Estudo da atividade ansiolítica e sedativa do óleo essencial das cascas de frutos de espécies do gênero Citrus

A ansiedade é um dos principais distúrbios do sistema nervoso central (SNC) de alta prevalência, cujas ferramentas terapêuticas disponíveis apresentam efeitos colaterais importantes ou não são ativas em parte dos pacientes. Deste modo, a avaliação de novos compostos com potencial atividade ansiolítica pode trazer novas alternativas de tratamento, mais eficazes e seguras. Preparações com base em informações quimiotaxonômicas podem ser uma promissora fonte de novas moléculas com atividade sobre o SNC. Assim, o objetivo deste trabalho foi avaliar, no modelo do teste de esconder esferas e caixa claro e escuro, um possível efeito ansiolítico das espécies de Citrus latifólia (CL) e Citrus reticulata (CR), atividade previamente detectada no Citrus aurantium (CA). O estudo do perfil da atividade farmacológica foi complementado com a avaliação dos possíveis efeitos sedativos, por meio do teste de sono induzido por éter etílico e a integridade do sistema motor foi avaliada com a utilização do teste da barra giratória. Os diferentes modelos experimentais estão relacionados a diferentes aspectos dos transtornos ansiosos. Assim, enquanto o procedimento do teste de esconder esferas parece refletir o transtorno obsessivo-compulsivo, a caixa claro escuro reflete o transtorno da ansiedade generalizada. Os tratamentos dos óleos essenciais (OE) foram todos realizados oralmente por gavagem e os resultados comparados com seus respectivos grupos-controle, tratados com o volume equivalente do veículo utilizado para a solubilização do OE. No procedimento do teste de esconder esferas, o OE das cascas dos frutos de CL apresentou atividade ansiolítica nas doses de 1,0 e 1,5g/kg e CR somente na maior dose de 1,5g/kg. No procedimento da caixa claro escuro, CL demonstrou atividade ansiolítica na dose de 0,5g/kg, porém CR não foi capaz de alterar os parâmetros avaliados...

The anxiety is one of the main disturb of the central nervous system (CNS) of high prevalence, whose available therapeutical tools present important collateral effect or are not active in part of the patients. In this way, the evaluation of new compounds with potential anxiolytic activity can bring new alternatives of treatment, more efficient and safe. Preparations with basis of quimiotaxonomy information can be a promising source of new molecule with activity on the CNS. Thus, the objective of this work was to evaluate, in the model of the marble burying test and light/dark box test, a possible anxiolytic effect of the species of Citrus latifolia (CL) and Citrus reticulata (CR), activity previously detected in the Citrus aurantium (CA). The study of the profile of the pharmacology activity was complemented with the evaluation of the possible sedative effect, by means of the sleep induced by diethyl ether test and the integrity of the motor system was evaluated with the use of the rota-rod test. The different experimental models are related with the different aspects of the anxious disturbs. Thus, while the procedure of the marble burying test seems to reflect the obsessive-compulsive disorder, the light/dark box test reflect the generalized anxiety disorder. The treatments of essential oils (EO) had been all realized orally by gavage and the results compared with its respective group-control, treated with the volume equivalent to the vehicle used for the dissolved of the EO. In the procedure of the marble burying test, the EO of the rinds of the fruits of CL only presented anxiolytic activity in the doses of 1,0 and 1,5 g/kg and CR in the high dose of 1,5g/kg. In the procedure of the light/dark box test, CL demonstrated anxiolytic activity in the dose of 0,5g/kg, however CR was not capable to modify the evaluated parameters.
