A good understanding of the population dynamics of algal communities is vital in many ecological and pollution studies of freshwater and oceanic systems. Present methods require manual counting and identification of algae and can take up to 90 min to obtain a statistically reliable count on a complex population. Several alternative techniques to accelerate the process have been tried on marine samples but none have been completely successful because insufficient effort has been put into verifying the technique before field trials. The objective of the present study has been to assess the potential of in vivo fluorescence of algal pigments as a means of automatically identifying algae. For this work total fluorescence spectroscopy was chosen as the observation technique.

Three direct plating methods and two most probable number (MPN) procedures were compared for the enumeration of Clostridium perfringens in seafoods the sulfitecycloserine (SC) agar, sulfite-polymyxin-sulfadiazine (SPS) agar, tryptone-sulfite- neomycin (TSN) agar, LS medium MPN procedure and iron milk MPN procedure. Isolates were confirmed as C. perfringens. The two MPN procedures compared very well with the three plating media tested with stock culture of C. perfringens from our laboratory collection and the reference strain NCIB 6125. But in fish samples, the two liquid media were found to be more sensitive and hence the MPN procedure using LS medium for the detection of C. perfringens in seafoods is suggested.

Samples of zooplankton and fish were collected from six sampling points in Nyanza Gulf, Lake Victoria in Kenya from June to December 1998, using 76 mu m memo-filament mesh size strainer and an 80 mu m mesh size plankton net. Identification and enumeration were done in the laboratory. Occurrence, numerical and points methods were used in stomach anlaysis of fish. Copepoda, Rotifera and Cladocera were the major zooplankton groups identified, zooplankton such as Moina, Daphnia and Caridina nilotica were important prey items for Lates niloticus, Oreochromis niloticus and Rastrineobola argentea

Improvements in methods for the detection and enumeration of microbes in water, particularly the application of techniques of molecular biology, have highlighted shortcomings in the ”standard methods” for assessing water quality. Higher expectations from the consumer and increased publicity associated with pollution incidents can lead to an uncoupling of the cycle which links methodological development with standard-setting and legislation. The new methodology has also highlighted problems within the water cycle, related to the introduction, growth and metabolism of microbes. A greater understanding of the true diversity of the microbial community and the ability to transmit genetic information within aquatic systems ensures that the subject of this symposium and volume provides an ideal forum to discuss the problems encountered by both researcher and practitioner.

Most microbiological methods require culture to allow organisms to recover or to selectively increase, and target organisms are identified by growth on specific agar media. Many cultural methods take several days to complete and even then the results require confirmation. Alternative techniques include the use of chromogenic and fluorogenic substances to identify bacteria as they are growing, selective capture using antibodies after short periods of growth, molecular techniques, and direct staining with or without flow cytometry for enumeration and identification. Future microbiologists may not use culture but depend on the use of specific probes and sophisticated detection systems.

Whilst current methods for the isolation and enumeration of Cryptosporidium spp. oocysts in water have provided some insight into their occurrence and significance, they are regarded as being inefficient, variable and time-consuming, with much of the interpretation being left to the expertise of the analyst. Two expectations of novel developments are to reduce the variability and subjectivity associated with the isolation and identification of oocysts. Flocculation, immunomagnetisable and flow cytometric techniques, for concentrating oocysts from water samples, should prove more reliable than current methods, whilst the development of more avid and specific monoclonal antibodies in conjunction with the use of nuclear fluorochromes will aid identification. Further insight into the viability, taxonomy, species identification, infectivity and virulence of the parasite should be forthcoming through the use of techniques such as the polymerase chain reaction, in situ hybridisation and non-uniform alternating current electrical fields. Such information is necessary in order to enable microbiologists, epidemiologists, engineers, utility operators and regulators to assess the safety of a water supply, with respect to Cryptosporidium contamination, more effectively.

A general survey carried out on several brands of frozen prawn products has shown that along with the standard plate count (SPC), the numbers of pathogenic organisms like Escherichia coli, enterococci and coagulase positive staphylococci have also to be taken into consideration for the evaluation of the quality of these products. No correlation could be established between the total plate count and the number of E. coli, enterococci or staphylococci. Enumeration of enterococci, however, is advocated as a better index of faecal contamination of the products than E. coli.

Bacteriological examination of the gastrointestinal microflora of 2 freshwater cichlid fish species (Sarotherodon mossambicus and Tilapia nilotica ) was performed, resulting in the bacteria enumeration of total viable counts of 1.06x10⁷/g and 7.75x10⁷/g of gastro-bacteria intestinal tract plus contents (wet weight) respectively, by aerobic incubation at 30+1°C. The majority (78%) of the total gut isolates from both fish species was Gram positive mesophilic which is characteristic of the higher ambient temperature in the tropics. These isolates were fastidious in their nutritional requirements and together with the rest are isogenous to bacteria autochthonous to soil and water. The occurrence of such organisms is attributed to the feeding habits of these fish. The gastrointestinal bacteria isolated in this study are transient residents but not "indigenous" in these cichlid fish.

Fish collected from local landing centres and also from local markets were examined for the presence and enumeration of Clostridium perfringens. A medium described by Beerens et al. (1982) was used for the detection and enumeration of C. perfringens. C. perfringens occurs in low numbers in fishes compared to prawns. Proper handling of fishes after landing can reduce the chance of any public health hazard by C. perfringens.
