Paramount to symbiotic nitrogen fixation (SNF) is the synthesis of a number of metalloenzymes that use iron as a critical component of their catalytical core. Since this process is carried out by endosymbiotic rhizobia living in legume root nodules, the mechanisms involved in iron delivery to the rhizobia-containing cells are critical for SNF. In order to gain insight into iron transport to the nodule, we have used synchrotron-based X-ray fluorescence to determine the spatio-temporal distribution of this metal in nodules of the legume Medicago truncatula with hitherto unattained sensitivity and resolution. The data support a model in which iron is released from the vasculature into the apoplast of the infection/differentiation zone of the nodule (zone II). The infected cell subsequently takes up this apoplastic iron and delivers it to the symbiosome and the secretory system to synthesize ferroproteins. Upon senescence, iron is relocated to the vasculature to be reused by the shoot. These observations highlight the important role of yet to be discovered metal transporters in iron compartmentalization in the nodule and in the recovery of an essential and scarce nutrient for flowering and seed production.

Symbiotic nitrogen fixation is a process that requires relatively high quantities of iron provided by the host legume. Using synchrotron-based X-ray fluorescence, we have determined that this iron is released from the vasculature into the apoplast of zone II of M. truncatula nodules. This overlaps with the distribution of MtNramp1, a plasma membrane iron importer. The importance of MtNramp1 in iron transport for nitrogen fixation is indicated by the 60% reduction of nitrogenase activity observed in knock-down lines, most likely due to deficient incorporation of this essential metal cofactor at the necessary levels.

Iron is critical for symbiotic nitrogen fixation (SNF) as a key component ofmultiple ferroproteins involved in this biological process. In the model legume Medicago truncatula, iron is delivered by the vasculature to the infection/maturation zone (zone II) of the nodule, where it is released to the apoplast. From there, plasma membrane iron transporters move it into rhizobia-containing cells, where iron is used as the cofactor of multiple plant and rhizobial proteins (e.g. plant leghemoglobin and bacterial nitrogenase). MtNramp1 (Medtr3g088460) is the M. truncatula Natural Resistance-Associated Macrophage Protein family member, with the highest expression levels in roots and nodules. Immunolocalization studies indicate that MtNramp1 is mainly targeted to the plasma membrane. A loss-of-function nramp1 mutant exhibited reduced growth compared with the wild type under symbiotic conditions, but not when fertilized with mineral nitrogen. Nitrogenase activity was low in the mutant, whereas exogenous iron and expression of wild-type MtNramp1 in mutant nodules increased nitrogen fixation to normal levels. These data are consistent with a model in which MtNramp1 is the main transporter responsible for apoplastic iron uptake by rhizobia-infected cells in zone II.

Amidases [EC 3.5.1.4] capable of converting indole-3-acetamide (IAM) into the major plant growth hormone indole-3-acetic acid (IAA) are assumed to be involved in auxin de novo biosynthesis. With the emerging amount of genomics data, it was possible to identify over forty proteins with substantial homology to the already characterized amidases from Arabidopsis and tobacco. The observed high conservation of amidase-like proteins throughout the plant kingdom may suggest an important role of theses enzymes in plant development. Here, we report cloning and functional analysis of four, thus far, uncharacterized plant amidases from Oryza sativa, Sorghum bicolor, Medicago truncatula, and Populus trichocarpa. Intriguingly, we were able to demonstrate that the examined amidases are also capable of converting phenyl-2-acetamide (PAM) into phenyl-2-acetic acid (PAA), an auxin endogenous to several plant species including Arabidopsis. Furthermore, we compared the subcellular localization of the enzymes to that of Arabidopsis AMI1, providing further evidence for similar enzymatic functions. Our results point to the presence of a presumably conserved pathway of auxin biosynthesis via IAM, as amidases, both of monocot, and dicot origins, were analyzed.

El uso intensivo de compuestos de cobre como herbicidas y fungicidas provoca la contaminación de suelos de uso agrícola debido a la acumulación de este metal en las capas más superficiales del suelo. Se sabe que la presencia de cobre y otros metales pesados afecta negativamente a las interacciones simbióticas que se establecen entre bacterias diazotróficas de los géneros Rhizobium, Sinorhizobium y Bradyrhizobium y leguminosas de interés agrícola (Laguerre et al., 2006). El objetivo de este trabajo es estudiar la diversidad de cepas endosimbióticas de leguminosas en suelos agrícolas chilenos que presentan un elevado contenido en cobre como resultado de la contaminación con residuos de extracciones mineras. Además, se pretende caracterizar el nivel de resistencia a cobre en las cepas aisladas con objeto de identificar aquellas altamente eficientes que puedan ser utilizadas como inoculantes microbianos. Para ello, se han prospectado 9 suelos agrícolas de las regiones III, V y VI de Chile con contenidos muy variables de metales. Utilizando estos suelos como inóculos de plantas trampa de leguminosas se ha obtenido una colección de 362 cepas aisladas de nódulos de guisante (Pisum sativum), judía (Phaseolus vulgaris) y alfalfa (Medicago sativa). Los análisis filogenéticos y los ensayos de resistencia a cobre realizados han permitido caracterizar y seleccionar aquellas cepas con mayores niveles de resistencia a este metal. Los resultados demuestran que los suelos altamente contaminados por cobre poseen una menor diversidad de bacterias endosimbióticas; las cepas más resistentes han sido aisladas de los suelos con niveles de contaminación intermedia. Los análisis fenotípicos y moleculares realizados sobre las cepas más resistentes han demostrado la existencia de sistemas de resistencia a cobre inducibles por este metal y potencialmente implicados en su homeostasis.
