Germination of macroconidia and/or microconidia of 24 strains of Fusarium solani, F. chlamydosporum, F. culmorum, F. equiseti, F. verticillioides, F. sambucinum, F. oxysporum and F. proliferatum isolated from fluvial channels and sea beds of the south-eastern coast of Spain, and three control strains (F. oxysporum isolated from affected cultures) was studied in distilled water in response to a range of water potentials adjusted with NaCI. (0, -13.79, -41.79, -70.37, -99.56 and -144.54 bars). The vialibility (UFC/ml) of suspension was also tested in three time periods (0,24 and 48h). Conidia always germinated in distilled water. The pattern of conidial germination obseved of F. verticillioides, F. oxysporum, F. proliferatum, F. chlamydosporum and F. culmorum was similar. A great diminution of spore germination was found in -13.79 bars solutions. Spore germination percentage for F. solani isolates was maximal at 48 h. and -13.79 bars with 21.33% spore germination, 16% higher than germination in distilled water. F. equiseti shows the maximum germination percentage in -144.54 bars solution in 24 h time with 12.36% germination. These results did not agree with those obtained in the viability test where maximum germination was found in distilled water. The viability analysis showed the great capacity of F. verticilloides strains to form viable colonies, even in such extreme conditions as -144,54 bars after 24 h F. proliferatum colony formation was prevented in the range of -70.37 bars. These results show the clear affectation of water potential to conidia germination of Fusaria. The ability of certain species of Fusarium to develop a saprophytic life in the salt water of the Mediterraneam Sea could be certain. Successful germination, even under high salty media conditions, suggests taht Fusarium spp. could have a competitive advantage over other soil fungi in crops irrigated with saline water. In the specific case of F. solani, water potential of -13.79 bars affected germination positively. It could indicate that F. solani has an special physiological mechanism of survival in low water potential environments.

Endo-β-mannanases (MAN; EC. 3.2.1.78) catalyze the cleavage of β1[RIGHTWARDS ARROW]4 bonds in mannan polymers and have been associated with the process of weakening the tissues surrounding the embryo during seed germination. In germinating Arabidopsis thaliana seeds, the most highly expressed MAN gene is AtMAN7 and its transcripts are restricted to the micropylar endosperm and to the radicle tip just before radicle emergence. Mutants with a T-DNA insertion in AtMAN7 have a slower germination than the wild type. To gain insight into the transcriptional regulation of the AtMAN7 gene, a bioinformatic search for conserved non-coding cis-elements (phylogenetic shadowing) within the Brassicaceae MAN7 gene promoters has been done, and these conserved motifs have been used as bait to look for their interacting transcription factors (TFs), using as a prey an arrayed yeast library from A. thaliana. The basic-leucine zipper TF AtbZIP44, but not the closely related AtbZIP11, has thus been identified and its transcriptional activation upon AtMAN7 has been validated at the molecular level. In the knock-out lines of AtbZIP44, not only is the expression of the AtMAN7 gene drastically reduced, but these mutants have a significantly slower germination than the wild type, being affected in the two phases of the germination process, both in the rupture of the seed coat and in the breakage of the micropylar endosperm cell walls. In the over-expression lines the opposite phenotype is observed.

Plants contain several genes encoding thioredoxins (Trxs), small proteins involved in redox regulation of many enzymes in different cell compartments. Among them, mitochondrial Trxo has been described to have a response in plants grown under salinity but there is scarce information about its functional role in abiotic stress or its gene regulation. In this work, the transcriptional regulation of the mitochondrial AtTrxo1 gene has been studied for the first time, by identifying functionally relevant cis- elements in its promoter: two conserved motives were found as positive and one as negative regulators. Using them as baits for the screening of an arrayed yeast library containing Arabidopsis Transcription Factors (TF) ORFs, two TFs were selected that are now being validated at the molecular level. We have also studied the response of T-DNA insertion mutant plants for AtTrxo1 to salt stress. The K.O. AtTrxo1 mutants presented several phenotypic changes including the time required to reach 50% germination under salinity, without affecting the final germination percentage.

La clasificación de las semillas de especies olerícolas se realiza principalmente por peso y tamaño, con criterios similares a los aplicados en cereales y leguminosas, en que se asocia positivamente estos atributos físicos con la calidad fisiológica. No obstante lo anterior, en diversas especies de hortalizas la información es escasa y contradictoria al respecto, lo que motiva la realización de la presente investigación. En semillas de tomate (Solanum lycopersicum L.) se determinó el efecto del peso y tamaño sobre la calidad fisiológica expresada como germinación y vigor. Además, se correlacionaron los resultados de las pruebas de evaluación de calidad fisiológica y se describieron variables del crecimiento y desarrollo. Se utilizaron lotes de diferentes variedades de semillas híbridas de cuatro temporadas, producidas en un clima templado cálido con lluvias invernales y estación seca prolongada (32º 54’ y 34° 21´ latitud Sur). Se midió peso y tamaño de semillas, además en dos temporadas se evaluaron las características internas de área y peso de embrión y área de endospermo. Se determinó la calidad de las semillas con la prueba de germinación y según fuera el año de estudio se midió vigor con las pruebas de envejecimiento acelerado, de plantas útiles al trasplante y de plántulas emergidas. Con análisis de imágenes y rayos X se extrajeron datos del tamaño externo e interno de las semillas y plántulas. Los lotes se compararon mediante análisis de varianza y las medias con la prueba de Tukey, la asociación entre dos variables se determinó con correlaciones de Pearson, las variables de peso y tamaño de la semilla y su relación con las pruebas de calidad, se analizaron mediante regresiones múltiples. Se utilizó un nivel de significación de 0,05 de probabilidad. Los resultados indicaron que el tamaño y no el peso de las semillas de tomate, diferenciaron calidad entre lotes en las diversas variedades. La prueba de germinación tuvo una baja sensibilidad para discriminar lotes, además de una escasa correlación con las características físicas de las semillas, cuando hubo asociación, la relación fue débil y negativa. La prueba de vigor de envejecimiento acelerado diferenció lotes y presentó escasa asociación con las características físicas de las semillas. El número de semillas germinadas en la prueba de envejecimiento acelerado se explicó por el efecto del tamaño de las semillas, mientras que las fracciones de descarte se asociaron con el peso de las mismas. La prueba de vigor de plantas útiles al trasplante no discriminó entre lotes. Tuvo una asociación débil con el peso y tamaño de las semillas. El modelo asociado a esta relación explicó con un alto coeficiente de determinación que el peso de la semilla influyó sobre la emergencia temprana, mientras que la relación fue menor y negativa con plantas de mayor desarrollo. La prueba de vigor de plántulas emergidas discriminó lotes de semillas con plántulas de 3 a 5 días después de siembra. Hubo escasa y débil asociación entre esta prueba y las características de peso y tamaño las semillas. El modelo de predicción de plántulas emergidas fue particular en cada temporada, cuando hubo un coeficiente de determinación alto influyó negativamente el peso o tamaño de la semilla. Entre las pruebas de calidad fisiológica evaluadas en semillas de tomate hubo escasas correlaciones significativas. Entre germinación y vigor las correlaciones significativas fueron débiles y sólo se encontraron en algunas temporadas de evaluación. Entre las pruebas de vigor no hubo asociación. En las pruebas de vigor de plantas útiles al trasplante y de plántulas emergidas, los cotiledones alcanzaron el mayor porcentaje de materia seca y se correlacionaron fuertemente con la materia seca total. En la prueba de plántulas emergidas la materia seca de las radículas diferenció parcialmente lotes de semillas al igual que la longitud total y de las radículas. La longitud de la radícula se correlacionó fuertemente con la longitud total de plántulas. ABSTRACT Seed selection for olericultural species is mainly carried out considering weight and size with similar criteria to those applied in cereals and legumes where size and physiological quality are favorably associated. However, information about several species is limited and contradictory regarding the above, leading to the present research. In tomato (Solanum lycopersicum L.) seeds, the effect of weight and size on the physiological quality expressed as germination and vigor was determined. In addition, results of quality evaluation tests were correlated and variables of growth and development were described. Batches of hybrid seeds from four seasons were used. These seeds were produced in a mild warm climate with winter rainfalls and long dry season (32º 54’ and 34° 21´South Latitude). Seed weight and size were determined, additionally internal characteristics such as embryo area and weight as well as endosperm area were evaluated in two seasons. The quality of seeds was established using the germination test and, depending on the year of the study, vigor was measured through accelerated aging tests for plants useful for transplanting and emerged seedlings. Using imaging analysis and X rays, data regarding external and internal size of seeds and seedlings were obtained. Batches were compared through ANOVA and means using Tukey’s test; the association between both variables was determined with Pearson correlations, whereas variables of seed weight and size and their relation to quality tests were analyzed through multiple regressions. A significance level of 0.05 probability was used. Results showed that the size (but not the weight) of tomatoes differentiates quality between batches from several seasons. The germination test was not sensitive enough to discriminate batches in addition to having a limited correlation with the characteristics of seeds, when they were associated, the relation was weak and unfavorable. Vigor test for accelerated aging made the difference between batches and presented low association with physical characteristics of the seeds. The number of germinated seeds in the accelerated aging test was explained by the effect of the seed size, whereas cull fractions were associated with their weight. The vigor test of plants useful for transplanting did not discriminate between batches. The association with seed weight and size was weak. The model associated to this relation explained, with a high coefficient determination, that the seed weight had influence on early emergence, whereas the relation was minor and unfavorable with more developed plants. Vigor test of emerged seedlings discriminated batches of seeds with seedlings of 3 to 5 days after sowing. There was a limited and weak association between this test and the characteristics of seed weight and size. The prediction model for seedlings emerged was particular in each season, when the determination coefficient was high, seed weight and size influenced negatively. Among the physiological quality tests evaluated in tomato seeds, significant correlations were negligible. Between germination and vigor, significant correlations were poor, being only found in some evaluation seasons. There was no association in the vigor tests. In vigor tests for plants useful for transplanting and emerged seedlings, cotyledons reached the highest percentage of dry matter and were strongly correlated with total dry matter. In the test of emerged seedlings, dry matter of radicles partially differentiated batches of seeds as well as total length and radicles. Radicle length was strongly correlated with total seedlings length.

Seed dormancy prevents seeds from germinating under environmental conditions unfavourable for plant growth and development and constitutes an evolutionary advantage. Dry storage, also known as after-ripening, gradually decreases seed dormancy by mechanisms not well understood. An Arabidopsis thaliana DOF transcription factor gene (DOF6) affecting seed germination has been characterized. The transcript levels of this gene accumulate in dry seeds and decay gradually during after-ripening and also upon seed imbibition. While constitutive over-expression of DOF6 produced aberrant growth and sterility in the plant, its over-expression induced upon seed imbibition triggered delayed germination, abscisic acid (ABA)-hypersensitive phenotypes and increased expression of the ABA biosynthetic gene ABA1 and ABA-related stress genes. Wild-type germination and gene expression were gradually restored during seed after-ripening, despite of DOF6-induced over-expression. DOF6 was found to interact in a yeast two-hybrid system andin planta with TCP14, a previously described positive regulator of seed germination. The expression of ABA1 and ABA-related stress genes was also enhanced in tcp14 knock-out mutants. Taken together, these results indicate that DOF6 negatively affects seed germination and opposes TCP14 function in the regulation of a specific set of ABA-related genes

Protein hydrolysis plays an important role during seed germination and post-germination seedling establishment. In Arabidopsis thaliana, cathepsin B-like proteases are encoded by a gene family of three members, but only the AtCathB3 gene is highly induced upon seed germination and at the early post-germination stage. Seeds of a homozygous T-DNA insertion mutant in the AtCathB3 gene have, besides a reduced cathepsin B activity, a slower germination than the wild type. To explore the transcriptional regulation of this gene, we used a combined phylogenetic shadowing approach together with a yeast one-hybrid screening of an arrayed library of approximately 1200 transcription factor open reading frames from Arabidopsis thaliana. We identified a conserved CathB3-element in the promoters of orthologous CathB3 genes within the Brassicaceae species analysed, and, as its DNA-interacting protein, the G-Box Binding Factor1 (GBF1). Transient overexpression of GBF1 together with a PAtCathB3::uidA (β-glucuronidase) construct in tobacco plants revealed a negative effect of GBF1 on expression driven by the AtCathB3 promoter. In stable P35S::GBF1 lines, not only was the expression of the AtCathB3 gene drastically reduced, but a significant slower germination was also observed. In the homozygous knockout mutant for the GBF1 gene, the opposite effect was found. These data indicate that GBF1 is a transcriptional repressor of the AtCathB3 gene and affects the germination kinetics of Arabidopsis thaliana seeds. As AtCathB3 is also expressed during post-germination in the cotyledons, a role for the AtCathB3-like protease in reserve mobilization is also inferred.

An HPLC/GC–MS/MS technique (high-pressure liquid chromatography in combination with gas chromatography–tandem mass spectrometry) has been worked out to analyze indole-3-acetamide (IAM) with very high sensitivity, using isotopically labelled IAM as an internal standard. Using this technique, the occurrence of IAM in sterile-grown Arabidopsis thaliana (L.) Heynh. was demonstrated unequivocally. In comparison, plants grown under non-sterile conditions in soil in a greenhouse showed approximately 50% higher average levels of IAM, but the differences were not statistically significant. Thus, microbial contributions to the IAM extracted from the tissue are likely to be minor. Levels of IAM in sterile-grown seedlings were highest in imbibed seeds and then sharply declined during the first 24 h of germination and further during early seedling development to remain below 20–30 pmol g–1 fresh weight throughout the rosette stage. The decline in indole-3-aetic acid (IAA) levels during germination was paralleled by a similar decline in IAM levels. Recombinant nitrilase isoforms 1, 2 and 3, known to synthesize IAA from indole-3-acetonitrile, were shown to produce significant amounts of IAM in vitro as a second end product of the reaction besides IAA. NIT2 was earlier shown to be highly expressed in developing and in mature A. thaliana embryos, and NIT3 is the dominantly active gene in the hypocotyl and the cotyledons of young, germinating seedlings. Collectively, these data suggest that the elevated levels of IAM in seeds and germinating seedlings result from nitrilase action on indole-3-acetonitrile, a metabolite produced in the plants presumably from glucobrassicin turnover.

During seed germination, the endosperm cell walls (CWs) suffer an important weakening process mainly driven by hydrolytic enzymes, such are endo-?- mannanases (MAN; EC. 3.2.1.78) that catalyze the cleavage of ?1?4 bonds in the mannan-polymers. In Arabidopsis thaliana seeds, endo-?-mannanase activity increases during seed imbibition, decreasing after radicle emergence1. AtMAN7 is the most highly expressed MAN gene in seeds upon germination and their transcripts are restricted to the micropylar endosperm and to the radicle tip just before radicle emergence. Mutants with a T-DNA insertion in this gene (K.O. MAN7) have a slower germination rate than the wild type (t50=34 h versus t50=25 h). To gain insight into the transcriptional regulation of the AtMAN7 gene, a bioinformatic search for conserved non-coding cis-elements (phylogenetic shadowing) within the Brassicaceae orthologous MAN7 gene promoters has been done and these conserved motives have been used as baits to look for their interacting transcription factors (TFs), using as a prey an arrayed yeast library of circa 1,200 TFs from A. thaliana. The basic leucine zipper AtbZIP44, but not its closely related ortholog AtbZIP11, has been thus identified and its regulatory function upon AtMAN7 during seed germination validated by different molecular and physiological techniques, such are RT-qPCR analyses, mRNA Fluorescence in situ Hybridization (FISH) experiments, and by the establishment of the germination kinetics of both over-expression (oex) lines and TDNA insertion mutants in AtbZIP44. The transcriptional combinatorial network through which AtbZIP44 regulates AtMAN7 gene expression during seed germination has been further explored through protein-protein interactions between AtbZIP44 and other bZIP members. In such a way, AtbZIP9 has been identified by yeast two-hybrid experiments and its physiological implication in the control of AtMAN7 expression similarly established.

En los suelos, el exceso de acidez lleva asociado deficiencias en ciertos nutrientes y una alta disponibilidad de aluminio, tóxico para los cultivos propios del ambiente mediterráneo. Su laboreo, provoca la pérdida de materia orgánica (MO), deteriora su estructura y reduce la actividad biológica, provocando en última instancia una menor calidad del suelo. Es de esperar pues que cuando se labran suelos ácidos, sus problemáticas particulares tiendan a agravarse. En nuestra zona de estudio, la “raña” de Cañamero (Extremadura, España), predominan los suelos muy ácidos y degradados por un laboreo inadecuado. Las rañas constituyen amplias plataformas casi horizontales, con unos suelos muy viejos (Palexerults), que se caracterizan por tener el complejo de cambio dominado por el aluminio, y un pH ácido que decrece en profundidad. Poseen un potente horizonte Bt rico en arcillas caoliníticas, que propicia que en periodos con exceso de lluvia, se generen capas colgadas de agua cercanas a la superficie. En torno a los años 1940’s estos suelos, que previamente sostenían un alcornocal, o su matorral de sustitución, se pusieron en cultivo. El laboreo aceleró la mineralización de la materia orgánica, agravó los problemas derivados del exceso de acidez y condujo al abandono de los campos cultivados por falta de productividad. Para recuperar la calidad de estos suelos degradados y obtener unos rendimientos compatibles con su uso agrícola es necesario, por un lado, aplicar enmiendas que eleven el pH y reduzcan la toxicidad del aluminio y, por otro, favorecer el incremento en el contenido en MO. En 2005 se implantó en esta raña un ensayo de campo para estudiar la influencia del no laboreo y de la utilización de una enmienda cálcica en parámetros relacionados con la calidad del suelo en un cultivo forrajero. El diseño experimental fue en parcelas divididas con cuatro repeticiones donde el factor principal fue el tipo de laboreo, no laboreo (NL) frente a laboreo convencional (LC), y el factor secundario el uso o no de una enmienda cálcica. La enmienda consistió básicamente en una mezcla de espuma de azucarería y yeso rojo y se incorporó al comienzo del ensayo hasta los 7 cm de profundidad. Desde el comienzo del ensayo el NL influyó positivamente en el contenido de carbono orgánico total (COT) y particulado (COP), mientras que la enmienda tuvo una ligera influencia al principio del ensayo en ambos pero su efecto positivo se desvaneció con el paso del tiempo. Los mayores contenidos en COT y POC se observaron cuando se combinó el NL con la enmienda. La enmienda incrementó con rapidez el pH, y el Ca, y disminuyó el contenido en aluminio hasta una profundidad de 50 cm, incluso en NL, y mejoró ligeramente la agregación del suelo. El NL por sí solo, gracias al aumento en POC, TOC y las proteínas del suelo relacionadas con la glomalina (PSRG), que son capaces de formar compuestos estables no tóxicos con el aluminio, también contribuyó a la reducción de la toxicidad de aluminio en la capa más superficial. Cuando en las campañas con exceso de precipitaciones se generaron capas colgadas de agua próximas a la superficie, el NL generó unas condiciones más favorables para la germinación y desarrollo del cultivo, resultando en una producción más alta que el LC. A ello contribuyó la mayor capacidad de almacenamiento de agua y la mayor transmisividad de esta hacia abajo, en la capa más superficial (0-5 cm) que propició una menor saturación por agua que el LC. Respecto a los parámetros relacionados con la agregación, el NL aumentó los macroagregados hasta los 10 cm de profundidad y favoreció la acumulación de CO y N en todas las fracciones de tamaño de agregados. Sin embargo, la recuperación del grado de macroagregación tras el cese del laboreo resulta lenta en comparación con otros suelos, posiblemente debido al bajo contenido en arcilla en el horizonte Ap. En comparación con el NL, la enmienda mostró también un efecto positivo, aunque muy ligero, en la agregación del suelo. En contradicción con otros estudios en suelos ácidos, nuestros resultados indican la existencia de una jerarquía de agregados, y destacan el papel importante de la MO en la mejora de la agregación. Tanto el NL como la enmienda favorecieron por separado varias propiedades químicas, físicas y biológicas del suelo, pero, en general, encontramos los mayores beneficios con su uso combinado. Además, a largo plazo el efecto positivo de NL en las propiedades del suelo fue en aumento, mientras que el efecto beneficioso de la enmienda se limitó básicamente a las propiedades químicas y se desvaneció en pocos años. Destacamos que las condiciones meteorológicas a lo largo del ensayo beneficiaron la producción de biomasa en NL, y en consecuencia las propiedades relacionadas con la materia orgánica, por lo que son un factor a tener en cuenta a la hora de evaluar los efectos de la enmienda y el laboreo sobre las propiedades del suelo, especialmente en zonas donde esas condiciones son muy variables entre una campaña y otra. Los resultados de este estudio han puesto de manifiesto que el NL no ha mermado la eficacia de la enmienda caliza, posiblemente gracias a la alta solubilidad de la enmienda aplicada, es más, el manejo con NL y enmienda es el que ha favorecido en mayor medida ciertos parámetros de calidad del suelo. Por el contrario el LC sí parece anular los beneficios de la enmienda en relación con las propiedades relacionadas con la MO. Por tanto, cabe concluir que la combinación de NL y la enmienda es una práctica adecuada para mejorar las propiedades químicas y físicas de suelos ácidos degradados por el laboreo. ABSTRACT Excessive acidity in soils is associated with deficiencies in certain nutrients and high concentrations of available aluminum, which is toxic for most Mediterranean crops. Tilling these soils results in the loss of soil organic matter (SOM), damages soil structure and reduces biological activity, ultimately degrading soil quality. It is expected, therefore, that when acid soils are tilled, their particular problems will tend to get worse. In our study area, the "Cañamero’s Raña” (Extremadura, Spain), acid soils degraded by an inappropriate tillage prevail. Rañas are large and flat platforms with very old soils (Palexerults), which are characterized by an exchange complex dominated by aluminum and an acid pH which decreases with depth. These soils have a strong Bt horizon rich in kaolinite clays, which encourages the formation of perched water-tables near the soil surface during periods of excessive rain. During the first third of the 20th century, these soils, that previously supported cork oak or its scrub replacement, were cultivated. Tillage accelerated the mineralization of the SOM, aggravating the problems of excessive acidity, which finally led to the abandonment of the land due to low productivity. To recover the quality of these degraded soils and to obtain consistent yields it is necessary, first, to apply amendments to raise the pH and reduce aluminum toxicity, and second to encourage the accumulation of SOM. In 2005 a field trial was established in the Raña to study the influence of no-tillage and the use of a Ca-amendment on soil quality related parameters in a forage crop agrosystem. The experimental design was a split-plot with four replicates where the main factor was tillage type, no-tillage (NT) versus traditional tillage (TT) and the secondary factor was the use or not of a Ca-amendment. The Ca-amendment was a mixture of sugar foam and red gypsum that was incorporated into the top 7 cm of the soil. Since the beginning of the experiment, NT had a positive influence on total and particulate organic carbon (TOC and POC, respectively), while the Ca-amendment had a small positive influence at the beginning of the study but its effect diminished with time. The highest TOC and POC contents were observed when NT and the Ca-amendment were combined. The Ca-amendment, even under NT, rapidly increased pH and Ca, and decreased the aluminum content to a depth of 50 cm, as well as improving soil aggregation slightly. NT, due to the increased POC, TOC and Glomalin-related soil proteins (GRSP), which can form stable non-toxic compounds with aluminum, also contributed to the reduction of aluminum toxicity in the upper layer. When perched water-tables near the soil surface were formed in campaigns with excessive rainfall, NT provided more favorable conditions for germination and crop development, resulting in higher yields compared with TT. This was directly related to the higher water storage capacity and the greater transmissivity of the water downwards from the upper layers, which led to lower water saturation under NT compared with TT. With regards to the aggregation-related parameters, NT increased macroaggregation to a depth of 10 cm and favored the accumulation of OC and N in all aggregate size fractions. However, the degree of recovery of macroaggregation after tillage ceased was slow compared with other soils, possibly due to the low clay content in the Ap horizon. Compared with NT, the Ca-amendment had a slight positive effect on soil aggregation. In contrast to other studies in acid soils, our results indicate the existence of an aggregate hierarchy, and highlight the important role of SOM in improving aggregation. Both NT and the Ca-amendment separately favored various chemical, physical and biological soil properties, but in general we found the greatest benefits when the two treatments were combined. In addition, the positive effect of NT on soil properties increased with time, while the beneficial effect of the Ca-amendment, which was limited to the chemical properties, vanished after a few years. It is important to note that the meteorological conditions throughout the experiment benefited biomass production under NT and, as a consequence, organic matter related properties. This suggests that meteorological conditions are a factor to consider when evaluating the effects of Ca-amendments and tillage on soil properties, especially in areas where such conditions vary significantly from one campaign to another. The results of this study show that NT did not diminish the effectiveness of the Ca-amendment, possibly due to the high solubility of the selected amendment. Moreover, the combination of NT and the Ca-amendment was actually the management that favored certain soil quality parameters the most. By contrast, TT seemed to nullify the benefits of the Ca-amendment with regards to the OM related properties. In conclusion, the combination of NT and the application of a Ca-amendment is an advisable practice for improving the chemical and physical properties of acid soils degraded by tillage.

1. Successful seed dispersal by animals is assumed to occur when undamaged seeds arrive at a favourable microsite. Most seed removal and dispersal studies consider only two possible seed fates, predation or escape intact. Whether partial consumption of seeds has ecological implications for natural regeneration is unclear. We studied partial consumption of seeds in a rodent-dispersed oak species. 2. Fifteen percent of dispersed acorns were found partially eaten in a field experiment. Most damage affected only the basal portion of the seeds, resulting in no embryo damage. Partially eaten acorns had no differences in dispersal distance compared to intact acorns but were recovered at farther distances than completely consumed acorns. 3. Partially eaten acorns were found under shrub cover unlike intact acorns that were mostly dispersed to open microhabitats. 4. Partially eaten acorns were not found buried proportionally more often than intact acorns, leading to desiccation and exposure to biotic agents (predators, bacteria and fungi). However, partial consumption caused more rapid germination, which enables the acorns to tolerate the negative effects of exposure. 5. Re-caching and shrub cover as microhabitat of destination promote partial seed consumption. Larger acorns escaped predation more often and had higher uneaten cotyledon mass. Satiation at seed level is the most plausible explanation for partial consumption. 6. Partial consumption caused no differences in root biomass when acorns experienced only small cotyledon loss. However, root biomass was lower when acorns experienced heavy loss of tissue but, surprisingly, they produced longer roots, which allow the seeds to gain access sooner to deeper resources. 7.Synthesis. Partial consumption of acorns is an important event in the oak regeneration process, both quantitatively and qualitatively. Most acorns were damaged non-lethally, without decreasing both dispersal distances and the probability of successful establishment. Faster germination and production of longer roots allow partially eaten seeds to tolerate better the exposure disadvantages caused by the removal of the pericarp and the non-buried deposition. Consequently, partially consumed seeds can contribute significantly to natural regeneration and must be considered in future seed dispersal studies.

Esta tesis se basa en la hipótesis de que la modernidad arquitectónica en México no es, como se ha pretendido, una historia homogénea, centrada en un puñado de figuras clave, sino una multiplicidad de narrativas complejas en las cuales el arte y los medios impresos juegan un papel esencial. Por tanto, se propone una nueva mirada sobre la arquitectura del siglo XX en México a partir de la relación con la fotografía, el dibujo, las ideas y los medios. La tesis se plantea con el fin de vincular la arquitectura con los movimientos artísticos relevantes, los autores con las publicaciones, las formas con los manifiestos. Asímismo, uno de los principales intereses es explorar los conceptos de modernidad y de identidad, como parte de la construcción misma de la arquitectura de dicha época y del concepto de “lo mexicano”. A pesar del énfasis que se ha dado en la construcción de un canon, muchas veces ligado a la noción de monumentalidad, regionalismo, y mestizaje, este trabajo parte de una mirada puesta no en las formas sino en los procesos. A partir de las conexiones entre distintas capas de información, se buscan nuevas maneras de abordar el proyecto arquitectónico. El crítico de arquitectura brasileño Hugo Segawa ha descrito la investigación sobre la arquitectura latinoamericana como “una tarea más de índole arqueológica que historiográfica”, sin embargo, también ha calificado a México como “el más vigoroso centro de debates teóricos en Latinoamérica a lo largo del siglo XX.” Ese descompas entre la ruina y el vigor, entre la abundancia de producción y la precariedad de su conservación, ha definido no solo el estudio de la arquitectura sino las propias formas de creación. Por tanto, la tesis se plantea como una nueva plataforma desde la cual sea posible reformular la arquitectura, lejos de su condición amnésica, pensada en cambio, como un sistema basado en una misma voluntad por indagar y crear. Se busca, siguiendo al crítico británico Anthony Vidler, “relacionar” la historia con el proyecto. Con el fin de quitarle lo escurridizo a una historia incompleta y sobre todo de poder entender la manera en que las ideas se convierten en forma o en objeto, la tesis se estructura a partir de 22 líneas de tiempo organizadas en tres recorridos que se cruzan: arquitectura; arte y pensamiento. A partir de referencias como el Atlas Mnemosyne de Aby Wargurg o la serie Asterisms del artista Gabriel Orozco, se crean nuevos dispositivos para ver. De tal manera, se desdoblan los distintos temas para trazar relaciones entre la ciudad, los edificios, las utopías, las publicaciones y la publicidad. El trabajo se construye como un nuevo instrumento de exploración articulado por medio de capas, como un mapa genealógico evolutivo. El objetivo es abarcar aquella arquitectura construida no sólo en la ciudad sino también en el papel. Iniciando con el trabajo de la generación que llevó la arquitectura al siglo XX, el estudio se extiende a manera de epílogo hasta la primera década del siglo XXI, reuniendo obras que normalmente se han visto de manera aislada para entenderlas en su contexto más amplio. Como escenario de búsquedas, esta tesis intenta provocar el cruce de significados, creyendo imprescindible una nueva reflexión en torno a la disciplina y a los escenarios en los cuales se inscribe. La arquitectura de México –un país que en el siglo XX pasó de tener 13 millones de habitantes a 100 millonescorresponde esencialmente a una producción anónima, o bien, fabricada a partir de estereotipos. Pero entre la mancha de desarrollo informal y el hito reconocible está un trabajo tan amplio como inexplorado. Por tanto, se ofrece una serie de nuevas constelaciones que comprenden desde la Revolución de 1910 a los Juegos Olímpicos de 1968; del terremoto de la ciudad de México en 1985 a los concursos internacionales de las últimas décadas. This thesis’ hypothesis states that architectural modernity in Mexico is not, as sometimes pretended, a homogeneous history, focused on some key figures, but rather a multiple and complex narrative, in which art and print media have played an essential role. Therefore, it proposes a new perspective on 20th century architecture in Mexico analized through the relationship between architecture and photography, art, theory and media. Its aim is to link architecture and artistic movements, authors and publications, forms and manifestos. What is intended here is to explore the concepts of ‘modernity’ and ‘identity’ as part of the construction of architecture and the concept of ‘Mexicanity’. Despite the emphasis that has been given to the construction of an architectural canon —mostly related to the notions of monumentality, regionalism and mestizaje/métissage— this thesis’ approach is focused mainly in processes and not in forms. Through connections between diverse layers of information, new ways of dealing with the architectural project are explored. Brazilian architecture critic Hugo Segawa has described the research on Latin American architecture as «more a task of archaeology than of historiography». Nonetheless, he has also described Mexico as «the most vigorous center of theoretical debates in Latin America throughout the 20th century». This acute discrepancy between decay and vigor, between abundance of production and precarious state of conservation has determined not only the ways in which architecture is studied and understood but also the process of architectural creation. This work is therefore outlined as a new platform in order to reformulate the discipline as a system based on a common will to research and create, far from the existing amnesiac attitude. Following British critic Anthony Vidler, the interest relies in the attempt to ‘relate’ History to project. In order to reduce the elusiveness of an incomplete history and, specially, to understand how ideas become forms and objects, this thesis is composed of 22 timelines organized in three intersecting itineraries: Architecture, Art and Theory. Drawing inspiration from Aby Warburg’s Atlas Mnemosyne and Gabriel Orozco’s series Asterisms, new exploration devices are created. In such a way, diverse topics unfold to draw connections between built environment, utopian projects, publications, photography and publicity. This work is developed as a new tool for exploration, articulated by layers, like an evolutionary genealogy map. Its objective is to analyze not only the architecture build in cities, but produced on paper. Starting with the work of the generation that led Mexican architecture into the 20th century, this research extends until the first decade of the 21st century (the epilogue), gathering together works which have been usually seen in isolation, and therefore making possible its understanding in a broader context. As a scenario for exploration, this work tries to prompt the crossing of meanings, in the belief that new approaches on the discipline and its context are needed. Architecture in Mexico — a country whose population grew in the 20th century form 13 to 100 million— is related essentially with an anonymous production, or else made from stereotypes. However, between the sprawl of informal urban developments and landmark buildings there is an architectural production as extensive as it is unexamined. This essay introduces a series of new constellations, ranging from the Revolution in 1910 to the Olympic Games in 1968; from the earthquake in Mexico City in 1985 to the international competitions of the last decade. These myriad perspectives present buildings that were never built, forgotten writings, iconic images and unpublished material.

Este trabajo aborda el estudio del uso de restos de poda, lodos de depuradora y diferentes biochares obtenidos a partir de dichos residuos, así como biochar comercial, como componentes de sustratos de cultivo en sustitución de la turba. Para ello, se han llevado a cabo varios experimentos usando dichos residuos originales y pirolizados, de forma individual o mezclándolos en distintas proporciones con turba con el fin de estudiar sus características químicas e hidrofísicas, su posible fitotoxicidad y el cultivo en macetas de Lactuca sativa. Los resultados obtenidos indican que es posible preparar sustratos de cultivo adecuados a partir de algunos de estos materiales, logrando así una forma de recuperación y valorización de los mismos, a la vez que se minimiza el consumo de turba. En general, la adición de biochar de restos de poda a la turba mejora la germinación y aumenta el crecimiento de Lactuca sativa respecto a la utilización de turba sola. ABSTRACT This research deals with the study of the use of pruning wastes, sewage sludges and different biochars obtained from these materials, as well as a commercial biochar, as growing media components to replace peat. Several experiments were conducted using original wastes and pyrolyzed ones, individually or mixed in different proportions with peat, in order to study their chemical and hydrophysical characteristics, its possible phytotoxicity and the pot cultivation of Lactuca sativa. The results obtained indicate that it is possible to prepare suitable growing media from some of these materials, not only achieving a recovery and valorisation of them, but also minimizing the consumption of peat. In general, the addition of pruning waste biochar to peat improves germination and increases the growth of Lactuca sativa in comparison with peat alone.

Biochar can provide multiple benefits in the ecosystem. However, the presence of phytotoxic compounds in some biochars is an important concern that needs to be addressed and that depends on the raw material and the pyrolysis conditions used in biochar production. For example, sewage sludge biochars can have elevated heavy metal con- tents as they were present in the feedstock and were enriched during pyrolysis. Also during carbonization, some phytotoxic compounds such as polycyclic aromatic hydrocarbons (PAHs), polyphenols or volatile organic com- pounds (VOCs) could be formed representing a risk of contamination to soils and crops. In this work we report the results from seed germination and plant development for three biochars prepared from wood, paper sludge plus wheat husks and sewage sludge. Five higher plant species (cress, lentils, cucumber, tomato and lettuce) were studied. Biochar from wood shows seed inhibition in several species and the paper sludge biochar on lettuce. For the rest, the effect on seed germination was positive. No inhibition of root growth was detected, but in some cases leaves and stems growth were inhibited. Our results are significant in terms of advancing or current understanding on the impacts of biochar on vegetative growth and linking those effects to biochar properties.

Cinchona officinalis (Rubiaceae), especie endémica del Valle de Loja, ubicado en la región sur del Ecuador, es un recurso forestal de importancia medicinal y ecológica, además la especie ha sido catalogada como planta nacional y es un ícono de la región sur por su aporte a la farmacopea mundial. Esta especie, entre los siglos XVII-XIX sufrió una gran presión en sus poblaciones debido a la extracción masiva de la corteza para la cura del paludismo. Aunque la actividad extractiva generó grandes ingresos a la Corona Española y a la región Sur del Ecuador, ésta fue poco o nada sustentable ecológicamente, provocando la desaparición de la especie en muchos sitios de la provincia, pues, en su momento, no se consideraron alternativas de recuperación de las poblaciones naturales. Actualmente la extracción y consumo de la corteza en la zona de origen es baja o nula, sin embargo esta zona enfrenta nuevas amenazas. La deforestación a causa de proyectos de desarrollo en infraestructuras, la práctica de actividades agrícolas y de ganadería, y los efectos del cambio climático han ocasionado, en estos últimos años, la fragmentación de los ecosistemas. La mayoría de los bosques del sur del Ecuador se han convertido en parches aislados (los bosques en los que se distribuye C. officinalis no son la excepción) siendo esta la principal causa para que la especie se encuentre en estado de amenaza. Los individuos de la especie tienen una alta capacidad de rebrote y producen semillas durante todo el año; sin embargo la capacidad germinativa y la tasa de sobrevivencia son bajas, además de estas dificultades la especie requiere de la asociación con otras especies vegetales para su desarrollo, lo cual ha limitado su distribución en pequeños parches aislados. Con esta problemática, la recuperación natural de las poblaciones es una necesidad evidente. Varios trabajos y esfuerzos previos se han realizado a nivel local: i. Identificación de la distribución actual y potencial; ii. Determinación de la fenología y fructificación iii. Programas de educación ambiental, iv. Análisis moleculares para determinar la diversidad genética. v. Ensayos de propagación vegetativa; y otras acciones de tipo cultural. No obstante, el estado de conservación y manejo de las poblaciones naturales no ha mejorado significativamente, siendo necesaria la aplicación de estrategias integradas de conservación in situ y ex situ, que permitan la recuperación y permanencia de las poblaciones naturales a largo plazo. El presente trabajo tiene como fin dar alternativas para el cultivo de tejidos in vitro de Cinchona officinalis centrados en la propagación masiva a partir de semillas, análisis de la fidelidad genética y alternativas de conservación de tejidos. Los objetivos específicos que se plantean son: i. Analizar el proceso de germinación y proliferación in vitro. ii. Evaluar la estabilidad genética en explantes cultivados in vitro, mediante marcadores ISSR. iii. Establecer protocolos de conservación in vitro mediante limitación del crecimiento y criopreservación de segmentos nodales y yemas. Los resultados más significativos de esta investigación fueron: i. El desarrollo de protocolos eficientes para mejorar los porcentajes de germinación y la proliferación de brotes en explantos cultivados in vitro. Para evaluar el efecto de los fenoles sobre la germinación, se determinó el contenido total de fenoles y el porcentaje de germinación en semillas de C. officinalis comparados con una especie de control, C. pubescens. Para inducir a proliferación, se utilizaron segmentos nodales de plántulas germinadas in vitro en medio Gamborg (1968) suplementado con diferentes combinaciones de reguladores de crecimiento (auxinas y citoquininas). Los resultados obtenidos sugieren que el contenido de compuestos fenólicos es alto en las semillas de C. officinalis en comparación con las semillas de C. pubescens. Estos fenoles pueden eliminarse con peróxido de hidrógeno o con lavados de agua para estimular la germinación. La formación de nuevos brotes y callos en la mayoría de las combinaciones de reguladores de crecimiento se observó en un período de 45 días. El mayor porcentaje de proliferación de brotes, formación de callos y presencia de brotes adventicios se obtuvo en medio Gamborg (B5) suplementado con 5.0 mg/l 6-bencil-aminopurina y 3.0 mg/l de ácido indol-3-butírico. ii. La evaluación de la fidelidad genética de los explantes obtenidos con distintas combinaciones de reguladores de crecimiento vegetal y diversos subcultivos. Se realizó el seguimiento a los explantes obtenidos de la fase anterior, determinando el índice de multiplicación y analizando la fidelidad genética de los tejidos obtenidos por las dos vías regenerativas: brotación directa y regeneración de brotes a partir de callos. Este análisis se realizó por amplificación mediante PCR de las secuencias ubicadas entre microsatélites-ISSR (Inter simple sequence repeat). El medio Gamborg (B5) con 3.0 mg/l de AIB y 5.0 mg/l de BAP usado como medio de inducción en la primera etapa de cultivo generó el mayor índice de proliferación (11.5). Un total de 13 marcadores ISSR fueron analizados, 6 de éstos fueron polimórficos. El mayor porcentaje de variación somaclonal fue inducido en presencia de 1.0 mg/l 2,4-D combinado con 0.2 mg/l Kin con un 1.8% en el segundo sub-cultivo de regeneración, la cual incrementó a 3.6% en el tercer sub-cultivo. Todas las combinaciones con presencia de 2,4-D produjeron la formación de callos y presentaron variación genética. Por su parte la fidelidad genética se mantuvo en los sistemas de propagación directa a través de la formación de brotes a partir de meristemos preformados. iii. El establecimiento de protocolos de conservación in vitro y crioconservación de segmentos nodales y yemas. Para la conservación limitando el crecimiento, se cultivaron segmentos nodales en los medios MS y B5 en tres concentraciones de sus componentes (25, 50 y 100%); y en medio B5 más agentes osmóticos como el manitol, sorbitol y sacarosa en diferentes concentraciones (2, 4 y 8%); los cultivos se mantuvieron por 12 meses sin subcultivos. Para el establecimiento de protocolos para la crioconservación (paralización del metabolismo) se usaron yemas axilares y apicales a las cuales se les aplicaron los métodos de encapsulación-deshidratación y vitrificación. La efectividad de los protocolos usados se determinó en función de la sobrevivencia, reducción del crecimiento y regeneración. Los resultados obtenidos en este apartado reflejan que un crecimiento limitado puede mantener tejidos durante 12 meses de almacenamiento, usando medio B5 más manitol entre 2 y 8%. En los protocolos de crioconservación, se obtuvo el mayor porcentaje de recuperación tras la congelación en NL en el tratamiento control seguido por el método crioprotector de encapsulación-deshidratación. Este trabajo brinda alternativas para la propagación de C. officinalis bajo condiciones in vitro, partiendo de material vegetal con alta diversidad genética. El material propagado puede ser fuente de germoplasma para la recuperación y reforzamiento de las poblaciones naturales así como una alternativa de producción para las comunidades locales debido a la demanda actual de corteza de la zona de origen para la elaboración de agua tónica. ABSTRACT Cinchona officinalis (Rubiaceae) is endemic to the Loja Valley, located in the southern area of Ecuador. The importance of this plant as medical and ecological resource is so great that it has been designated as the national flower and is an icon of the southern region for its contribution to the world pharmacopoeia. Between XVII-XIX centuries its population suffered great reduction due to massive harvesting of the bark to cure malaria. Although extraction activity generated large revenues to the Spanish Crown and the southern region of Ecuador, this was not ecologically sustainable, causing the disappearance of the species in many areas of the province, because during that time alternatives to prevent extinction and recover natural populations were not taken in account. Currently the extraction and consumption of bark in the area of origin is almost absent, but this species faces new threats. Deforestation due to infrastructure development, the practice of farming and ranching, and the effects of climate change had led to the fragmentation of ecosystems during the recent years. Most of the forests of southern Ecuador have become isolated patches, including those where C. officinalis is diffused. The lack of suitable habitat is today the main threat for the species. The species has a high capacity for regeneration and produces seeds throughout the year, but the germination rate is low and the growth is slow. In addition, the species requires the association with other plant species to develop. All these factors had limited its distribution to small isolated patches. The natural recovery of populations is essential to face this problem. Several studies and previous efforts had been made at local level: i. Identification of current and potential distribution; ii. Phenology determination. iii. Environmental education programs, iv. Molecular analisis to determine the genetic diversity. v. Testing of vegetative propagation; and other actions of cultural nature. Despite these efforts, the state of conservation and management of natural populations has not improved significantly. Implementation of integrated in situ and ex situ conservation strategies for the recovery and permanence of long-term natural populations is still needed. This work aims to provide alternatives for in vitro culture of tissue of Cinchona officinalis focused on mass propagation from seeds, genetic fidelity analysis and tissue conservation alternatives. The specific aims are: i. Analyze the process of germination and proliferation in vitro. ii. To evaluate the genetic stability of the explants cultured in vitro by ISSR markers. iii. Establish protocols for in vitro conservation by limiting growth and cryopreservation of nodal segments and buds. The most significant results of this research were: i. The development of efficient protocols to improve germination rates and proliferation of buds in explants cultured in vitro. To study the effect of phenols on germination, the total phenolic content and percentage germination was measured in C. officinalis and in a control species, C. pubescens, for comparison. The content of phenolic compounds in C. officinalis seeds is higher than in C. pubescens. These phenols can be removed with hydrogen peroxide or water washes to stimulate germination. To analyze the regeneration, we used nodal explants from seedlings germinated in vitro on Gamborg medium (1968) supplemented with different combinations of growth regulators (auxins and cytokinins) to induce proliferation. The formation of new shoots and calluses was observed within a period of 45 days in most combinations of growth regulators. The highest percentage of shoot proliferation, callus formation and adventitious buds were obtained in B5 medium supplemented with 5.0 mg/l 6-benzyl-aminopurine and 3.0 mg/l indole-3-butyric acid. ii. Evaluating genetic fidelity explants obtained with various combinations of plant growth regulators and different subcultures. The genetic fidelity was analyzed in tissues obtained by the two regenerative pathways: direct sprouting and shoot regeneration from callus. This analysis was performed by PCR amplification of the sequences located between microsatellite-ISSR (Inter Simple Sequence Repeat). Among a total of 13 ISSR markers analyzed, 6 were polymorphic. The highest percentage of somaclonal variation was induced in the presence of 1.0 mg/l 2,4-D combined with 0.2 mg/l Kin with 1.8% in the second round of regeneration, and increased to 3.6% in the third round. The presence of 2,4-D induced genetic variation in all the combinations of growth regulators. Meanwhile genetic fidelity remained systems propagation through direct shoot formation from meristems preformed. iii. Establishing conservation protocols in vitro and cryoconservation of nodal segments and buds. For medium-term conservation (limited growth) nodal segments were cultured in MS and B5 media at three concentrations (25, 50 and 100%); we tested B5 medium with different concentrations of osmotic agents such as mannitol, sorbitol and sucrose (2, 4 and 8%); cultures were maintained for 12 months with regular subculturing. To establish protocols for cryoconservation (cessation of metabolism) different methods of encapsulation-dehydration and vitrification were applied to axillary and apical buds. The effectiveness of the used protocols is determined based on the survival, growth and regeneration success. The results show that these tissues can be maintained in storage for 12 months, using B5 medium plus mannitol between 2 and 8%. The cryoconservation protocol with highest percentage of recovery was obtained by contral treatment, followed by freezing in NL with encapsulation-dehydration method. This work provides alternatives for the propagation in vitro of C. officinalis, starting from plant material with high genetic diversity. The obtained material represents a source of germplasm to support the recovery and strengthening of natural populations as well as a creation of alternative sources for local communities due to the current demand of bark for the preparation of tonic water.

La fase de establecimiento del regenerado es un proceso crítico para el desarrollo posterior de la masa tanto por las elevadas tasas de mortalidad que habitualmente lleva asociadas, como por proporcionar el material de partida del que van a disponer las fases subsiguientes. Las restricciones a la germinación y establecimiento de la regeneración del pino silvestre varían enormemente entre las distintas regiones de su extensa área de distribución geográfica. La región Mediterránea constituye un hábitat marginal de la especie en el que las condiciones ecológicas son muy diferentes a las del grueso de su área de distribución. Frente a otras limitaciones (frío, luz, encharcamiento…), en el entorno mediterráneo la tasa de mortalidad parece estar asociada a las condiciones micrometeorológicas del período estival - particularmente, a la sequía -, así como a la presencia excesiva de ganado o ungulados silvestres. No obstante, la mayoría de la información disponible sobre el proceso de regeneración de la especie procede del centro y norte de Europa, por lo que no es de aplicación directa en nuestra región, en la que los estudios de este tipo son mucho más escasos. El presente trabajo pretende contribuir a paliar esta relativa escasez a través del estudio del proceso de regeneración natural en el monte “Cabeza de Hierro”, masa irregular por bosquetes de pino silvestre, paradigma de gestión sostenible y uso múltiple. En este entorno, se pretende caracterizar y cuantificar tanto el proceso de germinación y supervivencia de la especie como la influencia de la cobertura vegetal (estratos arbóreo, arbustivo y herbáceo, y capa de restos vegetales) en su desarrollo. Se persigue así mismo analizar el efecto de la compactación del suelo sobre la persistencia de la masa y contrastar y comparar la eficacia de dos tratamientos edáficos de ayuda a la regeneración: escarificado y decapado+acaballonado. Con este fin se han planteado dos diseños experimentales consistentes en sendas redes de muestreo (Red de Muestreo I o RM I y Red de Muestreo II o RM II) integradas, respectivamente, por 192 y 24 parcelas de 1,5x1,5 m ubicadas bajo distintas condiciones de cobertura vegetal. Sobre una parte de estas parcelas (1/4 en la Red de Muestreo I; 1/2 en la Red de Muestreo II) se han aplicado tratamientos de ayuda a la regeneración (RM I: escarificado; RM II: decapado+acaballonado) y, tras llevar a cabo siembras controladas al inicio del período vegetativo, se han practicado controles periódicos de germinación y supervivencia durante uno (RM II) y tres años consecutivos (RM I). Se han realizado así mismo mediciones complementarias de variables micrometeorológicas, espesura, recubrimiento superficial del suelo y compactación. Los resultados obtenidos a partir de las experiencias realizadas en el monte objeto de estudio permiten concluir que, en relación con el proceso de regeneración natural de la especie en este tipo masa y entorno: 1) la regeneración del pino silvestre durante el primer período vegetativo presenta una tasa de éxito muy baja (1,4% de los sembrados), provocada por una elevada mortalidad durante el primer período estival (>92%) subsiguiente a una germinación de en torno al 17% de las semillas viables que llegan al suelo; 2) la mortalidad sigue siendo elevada hasta el tercer período vegetativo, en que comienza a reducirse significativamente hasta alcanzar el 45%; 3) la cobertura vegetal influye significativamente tanto en el proceso de germinación como en el de supervivencia, aunque ambos procesos presentan una baja correlación linear que pone de manifiesto que los lugares idóneos para la germinación no siempre son los más adecuados para la supervivencia; 4) la escarificación del suelo mejora las tasas iniciales de germinación y supervivencia, pero empeora la tasa de supervivencia posterior (años 2 y 3), por lo que su efecto a medio plazo no resulta significativo; 5) el decapado+acaballonado presenta mejores resultados que el escarificado durante el primer verano, aunque sólo resulta efectivo en condiciones intermedias de espesura de masa; 6) la compactación edáfica no resulta limitante para la productividad ni la persistencia de la masa considerada. ABSTRACT Seedling establishment is critical for later stand progress because it involves high mortality rates and the surviving saplings constitute the starting material for all the subsequent stages. Restrictions for Scots pine germination and seedling survival may vary greatly across its geographical range, as it is widely distributed within north latitudes. Mediterranean region is a marginal sector within this species range and its ecological conditions differ greatly from those of the bulk of the area. Mortality rates in Mediterranean environments seem to be related to summer weather (mainly drought) and high livestock stocking rather than to cold, light or flooding. Most available information on scots pine regeneration process comes from north European experiences and is not transferable to Spanish forests, whereas studies on Mediterranean region are much scarcer. The present work aims at broadening Scots pine regeneration knowledge within Mediterranean region by analyzing its establishment process in the “Cabeza de Hierro” forest: a Scots pine uneven-aged forest at blocklevel scale, exemplary managed for multi-services purpose. Germination and surviving processes are to be characterized and quantified as to vegetation cover both in trees, shrubs, grass and litter strata. Soil compaction effects on forest sustainability are also assessed and the efficacy of some site preparation techniques on regeneration success is tested and compared (scarification vs. scalping+mounding). Two sampling networks comprising respectively 198 (SN I) and 24 plots (SN II) of 1.5x1.5m have been established over a wide range of vegetal cover conditions within the forest. Soil preparation techniques have been applied only to some of the sampling points; namely, 1 out of 4 plots have been scarified within Sampling Network I , while 1 out of 2 plots have been object of scalping & mounding within Sampling Network II. After localized sowing prior to growing season, germination and surviving have been periodically sampled for either one (SN II) or three years (SN I). Supplementary measures for micrometeorological variables, stand density, ground vegetal cover and compaction have also been carried out. Results obtained for the studied forest lead to the following insights regarding Scots pine natural regeneration process within this sort of forest and environment: 1) seedling establishment success rate is quite low (0,15% of sowing seeds), due to high mortality during the first summer (>92%), following a prior 17% rate of germination over viable seeds reaching the soil; 2) mortality rate remains high until the third year after emergence and then decreases to the 50% of surviving; 3) although vegetal cover significantly affects both seedling germination and survival, lineal correlation between those two processes is rather low, which may indicate that places fit for emergence are not necessarily suitable for summer surviving; 4) soil scarification improves both germination and survival during the first growing season, but it is associated to higher mortality rates during the next two years; hence it has no significant medium term effect; 5) scalping & mounding treatment is more effective than scarification concerning establishment improving during the first summer; but its effects are only significant under intermediate stand density levels; 6) soil compaction does not restrict either forest productivity or persistence, despite the area’s long history of high livestock stocking rates and mechanized logging.
