ENGLISH: Tagging and the recovery of tagged yellowfin (Thunnus albacares) and skipjack (Katsuwonus pelamis) tunas are important aspects of the investigations conducted by the Inter-American Tropical Tuna Commission in the Eastern Tropical Pacific Ocean. The results of the tagging program provide information on population structures, migrations, mortality rates and growth rates of these two species. The present experimental program was undertaken to study the relationship between muscular fatigue and high tagging mortalities in yellowfin and skipjack. SPANISH: La marcación del atún aleta amarilla (Thunnus albacares) y del barrilete (Katsuwonus pelamis), y el recobro de estos atunes marcados, son aspectos importantes de la investigación que efectúa la Comisión Interamericana del Atún Tropical en el Océano Pacífico Oriental Tropical. Los resultados del programa de marcación proporcionan información sobre la estructura de las poblaciones, migraciones, tasas de mortalidad y tasas de crecimiento de estas dos especies. El programa experimental presente fue emprendido para estudiar la relación entre la fatiga muscular y la alta mortalidad causada por la marcación en el atún aleta amarilla y el barrilete. Oreochromis niloticus was subjected to transportation stress to investigate hepatic glycogen levels and mortality as indices of stress. Mortalities lasted up to three days after transportation, except in highly aerated samples. Heptic glycogen levels in transported fish were significantly lower in the controls. Stress appeared to be more intense when fish were transported at a high density and in a high salinity medium.

This dissertation: 1) determines the factor(s) responsible for spawning induction in NematosteJla vectensis; 2) isolates, describes, and documents the source of jelly from egg masses of N. vectensis; and 3) describes N. vectensis' early development. Namatostella vectensis were maintained on a 7-day mussel feeding/water change regime over 159 days. Within 36 hours of mussel feeding/water change. 69.1% of females and 78.5% of males spawned reliably. Through manipulation of feeding, water change, oxygen and nitrogenous waste concentrations, spawning induction was found to be triggered by the oxygen concentration associated with water change, and not by feeding. Ammonia, anemones' major waste product, inhibited this induction in a concentration-dependent manner. Female N. vectensis release eggs in a persistent jellied egg mass which is unique among the Actiniaria. The major component of this egg mass jelly was a positive periodic acid-Schiffs staining, 39.5-40.5 kD glycoprotein. Antibodies developed in rabbits against this glycoprotein bound to jelly of intact egg masses and to granules (~ 2.8 IJm in diameter) present in female anemone mesenteries and their associated filaments. Antibodies did not label male tissues. Nematostella vecfensis embryos underwent first karyokinesis -60 minutes following the addition of sperm to eggs. Second nuclear division took place, followed by first cleavage, 90-120 minutes later. Each of the 4 blastomeres that resulted from first cleavage contained a single nucleus. Arrangement of these blastomeres ranged from radial to pseudospiral. Embryonic development was both asynchronous and holoblastic. Following formation of the 4-cell stage, 71% of embryos proceeded to cleave again to form an 8-cell stage. In each of the remaining 29% of embryos, a fusion of from 2-4 blastomeres resulted in 4 possible patterns which had no affect on either cleavage interval timing or subsequent development. The fusion event was not due to ooplasmic segregation. Blastomeres isolated from 4-celled embryos were regulative and developed into normal planula larvae and juvenile anemones that were 1/4 the size of those that developed from intact 4-celled embryos. Embryos exhibiting the fusion phenomenon were examined at the fine structural level. The fusion phenomenon resulted in formation of a secondary syncytium and was not a mere compaction of blastomeres.

The procedure to conduct horizontal starch gel electrophoresis on enzymes is described in detail. Areas covered are (I) collection and storage of specimens, (2) preparation of tissues, (3) preparation of a starch gel, (4) application of enzyme extracts to a gel, (5) setting up a gel for electrophoresis, (6) slicing a gel, and (7) staining a gel. Sierra Leone is a tropical country where water temperatures are high throughout the year. Consequently the local oysters tend to spawn the year round, with one or two spawning peaks. The condition of such tropical oysters may not be as high as those oyesters in temperate countries since the stored glycogen is regularly utilized to form gonads. A high condition factor value indicates that the oysters have accumulated glycogen and or gonads, whereas a low condition factor value indicates that the oysters have spawned and are in the process of accumulating glycogen, which may later be utilized for gonad development. In oyster culture, condition factor studies may be supported by plankton and oyster spat settlement studies in the culture area. These studies give an indication of when oyster larvae and spat settlement are at their peak values. In Sierra Leone studies of the plankton and spat settlement are undertaken every week throughout the year. Conditions factor is obtained from the ratio weight of dry (oyster) meat x 1000/internal volume. Detailed condition factor values are shown in relation to salinity at two stations. Condition factor declines with reducing salinity, which principally occurs during the rainy season. The best times to collect spat are May to June and September to October

Recent histochemical and histophysiological research on the skin of fish has posed interesting problems both with regard to the presence of specialized types of cell and with regard to the function of this organ. The present paper intends to study the development and the functional differentiation of the trout's skin, both from the histomorphological and the histochemical point of view. The skin of Salmonids is devoid of specialized cells; the granular cells of Petromyzonti, the serous cells of Selacii and the clavate cells described in many other teleosts and it lacks keratin. As such it can be considered a good working model which can be used to show the eventual histomorphological and histochemical changes occurring both in the transformation from the endovular to the aqueous environment. Histomorphological observations were carried out demonstrate the great structural simplicity of the trout's skin at all stages. The article concludes that glycogen increases greatly when the epidermis thickens and therefore when it becomes necessary to guarantee resistance, amongst which is a support mechanism.

Most microbiological methods require culture to allow organisms to recover or to selectively increase, and target organisms are identified by growth on specific agar media. Many cultural methods take several days to complete and even then the results require confirmation. Alternative techniques include the use of chromogenic and fluorogenic substances to identify bacteria as they are growing, selective capture using antibodies after short periods of growth, molecular techniques, and direct staining with or without flow cytometry for enumeration and identification. Future microbiologists may not use culture but depend on the use of specific probes and sophisticated detection systems.

This chapter describes the procedures for determining the reproductive stage of oysters, mytilid mussels, and dreissenid mussels collected for NOAA’s National Status and Trends Mussel Watch Project. Analyses are conducted on paraffin-embedded tissues sectioned at a 5-μm thickness and stained using a pentachrome staining procedure. Each slide is examined microscopically to determine the animal’s sex and stage of gonadal development. A semi-quantitative ranking is assigned.

Living specimens of Barilius bendelisis, Crossocheilus latius latius, Torputitora, Glyptothorax pectinopterus and Pseudecheneis sulcatus were collected from the streams and rivers of Garhwal Himalaya. Histochemical localization of carbohydrates (1:2 glycol groups, glycogen, B-metachromasia and acid mucopolysaccharides), proteins and protein bound NH sub(2) group, bound lipids and the enzymes (acid and alkaline phosphatases) in the olfactory epithelium were studied. The receptor, supporting basal and mucous cells show varying degrees of distinction and distribution of these substances. The enzymes are present on the surface of the epithelium, sensory hairs and the boundaries of mucous cells only.

Some biochemical variations during ovarian vitellogenic growth in hill-stream teleost Garra mullya due to sublethal concentration of cadmium has been discussed. Total protein, cholesterol and glycogen in ovary and liver along with gonadosomatic index (GSI) and hepatosomatic index (HSI) in Cd-treated fish exhibited significant decrease while liver glycogen remained unaltered.

Preliminary investigations to study the lunar, diurnal and tidal periodicity in abundance and migrations of prawns were made in the Bairavapalam distributary of the Goutami branch of the Godavari estuary during the period of November 1961 to July 1962. The study was based on observation of the catches of a stake net (bag net) operated near the mouth of the estuary. Records of the catches were maintained tidewise daily. Comparative estimates of abundance were made on the basis of catch per hour's operation. It was generally observed that the catches were higher during the darker half of the month than in the brighter fortnight. The landings during low tides were generally higher than those during high tides and usually heavier catches were made during nocturnal low tides than during the day low tides. A continuous inward and outward (immigration and emigration) movement of prawns of all size groups was observed in the estuary from November to July. In general, more penaeid prawns were found to be immigrating at dawn than at dusk. Similarly, the number of emigrants was also found to be generally higher during the new moon period than during the full moon. Metapenaeus monoceros showed an almost distinct nocturnal periodicity in migration, while no such periodicity was observed, distinctly, in other species. In the case of Penaeus indicus the movement of migrants was prolonged. In M. brevicornis the migrants were scarce till March and thereafter increased numerically. The migrant forms of Metapenaeus dobsoni continued to be abundant till May with peak periods in January and February. The migratory pattern of Metapenaeus affinis was similar to that of Metapenaeus brevicornis, though the migrants of the former species appeared a month earlier than the latter. Intensive studies over extended areas for longer periods are required to understand clearly the migratory pattern of the various species. The phenomenon of immigration of prawns can be clearly understood only by vital staining or tagging studies. Perhaps the emigrants might be returning with the succeeding changing life. To verify this, laboratory experiments, by vital stains, were conducted. The marked specimens, if released during the low tides on a large scale, may be recaptured during the subsequent high tides and the duration also may be calculated. At least some percentage of the emigrants remains in the sea for maturity and breeding.

The object of this study was to determine the value of physical, bacteriological and chemical tests used to find out and compare the indices of quality of prawns stored at 0°C and at 18°C. pH value, nature of drip, the total bacterial count, presence or absence of tryptophan, trimethylamine content, glycogen, lactic acid, vitamins such as thiamin, riboflavin and niacin were estimated periodically to evaluate the quality of prawns stored at 0°C, whereas in addition to organoleptic changes, pH, bacterial count, nature of growth in peptone water, soluble protein in salt solution and loss of moisture, glycogen, lactic acid, and changes in vitamin B contents were noted periodically for prawns stored at -l8°C. Riboflavin and niacin were not affected appreciably but the retention of thiamin in prawns was very low.

The distribution pattern of exocrine pancreas in Labeo rohita besides its general location along the course of intestinal mesentery was studied. It is evenly distributed within the liver around portal vessels and also within the spleen near a blood vessel. On ultrastructure, two cell types of different degrees of staining intensities containing abundant rough endoplasmic reticulum, mitochondria, pre-zymogen and zymogen granules were marked. During aflatoxicosis, the mesenteric pancreas and hepatic pancreas were mostly affected revealing necrotic changes to acini. The zymogen granular activities were markedly reduced. Ultra structurally, the rough endoplasmic reticulum was fully dilated and formed whorled pattern. The damage to the exocrine pancreas might be affecting digestive enzymes' secretion which may be one of the causes of aflatoxin-induced anorexia in fish.

An outbreak of saprolegniasis in Catla catla in composite carp culture ponds were recorded during winter season. The typical cotton wool growths were observed on whole body surfaces of catla along with sporadic mortality. The fungal invasion was only restricted to skin and no fungal elements were visible in any internal organs after periodic acid schiff staining. On histology, periportal accumulation of mononuclear cells in liver, presence of myxosporidean cysts in antieror kidney, eosinophilic granular cells reaction in submucosa of stomach and intestine, dilated and engorged blood vessels of brain along with sloughing of epidermis and hyperplasia at gill lamellar base were pronounced changes. The possible role of release of Saprolegnia toxin in producing internal organs pathology has been discussed.

Rate and pattern of spoilage of some of the economically important edible species of shell fishes Mytilus edulis (Mussel), Villorita cornucopia (Clam), Neptunus pelagicus (Crab) and Scylla serrata (Crab) have been discussed in this communication. Chemical indices used for objective evaluation of quality were water extractable nitrogen (WEN), non-protein nitrogen (NPN), free α-amino nitrogen (α - NH2 -N), glycogen, lactic acid and inorganic phosphorus in addition to the subjective tests. No significant difference in the spoilage pattern of the species during ice storage was observed and these species could be preserved in ice in organoleptic acceptable condition up to 8 days, 9 days, 8 days and 11 days respectively.
