The rates of oximolysis of p-nitrophenyl diphenyl phosphate (PNPDPP) by Acetophenoxime; 10-phenyl-10-hydi-oxyiminodecanoic acid; 4-(9-carboxynonanyl)-1-(9-carboxy-1-hydroyiminononanyl) benzene; 1-dodecyl-2-[(hydroxyimino)methyl]-pyridinium chloride (IV) and N-methylpyridinium-2-aldoxime chloride were determined in micelles of N-hexadecyl-N,N,N-trimethylammonium chloride (CTAC), N-hexadecyl-N,N-dimethylammonium propanesulfonate and dioctadecyldimethylammonium chloride (DODAC) vesicles. The effects of CTAC micelles and DODAC vesicles on the rates of oxymolysis of O,O-Diethyl O-(4-nitrophenyl) phosphate (paraoxon) by oxime IV were also determined. Analysis of micellar and vesicular effects on oximolysis of PNPDPP, using pseudophase or pseudophase with explicit consideration of ion exchange models, required the determination of the aggregate`s effects on the pK(a), of oximes and on the rates of PNPDPP hydrolysis. All aggregates increased the rate of oximolysis of PNPDPP and the results were analyzed quantitatively. In particular, DODAC vesicles catalyzed the reaction and increased the rate of oximolysis of PNPDPP by IV several million fold at pH`s compatible with pharmaceutical formulations. The rate increase produced by DODAC vesicles on the rate of oximolysis paraoxon by IV demonstrates the pharmaceutical potential of this system, since the substrate is used as an agricultural defensive agent and the surfactant is extensively employed in cosmetic formulations. Background: Organophosphate (OP) pesticides are well-known developmental neurotoxicants that have been linked to abnormal cognitive and behavioral endpoints through both epidemiological studies and animal models of behavioral teratology, and are implicated in the dysfunction of multiple neurotransmitters, including dopamine. Chemical similarities between OP pesticides and organophosphate flame retardants (OPFRs), a class of compounds growing in use and environmental relevance, have produced concern regarding whether developmental exposures to OPFRs and OP pesticides may share behavioral outcomes, impacts on dopaminergic systems, or both. Methods: Using the zebrafish animal model, we exposed developing fish to two OPFRs, TDCIPP and TPHP, as well as the OP pesticide chlorpyrifos, during the first 5 days following fertilization. From there, the exposed fish were assayed for behavioral abnormalities and effects on monoamine neurochemistry as both larvae and adults. An experiment conducted in parallel examined how antagonism of the dopamine system during an identical window of development could alter later life behavior in the same assays. Finally, we investigated the interaction between developmental exposure to an OPFR and acute dopamine antagonism in larval behavior. Results: Developmental exposure to all three OP compounds altered zebrafish behavior, with effects persisting into adulthood. Additionally, exposure to an OPFR decreased the behavioral response to acute D2 receptor antagonism in larvae. However, the pattern of behavioral effects diverged substantially from those seen following developmental dopamine antagonism, and the investigations into dopamine neurochemistry were too variable to be conclusive. Thus, although the results support the hypothesis that OPFRs, as with OP pesticides such as chlorpyrifos, may present a risk to normal behavioral development, we were unable to directly link these effects to any dopaminergic dysfunction.

Concepts used in this chapter include: Thermoregulation:- Thermoregulation refers to the body’s sophisticated, multi-system regulation of core body temperature. This hierarchical system extends from highly thermo-sensitive neurons in the preoptic region of the brain proximate to the rostral hypothalamus, down to the brain stem and spinal cord. Coupled with receptors in the skin and spine, both central and peripheral information on body temperature is integrated to inform and activate the homeostatic mechanisms which maintain our core temperature at 37oC.1 Body heat is lost through the skin, via respiration and excretions. The skin is perhaps the most important organ in regulating heat loss. Hyporthermia:- Hypothermia is defined as core body temperature less than 350C and is the result of imbalance between the body’s heat production and heat loss mechanisms. Hypothermia may be accidental, or induced for clinical benefit i.e: neurological protection (therapeutic hypothermia). External environmental conditions are the most common cause of accidental hypothermia, but not the only causes of hypothermia in humans. Other causes include metabolic imbalance; trauma; neurological and infectious disease; and exposure to toxins such as organophosphates. Therapeutic Hypothermia:- In some circumstances, hypothermia can be induced to protect neurological functioning as a result of the associated decrease in cerebral metabolism and energy consumption. Reduction in the extent of degenerative processes associated with periods of ischaemia such as excitotoxic cascade; apoptotic and necrotic cell death; microglial activation; oxidative stress and inflammation associated with ischaemia are averted or minimised.2 Mild hypothermia is the only effective treatment confirmed clinically for improving the neurological outcomes of patient’s comatose following cardiac arrest.3

Laboratory experiments were conducted to determine the efficacy of spinosad (a biopesticide), chlorpyrifos-methyl (an organophosphorus compound (OP)) and s-methoprene (a juvenile hormone analogue) applied alone and in binary combinations against five stored-grain beetles in wheat. There were three strains of Rhyzopertha dominica, and one strain each of Sitophilus oryzae, Tribolium castaneum, Oryzaephilus surinamensis and Cryptolestes ferrugineus. These strains were chosen to represent a range of possible resistant genotypes, exhibiting resistance to organophosphates, pyrethroids or methoprene. Treatments were applied at rates that are registered or likely to be registered in Australia. Adults were exposed to freshly treated wheat for 2 weeks, and the effects of treatments on mortality and reproduction were determined. No single protectant or protectant combination controlled all insect strains, based on the criterion of >99% reduction in the number of live F1 adults relative to the control. The most effective combinations were spinosad at 1 mg kg-1+chlorpyrifos-methyl at 10 mg kg-1 which controlled all strains except for OP-resistant O. surinamensis, and chlorpyrifos-methyl at 10 mg kg-1+s-methoprene at 0.6 mg kg-1 which controlled all strains except for methoprene-resistant R. dominica. The results of this study demonstrate the difficulty in Australia, and potentially other countries which use protectants, of finding protectant treatments to control a broad range of pest species in the face of resistance development.

Fruit size and quality are major problems in early-season stonefruit cultivars grown in Australia and South-East Asia. In Australia, Thailand and Vietnam, new training and trellising systems are being developed to improve yield and fruit quality. Australian trials found that new training systems, such as the Open Tatura system, are more productive compared with standard vase-trained trees. We established new crop-loading indices for low-chill stonefruit to provide a guide for optimum fruit thinning based on fruit number per canopy surface and butt cross sectional area. Best management practices were developed for low-chill stonefruit cultivation using growth retardants, optimizing leaf nitrogen concentrations and controlling rates and timing of irrigation. Regulated deficit irrigation (RDI) improved fruit sugar concentrations by restricting water application during stage II of fruit growth. New pest and disease control measures are also being developed using a new generation of fruit fly baits. Soft insecticides such as (Spinosad) are used at significantly lower concentrations and have lower mammalian toxicity than the organophosphates currently registered in Australia. In addition, fruit fly exclusion netting effectively eliminated fruit fly and many other insect pests from the orchard with no increase in diseases. This netting system increased sugar concentrations of peach and nectarine by as much as 30%. Economic analyses showed that the break-even point can be reduced from 12 to 6 years Open Tatura trellising and exclusion netting.

Kasvinsuojeluaineita käytetään elintarviketuotannossa kasvitautien ehkäisemiseen sekä kasvintuhoojien vaikutusten ajoittamiseen ja kasvun säätelyyn. Käytettyjen tehoaineiden riskinarviointi yksi kerrallaan on tuottanut tärkeää perustietoa, mutta se ei ole antanut selkeää kokonaiskuvaa kuluttajien altistumisesta. Tämän vuoksi asetelmaa tarkasteltiin kokonaisvaltaisesti, ottaen kaikki elintarvikkeista havaitut tehoainejäämät mukaan arvioon kumulatiivisesti. Aikuisten lisäksi mukana on ensi kertaa myös lapsiryhmiä. Ensimmäistä kertaa Suomessa tarkastellaan sekä pitkäaikaista että akuuttia altistusta. Esitetty kumulatiivinen riskinarviointi perustuu vuosina 2002- 2008 kasvinsuojeluainejäämien valvonnassa yhteensä 10 565 elintarvikenäytteestä saatuihin tutkimustuloksiin. Lisäksi riskinarvioinnissa on käytetty Terveyden ja hyvinvoinnin laitoksen ja DIPP-konsortion tuottamia aikuisten ja lasten ruoankulutustietoja (Finravinto 2007 ja DIPP-ravintotutkimukset). Ravinnon välittämälle tehoainejäämien altistukselle on tunnusomaista matala perustaso, jossa esiintyy lyhytaikaisia altistushuippuja. Kun ravinnon välittämä pitkäaikainen altistus kasvinsuojeluaineille on hyväksyttävällä tasolla, ei lyhytaikaisen altistuksen tilanne kaikilta osin ole yhtä hyvä. Kolmivuotiailla lapsilla todennäköisyys aRfD:n ylittymiselle on organofosfaattien ja karbamaattien osalta ollut suurempi kuin 0,1 % eli enemmän kuin yksi tuhannesta, mikä ei vielä vastaa tavoitteita. Ylitykset aiheutuvat tuontituotteista,koska niiden taustalla olevia karbamaatteja ja organofosfaatteja ei enää käytetä Suomessa. Myönteistä kehitystä on tapahtunut, mutta tilannetta on aiheellista edelleen seurata.

A ação inibitória dos organofosforados sobre as esterases, por ser específica, pode ser empregada como um eficiente biomarcador da exposição de seres vivos aos organofosforados. A inibição da acetilcolinesterase (AChE; EC 3.1.1.7) provoca acúmulo do neurotransmissor acetilcolina nas fendas sinápticas colinérgicas, o que pode resultar na morte do indivíduo. Outra atividade também afetada por organofosforados é a da enzima carboxilesterase (CarbE; EC 3.1.1.1). CarbE estão envolvidas na fase I da biotransformação de xenobióticos e atuam como captadoras (scavengers) de organofosfatos, incluindo os formados pela biotransformação dos organofosforados. As CarbE estudadas até hoje se ligam com maior velocidade aos organofosfatos do que as colinesterases. Por isto se admite que CarbE possam diminuir, por captação estequiométrica, a ligação tóxica de moléculas de organofosfatos às acetilcolinesterases das sinapses colinérgicas e das placas motoras dos músculos. Experimentos realizados em nosso laboratório mostraram que a atividade da CarbE está aproximadamente 50% menor no soro e no fígado de pacus submetidos à hipoxia. Por causa disso, em razão de uma possível diminuição da capacidade captadora da CarbE, decidimos verificar se o pacu em hipoxia seria mais sensível aos agrotóxicos organofosforados. Para este propósito foram colocados seis pacus divididos em dois tanques. No primeiro tanque, os animais foram submetidos a 24 horas de hipoxia seguidos por mais 4 horas de exposição ao organofosforado metilparation em duas concentrações diferentes (0,02 ou 0,01 mg / L). No segundo tanque os animais permaneceram em normoxia durante o mesmo período de 24 horas e depois foram expostos ao metilparation como no primeiro tanque. As atividades da AChE ensaiada com acetiltiocolina, a da butirilcolinesterase (BChE) ensaiada com butiriltiocolina e a da CarbE ensaiada com p-nitrofenilacetato foram avaliadas no soro, fígado, cérebro, músculo e coração dos pacus. Houve redução de aproximadamente 35% da atividade de CarbE no soro dos pacus submetidos a 24 horas de hipoxia. Uma queda de 85% na atividade de CarbE do soro foi observada nos animais que sofreram hipoxia e subsequente exposição a 0,02 mg de metilparation por litro. Com metilparation a 0,01 mg/L a diminuição observada foi de 48,2%. No músculo dos pacus expostos a 0,02 mg/L, as atividades de AChE e BChE cairam pela metade quando os mesmos foram submetidos à hipoxia quando comparados a animais que permaneceram em normoxia. Nos diversos tecidos dos pacus expostos a 0,01 mg/L de metilparation não observamos diferenças significativas nas atividades de AChE, BChE ou CarbE. Concluímos que a duplicação da concentração de metilparation de 0,01 para 0,02 mg/L levou à atividade residual de CarbE do soro de 51,8% para 15%. A ausência de mudanças nas atividades das esterases dos tecidos de animais expostos a 0,01 mg/L entre os grupos hipoxia e normoxia deve ter ocorrido porque a concentração de organofosforado não foi suficiente para superar a primeira barreira de proteção das esterases séricas e atingir os tecidos. Mas, no experimento com 0,02 mg/L de metilparation, as inibições de AChE e de BChE no músculo dos animais em hipoxia podem ser explicadas pela diminuição da atividade de CarbE do soro dos pacus.

Quando as esterases acetilcolinesterase (AChE), butirilcolinesterase (BChE) e carboxilesterase (CarbE) hidrolisam ésteres de fosfato seus sítios ativos sofrem fosfatação inibitória. Por isto, tal fosfatação pode proteger seres vivos contra o espalhamento de xenobióticos organofosforados dentro de seus corpos, já que estas enzimas têm a capacidade de captar moléculas de pesticidas organofosforados estequiometricamente. Os organismos terrestres vivem em um ambiente com mais oxigênio do que os organismos aquáticos. Na água, quando o nível de oxigênio atinge aproximadamente 2,6 mg/L o ambiente está em hipoxia. Este fenômeno afeta ecossistemas aquáticos, uma vez que muitos organismos não conseguem se adaptar à baixa do oxigênio. Estudamos peixes em hipoxia e hiperoxia para entender melhor a bioquímica do funcionamento de suas enzimas captadoras de organofosforados quando eles estão expostos às variações físico-químicas de seus habitats. Dois grupos de no mínimo seis pacus (Piaractus mesopotamicus), seis peixes dourados (Carassius auratus auratus), seis tilápias (Oreochromis niloticus niloticus), seis piavussus (Leporinus macrocephalus), seis apaiaris (Astronotus ocellatus), ou seis carpas (Cyprinus carpio carpio) foram aclimatados à temperatura ambiente em dois aquários de 250 L. No primeiro aquário, pelo menos três animais ensaio de cada espécie sofreram hipoxia por diminuição da concentração de oxigênio até 0,5 mg/L através de borbulhamento de nitrogênio na água. Quando estes animais atingiram a hipoxia foram mantidos a 0,5 mg/L de oxigênio por 6, 8, 24 ou, no máximo, por 42 horas. Três peixes controle de cada espécie foram mantidos em normoxia (4,5 até 7,0 mg/L de oxigênio). Após estes tempos houve a retirada de cerca de 3,5 mL de sangue e dos fígados. Depois de coagular, o sangue foi centrifugado para retirada do soro sobrenadante, que foi usado como amostra para ensaios das esterases. Os fígados foram armazenados em freezer a -70 C e, no momento do ensaio, homogeneizados e centrifugados para obter as frações citosólica e microssomal. As atividades das esterases foram ensaiadas em espectrofotômetro com os substratos acetiltiocolina, butiriltiocolina ou p-nitrofenilacetato. As atividades sobre p-nitrofenilacetato (CarbE) do soro e do fígado sofreram queda em todos os exemplares das espécies submetidos à hipoxia. Tipicamente, esta atividade caiu cerca de 50% nos soros de pacus mantidos por 42 h sob concentrações de oxigênio abaixo de 1,0 mg/L. O tempo para que ocorresse a queda desta atividade enzimática variou de espécie para espécie.

Among the variety of applications for biosensors one of the exciting frontiers is to utilize those devices as post-synaptic sensing elements in chemical coupling between neurons and solid-state systems. The first necessary step to attain this challenge is to realize highly efficient detector for neurotransmitter acetylcholine (ACh). Herein, we demonstrate that the combination of floating gate configuration of ion-sensitive field effect transistor (ISFET) together with diluted covalent anchoring of enzyme acetylcholinesterase (AChE) onto device sensing area reveals a remarkable improvement of a four orders of magnitude in dose response to ACh. This high range sensitivity in addition to the benefits of peculiar microelectronic design show, that the presented hybrid provides a competent platform for assembly of artificial chemical synapse junction. Furthermore, our system exhibits clear response to eserine, a competitive inhibitor of AChE, and therefore it can be implemented as an effective sensor of pharmacological reagents, organophosphates, and nerve gases as well. © 2007 Materials Research Society.

Farming of salmon has become a significant industry in many countries over the past two decades. A major challenge facing this sector is infestation of the salmon by sea lice. The main way of treating salmon for such infestations is the use of medicines such as organophosphates, pyrethrins, hydrogen peroxide or benzoylphenyl ureas. The use of these medicines in fish farms is, however, highly regulated due to concerns about contamination of the wider marine environment. In this paper we report the use of photochemically active biocides for the treatment of a marine copepod, which is a model of parasitic sea lice. Photochemical activation and subsequent photodegradation of PDAs may represent a controllable and environmentally benign option for control of these parasites or other pest organisms in aquaculture.

The state of the art of voltammetric and amperometric methods used in the study and determination of pesticides in crops, food, phytopharmaceutical products, and environmental samples is reviewed. The main structural groups of pesticides, i.e., triazines, organophosphates, organochlorides, nitrocompounds, carbamates, thiocarbamates, sulfonylureas, and bipyridinium compounds are considered with some degradation products. The advantages, drawbacks, and trends in the development of voltammetric and amperometric methods for study and determination of pesticides in these samples are discussed.

In recent years, pollution in general and sea water pollution in particular, has become an important topic for national and international considerations. Because of its impact on society, marine pollution has attracted great attention from politicians, administrators, natural scientists and technologists all over the world. To save our environment from further deterioration, it is essential to have an assessment of this problem This thesis involves investigation of the lethal and sub lethal effects of four pesticides and two petroleum oil, individually and in combinations on two commercially important bivalves. Among the four pesticides used two are organophosphates and the other two are organochlorines. Synthetic Pesticides, especially organophosphates and organochlorines have become increasingly important additions to chemical wastes polluting natural aquatic Communities special attention is given in the present investigation to delineate the combined toxic effect of oil and pesticides. The results are presented under different sections to make the presentation meaningful.

Industrial pollutants, consisting of heavy metals, petroleum residues, petrochemicals, and a wide spectrum of pesticides, enter the marine environment on a massive scale and pose a very serious threat to all forms of aquatic life. Although, earlier, efforts were directed towards the identification of pollutants and their major sources, because of a growing apprehension about the potential harm that pesticides can inflict upon various aquatic fauna and flora, research on fundamental and applied aspects of pesticides in the aquatic environment has mushroomed to a point where it has become difficult to even keep track of the current advances and developments. The Cochin Estuarine System (CES), adjoining the Greater Cochin area, receives considerable amounts of domestic sewage, urban wastes, agricultural runoff as well as effluent from the industrial units spread all along its shores. Since preliminary investigations revealed that the most prominent of organic pollutants discharged to these estuarine waters were the pesticides, the present study was designed to analyse the temporal and spatial distribution profile of some of the more toxic, persistent pesticides ——— organochlorines such as DDT and their metabolites; HCH-isomers; a cyclodiene compound," Endosulfan and a widely distributed, easily degradable, organophosphorus compound, Malathion, besides investigating their sorptional and toxicological characteristics. Although, there were indications of widespread contamination of various regions of the CBS with DDT, HCH-isomers etc., due to inadequacies of the monitoring programmes and due to a glaring void of baseline data the causative factors could not identified authentically. Therefore, seasonal and spatial distributions of some of the more commonly used pesticides in the CES were monitored systematically, (employing Gas Chromatographic techniques) and the results are analysed.

Los organofosforados y carbamatos son ampliamente usados a nivel mundial como insecticidas en el hogar, jardinería y agricultura. La exposición tóxica a estos compuestos es un serio problema de salud pública, con más de 3 millones de intoxicaciones y 200.000 muertes reportadas por año; donde las intoxicaciones accidentales son más frecuentes en niños, agricultores y trabajadores de manufactura (floricultura, ganaderos, fumigadores).La idea de esta revisión es proveer información actualizada sobre la intoxicación por inhibidores de la colinesterasa a intensivistas y pediatras, con el fin de proporcionar una guía de manejo breve
