El objetivo de la presente tesis doctoral es el desarrollo de un nuevo concepto de biosensor óptico sin marcado, basado en una combinación de técnicas de caracterización óptica de interrogación vertical y estructuras sub-micrométricas fabricadas sobre chips de silicio. Las características más importantes de dicho dispositivo son su simplicidad, tanto desde el punto de vista de medida óptica como de introducción de las muestras a medir en el área sensible, aspectos que suelen ser críticos en la mayoría de sensores encontrados en la literatura. Cada uno de los aspectos relacionados con el diseño de un biosensor, que son fundamentalmente cuatro (diseño fotónico, caracterización óptica, fabricación y fluídica/inmovilización química) son desarrollados en detalle en los capítulos correspondientes. En la primera parte de la tesis se hace una introducción al concepto de biosensor, en qué consiste, qué tipos hay y cuáles son los parámetros más comunes usados para cuantificar su comportamiento. Posteriormente se realiza un análisis del estado del arte en la materia, enfocado en particular en el área de biosensores ópticos sin marcado. Se introducen también cuáles son las reacciones bioquímicas a estudiar (inmunoensayos). En la segunda parte se describe en primer lugar cuáles son las técnicas ópticas empleadas en la caracterización: Reflectometría, Elipsometría y Espectrometría; además de los motivos que han llevado a su empleo. Posteriormente se introducen diversos diseños de las denominadas "celdas optofluídicas", que son los dispositivos en los que se va a producir la interacción bioquímica. Se presentan cuatro dispositivos diferentes, y junto con ellos, se proponen diversos métodos de cálculo teórico de la respuesta óptica esperada. Posteriormente se procede al cálculo de la sensibilidad esperada para cada una de las celdas, así como al análisis de los procesos de fabricación de cada una de ellas y su comportamiento fluídico. Una vez analizados todos los aspectos críticos del comportamiento del biosensor, se puede realizar un proceso de optimización de su diseño. Esto se realiza usando un modelo de cálculo simplificado (modelo 1.5-D) que permite la obtención de parámetros como la sensibilidad y el límite de detección de un gran número de dispositivos en un tiempo relativamente reducido. Para este proceso se escogen dos de las celdas optofluídicas propuestas. En la parte final de la tesis se muestran los resultados experimentales obtenidos. En primer lugar, se caracteriza una celda basada en agujeros sub-micrométricos como sensor de índice de refracción, usando para ello diferentes líquidos orgánicos; dichos resultados experimentales presentan una buena correlación con los cálculos teóricos previos, lo que permite validar el modelo conceptual presentado. Finalmente, se realiza un inmunoensayo químico sobre otra de las celdas propuestas (pilares nanométricos de polímero SU-8). Para ello se utiliza el inmunoensayo de albumina de suero bovino (BSA) y su anticuerpo (antiBSA). Se detalla el proceso de obtención de la celda, la funcionalización de la superficie con los bioreceptores (en este caso, BSA) y el proceso de biorreconocimiento. Este proceso permite dar una primera estimación de cuál es el límite de detección esperable para este tipo de sensores en un inmunoensayo estándar. En este caso, se alcanza un valor de 2.3 ng/mL, que es competitivo comparado con otros ensayos similares encontrados en la literatura. La principal conclusión de la tesis es que esta tipología de dispositivos puede ser usada como inmunosensor, y presenta ciertas ventajas respecto a los actualmente existentes. Estas ventajas vienen asociadas, de nuevo, a su simplicidad, tanto a la hora de medir ópticamente, como dentro del proceso de introducción de los bioanalitos en el área sensora (depositando simplemente una gota sobre la micro-nano-estructura). Los cálculos teorícos realizados en los procesos de optimización sugieren a su vez que el comportamiento del sensor, medido en magnitudes como límite de detección biológico puede ser ampliamente mejorado con una mayor compactación de pilares, alcanzandose un valor mínimo de 0.59 ng/mL). The objective of this thesis is to develop a new concept of optical label-free biosensor, based on a combination of vertical interrogation optical techniques and submicron structures fabricated over silicon chips. The most important features of this device are its simplicity, both from the point of view of optical measurement and regarding to the introduction of samples to be measured in the sensing area, which are often critical aspects in the majority of sensors found in the literature. Each of the aspects related to the design of biosensors, which are basically four (photonic design, optical characterization, fabrication and fluid / chemical immobilization) are developed in detail in the relevant chapters. The first part of the thesis consists of an introduction to the concept of biosensor: which elements consists of, existing types and the most common parameters used to quantify its behavior. Subsequently, an analysis of the state of the art in this area is presented, focusing in particular in the area of label free optical biosensors. What are also introduced to study biochemical reactions (immunoassays). The second part describes firstly the optical techniques used in the characterization: reflectometry, ellipsometry and spectrometry; in addition to the reasons that have led to their use. Subsequently several examples of the so-called "optofluidic cells" are introduced, which are the devices where the biochemical interactions take place. Four different devices are presented, and their optical response is calculated by using various methods. Then is exposed the calculation of the expected sensitivity for each of the cells, and the analysis of their fabrication processes and fluidic behavior at the sub-micrometric range. After analyzing all the critical aspects of the biosensor, it can be performed a process of optimization of a particular design. This is done using a simplified calculation model (1.5-D model calculation) that allows obtaining parameters such as sensitivity and the detection limit of a large number of devices in a relatively reduced time. For this process are chosen two different optofluidic cells, from the four previously proposed. The final part of the thesis is the exposition of the obtained experimental results. Firstly, a cell based sub-micrometric holes is characterized as refractive index sensor using different organic fluids, and such experimental results show a good correlation with previous theoretical calculations, allowing to validate the conceptual model presented. Finally, an immunoassay is performed on another typology of cell (SU-8 polymer pillars). This immunoassay uses bovine serum albumin (BSA) and its antibody (antiBSA). The processes for obtaining the cell surface functionalization with the bioreceptors (in this case, BSA) and the biorecognition (antiBSA) are detailed. This immunoassay can give a first estimation of which are the expected limit of detection values for this typology of sensors in a standard immunoassay. In this case, it reaches a value of 2.3 ng/mL, which is competitive with other similar assays found in the literature. The main conclusion of the thesis is that this type of device can be used as immunosensor, and has certain advantages over the existing ones. These advantages are associated again with its simplicity, by the simpler coupling of light and in the process of introduction of bioanalytes into the sensing areas (by depositing a droplet over the micro-nano-structure). Theoretical calculations made in optimizing processes suggest that the sensor Limit of detection can be greatly improved with higher compacting of the lattice of pillars, reaching a minimum value of 0.59 ng/mL).

Light trapping is becoming of increasing importance in crystalline silicon solar cells as thinner wafers are used to reduce costs. In this work, we report on light trapping by rear-side diffraction gratings produced by nano-imprint lithography using interference lithography as the mastering technology. Gratings fabricated on crystalline silicon wafers are shown to provide significant absorption enhancements. Through a combination of optical measurement and simulation, it is shown that the crossed grating provides better absorption enhancement than the linear grating, and that the parasitic reflector absorption is reduced by planarizing the rear reflector, leading to an increase in the useful absorption in the silicon. Finally, electro-optical simulations are performed of solar cells employing the fabricated grating structures to estimate efficiency enhancement potential.

Increasing consumer dissatisfaction related with lack of ripeness in peach has been repeatedly reported since 1990 to the present day. There is thus, a great interest in improving the assessment of peach maturity, currently based on Magness Taylor firmness (destructive, highly variable, and time consuming) and colour (not reliable for highly coloured varieties). The present research studies as an alternative several non-destructive (ND) measurements, based on multispectral imaging, visible spectra, and low mass impact response. Their relationship with maturity, as well as the potential of their combination was studied. As a result, two rather independent (R2 = 0.3) groups of non-destructive measurements, chlorophyll related optical indexes and low mass impact (LMI) measurements, were identified. Optical measurements showed the best behaviour for assessing maturity at harvest, while LMI measurements reflected handling incidences, showing a promising potential to be used to control transport and postharvest handling.

A new method for measuring the linewidth enhancement factor (α-parameter) of semiconductor lasers is proposed and discussed. The method itself provides an estimation of the measurement error, thus self-validating the entire procedure. The α-parameter is obtained from the temporal profile and the instantaneous frequency (chirp) of the pulses generated by gain switching. The time resolved chirp is measured with a polarization based optical differentiator. The accuracy of the obtained values of the α-parameter is estimated from the comparison between the directly measured pulse spectrum and the spectrum reconstructed from the chirp and the temporal profile of the pulse. The method is applied to a VCSEL and to a DFB laser emitting around 1550 nm at different temperatures, obtaining a measurement error lower than ± 8%.

The technique is based on the detection of the temporal dispersion of a short laser pulse injected into the probed medium. The time-distribution of re-emitted photons interpreted with a solution of the Diffusion equation yields the mean values of the absorption and reduced scattering coefficients of the medium. The proposed technique proved valuable for the measurement of the absorption and scattering spectra of different varieties of apples. No major variations were observed in the experimental data when the fruit was peeled, proving that the measured optical properties are referred to the pulp. The depth of probed volume was determined to be about 2 cm. Finally, the technique proved capable to follow the change in chlorophyll absorption during storage.

ZnTe doped with high concentrations of oxygen has been proposed in previous works as intermediate band (IB) material for photovoltaic applications. The existence of extra optical transitions related to the presence of an IB has already been demonstrated in this material and it has been possible to measure the absorption coefficient of the transitions from the valence band (VB) to the IB. In this work we present the first measurement of the absorption coefficient associated to transitions from the IB to the conduction band (CB) in ZnTe:O. The samples used are 4 ?m thick ZnTe layers with or without O in a concentration ~ 1019 cm-3, which have been grown on semi-insulating GaAs substrates by molecular beam epitaxy (MBE). The IB-CB absorption coefficient peaks for photon energies ~ 0.4 eV. It is extracted from reflectance and transmittance spectra measured using Fourier Transform Infrared (FTIR) spectroscopy. Under typical FTIR measurement conditions (low light intensity, broadband spectrum) the absorption coefficient in IB-to-CB transitions reaches 700 cm-1. This is much weaker than the one observed for VB-IB absorption. This result is consistent with the fact that the IB is expected to be nearly empty of electrons under equilibrium conditions in ZnTe(:O). The absorption for VB to IB transitions is also observed in the same samples through reflectance measurements performed in the visible range using a monochromator. These measurements are compared with the quantum efficiency (QE) from solar cells fabricated under similar conditions.

We present improved experimental transition probabilities for the optical Ca I 4s4p-4s4d and 4s4p-4p2multiplets. The values were determined with an absolute uncertainty of 10%. Transition probabilities have been determined by the branching ratios from the measurement of relative line intensities emitted by laser-induced plasma (LIP). The line intensities were obtained with the target (leadcalcium) placed in argon atmosphere at 6 Torr, recorded at a 2.5 µs delay from the laser pulse, which provides appropriate measurement conditions, and analysed between 350.0 and 550.0 nm. They are measured when the plasma reaches local thermodynamic equilibrium (LTE). The plasma is characterized by electron temperature (T) of 11400 K and an electron number density (Ne) of 1.1 x 1016 cm-3. The influence self-absorption has been estimated for every line, and plasma homogeneity has been checked. The values obtained were compared with previous experimental values in the literature. The method for measurement of transition probabilities using laser-induced plasma as spectroscopic source has been checked.

Resumen El diseño de sistemas ópticos, entendido como un arte por algunos, como una ciencia por otros, se ha realizado durante siglos. Desde los egipcios hasta nuestros días los sistemas de formación de imagen han ido evolucionando así como las técnicas de diseño asociadas. Sin embargo ha sido en los últimos 50 años cuando las técnicas de diseño han experimentado su mayor desarrollo y evolución, debido, en parte, a la aparición de nuevas técnicas de fabricación y al desarrollo de ordenadores cada vez más potentes que han permitido el cálculo y análisis del trazado de rayos a través de los sistemas ópticos de forma rápida y eficiente. Esto ha propiciado que el diseño de sistemas ópticos evolucione desde los diseños desarrollados únicamente a partir de la óptica paraxial hasta lo modernos diseños realizados mediante la utilización de diferentes técnicas de optimización multiparamétrica. El principal problema con el que se encuentra el diseñador es que las diferentes técnicas de optimización necesitan partir de un diseño inicial el cual puede fijar las posibles soluciones. Dicho de otra forma, si el punto de inicio está lejos del mínimo global, o diseño óptimo para las condiciones establecidas, el diseño final puede ser un mínimo local cerca del punto de inicio y lejos del mínimo global. Este tipo de problemática ha llevado al desarrollo de sistemas globales de optimización que cada vez sean menos sensibles al punto de inicio de la optimización. Aunque si bien es cierto que es posible obtener buenos diseños a partir de este tipo de técnicas, se requiere de muchos intentos hasta llegar a la solución deseada, habiendo un entorno de incertidumbre durante todo el proceso, puesto que no está asegurado el que se llegue a la solución óptima. El método de las Superficies Múltiples Simultaneas (SMS), que nació como una herramienta de cálculo de concentradores anidólicos, se ha demostrado como una herramienta también capaz utilizarse para el diseño de sistemas ópticos formadores de imagen, aunque hasta la fecha se ha utilizado para el diseño puntual de sistemas de formación de imagen. Esta tesis tiene por objeto presentar el SMS como un método que puede ser utilizado de forma general para el diseño de cualquier sistema óptico de focal fija o v afocal con un aumento definido así como una herramienta que puede industrializarse para ayudar al diseñador a afrontar de forma sencilla el diseño de sistemas ópticos complejos. Esta tesis está estructurada en cinco capítulos: El capítulo 1, es un capítulo de fundamentos donde se presentan los conceptos fundamentales necesarios para que el lector, aunque no posea una gran base en óptica formadora de imagen, pueda entender los planteamientos y resultados que se presentan en el resto de capítulos El capitulo 2 aborda el problema de la optimización de sistemas ópticos, donde se presenta el método SMS como una herramienta idónea para obtener un punto de partida para el proceso de optimización. Mediante un ejemplo aplicado se demuestra la importancia del punto de partida utilizado en la solución final encontrada. Además en este capítulo se presentan diferentes técnicas que permiten la interpolación y optimización de las superficies obtenidas a partir de la aplicación del SMS. Aunque en esta tesis se trabajará únicamente utilizando el SMS2D, se presenta además un método para la interpolación y optimización de las nubes de puntos obtenidas a partir del SMS3D basado en funciones de base radial (RBF). En el capítulo 3 se presenta el diseño, fabricación y medidas de un objetivo catadióptrico panorámico diseñado para trabajar en la banda del infrarrojo lejano (8-12 μm) para aplicaciones de vigilancia perimetral. El objetivo presentado se diseña utilizando el método SMS para tres frentes de onda de entrada utilizando cuatro superficies. La potencia del método de diseño utilizado se hace evidente en la sencillez con la que este complejo sistema se diseña. Las imágenes presentadas demuestran cómo el prototipo desarrollado cumple a la perfección su propósito. El capítulo 4 aborda el problema del diseño de sistemas ópticos ultra compactos, se introduce el concepto de sistemas multicanal, como aquellos sistemas ópticos compuestos por una serie de canales que trabajan en paralelo. Este tipo de sistemas resultan particularmente idóneos para él diseño de sistemas afocales. Se presentan estrategias de diseño para sistemas multicanal tanto monocromáticos como policromáticos. Utilizando la novedosa técnica de diseño que en este capítulo se presenta el diseño de un telescopio de seis aumentos y medio. En el capítulo 5 se presenta una generalización del método SMS para rayos meridianos. En este capítulo se presenta el algoritmo que debe utilizarse para el diseño de cualquier sistema óptico de focal fija. La denominada optimización fase 1 se vi introduce en el algoritmo presentado de forma que mediante el cambio de las condiciones iníciales del diseño SMS que, aunque el diseño se realice para rayos meridianos, los rayos skew tengan un comportamiento similar. Para probar la potencia del algoritmo desarrollado se presenta un conjunto de diseños con diferente número de superficies. La estabilidad y potencia del algoritmo se hace evidente al conseguirse por primera vez el diseño de un sistema de seis superficies diseñado por SMS. vii Abstract The design of optical systems, considered an art by some and a science by others, has been developed for centuries. Imaging optical systems have been evolving since Ancient Egyptian times, as have design techniques. Nevertheless, the most important developments in design techniques have taken place over the past 50 years, in part due to the advances in manufacturing techniques and the development of increasingly powerful computers, which have enabled the fast and efficient calculation and analysis of ray tracing through optical systems. This has led to the design of optical systems evolving from designs developed solely from paraxial optics to modern designs created by using different multiparametric optimization techniques. The main problem the designer faces is that the different optimization techniques require an initial design which can set possible solutions as a starting point. In other words, if the starting point is far from the global minimum or optimal design for the set conditions, the final design may be a local minimum close to the starting point and far from the global minimum. This type of problem has led to the development of global optimization systems which are increasingly less sensitive to the starting point of the optimization process. Even though it is possible to obtain good designs from these types of techniques, many attempts are necessary to reach the desired solution. This is because of the uncertain environment due to the fact that there is no guarantee that the optimal solution will be obtained. The Simultaneous Multiple Surfaces (SMS) method, designed as a tool to calculate anidolic concentrators, has also proved useful for the design of image-forming optical systems, although until now it has occasionally been used for the design of imaging systems. This thesis aims to present the SMS method as a technique that can be used in general for the design of any optical system, whether with a fixed focal or an afocal with a defined magnification, and also as a tool that can be commercialized to help designers in the design of complex optical systems. The thesis is divided into five chapters. Chapter 1 establishes the basics by presenting the fundamental concepts which the reader needs to acquire, even if he/she doesn‟t have extensive knowledge in the field viii of image-forming optics, in order to understand the steps taken and the results obtained in the following chapters. Chapter 2 addresses the problem of optimizing optical systems. Here the SMS method is presented as an ideal tool to obtain a starting point for the optimization process. The importance of the starting point for the final solution is demonstrated through an example. Additionally, this chapter introduces various techniques for the interpolation and optimization of the surfaces obtained through the application of the SMS method. Even though in this thesis only the SMS2D method is used, we present a method for the interpolation and optimization of clouds of points obtained though the SMS3D method, based on radial basis functions (RBF). Chapter 3 presents the design, manufacturing and measurement processes of a catadioptric panoramic lens designed to work in the Long Wavelength Infrared (LWIR) (8-12 microns) for perimeter surveillance applications. The lens presented is designed by using the SMS method for three input wavefronts using four surfaces. The powerfulness of the design method used is revealed through the ease with which this complex system is designed. The images presented show how the prototype perfectly fulfills its purpose. Chapter 4 addresses the problem of designing ultra-compact optical systems. The concept of multi-channel systems, such as optical systems composed of a series of channels that work in parallel, is introduced. Such systems are especially suitable for the design of afocal systems. We present design strategies for multichannel systems, both monochromatic and polychromatic. A telescope designed with a magnification of six-and-a-half through the innovative technique exposed in this chapter is presented. Chapter 5 presents a generalization of the SMS method for meridian rays. The algorithm to be used for the design of any fixed focal optics is revealed. The optimization known as phase 1 optimization is inserted into the algorithm so that, by changing the initial conditions of the SMS design, the skew rays have a similar behavior, despite the design being carried out for meridian rays. To test the power of the developed algorithm, a set of designs with a different number of surfaces is presented. The stability and strength of the algorithm become apparent when the first design of a system with six surfaces if obtained through the SMS method.

The production and industry of paprika present several problems related to quality and to production costs. One of the main difficulties is to obtain an objective and quick method for predicting quality. Quality in powder paprika involves: quantity of carotenoids and the appearance and stability of colour. The method used currently for determining quality is the measurement of absorbance at 460 nm wavelength, of an acetone extract of carotenoids, but there is no information about the appearance of the paprika or the stability of its colour with time. " Another important problem is the presence of mixtures of powdered paprika produced in the Spanish region of "La Vera", which has a peculiar way of production, with a high '' quality and price, with other products of lower quality. It is necessary to obtain methods which are able to detect the fraud.

A fully automated instrumentation for time-resolved reflectance measurements was developed It is based on mode-locked laser sources and electronics for time-correlated single photon counting, and provides a time-resolution of 120-160 ps The system was used to probe the optical properties of several species and varieties of fruits and vegetables in the red and NIR range (650-1000 nm). In most fruits, the absorption line shape is dominated by the absorption peak of water, centred around 970 nm Generally, the absorption spectra also show the spectral features typical of chlorophyll, with maximum at 675 nm In particular, for what concerns apples, variations in peak intensity are observed depending on the variety, the degree of ripeness as well as the position on the apple. For all the species and varieties considered, the transport scattering coefficient decreases progressively upon increasing the wavelength.

Mealiness is a textural attribute related to an internal fruit disorder that involves quality loss. It is characterised by the combination of abnormal softness of the fruit and absence of free juiciness in the mouth when eaten by the consumer. Recent research concluded with the development of precise instrumental procedure to measure a scale of mealiness based on the combination of several rheological properties and empirical magnitudes. In this line, time-domain laser reflectance spectroscopy (TDRS) is a new medical technology, used to characterise the optical properties of tissues, and to locate affected areas like tumours. Among its advantages compared to more traditional spectroscopic techniques, there is the feasibility to asses simultaneously and independently two optical parameters: the absorption of the light inside the irradiated body, and the scattering of the photons across the tissues, at each wavelength, generating two coefficients (µa, absorption coeff.; and µ's, transport scattering coeff.). If it is assumed that they are related respectively to chemical components and to physical properties of the sample, TDRS can be applied to the quantification of chemicals and the measurement of the rheological properties (i.e. mealiness estimation) at the same time. Using VIS & NIR lasers as light sources, TDRS was applied in this work to Golden Delicious and Cox apples (n=90), conforming several batches of untreated samples and storage-treated (20°C & 95%RH) to promote the development of mealiness. The collected database was clustered into different groups according to their instrumental test values (Barreiro et al, 1998). The optical coefficients were used as explanatory variables when building discriminant analysis functions for mealiness, achieving a classification score above 80% of correctly identified mealy versus fresh apples.

In developing instrumentation for the measurement of fruit quality, there is the need for fast and non-destructive devices, based on sensors, to be installed on-line. In the case of some fruits, like peaches, post-harvest ripeness, which is closely related to high quality for the consumer, is a priority. During ripening, external appearance (colour) and internal mechanical (firmness) and chemical (sugars and acids) quality are main features that evolve rapidly from and unripe to a ripe (high quality) stage. When considering the evolution of fruit quality in this scheme, external colour and firmness are shown to evolve in a parallel pattern, if monitored from the time of harvest to full consumer ripeness ( Rood, 1957; Crisosto et al, 1995; Kader, 1996). The visible (VIS) reflectance spectrum is a fast and easy reference that can be used to estimate quality of peaches, if we could show it to be reliably correlated with peach ripening rate during postharvest (Genard et al. 1994; Moras, 1995; Delwiche and Baumgartner, 1983; Delwiche et al. 1987; Slaughter, 1995; Lleo et al., 1998). Taste, described as an expert acceptance score, improves with ripeness (firmness and colour evolution), when considering the fruits on the tree, and also post-harvest.
