Atualmente, uma organização industrial com vista a singrar no mercado global é fortemente influenciada por pressões que visam o aumento da eficiência global e consequente redução de custos operacionais. O desafio para as mesmas passa, portanto, por expurgar do produto tudo aquilo que não lhe acrescenta valor percetível pelo cliente e por maximizar a utilização dos vários recursos industriais instalados. No seguimento deste desafio, surge o Problema de Planeamento e Programação da Produção, ao qual é necessário dar uma resposta eficiente. Este projeto tem como objetivo estudar o problema da Programação da Produção numa indústria de pavimentos e revestimentos cerâmicos, desenvolvendo uma heurística construtiva capaz de traduzir com fiabilidade a realidade do processo produtivo da mesma e, se possível, auxiliar na sua resolução. O problema da programação da produção em estudo visa responder às questões: o quê, em que quantidade, quando e em que linha produzir, por forma a satisfazer as necessidades dos clientes num prazo previamente estipulado como admissível, garantindo o enchimento dos fornos ligados. Sem grandes constrangimentos ao normal lavor da Produção, pretende obter-se com a heurística planos de produção viáveis, que minimizem o tempo necessário para a conclusão do conjunto de referências com necessidades produtivas. O problema é também abordado através de um modelo exato como um problema de máquinas paralelas idênticas capacitado, com matriz de compatibilidades, setups de família e de subfamília e com lotes mínimos de produção. Quer a heurística quer o modelo de programação inteira mista desenvolvidos permitem obter planos de produção válidos, equivalentes aos obtidos atualmente pela empresa através dos meios de programação atuais, embora com um dispêndio de tempo muito inferior.

O fator de transcrição OCT4 é um importante marcador de células tronco e tem sido relacionado com o conceito de células tronco tumorais (CTTs). Recentemente, ele tem sido também relacionado ao fenótipo de resistência a múltiplas drogas (MDR). O objetivo deste trabalho foi testar se o OCT4 está ligado ao fenótipo MDR em células eritroleucêmicas derivadas da linhagem LMC-K562. Para isso, foi realizada a análise de expressão de genes associados à superfamília de transportadores ABC (ATP Binding Cassette) e, também, de fatores de transcrição relacionados a células tronco. Os primeiros resultados apontaram uma relação direta entre OCT4 e transportadores ABC na linhagem MDR derivada de K562 (Lucena). O sequenciamento de promotores ABC não revelou qualquer mutação que pudesse explicar a expressão diferenciada do OCT4 na linhagem MDR. Posteriormente, o sequenciamento da região contendo o domínio homeobox do gene OCT4 de ambas as linhagens celulares evidenciou, pela primeira vez, que este fator de transcrição é alvo de mutações que podem estar relacionados com o fenótipo MDR. As mutações encontradas implicam substituições de vários aminoácidos em ambas as linhagens celulares. K562 teve sete substituições (três exclusivas), enquanto Lucena teve 13 (nove exclusivas). Além disso, um busca in silico por motivos de fosforilação dentro da sequência de aminoácidos comparada, revelou que o OCT4 humano normal possui sete motivos potenciais de fosforilação. Entretanto, K562 perdeu um motivo e Lucena dois deles. Moléculas com diferentes padrões de fosforilação podem ter sua função modificada. Estes resultados indicam o OCT4 com uma alvo potencial para o tratamento do câncer, especialmente aqueles resistentes à quimioterapia.

The Landless Rural Workers Movement (MST) from its educational process and the path taken in education, started in the year 2013, with an experimental curricular proposal by Complexes of study the School Base and the Itinerant Schools, the MST, in Paraná state. For the construction of this proposed curriculum, the movement takes the historical background and the experience of the original foundations of the single school of labor started by the working class in revolutionary Soviet period as a training proposal and school organization that comes close to the creation of collective subjects, fighters and builders of a new society and sociopolitical objectives to this social movement. The Soviet experiment reference required a work of a critical appropriation for the Brazilian context. The curriculum prescription, called Study Plan, in its introduction, includes elements of design fundamentals like: the Education Eesign and Training Matrix; Matrix detailing: School and Life, School and Labor Formative Matrix, School and Social Struggle Formative Matrix, School and Collective Organization Formative Matrix, School and Culture Formative Matrix and School and History Formative Matrix; general school guidelines: a pedagogical function of the environment, the school's political organization, school times, specific methodological aspects, sequencing and duration of Complexes of Study and the evaluation process. The Study Plan contemplates the complexes, the disciplines, the portions of reality / practice categories present in complexes, organized by semester and year, i. e., from 6th to 9th grade in elementary school. Given the the presented context, this research aims to analyze the process of testing the proposed curriculum for the Complexes of Study in Primary School of Iraci Salette Strozak State School, located in the Marcos Freire Settlement, in Rio Bonito do Iguaçu in Paraná state. As a methodological approach, we chose the qualitative approach and analysis were conducted under the Marxism perspective. Library research and field research, semi-structured interviews and analysis of documents generated in the process of construction of the proposal were made.Initially, in this paper, we discus about the path followed by the MST in the fight for schools and public education; highlighting elements of the process which led to the understanding, by the Movement, of which only the struggle for land is not enough for the realization of Agrarian Reform. Then discuss whether some elements of Pedagogy of the Movement, the concept of education and the goals for education that Social Movement, the training matrices and the potential to transform the school from a pedagogical proposal that has these elements as guiding. They present also the original foundations of Complexes Study in its historical origin and design. Is discussed about the changes and curriculum innovations, curriculum as schooling as social reproduction and presents the structure of Curriculum Proposal by Complex of Study. Forth, it is shown how the experiment occurred in basis School Iraci Salette Strozak. At this point, we propose a dialogue on the transformations in the organization of pedagogical work, discussing the elements of the proposal that are being experienced and the changes already perceived. Still, we address the issue of formation of educators and also elements relating to the challenges andadvancements encountered by the school in this area, and possible implications for the experiment.

Flowering is a fundamental process in the life cycle for plant. This process is marked by vegetative to reproductive apical meristem conversion, due to interactions between several factors, both internal and external to plant. Therefore, eight subtractive libraries were constructed using apical meristem induced or not induced for two contrasting species: Solanum lycopersicum cv. Micro-Tom and Solanum pimpinellifolium. Several cDNAs were identified and among these, were selected two cDNAs: one homologous cDNA to cyclophilin (LeCYP1) and the other to Auxin repressed protein (ARP). It has observed that LeCYP1 and ARP genes are important in the developmental process to plants. In silico analysis, were used several databases with the exclusion criterion E-value <1.0x10-15. As a result, conservation was observed for proteins analyzed by means of multiple alignments and the presence of functional domains. Then, overexpression cassettes were constructed for the ARP cDNA in sense and antisense orientations. For this step, it was used the CaMV35S promoter. The cDNA orientation (sense or antisense) in relation to the promoter was determined by restriction enzymes and sequencing. Then, this cassette was transferred to binary vector pZP211 and these cassettes were transferred into Agrobacterium tumefaciens LBA4404. S. lycopersicum cv. Micro-Tom (MT) and MT-Rg1 plants were transformed. In addition, seedlings were subjected to hormone treatments using a synthetic auxin (- naphthalene acetic acid) and cyclosporin A (cyclophilin inhibitor) treatments and it was found that the hormone treatment there were changes in development of lateral roots pattern, probably related to decreases in auxin signaling caused by reduction of LeCYP1 in MT-dgt plants while cyclosporin A treatments, there was a slight delay in flowering in cv. MT plants. Furthermore, assay with real-time PCR (RT-qPCR) were done for expression level analysis from LeCYP1 and ARP in order to functionally characterize these sequences in tomato plants.

Sugarcane (Saccharum spp.) is a plant from Poaceae family that has an impressive ability to accumulate sucrose in the stalk, making it a significant component of the economy of many countries. About 100 countries produce sugarcane in an area of 22 million hectares worldwide. For this reason, many studies have been done using sugarcane as a plant model in order to improve production. A change in gravity may be one kind of abiotic stress, since it generates rapid responses after stimulation. In this work we decided to investigate the possible morphophysiological, biochemical and molecular changes resulting from microgravity. Here, we present the contributions of an experiment where sugarcane plants were submitted to microgravity flight using a vehicle VSB-30, a sounding rocket developed by Aeronautics and Space Institute teams, in cooperation with the German Space Agency. Sugarcane plants with 10 days older were submitted to a period of six minutes of microgravity using the VSB-30 rocket. The morphophysiological analyses of roots and leaves showed that plants submitted to the flight showed changes in the conduction tissues, irregular pattern of arrangement of vascular bundles and thickening of the cell walls, among other anatomical changes that indicate that the morphology of the plants was substantially influenced by gravitational stimulation, besides the accumulation of hydrogen peroxide, an important signaling molecule in stress conditions. We carried out RNA extraction and sequencing using Illumina platform. Plants subjected to microgravity also showed changes in enzyme activity. It was observed an increased in superoxide dismutase activity in leaves and a decreased in its activity in roots as well as for ascorbate peroxidase activity. Thus, it was concluded that the changes in gravity were perceived by plants, and that microgravity environment triggered changes associated with a reactive oxygen specie signaling process. This work has helped the understanding of how the gravity affects the structural organization of the plants, by comparing the anatomy of plants subjected to microgravity and plants grown in 1g gravity

In vitro and in animal models, APE1, OGG1, and PARP-1 have been proposed as being involved with inflammatory response. In this work, we have investigated if the SNPs APE1 Asn148Glu, OGG1 Ser326Cys, and PARP-1 Val762Ala are associated to meningitis and also developed a system to enable the functional analysis of polymorphic proteins. Patients with bacterial meningitis (BM), aseptic meningitis (AM) and controls (non-infected) genotypes were investigated by PIRA-PCR or PCR-RFLP. DNA damages were detected in genomic DNA by Fpg treatment. IgG and IgA were measured from plasma and the cytokines and chemokines were measured from cerebrospinal fluid samples using Bio-Plex assays. The levels of NF-κB and c-Jun were measured in CSF by dot blot assays. A significant (P<0.05) increase in the frequency of APE1 148Glu allele in BM and AM patients was observed. A significant increase in the genotypes Asn/Asn in control group and Asn/Glu in BM group was also found. For the SNP OGG1 Ser326Cys, the genotype Cys/Cys was more frequent (P<0.05) in BM group. The frequency of PARP-1 Val/Val genotype was higher in control group (P<0.05). The occurrence of combined SNPs increased significantly in BM patients, indicating that these SNPs may be associated to the disease. Increasing in sensitive sites to Fpg was observed in carriers of APE1 148Glu allele or OGG1 326Cys allele, suggesting that SNPs affect DNA repair activity. Alterations in IgG production were observed in the presence of SNPs APE1Asn148Glu, OGG1Ser326Cys or PARP-1Val762Ala. Reductions in the levels ofIL-6, IL-1Ra, MCP-1/CCL2and IL-8/CXCL8 were observed in the presence of APE1148Glu allele in BM patients, however no differences were observed in the levels of NF-κB and c-Jun considering genotypes and analyzed groups. Using APE1 as model, a system to enable the analysis of cellular effects and functional characterization of polymorphic proteins was developed using strategies of cloning APE1 cDNA in pIRES2-EGFP vector, cellular transfection of the construction obtained, siRNA for endogenous APE1 and cellular cultures genotyping. In conclusion, we obtained evidences of an effect of SNPs in DNA repair genes on the regulation of immune response. This is a pioneering work in the field that shows association of BER variant enzymes with an infectious disease in human patients, suggesting that the SNPs analyzed may affect immune response and damage by oxidative stress level during brain infection. Considering these data, new approaches of functional characterization must be developed to better analysis and interactions of polymorphic proteins in response to this context

Members of the oomycete cause extensive losses in agriculture and widespread degradation in natural plant communities, being responsible for the death of thousands of trees every year. Two of the representative species are Phytophthora infestans, which causes late blight of potato, and Phytophthora cinnamomi, which causes chestnut ink disease, responsible for losses on sweet chestnut production in Europe. Genome sequencing efforts have been focused on the study of three species: P. infestans, P. sojae and P. ramorum. Phytophthora infestans has been developed as the model specie for the genus, possessing excellent genetic and genomics resources including genetic maps, BAC libraries, and EST sequences. Our research team is trying to sequence the genome of P. cinnamomi in order to gain a better understanding of this oomycete, to study changes in plant-pathogen relationships including those resulting from climate change and trying to decrease the pathogen’s impact on crops and plants in natural ecosystems worldwide. We present here a preliminary report of partially sequenced genomic DNA from P. cinnamomi encoding putative protein-coding sequences and tRNAs. Database analysis reveals the presence of genes conserved in oomycetes.

Introducción: A mediados de los años 70’s del siglo pasado el descubrimiento de la tecnología del ADN recombinante marca el inicio de la era de la biotecnología moderna. La implementación de estas tecnologías permitió la utilización de organismos como sistemas de expresión que a lo largo de los años ha generado la producción de una gran variedad de productos biológicos. Dentro de estos sistemas Pichia pastoris es un sistema de expresión ampliamente utilizado debido a sus características tales como la producción de proteínas en grandes cantidades, la liberación de los productos al medio de cultivo, la obtención de productos complejos que requieren modificaciones postraduccionales típicas de los eucariotas o que contienen puentes disulfuro, entre otras. Nocardia brasiliensis es una bacteria parcialmente ácido-alcohol resistente la cual forma colonias granulares, con hifas aéreas escasas, sus colonias exhiben un color anaranjado pardo con bordes en blanco. N. brasiliensis es patógena para el ser humano y es el agente causal del actinomicetoma. El actinomicetoma es una enfermedad crónica generalmente localizada en las extremidades. Se caracteriza por ser un proceso lento de tumefacción con nódulos, abscesos y fístulas.La Superóxido Dismutasa (SOD) es una enzima reductora polimérica que cataliza la conversión del ión superóxido a peróxido de hidrógeno y oxígeno molecular. La SOD ha sido propuesta como un factor de virulencia de microorganismos patógenos, cuya acción consiste en bloquear los efectores oxidativos del estallido respiratorio iniciado por los fagocítos en el fagolisosoma. Este mecanismo ha sido descrito para bacterias de los géneros Mycobacterium, Rhodococcus y Nocardia. Objetivo: producir y caracterizar la Superóxido Dismutasa A (SODA) de Nocardia brasiliensis en Pichia pastoris. Metodología: se realizó el diseño de primers adicionando secuencias de sitios de corte para las enzimas XhoI y AvrII, así como una cola de histidinas en el extremo 5’ para la amplificación del gen sodA de N. brasiliensis a partir del ADN genómico de Nocardia brasiliensis. El amplicón se clonó en el vector de expresión pPIC9. Se llevó a cabo la transformación por electroporación de levaduras Pichia pastoris GS115. La producción de SOD se llevó a cabo en inducciones de 96 h con metanol como agente inductor. Los sobrenadantes se dializaron con membranas de celulosa. Los dializados se observaron por SDS-PAGE y western blot. Se analizó la actividad funcional de la enzima con el SOD Assay kit de Sigma Aldrich. Resultados: Por reacción en cadena de la polimerasa se obtuvo una secuencia de 625 pb correspondiente al gen sodA. El fragmento se ligó al vector de expresión pPIC9 y fue caracterizado con las enzimas de restricción XhoI y AvrII. Las cepas trasformadas de P. pastoris GS115 se caracterizaron con el gen aox1 obteniendo cepas Mut+ y Muts. Los análisis por SDSPAGE mostraron bandas no observadas en el control negativo de expresión mientras en los western blot solo una de las clonas mostró señal. Los análisis de actividad funcional sugieren inhibición de la reacción enzimática infiriendo presencia de la proteína SOD en el medio dializado. Conclusiones: Se logró la construcción del sistema de expresión Pichia pastoris con el casete de expresión de la SOD de N. brasiliensis. Así como la generación de cepas Mut+y Muts. En los ensayos de actividad funcional se observó inhibición de la reacción enzimática.

Este comunicado descreve os procedimentos de coleta e processamento de líquido sinovial e sangue seguido pela extração de RNA genômico e, finalmente, o diagnóstico molecular do vírus pela técnica de RT-nested PCR.
