Histopathologic studies of lesions found in commercially important North Atlantic marine fishes are uncommon. As part of a comprehensive Northeast Fisheries Center program ("Ocean Pulse") to evaluate environmental and resource health on the U.S. Continental Shelf from Cape Hatteras to Nova Scotia, grossly visible lesions of the gills, integument, muscle, and viscera of primarily bottom-dwelling fishes were excised and examined using light microscopy. Several gadid and pleuronectid fishes accounted for most of the lesions observed. Most pathological examinations were incidental to samples taken for age and growth determination and evaluation of predator/prey relationships. Several gadids, with either gill, heart, or spleen lesions, were sampled more intensively. Gill lesions principally affected gadids and were caused by either microsporidans or an unidentified oocyte-like cell. The majority of gastrointestinal lesions consisted of encapsulated or encysted larval worms or microsporidan-induced cysts. Few heart lesions were found. Integumental lesioos included ulcers, lymphocystis, and trematode metacercariae. Liver lesions almost always consisted of encapsulated or encysted larval helminths. Necrotic granulomata were seen in muscle and microsporidan-induced granulomata in spleen. Although not numerous, histologically interesting lesions were noted in integument, heart, liver, spleen, and muscle of several fish species. Histologic study of tissues excised from a variety of demersal and pelagic fishes from the eastern North Atlantic (France, Germany, Spain) revealed assorted integumental, renal, hepatic, and splenic lesions. Small sample size and non-random sampling precluded obtaining a meaningful quantitative estimate of the prevalence of the observed lesions in the population at risk; however, a useful census has been made of the types of lesions present in commercially important marine fishes. Abrasion, feeding, injection and immersion methods were used to evaluate the pathogenicity of five different strains of Aeromonas hydrophila viz. RG (rui gill), ML (mrigal lesion), SG (sharpunti gill), F1K (mrigal kidney), GFL (gold fish lesion) and Ah-19 (Aeromonas hydrophila-19, Ref. Strain) against C. mrigala H. Bacterial suspension containing viable cells of 7.5x 10⁵ per ml was found to be very effective in intramuscular injection and feeding resulting 100% mortality after 96hr of inoculation. The strain RG, ML and F1K produced scale loss with erosion of the skin surface with/without hemorrhagic lesion after 48hr of inoculation following abrasion method. The strains SG and Ah-19 resulting scale loss with erosion of the skin surface with/without hemorrhagic lesion after 72hr of inoculation following abrasion and injection methods. SG and F1K caused reddening in mouth region after 72hr of feeding inoculation, whereas RG resulted frank ulcers from eroded dermal layer exposing underlying musculature which was hemorrhagic after 96hr of inoculation by abrasion method.

Bacterial flora associated with tail rot/fin rot of Carassius auratus, Xiphophorus helleri and hemorrhagic ulcers of Clarias spp were studied. Sensitivity pattern of 33 isolates comprising Aeromonas spp, Pseudomonas spp and Gram-positive rods from diseased C. auratus, X. helleri and Clarias spp were screened against six broad-spectrum antibiotics viz. ciprofloxacin, chloramphenicol, co-trimoxazole, gentamycin, nitro-furantoin and oxytetracycline. Ciprofloxacin was the most effective in inhibiting bacteria at 0.05-0.10 µg/ml level. About 44% of Pseudomonas spp. was resistant to nitrofurantoin. Resistance to oxytetracycline was seen in 27% of Aeromonas spp Gram-positive rods were comparatively more resistant to antibiotics. The multiple antibiotic resistances were seen in 21% of the bacterial isolates of diseased fish.

Motile Aeromonas are the most common bacteria of freshwater in the world that cause disease in fish and other cold-blooded and warm-blooded hosts. Among this group of bacteria, Aeromonas hydrophila is important in causing complications such as fin rot, skin ulcers and lethal hemorrhagic septicemia in fish. Several virulence factors involved in the pathogenesis of Aeromonas hydrophila, including extracellular enzymes (protease, lipase, elastase, gelatinase and nuclease) and toxins. From the exotoxins, hemolysin, aerolysin and cytolytic enterotoxin play an important role in pathogenesis. Detection of virulence markers by PCR as a key component of determining the pathogenesis of the bacteria and using indigenous vaccines for better immunization against this disease is important. In this study, a total of 200 fanned carps (126 common carp. 39 silver carp and 35 of grass carp) with symptoms suspected aeromonas septicemia were isolated from Khouzestan province farms. 125 bacteria belong to Aeromonas genus detected by biochemical and PCR methods. 31 of all isolates recognized as Aeromonas hydrophila with biochemical methods, I6srRNA detection and Lipase genes. Results showed that the role of Aeromonas sp. and Aeromonas hydrophila in fish with disease symptoms were 62.5% and 15.5% respectively. By using specific primers, three virulence genes including hemolysin, aerolysine and cytolytic enterotoxin were detected in these confirmed isolates, that 18 isolates (58/06%) hemolysin positive (hlyA +), 16 isolates (51/61%) aerolysine positive (aerA+) and 23 isolates (74/19%) for cytolytic enterotoxin gene (act+) were positive. The result of present study showed that most of the confirmed isolates genotype was hlyA+ act- with frequency equal to 51/61%. For investigating the protection effect of acut strain of bacteria, UV inactivated bacterin was used.
