Three enrichment broths and six plating media were compared for efficiency of detection Salmonella in the presence of numbers of Coliforms (10super(5)/ml) and proteus (10super(3)/ml) from artificially inoculated fish samples. Recovery experiments Salmonella anatum, S. typhimurium and S. enteritidis indicated that the two enrichment broths Dulcitol Selinite (DSE) and Selinite Cystine (SC) were equally efficient. Further, the viability of Salmonella, inoculated into fish muscle and kept at 4°C for 48 hours, was found to be not affected by the low temperature storage. Selective plating media like Xylose Lysine Deoxycholate agar (XLD), Brilliant Green Sulphadiazine agar (BGS) and Brilliant Green agar (BG) were found to be superior in performance to Salmonella-Shigella agar: (SS) and Bismuth Sui phite agar (BiS).

The paper deals with the collection of gall bladders, isolation of bile and identification of the constituents of the bile salts from different fishes. The yield of bile contents from fresh water fishes rohu, mrigal and catla was compared with that from marine fishes seer, tuna, shark and sardine. Considerable variation in yield was showed between marine and fresh water fish as well as between the species in both groups. It ranged from 0.04 to 0.06% of the body weight of fish in calla, mrigal and rohu. The bile constituents from rohu and mrigal were analysed by thin layer chromatography. The result showed that bile of rohu and mrigal contains mainly taurine derivative of lithocholic acid.

ENGLISH: The spawning of yellowfin tuna (Thunnus albacares) in the eastern Pacific Ocean was examined to ascertain the existence of separate subpopulations within this area. Investigations of biochemical genetics of yellowfin indicate that there are a number of genetically distinct groups in the eastern Pacific. In addition, yellowfin belong to two recruitment cohorts, X and Y, which are composed of a mixture of these genetically different groups. Spawning data were collected from 1956 through 1961 from the coastal fishing grounds, and from 1970 through 1973 from the offshore fishing areas. Temporal and spatial aspects of spawning of the fish of the two cohorts were analyzed to determine if yellowfin spawning behavior supports the existence of genetically separate subpopulations. Spawning condition was inferred from the maturity of the ovaries. It was found that the coastal fish of each cohort exhibit at least two spawning periods per year which vary in length and time of occurrence from year to year. Fish taken from the offshore fishing grounds did not exhibit this variable spawning pattern. Although samples were not available for all months, the data showed that each cohort has a spawning period of at least 7 months and may spawn year around. Samples from offshore areas also had much higher percentages of spawners than those from the coastal areas. Temporal differences in spawning are not maintaining the genetically separate groups found in the fishery, since fish of both recruitment cohorts spawn at the same time. Also, fish of both the X and Y cohorts spawned in all areas examined; however, these data are insufficient to determine whether spatial isolation of spawning groups is occurring within the areas. High suspended sediment loads may be deleterious to adult salmonids and invertebrates in gravel-bedded streams. Further, the accumulation of fine material in the interstices of the gravel may have an adverse impact on the recruitment of the young stages of salmonids. It is important therefore not only to quantify the rates and degrees of silting but also to identify sediment sources and to determine both, the frequency of sediment inputs to the system and the duration of high sediment concentrations. This report explores the application of variance spectrum analysis to the isolation of sediment periodicities. For the particular river chosen for examination the method demonstrated the essentially undisturbed nature of the catchment. The regulated river chosen for examination is the River Tees in Northern England. Variance spectrum analysis was applied to a series of over 4000 paired daily turbidity and discharge readings.

Genetic variation of Contracaecum ogmorhini (sensu lato) populations from different otariid seals of the northern and southern hemisphere was studied on the basis of 18 enzyme loci as well as preliminary sequence analysis of the mitochondrial cyt b gene (260 bp). Samples were collected from Zalophus californianus in the boreal region and from Arctocephalus pusillus pusillus, A. pusillus doriferus and A. australis from the austral region. Marked genetic heterogeneity was found between C. ogmorhini (sensu lato) samples from the boreal and austral region, respectively. Two loci (Mdh-2 and NADHdh) showed fixed differences and a further three loci (Iddh, Mdh-1 and 6Pgdh) were highly differentiated between boreal and austral samples. Their average genetic distance was DNei = 0.36 at isozyme level. At mitochondrial DNA level, an average proportion of nucleotide substitution of 3.7% was observed. These findings support the existence of two distinct sibling species, for which the names C. ogmorhini (sensu stricto) and C. margolisi n. sp., respectively, for the austral and boreal taxon, are proposed. A description for C. margolisi n. sp. is provided. No diagnostic morphological characters have so far been detected; on the other hand, two enzyme loci, Mdh-2 and NADHdh, fully diagnostic between the two species, can be used for the routine identification of males, females and larval stages. Mirounga leonina was found to host C. ogmorhini (s.s.) inmixed infections with C. osculatum (s.l.) (of which C. ogmorhini (s.l.) was in the past considered to be a synonym) and C. miroungae; no hybrid genotypes were found, confirming the reproductive isolation of these three anisakid species. The hosts and geographical range so far recorded for C. margolisi n. sp. and C. ogmorhini (s.s.) are given.

The continental shelf adjacent to the Mississippi River is a highly productive system, often referred to as the fertile fisheries crescent. This productivity is attributed to the effects of the river, especially nutrient delivery. In the later decades of the 2oth century, though, changes in the system were becoming evident. Nutrient loads were seen to be increasing and reports of hypoxia were becoming more frequent. During most recent summers, a broad area (up to 20,000 krn2) of near bottom, inner shelf waters immediately west of the Mississippi River delta becomes hypoxic (dissolved oxygen concentrations less than 2 mgll). In 1990, the Coastal Ocean Program of the National Oceanic and Atmospheric Administration initiated the Nutrient Enhanced Coastal Ocean Productivity (NECOP) study of this area to test the hypothesis that anthropogenic nutrient addition to the coastal ocean has contributed to coastal eutrophication with a significant impact on water quality. Three major goals of the study were to determine the degree to which coastal productivity in the region is enhanced by terrestrial nutrient input, to determine the impact of enhanced productivity on water quality, and to determine the fate of fixed carbon and its impact on living marine resources. The study involved 49 federal and academic scientists from 14 institutions and cost $9.7 million. Field work proceeded from 1990 through 1993 and analysis through 1996, although some analyses continue to this day. The Mississippi River system delivers, on average, 19,000 m3/s of water to the northern Gulf of Mexico. The major flood of the river system occurs in spring following snow melt in the upper drainage basin. This water reaches the Gulf of Mexico through the Mississippi River birdfoot delta and through the delta of the Atchafalaya River. Much of this water flows westward along the coast as a highly stratified coastal current, the Louisiana Coastal Current, isolated from the bottom by a strong halocline and from mid-shelf waters by a strong salinity front. This stratification maintains dissolved and particulate matter from the rivers, as well as recycled material, in a well-defined flow over the inner shelf. It also inhibits the downward mixing of oxygenated surface waters from the surface layer to the near bottom waters. This highly stratified flow is readily identifiable by its surface turbidity, as it carries much of the fine material delivered with the river discharge and resuspended by nearshore wave activity. A second significant contribution to the turbidity of the surface waters is due to phytoplankton in these waters. This turbidity reduces the solar radiation penetrating to depth through the water column. These two aspects of the coastal current, isolation of the inner shelf surface waters and maintenance of a turbid surface layer, precondition the waters for the development of near bottom summer hypoxia.

Isolation of Lactic Acid Bacteria (LAB) was carried out from gastro intestinal tract of beluga and Persian sturgeon at international sturgeon research institute and PCR has been used for bacteria Identification. Two species of LAB including Enterococcus seriolicida and Leuconostoc mesenteroides were isolated from Gastrointestinal tract (GI) of persian sturgeon in this study and the counts of Leu. mesenteroides (4.63×102 CFU/gr of GI) was significantly higher than other species. Lactobacillus curvatus, Lactococcus raffinolactis, Lactococcus lactis and Streptococcus sp. were also isolated from GI of beluga and maximum counts was belonged to Lb. curvatus (4.63×102 CFU/gr of GI) in this species. Dominant species were lyophilized and adding to the water since start of mix feeding of sturgeon with different counts including 2×109, 5×109 and 9×109 CFU/gr of live food, 4 times a day. The results revealed that the maximum and minimum growth rate and protease, amylase, and lipase activity in beluga was gained by using of Lb. curvatus with total viable count of 9×10 9 CFU/gr of live food and Leu. mesenteroides with total viable count of 9×109 CFU/gr of live food. According to the results of this study, the maximum and minimum growth rate and protease, amylase, and lipase activity in Persian sturgeon was gained by using of Leu. mesenteroides with total viable count of 2×10 9 CFU/gr of live food and Lb. curvatus with total viable count of 9×109 CFU/gr of live food. Histological study showed that gastrointestinal development was same during larva rearing in control and other treatments but the size of liver was bigger in treatments that received nonspecific LAB in both species. According to the results, positive effects of using dominant specific LAB bacteria for larviculture of sturgeon has been proved in this study.

Moon oxygenases related to cytochrome P450s are the molecular Biomarkers which have important role in Biotransformation of endogenous and exogenous compounds and catalazyin of many biological reactions. One of the important isoenzyme is cytochrome P4501A. This isoenzyme involved in metabolism of environment pollutnts such as PAHs. Because of its inducibility, it has a key tool for impact assesment of contaminants in aquatic environment. In this study, at first, that fractions containing Acipenser persicus and Huso huso isoenzyme were purified, and after that Antibodies against them were prepared. For isolation of isoenzyme fraction, Microsomes were prepared from fish liver using differential centrifugation at high speeds. microsomes were solubiized by cholat sodium and Emulgen. Extraction of this isoenzyme was done with the combinatuion of ionexchange chromatography and gelfiltration or chromatofocusing chromatography. Ion exchange chromatography and gel filtration were applied in DEAE sepharose fast flow and sephacryl S200 respectively and chromatofocusing was done at poly buffer 74 and 94 exchanger. The results of SDS-PAGE Showed that the molecular weight of isoenzyme was about 58±1 KDa. Furthermore the inmunoblotting results confirmed this subject. Isoenzyme activigy based on EROD (Ethoxyresorofin o-deethylase) reaction showed about 20-26 fold increase in enzyme activity of treated fish than control fish. The results of Elisa, Using monoclonal anti cod P4501A demonstrated the inducibility and highly elevated of its activity in treated sample more than the control fish. Mean while, the fish sample were showed the strong reaction to polyclonal antibody against beluga P4501A1 prepared in our Lab compared to monoclonal anti body.
