The purpose of the investigation is to make a detailed study of the geology and ground water in the Ruskin area, especially as related to the problem of salt-water encroachment. The major objectives of the program includes: (1) An inventory of wells to determine their number and distribution, their depths and diameters, and other pertinent information. (2) A study of artesian pressures. (3) Analyses of water from selected wells to determine the location and extent of any areas in which the artesian water is salty. (4) A study of the surface and subsurface geology as related to the occurrence and movement of ground water. (5) An estimate of the quantity of ground water withdrawn. (PDF contains 24 pages.)

A large part of western Manatee County is devoted to the growing of winter vegetables and citrus fruits. As in most of peninsular Florida, rainfall in the county during the growing season is not sufficient for crop production and large quantites of artesian water are used for irrigation. The large withdrawals of artesian water for irrigation result in a considerable decline of the artesian head in the western part of the county. This seasonal decline of the artesian head has become larger as the withdrawal of artesian water has increased. The lowering of the fresh-water head in some coastal areas in the State has resulted in an infiltration of sea water into the water-bearing formations. The presence of salty water in the artesian aquifer in parts of the coastal area of Manatee County indicates that sea water may also have entered the waterbearing formations in this area as a result of the decline of artesian pressure during the growing season. The purpose of the investigation is to make a detailed study of the geology and ground-water resources of the county, primarily to determine whether salt-water encroachment has occurred or is likely to occur in the coastal area. A detailed study of the geology and ground-water resources of the Ruskin area (fig. 1) was made during the period from 1950 to 1955, by the U. S. Geological Survey in cooperation with the Florida Geological Survey and the Board of County Commissioners of Hillsborough County. The results of this study are given in a report by Harry M. Peek entitled "The artesian water of the Ruskin area of Hillsborough County, Florida" and published by the Florida Geological Survey as Report of Investigations No. 21. This report contains tables of well records that were compiled from data collected during that investigation. The well-numbering system used in the tables is based on latitude and longitude. This report describes FORTH software written for several instruments used in the NASA-sponsored project to design and build Marine Optical Buoy System (MOBS) and in the NOAA-sponsored project "EOS MODIS Execution: Oceanographic Profiling, Data Acquisition and Management for the Marine Optical Buoy System·'. In the NOAA project MLML and NOAA personnel will participate in quarterly cruises at the MOBS Hawaiian site to validate performance of SeaWiFS and will participate in several extended "process" cruises to provide wide geographic surface truthing investigations similar to those lead by Dennis Clark (NOAA) following the "launch of CZCS in 1979. In the NASA project we are designing and building MOBS, a high resolution spectroradiometer that will operate autonomously in a buoy moored west of Lanai in the Hawaiian Islands. That instrument, the "Marine Optical System" (MOS), will transmit by cellular phone in near real time observations of upwelled radiance and downwelled irradiance from three depths. Rockfish (Sebastes spp.) juveniles are often difficult to identify by using morphological characters. This study independently applies morphological characters and a key based on mitochondrial restriction site variation to identify juvenile rockf ishes collected in southern California during juvenile rockfish surveys. Twenty-four specimens of Sebastes were examined genetically without knowledge of the morphological assignment. Seventeen fish were identified genetically as S. semicinctus, S. goodei, S. auriculatus, S. jordani, S. levis, S. rastrelliger, and S. saxicola. Identities for the remaining fish were narrowed to two or three species: 1) three fish were either S. carnatus or S. chrysomelas; 2) one fish was either S. chlorosticus, S. eos, or S. rosenblatti; and 3) three fish could have been either S. hopkinsi or S. ovalis, the latter for which we now have distinguishing mitochondrial markers. The genetic and morphological assignments concurred except for the identity of one fish that could only be narrowed down to S. hopkinsi or S. semicinctus by using morphological characters. Genetics excluded more species from multispecies groupings than did the morphological approach, especially species within the subgenus Sebastomus. Species in the genetically unresolvable groups may be similar because of recent divergence or because of interspecies introgression.

Larval and juvenile rockfishes (Sebastes spp.) are difficult to identify using morphological characters. We developed a key based on sizes of restriction endonuclease fragments of the NADH dehydrogenase-3 and -4 (ND3/ND4) and 12S and 16S ribosomal RNA (12S/16S) mitochondrial regions. The key makes use of variation in the ND3/ND4 region. Restriction endonuclease Dde I variation can corroborate identifications, as can 12S/16S variation. The key, based on 71 species, includes most North American taxa, several Asian species, and Sebastolobus alascanus and Helicolenus hilgendorfi that are closely related to rockfishes. Fifty-eight of 71 rockfish species in our database can be distinguished unequivocally, using one to five restriction enzymes; identities of the remaining species are narrowed to small groups: 1) S. polyspinis, S. crameri, and S. ciliatus or variabilis (the two species could not be distinguished and were considered as a single species) ; 2) S. chlorostictus, S. eos, and S. rosenblatti; 3) S. entomelas and S. mystinus; 4)S. emphaeus, S. variegatus, and S. wilsoni; and 5) S. carnatus and S. chrysomelas.

The first studies on the problems related to the reproduction of marine fish in Argentina (SW Atlantic ocean) have been carried out with a species of great economic importance, the anchovy (Engraulis anchoita Hubbs and Marini). The spawning period of this species during the period 1963-64 has been determined. By the application of the quantitative collection method, it has been established that spawning commenced in the areas close to the coast during the first few days of September and at a water temperature of 10,3° C and reached its greatest intensity in October (up 1569 eggs/1 square metre surface water) at a temperature varying between 11,5-13,8° C. From the middle of November the anchovy continues to reproduce in a less intensive form and further out to Sea, up to at least the month of May. The intensity of reproduction reaches a peek in February, and at a water temperature of 20° C. It is believed that two physiologically distinct populations of Engraulis anchoita may exist, the first reproducing in Spring and the second in Autumn. It has been established that a daily spawning rhythm occurs, between the hours of 8 and 12 p. m. During this period it was possible to obtain mature females with which artificial fertilization was performed. The rate of development was determined, which at a temperature of 14-15° C is from 69-72 hours, and at a temperature of 19-20° C from 50-53 hours. A temperature of 49° C was found be lethal. The different embryonic and larval stages of development are illustrated diagramatically and individually described. The preliminary studies on the larvae and juveniles caught in the Sea during the period of reproduction led to the formulation of certain conclusions whith reference that the juveniles, as yet larvae, begin to group together at an early age, and the younger individuals the more uniform are the schoals whith regards to the total size of the specimens. It has shown that the anchovy during its first year of life tends to display littoral behaviour. RESUMEN EN ESPAÑOL: Los primeros estudios de los problemas referentes a la reproducción de los peces marinos en la Argentina (océano Atlántico sudoccidental) se han efectuado sobre una especie de gran importancia económica, Engraulis anchoita Hubbs y Marini. Se ha determinado la época de desove de la anchoíta en un período anual 1963-64. Aplicando el método de recolección cuantitativo se ha establecido que el desove de esta especie ha empezado en las zonas muy cercanas a la costa, en los primeros días de setiembre a la temperatura 10,3° C y ha alcanzado mayor intensidad en octubre (hasta 1569 huevos en 1 m2 de la superficie del agua) a la temperatura 11,5°-13,8° C. Desde mediados de noviembre la anchoíta sigue reproduciéndose en forma poco intensiva y más mar afuera, hasta por lo menos el mes de mayo. La intensidad de reproducción para este segundo período alcanza un pico, aunque muy pequeño en febrero a la temperatura 20° C. Se hace la suposición de que pueden existir dos de distintas características fisiológicas poblaciones de Engraulis anchoita una de reproducción primaveral y otra de reproducción otoñal. Además se ha establecido que existe un ritmo diario de desove que comprende las horas 20-24. En las horas de postura se pudo conseguir hembras maduras y con sus productos sexuales se efectuó la fecundación artificial. Se determinó la velocidad de desarrollo que a la temperatura 14°-15° C es de 69-72 horas y en la temperatura 19°- 20° C es de 50-53 horas. La temperatura 4° resultó ser letal. Se realizaron dibujos y descripciones correspondientes a los distintos estadios embrionarios y larvales. Los estudios preliminares de las larvas y juveniles de la anchoíta, capturados en el mar en la época de reproducción, permitieron sacar ciertas conclusiones sobre el crecimiento en sus primeros meses de vida. Se observó que los juveniles, larvas todavía, empiezan a agruparse muy temprano y cuanto más jóvenes son los individuos, tanto más uniformes son los cardúmenes en las dimensiones de los ejemplares. Se demostró que la anchoíta en su primer año de vida tiene costumbres muy costeras.

In the present study, natural occurrence of fungi and aflatoxin B1 (AFB1) in pellet feed and feed ingredients used for rainbow trout was investigated with emphasis to Aspergillus section Flavi members and medicinal plants inhibitory to Aspergillus growth and/or AF production. The feed samples were cultured on the standard isolation media including dichloran rosebengal chloramphenicol agar (DRCA) and Aspergillus flavus/parasiticus agar (AFPA) for 2 weeks at 28 °C. Identification of fungal isolates was implemented based on the macro- and microscopic morphological criteria. AFs were detected using high performance liquid chromatography (HPLC). Based on the results obtained, a total of 109 fungal isolates were identified of which Aspergillus was the prominent genus (57.0%), followed by Penicillium (12.84%), Absidia (11.01%) and Pseudallscheria (10.10%). The most frequent Aspergillus species was A. flavus (60.66%) isolated from all the feed ingredients as well as pellet feed. Among 37 A. flavus isolates, 19 (51.35%) were able to produce AFB1 on yeast extract-sucrose (YES) broth in the range of 10.2 to 612.8 [tg/g fungal dry weight. HPLC analyses of trout feed showed that pellet feed and all feed ingredients tested except gluten were contaminated with different levels of AFB1 in the range of 1.83 to 67.35 lig/kg. In order to finding natural inhibitors of fungal growth and/or AF production, essential oils (EOs) and extracts of 49 medicinal plants were studied against an aflatoxin-producing A. parasiticus using a microbioassay technique. The EOs was analyzed by gas chromatography/mass spectrometry (GC/MS). Based on the results obtained, Achillea millefolium sub sp. elborsensis, Ferula gummosa, Mentha spicata, Azadirachta indica, Conium maculatum and Artemisia dracunculus remarkably inhibited A. parasiticus growth without affecting AF production by the fungus. Besides of Thymus vulgaris and Citrus aurantifolia, the EO of Foeniculum vulgare significantly inhibited both fungal growth (-70.0%) and AFs B1 and G1 (-99.0%) production. The EO of Carum carvi and ethyl acetate extract of Platycladus orientalis suppressed AFs B1 and G1 by more than 90.0%, without any obvious effect on fungal growth. The IC50 values of bioactive plants for AFs B1 and G1 were determined in the ranges of 90.6 to 576.2 and 2.8 to 61.9 µg/ml, respectively. Overall, results of the present study indicate the importance of AF contamination of trout feed as a risk factor for fish farming and thus, an urgent necessity for constant monitoring of trout feed for any unacceptable levels of AF contamination. Likewise, antifungal activities of bioactive plants introduced here would be an important contribution to explain the use of these plants as effective antimicrobial candidates to protect feeds from toxigenic fungus growth and subsequent AF contamination.
