transects per site. Thirteen of 26 finfish taxa observed were selected for quantitative evaluation over the time series based on a minimum criterion of abundance (0.05/100 m2). Ten of these 13 finfish appear to be more abundant at the state marine reserves relative to fished areas when densities were averaged across the 2005 to 2007 period. One of the species that appears to be more abundant in fished areas was señorita, a relatively small prey species that is not a commercial or recreational target. (PDF contains 83 pages.)

Red drum is one ofthe most popular species sought by anglers in Florida Bay, yet juveniles are rarely encountered. We evaluated Florida Bay as a nursery area for red drum by sampling for recently-settled late larvae in basin areas within the bay with an epi-benthic sled at six stations in November 2000, and at seven stations during December 2000 through February 2001. In November 2000 we surveyed potential sampling sites in quiet backwaters adjacent to mangroves for juvenile red drum. A total of 202 sites were sampled mainly in northern Florida Bay and adjacent waters with a cast net. We collected only one recently-settled red drum larvae and no juveniles. Obviously the sites that we sampled in Florida Bay and adjacent waters are not nursery habitat for this valuable species. Sled collections were dominated by bay anchovy, Anchoa mitchilli, but densities were biased by one collection. Five small resident species were among the dominant species: rainwater killifish, Lucania parva; dusky pipefish, Syngnathus floridae; dwarf seahorse, Hippocampus zosterae; and clown goby, Microgobius gulosus. Three species that spawn outside Florida Bay in the GulfofMexico were common: pinfish, Lagodon rhomboides; pigfish, Orthopristis chrysoptera; and silver perch, Bairdiella chrysoura. Twenty-seven species were collected with the cast net. Hardhead silversides (Atherinomorus stipes), bay anchovy, tidewater mojarra (Eucinostomus harengulus), silver jenny (Eucinostomus gula), and goldspotted killifish (Floridichthys carpio) were the most common in cast net collections. Although only one red drum was collected, we were able to: (1) identify mesohaline waters from our cast net sites to test our preliminary assessment that mesohaline habitat might be limited in Florida Bay, (2) document the distribution and abundance of fishes collected by cast net that should enhance our understanding of ichthyofauna in the Northern Subdivision ofFlorida Bay and adjacent waters, and (3) from epibenthic sled collections, describe the habitats, abundance and distribution of recently settled larvae/small juveniles/small resident fishes during late fall and winter. Since 1999, NOAA’s Biogeography Branch of the Center for Coastal Monitoring and Assessment (CCMA-BB) has been working with federal and territorial partners to characterize, monitor, and assess the status of the marine environment around northeastern St. Croix, U.S. Virgin Islands. This effort is part of the broader NOAA Coral Reef Conservation Program’s (CRCP) National Coral Reef Ecosystem Monitoring Program (NCREMP). With support from CRCP’s NCREMP, CCMA conducts the “Caribbean Coral Reef Ecosystem Monitoring project” (CREM) with goals to: (1) spatially characterize and monitor the distribution, abundance, and size of marine fauna associated with shallow water coral reef seascapes (mosaics of coral reefs, seagrasses, sand and mangroves); (2) relate this information to in situ fine-scale habitat data and the spatial distribution and diversity of habitat types using benthic habitat maps; (3) use this information to establish the knowledge base necessary for enacting management decisions in a spatial setting; (4) establish the efficacy of those management decisions; and (5) develop data collection and data management protocols. The monitoring effort in northeastern St. Croix was conducted through partnerships with the National Park Service (NPS) and the Virgin Islands Department of Planning and Natural Resources (VI-DPNR). The geographical focal point of the research is Buck Island Reef National Monument (BIRNM), a protected area originally established in 1961 and greatly expanded in 2001; however, the work also encompassed a large portion of the recently created St. Croix East End Marine Park (EEMP). Project funding is primarily provided by NOAA CRCP, CCMA and NPS. In recent decades, scientific and non-scientific observations have indicated that the structure and function of the coral reef ecosystem around northeastern St. Croix have been adversely impacted by a wide range of environmental stressors. The major stressors have included the mass Diadema die off in the early 1980s, a series of hurricanes beginning with Hurricane Hugo in 1989, overfishing, mass mortality of Acropora corals due to disease and several coral bleaching events, with the most severe mass bleaching episode in 2005. The area is also an important recreational resource supporting boating, snorkeling, diving and other water based activities. With so many potential threats to the marine ecosystem and a dramatic change in management strategy in 2003 when the park’s Interim Regulations (Presidential Proclamation No. 7392) established BIRNM as one of the first fully protected marine areas in NPS system, it became critical to identify existing marine fauna and their spatial distributions and temporal dynamics. This provides ecologically meaningful data to assess ecosystem condition, support decision making in spatial planning (including the evaluation of efficacy of current management strategies) and determine future information needs. The ultimate goal of the work is to better understand the coral reef ecosystems and to provide information toward protecting and enhancing coral reef ecosystems for the benefit of the system itself and to sustain the many goods and services that it offers society. This Technical Memorandum contains analysis of the first six years of fish survey data (2001-2006) and associated characterization of the benthos (1999-2006). The primary objectives were to quantify changes in fish species and assemblage diversity, abundance, biomass and size structure and to provide spatially explicit information on the distribution of key species or groups of species and to compare community structure inside (protected) versus outside (fished) areas of BIRNM. ENGLISH: A two-stage sampling design is used to estimate the variances of the numbers of yellowfin in different age groups caught in the eastern Pacific Ocean. For purse seiners, the primary sampling unit (n) is a brine well containing fish from a month-area stratum; the number of fish lengths (m) measured from each well are the secondary units. The fish cannot be selected at random from the wells because of practical limitations. The effects of different sampling methods and other factors on the reliability and precision of statistics derived from the length-frequency data were therefore examined. Modifications are recommended where necessary. Lengths of fish measured during the unloading of six test wells revealed two forms of inherent size stratification: 1) short-term disruptions of existing pattern of sizes, and 2) transition zones between long-term trends in sizes. To some degree, all wells exhibited cyclic changes in mean size and variance during unloading. In half of the wells, it was observed that size selection by the unloaders induced a change in mean size. As a result of stratification, the sequence of sizes removed from all wells was non-random, regardless of whether a well contained fish from a single set or from more than one set. The number of modal sizes in a well was not related to the number of sets. In an additional well composed of fish from several sets, an experiment on vertical mixing indicated that a representative sample of the contents may be restricted to the bottom half of the well. The contents of the test wells were used to generate 25 simulated wells and to compare the results of three sampling methods applied to them. The methods were: (1) random sampling (also used as a standard), (2) protracted sampling, in which the selection process was extended over a large portion of a well, and (3) measuring fish consecutively during removal from the well. Repeated sampling by each method and different combinations indicated that, because the principal source of size variation occurred among primary units, increasing n was the most effective way to reduce the variance estimates of both the age-group sizes and the total number of fish in the landings. Protracted sampling largely circumvented the effects of size stratification, and its performance was essentially comparable to that of random sampling. Sampling by this method is recommended. Consecutive-fish sampling produced more biased estimates with greater variances. Analysis of the 1988 length-frequency samples indicated that, for age groups that appear most frequently in the catch, a minimum sampling frequency of one primary unit in six for each month-area stratum would reduce the coefficients of variation (CV) of their size estimates to approximately 10 percent or less. Additional stratification of samples by set type, rather than month-area alone, further reduced the CV's of scarce age groups, such as the recruits, and potentially improved their accuracy. The CV's of recruitment estimates for completely-fished cohorts during the 198184 period were in the vicinity of 3 to 8 percent. Recruitment estimates and their variances were also relatively insensitive to changes in the individual quarterly catches and variances, respectively, of which they were composed. Para barcos cerqueros, la unidad primaria de muestreo (n) es una bodega de salmuera que contenía peces de un estrato de mes-área; el numero de ta11as de peces (m) medidas de cada bodega es la unidad secundaria. Limitaciones de carácter practico impiden la selección aleatoria de peces de las bodegas. Por 10 tanto, fueron examinados los efectos de distintos métodos de muestreo y otros factores sobre la confiabilidad y precisión de las estadísticas derivadas de los datos de frecuencia de ta11a. Se recomiendan modificaciones donde sean necesarias. Las ta11as de peces medidas durante la descarga de seis bodegas de prueba revelaron dos formas de estratificación inherente por ta11a: 1) perturbaciones a corto plazo en la pauta de ta11as existente, y 2) zonas de transición entre las tendencias a largo plazo en las ta11as. En cierto grado, todas las bodegas mostraron cambios cíclicos en ta11a media y varianza durante la descarga. En la mitad de las bodegas, se observo que selección por ta11a por los descargadores indujo un cambio en la ta11a media. Se uso el contenido de las bodegas de prueba para generar 25 bodegas simuladas y comparar los resultados de tres métodos de muestreo aplicados a estas. Los métodos fueron: (1) muestreo aleatorio (usado también como norma), (2) muestreo extendido, en el cual el proceso de selección fue extendido sobre una porción grande de una bodega, y (3) medición consecutiva de peces durante la descarga de la bodega. EI muestreo repetido con cada método y distintas combinaciones de n y m indico que, puesto que la fuente principal de variación de ta11a ocurría entre las unidades primarias, aumentar n fue la manera mas eficaz de reducir las estimaciones de la varianza de las ta11as de los grupos de edad y el numero total de peces en los desembarcos. El muestreo extendido evito mayormente los efectos de la estratificación por ta11a, y su desempeño fue esencialmente comparable a aquel del muestreo aleatorio. Se recomienda muestrear con este método. El muestreo de peces consecutivos produjo estimaciones mas sesgadas con mayores varianzas. Un análisis de las muestras de frecuencia de ta11a de 1988 indico que, para los grupos de edad que aparecen con mayor frecuencia en la captura, una frecuencia de muestreo minima de una unidad primaria de cada seis para cada estrato de mes-área reduciría los coeficientes de variación (CV) de las estimaciones de ta11a correspondientes a aproximadamente 10% 0 menos. Una estratificación adicional de las muestras por tipo de lance, y no solamente mes-área, redujo aun mas los CV de los grupos de edad escasos, tales como los reclutas, y mejoró potencialmente su precisión. Los CV de las estimaciones del reclutamiento para las cohortes completamente pescadas durante 1981-1984 fueron alrededor de 3-8%. Las estimaciones del reclutamiento y sus varianzas fueron también relativamente insensibles a cambios en las capturas de trimestres individuales y las varianzas, respectivamente, de las cuales fueron derivadas. The extent of rural women fisherfolks contribution to fisheries was grossly under estimated and certainly under valued. The basis of their involvement in fisheries activities is to make themselves equal partners to men productive and self reliant participants in the process of improving their own and their family living standard and to enable them realize their full potentials. This survey is informed a system of data collection with the aid of questionnaires and analysis. Ninety-six of the questionnaires were administered to women fisher folks in eight fishing communities selected at random. Thirty seven (97) percent of the women fisher folks are between the ages of 25-35 years, 59.4% of them can only read and write in Arabic language and 21.9% only are literature in western education, which is a general characteristic of the rural population in Nigeria. 24.0% of the respondents are in full time fishing activities while 65-5% are fishmongers. They belong to cooperative societies but only 26% indicated to have benefited from loan and credit facilities. 84.4% of the respondents are sustained by the business. The major problems facing women fisherfolks includes poor market price, fish spoilage, high transport cost and lack of access to loan and credit facilities and extension assistance. Solution to these problems will increase their status benefit and development

A reoccurring goal listed during the creation of Marine Protected Areas (MPAs) is to return the region to a former state. However, limited data is available that describes or characterizes this former condition. Data collected from ecosystems with comparatively limited anthropogenic impacts, can provide invaluable information in suggesting what former states may have looked like. One example is the Flower Garden Banks National Marine Sanctuary which is located 180 kilometers off the coast of Texas. These relatively isolated and pristine banks are capped by substantial scleractinian coral communities, forming excellent habitat for over 200 species of fish. While fishing is permitted, it is limited by difficulty of access. In 2006, NOAA’s Biogeography Branch, in collaboration with the Sanctuary, initiated the first quantitative assessment of fish resources throughout the diveable portions of the Sanctuary. The sampling design and methodologies employed were identical to those that the Branch has utilized in other more impacted regions of the US Caribbean. Initial analyses reveal that fish density and species richness at the Sanctuary were almost two times greater than that found within the US Caribbean and biomass was approximately six times higher. This was due in large part to the presence of sizeable piscivores of the genera Mycteroperca and Dermatolepis. The Sanctuary is one of few minimally impacted locations remaining within the Tropical Western Atlantic. As such, these findings should be considered when attempting to establish a former state or evaluate effectiveness of an MPA in meeting its management goals.

The primary objective in doing this work was to become acquainted with as many forms as possible of the marine fauna of the intertidal zone and if possible to determine some of the environmental relationships which exist in as many different types of habitats as possible. Due to limited amount of time spent in this study no very intensive work could be done and only a general survey was made of the more conspicuous forms of life which were encountered. Most of the work consisted of collecting and observing animals in the tide pools during periods of low tides. The animals collected were then taken to the laboratory and observed and determined as to species. Notes were taken as to place, time, and situation under which the animals were found. As many different types of habitats as possible were visited which included rocky intertidal areas of Mussel Point, Point Pinos, Lighthouse Point, Pescadero Point and Carmel Point just east of Carmel Beach. Sandy beaches were visited at Monterey Beach, Carmel Beach and Asilomar Beach. A marine estuary habitat was visited at Elkhorn Slough. More than two hundred species were identified and observed during this six-week period. A rather hasty population study was made of the eelgrass, Phyllospadix, of the intertidal zone at Mussel Point and of an algae, Gigartina caniculata, which grows at the level just above the eelgrass.

Frozen storage characteristics of fillets from six major species of fresh water fishes namely, Labeo rohita, Catla catla, Cirrhina mrigala, Labeo calbasu, Mystus seenghala, and Wallago attu are reported. The biochemical, bacteriological and organoleptic changes in the frozen fillets during storage at -18°C have been followed systematically. Compared to the two species of fresh water cat fishes, the four species of carps studied, had a slightly better storage life. From the organoleptic point of view, fillets of Cirrhina mrigala had the best shelf life.

The culture of Penaeus monodon has explicitly defined the need for diet formulations or supplementary feeds that would promote optimum growth and survival of the animal. A total of 28 feed combinations were developed for P. monodon. Fish meal, shrimp head meal, squid head meal, Ascetes spp. rice bran, and soybean cake were used as primary ingredients in these feeds. The commercial vitamin mix No. 22 was added to the dry ingredients. Gelatinized corn starch and wheat flour were used as binders. The pellets were extruded using a portable kitchen grinder with a diameter of 4 mm. The products were either sun-dried for 8 hours or oven-dried overnight at 50 degree C to stabilize moisture at 8-10%. The pellets were then kept in covered glass bottles and stored in the laboratory at room temperature. The cost of the feeds excluding labour were also computed. The pellets were analyzed for protein, fat, carbohydrate, crude fiber, ash, and moisture contents using standard procedures. They were also analyzed for water stability. To test the stability of pellets in water, 2-g samples were placed in plankton nets (mesh #40) and suspended in water for two, and six hours. The undissolved samples were then vacuum-dried and the moisture determined. Cost of the feeds ranged from P1.10 to P2.60 per kg depending on the feed ingredient. Squid and Ascetes spp. were rather expensive for use as basic ingredients. Proximate analysis of dry weight showed percentage protein content ranged from 20-63 g; fat, 8-20 g; carbohydrate (by difference), 11-36 g; ash, 8-28 g; moisture, 6-11 g; and crude fiber, 5 . 13 g. Stability tests showed that after two hours, 35-88% of solids remained intact and after 6 hours, 20-55% of the pellets remained undissolved. When a pellet disintegrates easily, pollution of the water occurs. Chances for the shrimp to feed on the pellet is minimized when the pellet is unstable. Thus, the search for a more compact feed pellet has to be continued.

Rice bran is widely used by fish farmers as supplementary feed while soybean cake is used both as feed and as fertilizer in fishponds. Both fish meal and shrimp head have been found acceptable as feed ingredients. However, not much is known of the acceptability and efficiency of a mixture of these ingredients as feed for Penaeus monodon larvae. Ninety 127-day old P. monodon were measured for length and weight and were randomly divided into nine aquaria each containing 20 liters of water. These were fed 'lampirong' for two months previous to the study. There were three replications for each treatment. Length, weight, and survival rates were used to compare the efficiency of the diets. Weighed amounts of pellets equivalent to 100% of the body weight were fed during the first three days and reduced to 50% thereafter. A stopwatch was used to determine the length of time that elapsed before the shrimps would approach the pellet. Ten shrimps approximately 4 months in age were placed in 10 liters of water in a 25-liter aquarium. Two grams of each pellet type were placed simultaneously on opposite sides of the aquarium. The time that elapsed from the moment the pellets sunk to the bottom up to the time that any one shrimp approached the pellets was recorded. The group fed the imported pellets gained the most. Those fed FP-2s-77 elongated faster than those fed FP-1s-77. Survival rate of those fed FP-2s-77 was 37% while those fed imported pellets was 73%. Both 1s and 2s pellets disintegrated in water easily but the imported pellets were stable even after six hours in water. The attractability test for the pellets showed that the prawns were more readily attracted to the pellets 1s and 2s than to the imported pellets.

Aquaculture has been expanded rapidly to become a major commercial and food-producing sector worldwide in recent decade. In parallel, viral diseases rapidly spread among farms causing enormous economic losses. The accurate detection of pathogens at early stages of infection is a key point for disease control in aquaculture. Spring Viraemia of Carp Virus (SVCV) is a very severe pathogen of carp fishes in different parts of the world and is categorized as a reportable listed disease in the annual published list of World Organization for animal Health (OIE). The objective of this study was to develop and evaluate RT- PCR test for detecting SVC virus and also the sensitivity and specificity of this test. A semi nested RT- PCR was designed using combination of three primers: two external (SVCF , SVCR) and one internal (SVCS) primers which based on conserved region of G gen. The specificity of designed primers (only external ones) by examination on Viral Hemorrhagic Septicemia Virus (VHSV) and Infectious Hematopoietic Necrosis Virus (IHNV) was confirmed. For optimizing of the PCR test, primer concentration, primer annealing temperature, cycle number and Mgcl2 concentration were surveyed. Also for validity test, prevention of false negative and Assurance of its accuracy, a competitive internal control (mimic) designed and its suitable concentration was defined. Evaluation of the sensitivity of designed test were conducted first by comparing the different commercially available RNA isolation guidelines, two guidelines: isotiocyanate phenol–chloroform based protocols (RNX–Plus Iran, Iq2000 kit Taiwan ) and two column based protocols (Cinna pure RNA Iran , high pure viral RNA kit, Roche Germany ). The results indicated that the column based protocols (Roche method and Cinna pure), yield 36.77 ng/μl and 16/47 ng/μl RNA concentration respectively, which were significantly higher than other protocols(P<0.05). Then for evaluation of extracted RNA sensitivity, Serial dilution of SVCV strain 56.70 grown in EPC (1.9×105 TCID50/ml) was examined To compare sensitivity. Extracted RNA from serial dilution with stone's primers and commercial IQ-2000 kit were examined simultaneously. The result indicated that designed semi- nested RT- PCR was able to recognize SVC virus to 10-4 dilution and stone's primer recognize to 10-3 dilution whereas Iq-2000 commercial kit did not recognized in any dilution. In high virus titer in designed test two DNA band (462 bp and 266 bp) produced, and by decreasing virus titer 462 bp was omitted. In low virus titer or lack of virus, just DNA band (mimic) 729 bp can propagate. After designing and optimizing PCR test, a total of 400 suspected cultured Cyprinus carpio with high mortality from 4 aquaculture zone of Khuzestan province were collected and tested for SVCV during 2012- 2013 using developed PCR method and IQ- 2000. The results indicated that SVC virus was not observed in samples using both methods.
