The tropical montane forests of the E Andean cordillera in Ecuador receive episodic Sahara- dust inputs particularly increasing Ca deposition. We added CaCl2 to isolate the effect of Ca deposition by Sahara dust to tropical montane forest from the simultaneously occurring pH effect. We examined components of the Ca cycle at four control plots and four plots with added Ca (2 × 5 kg ha?1 Ca annually as CaCl2) in a random arrangement. Between August 2007 and December 2009 (four applications of Ca), we determined Ca concentrations and fluxes in litter leachate, mineral soil solution (0.15 and 0.30 m depths), throughfall, and fine litterfall and Al con- centrations and speciation in soil solutions. After 1 y of Ca addition, we assessed fine-root bio- mass, leaf area, and tree growth. Only < 3% of the applied Ca leached below the acid organic layer (pH 3.5?4.8). The added CaCl2 did not change electrical conductivity in the root zone after 2 y. In the second year of fertilization, Ca retention in the canopy of the Ca treatment tended to decrease relative to the control. After 2 y, 21% of the applied Ca was recycled to soil with throughfall and litterfall. One year after the first Ca addition, fine-root biomass had decreased significantly. Decreasing fine-root biomass might be attributed to a direct or an indirect beneficial effect of Ca on the soil decomposer community. Because of almost complete association of Al with dissolved organic matter and high free Ca2+ : Al3+ activity ratios in solution of all plots, Al toxicity was unlikely. We conclude that the added Ca was retained in the system and had benefi- cial effects on some plants.

We have determined matrix elements for all experimental configurations of Ca III, including the 3s3p63d configuration. These values have been obtained using intermediate coupling (IC). For these IC calculations, we have used the standard method of least-squares fitting from the experimental energy levels, using the computer code developed by Robert Cowan. In this paper, using these matrix elements, we report the calculated values of the Ca III Stark widths and shifts for 148 spectral lines, of 56 Ca III spectral line transition probabilities and of eight radiative lifetimes of Ca III levels. The Stark widths and shifts, calculated using the Griem semi-empirical approach, correspond to the spectral lines of Ca III and are presented for an electron density of 1017 cm?3 and temperatures T = 1.0?10.0 (×104 K). The theoretical trends of the Stark broadening parameter versus the temperature are presented for transitions that are of astrophysical interest. There is good agreement between our calculations, for transition probabilities and radiative lifetimes, and the experimental values presented in the literature. We have not been able to find any values for the Stark parameters in the references.

Ca-amendments are routinely applied to improve acid soils, whilst no-tillage (NT) has been widely recommended in soils where traditional tillage (TT) has led to losses of organic matter. However, the potential interactions between the two treatments are only partially known. Our study was conducted on an annual forage crop agrosystem with a degraded Palexerult soil located in SW Spain, in order to assess if the combination of NT plus a Ca-amendment provides additional benefits to those of their separate use. To this end we analysed the effects of four different combinations of tillage and Ca-amendment on selected key soil properties, focusing on their relationships. The experimental design was a split-plot with four replicates. The main factor was tillage (NT versus TT) and the second factor was the application or not of a Ca-amendment, consisting of a mixture of sugar foam (SF) and red gypsum (RG). Soil samples were collected from 3 soil layers down to 50 cm after four years of treatment (2009). The use of the Ca-amendment improved pH and Al-toxicity down to 25 cm and increased exchangeable Ca2+ down to 50 cm, even under NT due to the combined effect of SF and RG. Both NT and the Ca-amendment had a beneficial effect on total organic carbon (TOC), especially on particulate organic carbon (POC), in the 0–5 cm layer, with the highest contents observed when both practices were combined. Unlike NT, the Ca-amendment failed to improve soil aggregation in spite of the carbon supplied. This carbon was not protected within the stable aggregates in the medium term, making it more susceptible to mineralization. We suggest that the fraction of Al extracted by oxalate from solid phase (AlOxa-Cu-K) and the glomalin-related soil proteins (GRSPs) are involved in the accumulation of carbon within water stable aggregates, probably through the formation of non-toxic stable Al-OM compounds, including those formed with GRSPs. NT alone decreased AlK in the 0–5 cm soil layer, possibly by increasing POC, TOC and GRSPs, which were observed to play a role in reducing Al toxicity. From our findings, the combination of NT and Ca-amendment appears to be the best management practice to improve chemical and physical characteristics of acid soils degraded by tillage.

En los suelos, el exceso de acidez lleva asociado deficiencias en ciertos nutrientes y una alta disponibilidad de aluminio, tóxico para los cultivos propios del ambiente mediterráneo. Su laboreo, provoca la pérdida de materia orgánica (MO), deteriora su estructura y reduce la actividad biológica, provocando en última instancia una menor calidad del suelo. Es de esperar pues que cuando se labran suelos ácidos, sus problemáticas particulares tiendan a agravarse. En nuestra zona de estudio, la “raña” de Caamero (Extremadura, España), predominan los suelos muy ácidos y degradados por un laboreo inadecuado. Las rañas constituyen amplias plataformas casi horizontales, con unos suelos muy viejos (Palexerults), que se caracterizan por tener el complejo de cambio dominado por el aluminio, y un pH ácido que decrece en profundidad. Poseen un potente horizonte Bt rico en arcillas caoliníticas, que propicia que en periodos con exceso de lluvia, se generen capas colgadas de agua cercanas a la superficie. En torno a los años 1940’s estos suelos, que previamente sostenían un alcornocal, o su matorral de sustitución, se pusieron en cultivo. El laboreo aceleró la mineralización de la materia orgánica, agravó los problemas derivados del exceso de acidez y condujo al abandono de los campos cultivados por falta de productividad. Para recuperar la calidad de estos suelos degradados y obtener unos rendimientos compatibles con su uso agrícola es necesario, por un lado, aplicar enmiendas que eleven el pH y reduzcan la toxicidad del aluminio y, por otro, favorecer el incremento en el contenido en MO. En 2005 se implantó en esta raña un ensayo de campo para estudiar la influencia del no laboreo y de la utilización de una enmienda cálcica en parámetros relacionados con la calidad del suelo en un cultivo forrajero. El diseño experimental fue en parcelas divididas con cuatro repeticiones donde el factor principal fue el tipo de laboreo, no laboreo (NL) frente a laboreo convencional (LC), y el factor secundario el uso o no de una enmienda cálcica. La enmienda consistió básicamente en una mezcla de espuma de azucarería y yeso rojo y se incorporó al comienzo del ensayo hasta los 7 cm de profundidad. Desde el comienzo del ensayo el NL influyó positivamente en el contenido de carbono orgánico total (COT) y particulado (COP), mientras que la enmienda tuvo una ligera influencia al principio del ensayo en ambos pero su efecto positivo se desvaneció con el paso del tiempo. Los mayores contenidos en COT y POC se observaron cuando se combinó el NL con la enmienda. La enmienda incrementó con rapidez el pH, y el Ca, y disminuyó el contenido en aluminio hasta una profundidad de 50 cm, incluso en NL, y mejoró ligeramente la agregación del suelo. El NL por sí solo, gracias al aumento en POC, TOC y las proteínas del suelo relacionadas con la glomalina (PSRG), que son capaces de formar compuestos estables no tóxicos con el aluminio, también contribuyó a la reducción de la toxicidad de aluminio en la capa más superficial. Cuando en las campañas con exceso de precipitaciones se generaron capas colgadas de agua próximas a la superficie, el NL generó unas condiciones más favorables para la germinación y desarrollo del cultivo, resultando en una producción más alta que el LC. A ello contribuyó la mayor capacidad de almacenamiento de agua y la mayor transmisividad de esta hacia abajo, en la capa más superficial (0-5 cm) que propició una menor saturación por agua que el LC. Respecto a los parámetros relacionados con la agregación, el NL aumentó los macroagregados hasta los 10 cm de profundidad y favoreció la acumulación de CO y N en todas las fracciones de tamaño de agregados. Sin embargo, la recuperación del grado de macroagregación tras el cese del laboreo resulta lenta en comparación con otros suelos, posiblemente debido al bajo contenido en arcilla en el horizonte Ap. En comparación con el NL, la enmienda mostró también un efecto positivo, aunque muy ligero, en la agregación del suelo. En contradicción con otros estudios en suelos ácidos, nuestros resultados indican la existencia de una jerarquía de agregados, y destacan el papel importante de la MO en la mejora de la agregación. Tanto el NL como la enmienda favorecieron por separado varias propiedades químicas, físicas y biológicas del suelo, pero, en general, encontramos los mayores beneficios con su uso combinado. Además, a largo plazo el efecto positivo de NL en las propiedades del suelo fue en aumento, mientras que el efecto beneficioso de la enmienda se limitó básicamente a las propiedades químicas y se desvaneció en pocos años. Destacamos que las condiciones meteorológicas a lo largo del ensayo beneficiaron la producción de biomasa en NL, y en consecuencia las propiedades relacionadas con la materia orgánica, por lo que son un factor a tener en cuenta a la hora de evaluar los efectos de la enmienda y el laboreo sobre las propiedades del suelo, especialmente en zonas donde esas condiciones son muy variables entre una campaña y otra. Los resultados de este estudio han puesto de manifiesto que el NL no ha mermado la eficacia de la enmienda caliza, posiblemente gracias a la alta solubilidad de la enmienda aplicada, es más, el manejo con NL y enmienda es el que ha favorecido en mayor medida ciertos parámetros de calidad del suelo. Por el contrario el LC sí parece anular los beneficios de la enmienda en relación con las propiedades relacionadas con la MO. Por tanto, cabe concluir que la combinación de NL y la enmienda es una práctica adecuada para mejorar las propiedades químicas y físicas de suelos ácidos degradados por el laboreo. ABSTRACT Excessive acidity in soils is associated with deficiencies in certain nutrients and high concentrations of available aluminum, which is toxic for most Mediterranean crops. Tilling these soils results in the loss of soil organic matter (SOM), damages soil structure and reduces biological activity, ultimately degrading soil quality. It is expected, therefore, that when acid soils are tilled, their particular problems will tend to get worse. In our study area, the "Caamero’s Raña” (Extremadura, Spain), acid soils degraded by an inappropriate tillage prevail. Rañas are large and flat platforms with very old soils (Palexerults), which are characterized by an exchange complex dominated by aluminum and an acid pH which decreases with depth. These soils have a strong Bt horizon rich in kaolinite clays, which encourages the formation of perched water-tables near the soil surface during periods of excessive rain. During the first third of the 20th century, these soils, that previously supported cork oak or its scrub replacement, were cultivated. Tillage accelerated the mineralization of the SOM, aggravating the problems of excessive acidity, which finally led to the abandonment of the land due to low productivity. To recover the quality of these degraded soils and to obtain consistent yields it is necessary, first, to apply amendments to raise the pH and reduce aluminum toxicity, and second to encourage the accumulation of SOM. In 2005 a field trial was established in the Raña to study the influence of no-tillage and the use of a Ca-amendment on soil quality related parameters in a forage crop agrosystem. The experimental design was a split-plot with four replicates where the main factor was tillage type, no-tillage (NT) versus traditional tillage (TT) and the secondary factor was the use or not of a Ca-amendment. The Ca-amendment was a mixture of sugar foam and red gypsum that was incorporated into the top 7 cm of the soil. Since the beginning of the experiment, NT had a positive influence on total and particulate organic carbon (TOC and POC, respectively), while the Ca-amendment had a small positive influence at the beginning of the study but its effect diminished with time. The highest TOC and POC contents were observed when NT and the Ca-amendment were combined. The Ca-amendment, even under NT, rapidly increased pH and Ca, and decreased the aluminum content to a depth of 50 cm, as well as improving soil aggregation slightly. NT, due to the increased POC, TOC and Glomalin-related soil proteins (GRSP), which can form stable non-toxic compounds with aluminum, also contributed to the reduction of aluminum toxicity in the upper layer. When perched water-tables near the soil surface were formed in campaigns with excessive rainfall, NT provided more favorable conditions for germination and crop development, resulting in higher yields compared with TT. This was directly related to the higher water storage capacity and the greater transmissivity of the water downwards from the upper layers, which led to lower water saturation under NT compared with TT. With regards to the aggregation-related parameters, NT increased macroaggregation to a depth of 10 cm and favored the accumulation of OC and N in all aggregate size fractions. However, the degree of recovery of macroaggregation after tillage ceased was slow compared with other soils, possibly due to the low clay content in the Ap horizon. Compared with NT, the Ca-amendment had a slight positive effect on soil aggregation. In contrast to other studies in acid soils, our results indicate the existence of an aggregate hierarchy, and highlight the important role of SOM in improving aggregation. Both NT and the Ca-amendment separately favored various chemical, physical and biological soil properties, but in general we found the greatest benefits when the two treatments were combined. In addition, the positive effect of NT on soil properties increased with time, while the beneficial effect of the Ca-amendment, which was limited to the chemical properties, vanished after a few years. It is important to note that the meteorological conditions throughout the experiment benefited biomass production under NT and, as a consequence, organic matter related properties. This suggests that meteorological conditions are a factor to consider when evaluating the effects of Ca-amendments and tillage on soil properties, especially in areas where such conditions vary significantly from one campaign to another. The results of this study show that NT did not diminish the effectiveness of the Ca-amendment, possibly due to the high solubility of the selected amendment. Moreover, the combination of NT and the Ca-amendment was actually the management that favored certain soil quality parameters the most. By contrast, TT seemed to nullify the benefits of the Ca-amendment with regards to the OM related properties. In conclusion, the combination of NT and the application of a Ca-amendment is an advisable practice for improving the chemical and physical properties of acid soils degraded by tillage.

Experiments to evaluate the effect of in-season calcium (Ca) sprays on late-season peach (Prunus persica L. Batsch cv. Calrico) were carried out for a 2-year period. Calcium formulations (0.5% and 1.0% in 2008 and only 0.5% tested in 2009) supplied either as CaCl2 or Ca propionate in combination with two or three adjuvants (0.05% of the nonionic surfactants Tween 20 and Break Thru, and 0.5% carboxymethylcellulose, CMC) were sprayed four to five times over the growing season. Peach mesocarp and endocarp Ca concentrations were determined on a 15-day basis from the beginning of May until the end of June. Further tissue analyses were performed at harvest. A decreasing trend in fruit Ca concentrations over the growing season was always observed regardless of the Ca treatments. Both in 2008 and 2009, significant tissue Ca increments associated with the application of Ca-containing sprays in combination with adjuvants were only observed in June, which may be coincident with the period of pit hardening. In 2008, both at harvest and after cold storage, the total soluble-solids concentration (° Brix) of fruits supplied with Ca propionate (0.5% and 1.0% Ca) was always lower as compared to the rest of treatments. The application of multiple Ca-containing sprays increased firmness at harvest and after cold storage, especially when CaCl2 was the active ingredient used. Supplying the adjuvants Tween 20 and CMC increased fruit acidity both at harvest and after cold storage. Evaluation of the development of physiological disorders after cold storage (2 weeks at 0°C) indicated a lower susceptibility of Ca-treated fruits to internal browning. Fruits treated with multiple CaCl2-, CMC-, and Break Thru®-containing sprays during the growing season were significantly less prone to the development of chilling injuries as compared to untreated peaches.

The effects of fire ( Control burned soil) and two emergency stabilisation techniques (grass Seeding and straw Mulching ) on 20 chemical characteristics were evaluated on 0 – 5 cm top-soils sampled 1, 90, 180 and 365 days after an experimental fi re in a steep shrubland of a temperate-humid region (NW Spain). Most part of pH (in H 2 O and KCl) variance was explained by the sampling date. No clear temporal trends were identi fi able for total soil C and N content, likely due to the large SOM pool in these soils; however, changes on soil δ 13 C were explained by the deposition of 13 C-depleted ashes, followed by its progressive erosion, while those on soil δ 15 N were a consequence of fi re induced N outputs. After the fi re, NH 4 + – N, P, Na, K, Mg, Ca, Mn, Cu, Zn and B concentrations increased, while those of NO 3 − – N, Al, Fe and Co did not vary significantly. Despite a significant decline with time, concentrations of Mg, Ca and Mn at the end of the study were still higher than in unburned soil, while those of K, Cu, Zn and B were similar to the pre-fire levels and those of NH 4 + – N, P and Na were below pre-fire values. Mulching and Seeding treatments for burned soil emergency stabilisation had significant effects on soil δ 15 N and extractable K, Mg and Ca, while data were inconclusive for their possible effects on the extractable Al, Fe and Co

Las cascadas de señalización mediadas por proteína quinasas activadas por mitógeno (MAP quinasas) son capaces de integrar y transducir señales ambientales en respuestas celulares. Entre estas señales se encuentran los PAMPs/MAMPs (Pathogen/Microbe-Associated Molecular Patterns), que son moléculas de patógenos o microorganismos, o los DAMPs (Damaged-Associated Molecular Patterns), que son moléculas derivadas de las plantas producidas en respuesta a daño celular. Tras el reconocimiento de los PAMPs/DAMPs por receptores de membrana denominados PRRs (Pattern Recognition Receptors), como los receptores con dominio quinasa (RLKs) o los receptores sin dominio quinasa (RLPs), se activan respuestas moleculares, incluidas cascadas de MAP quinasas, que regulan la puesta en marcha de la inmunidad activada por PAMPs (PTI). Esta Tesis describe la caracterización funcional de la MAP quinasa quinasa quinasa (MAP3K) YODA (YDA), que actúa como un regulador clave de la PTI en Arabidopsis. Se ha descrito previamente que YDA controla varios procesos de desarrollo, como la regulación del patrón estomático, la elongación del zigoto y la arquitectura floral. Hemos caracterizado un alelo mutante hipomórfico de YDA (elk2 o yda11) que presenta una elevada susceptibilidad a patógenos biótrofos y necrótrofos. Notablemente, plantas que expresan una forma constitutivamente activa de YDA (CA-YDA), con una deleción en el dominio N-terminal, presentan una resistencia de amplio espectro frente a diferentes tipos de patógenos, incluyendo hongos, oomicetos y bacterias, lo que indica que YDA juega un papel importante en la regulación de la resistencia de las plantas a patógenos. Nuestros datos indican que esta función es independiente de las respuestas inmunes mediadas por los receptores previamente caracterizados FLS2 y CERK1, que reconocen los PAMPs flg22 y quitina, respectivamente, y que están implicados en la resistencia de Arabidopsis frente a bacterias y hongos. Hemos demostrado que YDA controla la resistencia frente al hongo necrótrofo Plectosphaerella cucumerina y el patrón estomático mediante su interacción genética con la RLK ERECTA (ER), un PRR implicado en la regulación de estos procesos. Por el contrario, la interacción genética entre ER y YDA en la regulación de otros procesos de desarrollo es aditiva en lugar de epistática. Análisis genéticos indicaron que MPK3, una MAP quinasa que funciona aguas abajo de YDA en el desarrollo estomático, es un componente de la ruta de señalización mediada por YDA para la resistencia frente a P. cucumerina, lo que sugiere que el desarrollo de las plantas y la PTI comparten el módulo de transducción de MAP quinasas asociado a YDA. Nuestros experimentos han revelado que la resistencia mediada por YDA es independiente de las rutas de señalización reguladas por las hormonas de defensa ácido salicílico, ácido jasmónico, ácido abscísico o etileno, y también es independiente de la ruta de metabolitos secundarios derivados del triptófano, que están implicados en inmunidad vegetal. Además, hemos demostrado que respuestas asociadas a PTI, como el aumento en la concentración de calcio citoplásmico, la producción de especies reactivas de oxígeno, la fosforilación de MAP quinasas y la expresión de genes de defensa, no están afectadas en el mutante yda11. La expresión constitutiva de la proteína CA-YDA en plantas de Arabidopsis no provoca un aumento de las respuestas PTI, lo que sugiere la existencia de mecanismos de resistencia adicionales regulados por YDA que son diferentes de los regulados por FLS2 y CERK1. En línea con estos resultados, nuestros datos transcriptómicos revelan una sobre-representación en plantas CA-YDA de genes de defensa que codifican, por ejemplo, péptidos antimicrobianos o reguladores de muerte celular, o proteínas implicadas en la biogénesis de la pared celular, lo que sugiere una conexión potencial entre la composición e integridad de la pared celular y la resistencia de amplio espectro mediada por YDA. Además, análisis de fosfoproteómica indican la fosforilación diferencial de proteínas relacionadas con la pared celular en plantas CA-YDA en comparación con plantas silvestres. El posible papel de la ruta ER-YDA en la regulación de la integridad de la pared celular está apoyado por análisis bioquímicos y glicómicos de las paredes celulares de plantas er, yda11 y CA-YDA, que revelaron cambios significativos en la composición de la pared celular de estos genotipos en comparación con la de plantas silvestres. En resumen, nuestros datos indican que ER y YDA forman parte de una nueva ruta de inmunidad que regula la integridad de la pared celular y respuestas defensivas, confiriendo una resistencia de amplio espectro frente a patógenos. ABSTRACT Plant mitogen-activated protein kinase (MAPK) cascades transduce environmental signals and developmental cues into cellular responses. Among these signals are the pathogen- or microbe-associated molecular patterns (PAMPs or MAMPs) and the damage-associated molecular patterns (DAMPs). These PAMPs/DAMPs, upon recognition by plant pattern recognition receptors (PRRs), such as Receptor-Like Kinases (RLKs) and Receptor-Like Proteins (RLPs), activate molecular responses, including MAPK cascades, which regulate the onset of PAMP-triggered immunity (PTI). This Thesis describes the functional characterization of the MAPK kinase kinase (MAP3K) YODA (YDA) as a key regulator of Arabidopsis PTI. YDA has been previously described to control several developmental processes, such as stomatal patterning, zygote elongation and inflorescence architecture. We characterized a hypomorphic, non-embryo lethal mutant allele of YDA (elk2 or yda11) that was found to be highly susceptible to biotrophic and necrotrophic pathogens. Remarkably, plants expressing a constitutive active form of YDA (CA-YDA), with a deletion in the N-terminal domain, showed broad-spectrum resistance to different types of pathogens, including fungi, oomycetes and bacteria, indicating that YDA plays a relevant function in plant resistance to pathogens. Our data indicated that this function is independent of the immune responses regulated by the well characterized FLS2 and CERK1 RLKs, which are the PRRs recognizing flg22 and chitin PAMPs, respectively, and are required for Arabidopsis resistance to bacteria and fungi. We demonstrate that YDA controls resistance to the necrotrophic fungus Plectosphaerella cucumerina and stomatal patterning by genetically interacting with ERECTA (ER) RLK, a PRR involved in regulating these processes. In contrast, the genetic interaction between ER and YDA in the regulation of other ER-associated developmental processes was additive, rather than epistatic. Genetic analyses indicated that MPK3, a MAP kinase that functions downstream of YDA in stomatal development, also regulates plant resistance to P. cucumerina in a YDA-dependent manner, suggesting that the YDA-associated MAPK transduction module is shared in plant development and PTI. Our experiments revealed that YDA-mediated resistance was independent of signalling pathways regulated by defensive hormones like salicylic acid, jasmonic acid, abscisic acid or ethylene, and of the tryptophan-derived metabolites pathway, which are involved in plant immunity. In addition, we showed that PAMP-mediated PTI responses, such as the increase of cytoplasmic Ca2+ concentration, reactive oxygen species (ROS) burst, MAPK phosphorylation, and expression of defense-related genes are not impaired in the yda11 mutant. Furthermore, the expression of CA-YDA protein does not result in enhanced PTI responses, further suggesting the existence of additional mechanisms of resistance regulated by YDA that differ from those regulated by the PTI receptors FLS2 and CERK1. In line with these observations, our transcriptomic data revealed the over-representation in CA-YDA plants of defensive genes, such as those encoding antimicrobial peptides and cell death regulators, and genes encoding cell wall-related proteins, suggesting a potential link between plant cell wall composition and integrity and broad spectrum resistance mediated by YDA. In addition, phosphoproteomic data revealed an over-representation of genes encoding wall-related proteins in CA-YDA plants in comparison with wild-type plants. The putative role of the ER-YDA pathway in regulating cell wall integrity was further supported by biochemical and glycomics analyses of er, yda11 and CA-YDA cell walls, which revealed significant changes in the cell wall composition of these genotypes compared with that of wild-type plants. In summary, our data indicate that ER and YDA are components of a novel immune pathway that regulates cell wall integrity and defensive responses, which confer broad-spectrum resistance to pathogens.
