El Cementerio Este de Malmo (1916-1973) ha sido analizado en el ámbito internacional sólo en contadas ocasiones, aunque en este lugar el arquitecto sueco Sigurd Lewerentz (1885-1975) realizó un proyecto experimental a gran escala, rechazando los trazados de cementerios pintorescos que se habían popularizado en Europa en aquella época y proponiendo una disposición más ortogonal, vinculada con el lugar de distintas maneras. Después de que Lewerentz ganase el concurso, el proyecto se desarrolló durante casi sesenta años. El Cementerio Este de Malmo ofrece una lente única a través de la cual se puede examinar la forma en la que se establecieron relaciones entre el proyecto de arquitectura y diversos aspectos del paisaje: con el entorno urbano y diversas circunstancias del entorno social mediante la vegetación como límite, con la topografía a través de la escala y los materiales de construcción, con la tradición organizando plantas, secciones y construcción, con el ámbito internacional a través de los paramentos constructivos, con las técnicas constructivas habituales en Suecia por medio de materiales cotidianos y con los alrededores a través del planteamiento de los detalles. La hipótesis de trabajo plantea que el concepto de límite en la obra de Lewerentz y en el Cementerio Este de Malmo en particular tiene un carácter dual, produciendo simultáneamente separaciones y relaciones con distintos aspectos del lugar y del tiempo. Esta investigación documenta y examina la construcción del Cementerio Este de Malmo apoyándose en planos y escritos originales de distintos archivos, datos recogidos en el proyecto así como en otros cementerios de la época y en análisis previos ya publicados. Proporcionando un estudio detallado de la evolución del enfoque de Lewerentz respecto a la relación entre arquitectura y paisaje, se define un marco en el que la obra de este arquitecto puede entenderse de una manera más completa. Lewerentz mantuvo una postura crítica con algunos modelos de su época, al tiempo que planteó vínculos con aspectos de la vida cotidiana de las personas. Este conjunto de referencias redefine, un conjunto de circunstancias vinculadas con el proyecto, que redefinen el lugar de forma abstracta. En esta doble cualidad del límite se da lugar a un espacio de intersección, entendido como un lugar de convergencia de personas. En estas relaciones, el usuario siempre es el centro, ya sea para crear un pórtico para resguardar, una avenida de árboles para crear un espacio más tranquilo o una ventana simplificada. Esta tesis contribuye a la investigación en el ámbito de la integración del proyecto de arquitectura en el lugar, describiendo una metodología de proyectar, en la que los elementos que sirven para separar espacios, también se emplean para vincularlos a un conjunto seleccionado de referencias a distintas escalas, lugares y tiempos. Este doble proceso que implica la construcción de los límites permite favorecer la proximidad a algunos aspectos del entorno y al mismo tiempo rechazar otros, permitiendo una perspectiva crítica y pudiendo proponer situaciones alternativas. ABSTRACT Malmó Eastern Cemetery (1916-1973) has received little international attention, despite the fact that Swedish architect Sigurd Lewerentz (1885-1975) led a large-scale experimental project, rejecting picturesque cemetery plans, popular in Europe at that time, and developing a more orthogonal layout, which was linked to the site in different ways. After Lewerentz won first prize in the competition to develop Malmó Eastern Cemetery, the project unfolded over a period of nearly sixty years. Malmó Eastern Cemetery project offers a unique lens through which to examine how different relationships were established between the architectural project and various features of the landscape: with the urban environment and various social circumstances using greenery as a boundary; with topography using the scale of construction and materials; with tradition by designing plans, sections and construction; with the international architectural arena by building walls; with common construction methods in Sweden using ordinary materials and with the surroundings by using detail. The hypothesis of this thesis explores the dual nature of the notion of limit in Lewerentz's work, and in Malmó Eastern Cemetery in particular, through the simultaneous creation of separations and connections with various aspects of place and time. This research documents and examines the construction of Malmó Eastern Cemetery by examining original drawings and writings from various archives, field exploration into the project and other cemeteries of the time, and the existing literature. By providing a detailed study of Lewerentz's evolving approach to the relationship between architecture and landscape, this thesis defines a framework in which Lewerentz's work may be understood in a more comprehensive way. Lewerentz was critical of some of the models of his time, although he established different connections with aspects of people's everyday lives. This set of connections defines a particular setting or set of circumstances for the project, which are more of an abstract notion. Within this double role of limits, a space of intersection is generated, which can be understood as a place of convergence for people. In this set of connections, the user is always at the centre of the design. For example, Lewerentz designed a portico that would provide shelter, an avenue of trees to produce a tranquil space of commemoration or a simplified window. This thesis contributes to research in the field of integration of an architectural project in a site, by describing a methodology of design, in which the elements used to subdivide spaces are also used to connect a set of selected references for different scales, locations and times. The dual process involved in building limits encourages proximity to some aspects of the environment, while rejecting others, thereby allowing for a critical perspective and the proposal of alternative situations.

La Tesis Doctoral que se presenta trata de profundizar en el conocimiento del patrimonio arbóreo, en la cuestión de la evaluación y la singularidad a nivel de ejemplares y agrupaciones. La metodología incorpora nuevas herramientas, modelos y criterios utilizados en la valoración del paisaje y de los recursos naturales. Siendo el árbol un generador de espacio y habitats, solo o en masa, es vínculo entre la naturaleza y el ser humano, las comunidades y sus costumbres. Desde estos parámetros se indagan los procesos que permiten estimar el significado, la importancia y el valor del árbol para llevarlo a una consideración de Singular y/o Monumental. El estudio se basa en los sistemas de catalogación, tras el reconocimiento, localization y selección de ejemplares. Así mismo, se explora la relación sistémica entre árbol y entorno para poner de relieve la importancia del árbol en la configuración de determinados paisajes culturales y ecológicos -como son los robledales de antiguos trasmochos en Euskadi-. Sobre un primer inventario se realiza un estudio pormenorizado de cada árbol registrado y, en un procedimiento paramétrico, se definen criterios -ecológicos y paisajísticos, etnográficos y culturales- de selección de elementos y de evaluación. La obtención de distintos índices de singularidad para los árboles, utilizando modelos tanto cualitativos como cuantitativos, sirve como vía hacia una categorization de los árboles muestreados. A partir de la figura de "Árbol Singular", recogida en la Ley 16/ 1994, de Conservación de la Naturaleza del País Vasco se realiza una revisión del marco legislativo y el régimen de protección, haciendo un análisis a nivel local, autonómico y estatal. Dicho examen pone de manifiesto la diversidad de contextos y significados bajo los que se presentan los árboles. Se muestra también una (in)definición: cierta ambigüedad en torno a la definición que induce a diferentes interpretaciones y nomenclaturas en un intento de delimitar la categoría para regularlo jurídicamente. Estas figuras concebidas desde las políticas de protección ambiental, no siempre resultan del todo efectivas. El Catálogo de Árboles Singulares del País Vasco, creado por Decreto como instrumento para poner en valor estos recursos naturales, no ha sido actualizado desde hace casi veinte años. Sin embargo, se han llevado a cabo iniciativas de ampliación como el trabajo impulsado por el Departamento de Medio Ambiente y Biodiversidad de la Diputación Foral de Álava para el inventario de los árboles singulares del Territorio Histórico de Álava y la propuesta de catálogo a partir del cual se desarrolla esta Tesis Doctoral. Desde estas reflexiones y el desarrollo de modelos para la evaluación y catalogación de los ejemplares registrados, la investigación trata de descifrar cómo observamos a los árboles con los que nos vinculamos, cómo son identificados, a través de qué otros parámetros intangibles les damos valor, y por qué necesitamos clasificarlos. El trabajo concluye con propuestas y acciones alternativas para la conservación y mejora de los árboles que se proponen como singulares, entre ellas, la divulgación y la sensibilización para garantizar el compromiso y la ampliación en el futuro de un catálogo abierto para los árboles de interés. ABSTRACT The PhD thesis here presented tries to deepen the knowledge of tree heritage, the issue of evaluation and singularity where it comes to either specimen or groups. The methodology includes new tools, models and criteria to be used in the assessment of landscape and natural resources. With the tree being a creator of space and habitats, alone or in groups, it is a link between nature and humans, societies and their habits. Using these parameters processes are being sought after: processes that allow us to assess the meaning, the importance and the value of trees in order to lead us to considering a tree as being a 'Singular tree' and/or 'Heritage tree'. The research is based on cataloging systems, after recognizing, localizing and selecting of specimen. This way, the systemic relation between the tree and its surrounding is being explored to get a view on the importance of trees in certain cultural and ecological landscapes -such as the oak fields consisting of ancient pollards in the Basque Country-. After a first inventory a detailed study is performed of each registered tree and, using a parametric method, criteria for selection of elements and evaluation are defined -ecological and those concerning the landscape, as well as ethnographical and cultural-. The creation of different indexes of singularity for trees, using qualitative as well as quantitative models, serves as a way to categorize the selected trees. A revision is done on legislation and the protection regimen, analyzing on a local, autonomic state and national level, parting from the concept of the Singular Tree, as included in the Law 16/1994, of Nature Conservation of Basque Coutry. This review proves the diversity of contexts and meanings in which the trees are being presented. Also an (un)definition appears; certain ambiguity of the definition which induces different interpretations and nomenclatures in an attempt to limit categorization in order to legally regulate. These concepts created out of environment protection politics do not always turn out to be completely effective. The Catalogue of Singular Trees in the Basque Country, created by decree as an instrument to value these natural resources, has not been updated since almost twenty years. However, there have been initiatives of amplification such as the work promoted by the Environment and Biodiversity Service of Provincial Council of Alava to create the inventory of singular trees of the Historical Territory of Álava and the catalogue proposal which forms the starting point of this PhD thesis. Parting from these considerations and the development of models for evaluation and cataloging of the registered specimen, the investigation attempts to unravel the way we observe the trees with which we link, how they are identified, by which intangible parameters we assess them and why we need to classify them. This study ends with proposals and alternative actions for the conservation and improvement of the trees that are being proposed as being singular. Among them are the publication and creating of awareness to guarantee the commitment and the development of an open catalogue for significant trees.

Old-growth trees play a very important role in the maintenance of biodiversity in forests. However, no clear definition is yet available to help identify them since tree age is usually not recorded in National Forest Inventories. To develop and test a new method to identify old-growth trees using a species-specific threshold for tree diameter in National Forest Inventories. Different nonlinear mixed models for diameter ? age were generated using data from the Spanish Forest Inventory in order to identify the most appropriate one for Aleppo pine in its South-western distribution area. The asymptote of the optimal model indicates the threshold diameter for defining an old-growth tree. Additionally, five site index curves were examined to analyze the influence of site quality on these models.

Persistence and abundance of species is determined by habitat availability and the ability to disperse and colonize habitats at contrasting spatial scales. Favourable habitat fragments are also heterogeneous in quality, providing differing opportunities for establishment and affecting the population dynamics of a species. Based on these principles, we suggest that the presence and abundance of epiphytes may reflect their dispersal ability, which is primarily determined by the spatial structure of host trees, but also by host quality. To our knowledge there has been no explicit test of the importance of host tree spatial pattern for epiphytes in Mediterranean forests. We hypothesized that performance and host occupancy in a favourable habitat depend on the spatial pattern of host trees, because this pattern affects the dispersal ability of each epiphyte and it also determines the availability of suitable sites for establishment. We tested this hypothesis using new point pattern analysis tools and generalized linear mixed models to investigate the spatial distribution and performance of the epiphytic lichen Lobaria pulmonaria, which inhabits two types of host trees (beeches and Iberian oaks). We tested the effects on L. pulmonaria distribution of tree size, spatial configuration, and host tree identity. We built a model including tree size, stand structure, and several neighbourhood predictors to understand the effect of host tree on L. pulmonaria. We also investigated the relative importance of spatial patterning on the presence and abundance of the species, independently of the host tree configuration. L. pulmonaria distribution was highly dependent on habitat quality for successful establishment, i.e., tree species identity, tree diameter, and several forest stand structure surrogates. For beech trees, tree diameter was the main factor influencing presence and cover of the lichen, although larger lichen-colonized trees were located close to focal trees, i.e., young trees. However, oak diameter was not an important factor, suggesting that bark roughness at all diameters favoured lichen establishment. Our results indicate that L. pulmonaria dispersal is not spatially restricted, but it is dependent on habitat quality. Furthermore, new spatial analysis tools suggested that L. pulmonaria cover exhibits a distinct pattern, although the spatial pattern of tree position and size was random.

We investigated how richness and composition of vascular plant species in the understory of a mixed hardwood forest stand varied with respect to the abundance and composition of the overstory. The stand is in central Spain and represents the southernmost range of distribution of several tree and herbaceous species in Europe. Understory species were identified in 46 quadrats (0.25 m2) where variables litter depth and light availability were measured. In addition, we estimated tree density, basal area, and percent basal area by tree species within 6-m-radius areas around each plot. Species richness and composition were studied using path analysis and scale-dependent geostatistical methods, respectively. We found that the relative abundance of certain trees species in the overstory was more important than total overstory abundance in explaining understory species richness. Richness decreased as soil litter depth increased, and soil litter increased as the relative proportion of Fagus sylvatica in the overstory increased, which accounted for a negative, indirect effect of Fagus sylvatica on richness. Regarding understory species composition, we found that some species distributed preferentially below certain tree species. For example, Melica uniflora was most frequent below Fagus sylvatica and Quercus petraea while the increasing proportion of Q. pyrenaica in the overstory favored the presence of Cruciata glabra, Arenaria montana, Prunus avium, Conopodium bourgaei, Holcus mollis, Stellaria media and Galium aparine in the understory. Overall, these results emphasize the importance of individual tree species in controlling the assemblage and richness of understory species in mixed stands. We conclude that soil litter accumulation is one way through which overstory composition shapes the understory community.

Abstract The cloud forest is a special type of forest ecosystem that depends on suitable conditions of humidity and temperature to exist; hence, it is a very fragile ecosystem. The cloud forest is also one of the richest ecosystems in terms of species diversity and rate of endemism. However, today, it is one of the most threatened ecosystems in the world. Little is known about tree species distribution and coexistence among cloud forest trees. Trees are essential to understanding ecosystem functioning and maintenance because they support the ecosystem in important ways. For this dissertation, an analysis of woody plant species distribution at a small scale in a north-Peruvian Andean cloud forest was performed, and some of the factors implicated in the observed patterns were identified. Towards that end, different natural factors acting on species distribution within the forest were investigated: (i) intra-specific arrangements, (ii) heterospecific spatial relationships and (iii) relationships with external environmental factors. These analyses were conducted first on standing woody plants and then on seedlings. The woody plants were found to be clumped in the forest, either considering all the species together or each species separately. However, each species presented a specific pattern and specific spatial relationship among different-age individuals. Dispersal mode, growth form and shade tolerance played roles in the final distribution of the species. Furthermore, spatial associations among species, either positive or negative, were observed. These associations were more numerous when considering individuals of the interacting species at different developmental stages, i.e., younger individuals from one species and older individuals from another. Accordingly, competition and facilitation are asymmetric processes and vary throughout the life of an individual. Moreover, some species appear to prefer certain habitat conditions and avoid other habitats. The habitat definition that best explains species distribution is that which includes both environmental and stand characteristics; thus, a combination of these factors is necessary to understanding species' niche preferences. Seedling distribution was also associated with habitat conditions, but these conditions explained less than the 30% of the spatial variation. The position of conspecific adult individuals also affected seedling distribution; although the seedlings of many tree species avoid the vicinity of conspecifics, a few species appeared to prefer the formation of cohorts around their parent trees. The importance of habitat conditions and distance dependence with conspecifics varied among regions within the forest as well as on the developmental stage of the stand. The results from this thesis suggest that different species can coexist within a given space, forming a “puzzle” of species as a result of the intra- and interspecific spatial relationships along with niche preferences and adaptations that operate at different scales. These factors not only affect each species in a different way, but specific preferences also vary throughout species' lifespans. Resumen Resumen El bosque de niebla es uno de los ecosistemas más amenazados del mundo además de ser uno de los más frágiles. Son formaciones azonales que dependen de la existencia de unas condiciones de humedad y temperatura que permitan la formación de nubes que cubran el bosque; lo que dificulta en gran medida su conservación. También es uno de los ecosistemas con mayor riqueza de especies además de tener uno de los mayores porcentajes de endemismos. Uno de los aspectos más importantes para entender el ecosistema, es identificar y entender los elementos que lo componen y los mecanismos que regulan las relaciones entre ellos. Los árboles son el soporte del ecosistema. Sin embargo, apenas hay información sobre la distribución y coexistencia de los árboles en los bosques de niebla. Esta tesis presenta un análisis de la distribución a pequeña escala de las plantas leñosas en un bosque de niebla situado en la cordillera andina del norte de Perú; así como el análisis de algunos de los factores que pueden estar implicados en que se origine la distribución observada. Para este propósito se estudia cómo influyen factores de diferente naturaleza en la distribución de las especies (i) organización intra-específica (ii) relaciones espaciales heterospecíficas y (iii) relación con factores ambientales externos. En estos análisis se estudiaron primero las plantas jóvenes y las adultas, y después las plántulas. Los árboles aparecieron agregados en el bosque, tanto considerando todos a la vez como cuando se estudió cada especie por separado. Sin embargo, cada especie mostró un patrón distinto así como una particular relación espacial entre individuos jóvenes y adultos. El modo de dispersión, la forma de vida y la tolerancia de la especies estuvieron relacionados con el patrón general observado. Se vio también que ciertas especies aparecían relacionadas con otras, tanto de forma positiva (compartiendo zonas) como negativa (apareciendo en áreas distintas). Las asociaciones fueron mucho más numerosas cuando se consideraron los pares de especies en diferente estado de desarrollo, es decir, individuos jóvenes de una especie e individuos mayores de la otra. Eso indicaría que los procesos de competencia y facilitación son asimétricos y además varían durante la vida de la planta. Por otro lado, algunas especies aparecen preferentemente bajo ciertas condiciones de hábitat y evitan otras. La definición de hábitat a la que mejor responden las especies es cuando se incluyen tanto variables ambientales como de masa; así que ambos tipos de variables son necesarias para entender la preferencia de las especies por ciertos nichos. La distribución de las plántulas también estuvo relacionada con condiciones de hábitat, pero eso sólo llegaba a explicar hasta un 30% de la variabilidad espacial. La posición de los adultos de la misma especie también afectó a la distribución de las plántulas. En bastantes especies las plántulas evitan la cercanía de adultos de su misma especie, padres potenciales, aunque algunas especies aisladas mostraron el patrón contrario y aparecieron preferentemente en las mismas áreas que sus padres. La importancia de las condiciones de hábitat y posición de los adultos en la disposición de las plántulas varía de una zona a otra del bosque y además también varía según el estado de desarrollo de la masa.

The development of reliable clonal propagation technologies is a requisite for performing Multi-Varietal Forestry (MVF). Somatic embryogenesis is considered the tissue culture based method more suitable for operational breeding of forest trees. Vegetative propagation is very difficult when tissues are taken from mature donors, making clonal propagation of selected trees almost impossible. We have been able to induce somatic embryogenesis in leaves taken from mature oak trees, including cork oak (Quercus suber). This important species of the Mediterranean ecosystem produces cork regularly, conferring to this species a significant economic value. In a previous paper we reported the establishment of a field trial to compare the growth of plants of somatic origin vs zygotic origin, and somatic plants from mature trees vs somatic plants from juvenile seedlings. For that purpose somatic seedlings were regenerated from five selected cork oak trees and from young plants of their half-sib progenies by somatic embryogenesis. They were planted in the field together with acorn-derived plants of the same families. After the first growth period, seedlings of zygotic origin doubled the height of somatic seedlings, showing somatic plants of adult and juvenile origin similar growth. Here we provide data on height and diameter increases after two additional growth periods. In the second one, growth parameters of zygotic seedlings were also significantly higher than those of somatic ones, but there were not significant differences in height increase between seedlings and somatic plants of mature origin. In the third growth period, height and diameter increases of somatic seedlings cloned from the selected trees did not differ from those of zygotic seedlings, which were still higher than data from plants obtained from somatic embryos from the sexual progeny. Therefore, somatic seedlings from mature origin seem not to be influenced by a possible ageing effect, and plants from somatic embryos tend to minimize the initial advantage of plants from acorns

A series of numerical simulations of the flow over a forest stand have been conducted using two different turbulence closure models along with various levels of canopy morphology data. Simulations have been validated against Stereoscopic Particle Image Velocimetry measurements from a wind tunnel study using one hundred architectural model trees, the porosities of which have been assessed using a photographic technique. It has been found that an accurate assessment of the porosity of the canopy, and specifically the variability with height, improves simulation quality regardless of the turbulence closure model used or the level of canopy geometry included. The observed flow field and recovery of the wake is in line with characteristic canopy flows published in the literature and it was found that the shear stress transport turbulence model was best able to capture this detail numerically.

La encina (Quercus ilex L.) es una de las especies forestales mediterráneas más importantes. Constituye gran parte del estrato arbóreo de dehesas o montados, produce bellota como alimento del ganado y establece simbiosis con hongos micorrizógenos de gran valor económico. La encina está considerada como una especie recalcitrante en términos de conservación de semillas y capacidad morfogénica, lo que dificulta los programas de conservación de recursos genéticos y la mejora de la especie. La propagación vegetativa es una potente herramienta de los programas de mejora, por lo que es preciso desarrollar protocolos de regeneración somática en encina. La embriogénesis somática está considerada como la modalidad más adecuada de regeneración basada en técnicas de cultivo de tejidos vegetales utilizada en biotecnología forestal. Este trabajo se centra en el estudio de determinados aspectos de la embriogénesis somática para la regeneración clonal de encinas adultas. La memoria de esta tesis se ha dividido en capítulos que se corresponden con diferentes aspectos del sistema embriogénico. La embriogénesis somática se indujo en tegumentos maternos de óvulos en desarrollo procedentes de bellotas inmaduras de encinas adultas. A pesar de las bajas frecuencias de inducción, las líneas embriogénicas generadas se amplificaron mediante embriogénesis secundaria observándose cierta pérdida de la capacidad de diferenciación con el tiempo. Tanto el genotipo como la formulación del medio de cultivo influyeron en la respuesta embriogénica, concluyendo que la formulación de macronutrientes de Schenk y Hildebrant del medio sin reguladores de crecimiento fue la combinación más efectiva en la inducción. Los resultados sugirieron la existencia de una ventana en el desarrollo del óvulo más sensible a la inducción. El genotipo in[luyó en la capacidad proliferativa de los cultivos y en la conversión de los embriones somáticos, que se incrementó suplementando el medio con ácido indol-3-butírico y 6-benciladenina. El cultivo en medio líquido de líneas embriogénicas en condiciones de inmersión transitoria incrementó el crecimiento, dependiendo del genotipo, con respecto al cultivo en medio semisólido. Sin embargo, no mejoró la capacidad de diferenciar embriones cotiledonares aislados. Se estableció un protocolo de inicio y mantenimiento de cultivos en suspensión para varias líneas embriogénicas mediante inoculación en alta densidad de agregados embrionarios procedentes del medio semisólido. Para evitar la pérdida de vigor y la capacidad morfogénica debida al cultivo prolongado se desarrolló un protocolo de crioconservación de líneas embriogénicas mediante vitrificación. Al determinar la influencia de los agentes crioprotectores antes y después de su inmersión en nitrógeno líquido se concluyó que las respuestas de capacidad de crecimiento y de diferenciación del material embriogénico son independientes, además de estar bajo influencia del genotipo y el tipo de material crioconservado. La combinación de sacarosa y PVS2 previa a la inmersión en nitrógeno líquido proporcionó la mayor tasa de recuperación. Cuando las líneas fueron crioconservadas 30 días la capacidad de diferenciación se perdió en todas ellas. El análisis de SSR detectó variación somaclonal en el material crioconservado a corto plazo. SSR y RAPD mostraron importantes diferencias genéticas entre los árboles donantes y el material embriogénico que dependieron del genotipo. El grado de detección dependió del marcador empleado. Ambos marcadores revelaron baja inestabilidad intraclonal. Los RAPD revelaron variación genética intra-individuo en las encinas donantes. Se discuten la variación genética pre-existente en encina, su aparición durante las primeras fases de la inducción de embriogénesis, y la presencia de tejidos provenientes de la fertilización en el explanto materno. Esto hace preciso definir la identidad genética del material donante y acometer ensayos de detección precoz de variación somaclonal. ABSTRACT Holm oak (Quercus ilex L.) is one of the most important Mediterranean forest species. It conforms the tree layer of dehesas or montados, it produces acorns to feed the livestock and it establishes symbiosis with profitable mycorrhizal fungi. Holm oak is considered as recalcitrant species in terms of seed conservation and morphogenic capacities, which complicates the development of genetic conservation and improvement programs. Vegetative propagation is one of the mightiest tools for breeding programs therefore; developing protocols for clonal regeneration of holm oak is essential. Somatic embryogenesis is considered the best tissue culture-based way of plant regeneration in forest biotechnology. The present study is focused on the study of certain aspects of somatic embryogenesis for clonal regeneration of mature holm oak. This thesis manuscript is divided into several chapters that match with different aspects of the embryogenic system. Somatic embryogenesis induction was achieved on maternal teguments of developing ovules from immature acorns of adult holm oak trees. Despite the low induction frequencies, the generated embryogenic lines were amplified by secondary embryogenesis. A decline in the differentiation capacity over time was also observed. It was concluded that both genotype and culture media formulation influenced the embryogenic response, being the Schenk and Hildebrandt´s macronutrients formulation from culture medium and the lack of plant growth regulators the most effective combination for the induction of the embryogenic response. It has been suggested the existence of a developmental window in which ovules are prone to induction. Genotype influenced the proliferation capacity and the plant conversion of somatic embryos, which was also favoured by the presence of indol-3-butyric acid and 6-bencyladenine. The use of temporary immersion systems as proliferation in liquid culture of the embryogenic lines increased the growth depending on genotype, when compared to semisolid cultures. However, it did not improve the differentiation of single cotyledonary embryos. A protocol for the initiation and maintenance of embryogenic suspension cultures was established for several embryogenic lines with highly dense inoculi of embryogenic clusters from proliferating semisolid cultures. In order to avoid the loss of vigour and morphogenic ability of embryogenic lines due to prolonged cultures, a cryopreservation protocol for embryogenic lines of holm oak has been developed. During the determination of the influence of cryoprotective agents on the growth and differentiation capacities before and after liquid nitrogen immersion, it was concluded that both responses were independent from each other and also under the influence of genotype and the type of cryopreserved material. The combination of sucrose and PVS2 prior liquid nitrogen immersion provided higher recovery rates. When the same embryogenic lines were cryopreserved for 30 days, none was able to differentiate. The SSRs analysis of the short-term cryopreserved material detected somaclonal variation. Both SSR and RAPD markers showed high sensitivity to detect genetic differences between the donor trees and the generated embryogenic material. Nevertheless, the degree of instability detection depended on the marker. The SSR analysis indicated a relationship between genotype, the studied loci and the located polymorphisms. Also, both markers revealed low intraclonal genetic variation. The RAPD detected genetic variation within the donor trees. The presence of pre-existent genetic variation within mature trees, in addition to its occurrence during the early stages of the embryogenic induction, and the presence of tissues of fertilisation origin within the maternal explants are all discussed. Nonetheless, the determination of the genetic identity of donor material is required, in addition to early detection methods of somaclonal variation.

La encina (Quercus ilex L.) es una de las especies forestales mediterráneas más importantes. Constituye gran parte del estrato arbóreo de dehesas o montados, produce bellota como alimento del ganado y establece simbiosis con hongos micorrizógenos de gran valor económico. La encina está considerada como una especie recalcitrante en términos de conservación de semillas y capacidad morfogénica, lo que dificulta los programas de conservación de recursos genéticos y la mejora de la especie. La propagación vegetativa es una potente herramienta de los programas de mejora, por lo que es preciso desarrollar protocolos de regeneración somática en encina. La embriogénesis somática está considerada como la modalidad más adecuada de regeneración basada en técnicas de cultivo de tejidos vegetales utilizada en biotecnología forestal. Este trabajo se centra en el estudio de determinados aspectos de la embriogénesis somática para la regeneración clonal de encinas adultas. La memoria de esta tesis se ha dividido en capítulos que se corresponden con diferentes aspectos del sistema embriogénico. La embriogénesis somática se indujo en tegumentos maternos de óvulos en desarrollo procedentes de bellotas inmaduras de encinas adultas. A pesar de las bajas frecuencias de inducción, las líneas embriogénicas generadas se amplificaron mediante embriogénesis secundaria observándose cierta pérdida de la capacidad de diferenciación con el tiempo. Tanto el genotipo como la formulación del medio de cultivo influyeron en la respuesta embriogénica, concluyendo que la formulación de macronutrientes de Schenk y Hildebrant del medio sin reguladores de crecimiento fue la combinación más efectiva en la inducción. Los resultados sugirieron la existencia de una ventana en el desarrollo del óvulo más sensible a la inducción. El genotipo in[luyó en la capacidad proliferativa de los cultivos y en la conversión de los embriones somáticos, que se incrementó suplementando el medio con ácido indol-3-butírico y 6-benciladenina. El cultivo en medio líquido de líneas embriogénicas en condiciones de inmersión transitoria incrementó el crecimiento, dependiendo del genotipo, con respecto al cultivo en medio semisólido. Sin embargo, no mejoró la capacidad de diferenciar embriones cotiledonares aislados.
