Afin de mieux comprendre l’évolution des fonctions du récepteur EphA4 pendant le développement du système nerveux central (SNC), nous avons étudié sa localisation cellulaire et subcellulaire dans l’hippocampe du rat, d’abord chez l’adulte, puis pendant le développement postnatal, ainsi que ses rôles potentiels dans la genèse, la migration ou la maturation des cellules granulaires dans l’hippocampe adulte. Pour ce faire, nous avons utilisé la méthode d’immunocytochimie en microscopie photonique, électronique et confocale. En microscopie photonique, une forte immunoréactivité (peroxydase/DAB) pour EphA4 est observée aux jours 1 et 7 suivant la naissance (P1 et P7) dans les couches de corps cellulaires, avec un marquage notamment associé à la surface des corps cellulaires des cellules granulaires et pyramidales, ainsi que dans les couches de neuropile du gyrus dentelé et des secteurs CA3 et CA1. L’intensité du marquage diminue progressivement dans les couches de corps cellulaires, entre P7 et P14, pour devenir faible à P21 et chez l’adulte, tandis qu’elle persiste dans les couches de neuropile, sauf celles qui reçoivent des afférences du cortex entorhinal. En microscopie électronique, après marquage à la peroxydase/DAB, EphA4 décore toute la surface des cellules pyramidales et granulaires, du corps cellulaire jusqu’aux extrémités distales, entre P1 et P14, pour devenir confiné aux extrémités synaptiques, c’est-à-dire les terminaisons axonales et les épines dendritiques, à P21 et chez l’adulte. À la membrane plasmique des astrocytes, EphA4 est redistribué comme dans les neurones, marquant le corps cellulaire et ses prolongements proximaux à distaux, à P1 et P7, pour devenir restreint aux prolongements périsynaptiques distaux, à partir de P14. D’autre part, des axones en cours de myélinisation présentent souvent une forte immunoréactivité punctiforme à leur membrane plasmique, à P14 et P21. En outre, dans les neurones et les astrocytes, le réticulum endoplasmique, l’appareil de Golgi et les vésicules de transport, organelles impliquées dans la synthèse, la modification posttraductionnelle et le transport des protéines glycosylées, sont aussi marqués, et plus intensément chez les jeunes animaux. Enfin, EphA4 est aussi localisé dans le corps cellulaire et les dendrites des cellules granulaires générées chez l’adulte, au stade de maturation où elles expriment la doublecortine (DCX). De plus, des souris adultes knockouts pour EphA4 présentent des cellules granulaires DCX-positives ectopiques, c’est-à-dire positionnées en dehors de la zone sous-granulaire, ce qui suggère un rôle d’EphA4 dans la régulation de leur migration. Ces travaux révèlent ainsi une redistribution d’EphA4 dans les cellules neuronales et gliales en maturation, suivant les sites cellulaires où un remodelage morphologique s’effectue : les corps cellulaires lorsqu’ils s’organisent en couches, les prolongements dendritiques et axonaux pendant leur croissance, guidage et maturation, puis les épines dendritiques, les terminaisons axonales et les prolongements astrocytaires distaux associés aux synapses excitatrices, jusque chez l’adulte, où la formation de nouvelles synapses et le renforcement des connexions synaptiques existantes sont exercés. Ces localisations pourraient ainsi correspondre à différents rôles d’EphA4, par lesquels il contribuerait à la régulation des capacités plastiques du SNC, selon le stade développemental, la région, l’état de santé, ou l’expérience comportementale de l’animal.

Mice that lack all beta1-class integrins in neurons and glia die prematurely after birth with severe brain malformations. Cortical hemispheres and cerebellar folia fuse, and cortical laminae are perturbed. These defects result from disorganization of the cortical marginal zone, where beta1-class integrins regulate glial endfeet anchorage, meningeal basement membrane remodeling, and formation of the Cajal-Retzius cell layer. Surprisingly, beta1-class integrins are not essential for neuron-glia interactions and neuronal migration during corticogenesis. The phenotype of the beta1-deficient mice resembles pathological changes observed in human cortical dysplasias, suggesting that defective integrin-mediated signal transduction contributes to the development of some of these diseases.

In the CNS, myelinating oligodendrocytes and axons form a functional unit based on intimate cell-cell interactions. In addition to axonal insulation serving to increase the conduction velocity of electrical impulses, oligodendrocytes provide trophic support to neurons essential for the long-term functional integrity of axons. The glial signals maintaining axonal functions are just at the beginning to become uncovered. Yet, their determination is highly relevant for all types of demyelinating diseases, where lack of glial support significantly contributes to pathology. rnThe present PhD thesis uncovers exosomes as a novel signaling entity in the CNS by which cargo can be transferred from oligodendrocytes to neurons. Exosomes are small membranous vesicles of endocytic origin, which are released by almost every cell type and have been implicated in intercellular communication. Oligodendrocytes secrete exosomes containing a distinct set of proteins as well as mRNA and microRNA. Intriguingly, oligodendroglial exosome release is stimulated by the neurotransmitter glutamate indicating that neuronal electrical activity controls glial exosome release. In this study, the role of exosomes in neuron-glia communication and their implications on glial support was examined. Cortical neurons internalized and accumulated oligodendroglial exosomes in the neuronal cell soma in a time-dependent manner. Moreover, uptake occurred likewise at the somatodendritic and axonal compartment of the neurons via dynamin and clathrin dependent endocytosis. Intriguingly, neuronal internalization of exosomes resulted in functional retrieval of exosomal cargo in vitro and in vivo upon stereotactic injection of Cre recombinase bearing exosomes. Functional recovery of Cre recombinase from transferred exosomes was indicated by acquired reporter recombination in the target cell. Electrophysiological analysis showed an increased firing rate in neurons exposed to oligodendroglial exosomes. Moreover, microarray analysis revealed differentially expressed genes after exosome treatment, indicating functional implications on neuronal gene expression and activity. rnTaken together, the results of this PhD thesis represent a proof of principle for exosome transmission from oligodendrocytes to neurons suggesting a new route of horizontal transfer in the CNS.rn

RESUMO: A isquémia cerebral é uma das doenças mais predominantes a nivel mundial, sendo uma das principais causas de mortalidade e invalidez. Parte da propagação de dano no cérebro é causado por inflamação descontrolada, causada principalmente por disfunção da microglia. Desta forma, existe a necessidade de tentar desenvolver estratégias para melhor compreender e modular as acções destas células. O monóxido de carbono (CO), é uma molécula endógena com provas dadas como anti-neuroinflamatório em vários modelos. Assim, o principal objectivo do trabalho foi o estudo do CO como um modulador da acção da microglia, com principal foco dado à comunicação entre estas células e neurónios, tentando entender se existe um efeito neuroprotector por inibição da inflamação. Um protocolo de meio condicionado foi estabelecido usando as linhas celulares BV2 e SH-SY5Y, de microglia e neurónio. A molécula CORM-A1, que liberta expontaniamente CO, foi usada como método de entrega da molécula às celulas. Demonstrámos que o pre-tratamento de células BV2 com CORM-A1 gera neuroprotecção já que reduz a morte celular de neurónios SH-SY5Y quando são incubados com meio condicionado de microglia activada em conjunto com o pró-oxidante t-BHP (tert-butil hidroperóxido). Assim, considerámos que o CO promove neuroprotecção ao inibir as acções inflamatórias da microglia. O papel anti-inflamatório da molécula CORM-A1 foi confirmado quando se verificou que pré-tratamento desta molécula em microglia BV2 limita a secreção de TNF-α mas estimula a secreção de IL-10. Por último, a CORM-A1 induziu a expressão do receptor da microglia CD200R1, molécula que participa na comunicação neurónio-microglia e fundamental para a modulação das acções inflamatórias destas últimas. Em suma, o nosso trabalho reforçou as propriedades anti-neuroinflamatórias do CO e uma capacidade de modular viabilidade neuronal através do seu efeito a nível de comunicação célula-célula. ---------------------------- ABSTRACT: Brain ischemia is a widespread disease worldwide, being one of the main causes of mortality and permanent disability. A portion of the damage that ensues following the ischemic event is caused by unrestrained inflammation, which is mainly orchestrated by exacerbated microglial activity. Hence, developing strategies for modulating microglial inflammation is a major concern nowadays. The endogenous molecule carbon monoxide (CO) has been shown to possess anti-neuroinflammatory properties using in vitro and in vivo approaches. Thus, our objective was to study CO as modulator of microglial activity, in particular in what concerns their communication with neurons, by promoting neuronal viability and limiting inflammatory output of activated microglia. A conditioned media strategy was established with BV2 microglia and SH-SY5Y neurons as cell models. CO-releasing molecule A1 (CORM-A1), a compound that releases CO spontaneously, was used as method of CO delivery to cells. We found that CORM-A1 pre-treatment in BV2 cells yields neuroprotective results, as it limits cell death when SH-SY5Y neurons are challenged with conditioned media from LPS-activated microglia and the pro-oxidant t-BHP (tert-butyl-hydroperoxide). Thus, we assumed carbon monoxide promotes neuroprotection via inhibition of microglial inflammation, displaying a non-cell autonomous role. CORM-A1 pre-treatment limited inflammation by inhibiting BV2 secretion of TNF-α and stimulating IL-10 production. These results reinforce that CO’s anti-inflammatory role confers neuroprotection, as the alterations in these cytokines occur concurrently with the increase in SH-SY5Y viability. Finally, we showed for the first time that carbon monoxide promotes the expression of CD200R1, a microglial receptor involved in neuron-glia communication and modulation of microglia inflammation. Further studies are necessary to clarify this role. Altogether, other than just highlighting CO as an anti-inflammatory and neuroprotective molecule, this work set the foundation for disclosing its involvement in cell-to-cell communication.

There has been tremendous progress in understanding neural stem cell (NSC) biology, with genetic and cell biological methods identifying sequential gene expression and molecular interactions guiding NSC specification into distinct neuronal and glial populations during development. Data has emerged on the possible exploitation of NSC-based strategies to repair adult diseased brain. However, despite increased information on lineage specific transcription factors, cell-cycle regulators and epigenetic factors involved in the fate and plasticity of NSCs, understanding of extracellular cues driving the behavior of embryonic and adult NSCs is still very limited. Knowledge of factors regulating brain development is crucial in understanding the pathogenetic mechanisms of brain dysfunction. Since injury-activated repair mechanisms in adult brain often recapitulate ontogenetic events, the identification of these players will also reveal novel regenerative strategies. Here, we highlight the purinergic system as a key emerging player in the endogenous control of NSCs. Purinergic signalling molecules (ATP, UTP and adenosine) act with growth factors in regulating the synchronized proliferation, migration, differentiation and death of NSCs during brain and spinal cord development. At early stages of development, transient and time-specific release of ATP is critical for initiating eye formation; once anatomical CNS structures are defined, purinergic molecules participate in calcium-dependent neuron-glia communication controlling NSC behaviour. When development is complete, some purinergic mechanisms are silenced, but can be re-activated in adult brain after injury, suggesting a role in regeneration and self-repair. Targeting the purinergic system to develop new strategies for neurodevelopmental disorders and neurodegenerative diseases will be also discussed.

Information processing in the human brain has always been considered as a source of inspiration in Artificial Intelligence; in particular, it has led researchers to develop different tools such as artificial neural networks. Recent findings in Neurophysiology provide evidence that not only neurons but also isolated and networks of astrocytes are responsible for processing information in the human brain. Artificial neural net- works (ANNs) model neuron-neuron communications. Artificial neuron-glia networks (ANGN), in addition to neuron-neuron communications, model neuron-astrocyte con- nections. In continuation of the research on ANGNs, first we propose, and evaluate a model of adaptive neuro fuzzy inference systems augmented with artificial astrocytes. Then, we propose a model of ANGNs that captures the communications of astrocytes in the brain; in this model, a network of artificial astrocytes are implemented on top of a typical neural network. The results of the implementation of both networks show that on certain combinations of parameter values specifying astrocytes and their con- nections, the new networks outperform typical neural networks. This research opens a range of possibilities for future work on designing more powerful architectures of artificial neural networks that are based on more realistic models of the human brain.

Evidence concerning the presence or absence of common neuronglia lineages in the postnatal mammalian central nervous system is still a matter of speculation. We address this problem using optic nerve explants, which show an extremely long survival in culture. Morphological, immunocytochemical and immunochemical methods were applied. The results obtained from in vitro tissue were compared with optic nerves (ONs) and whole-brain samples from animals of different ages. Newborn rat ONs represented the starting material of our tissue culture; they are composed of unmyelinated axons, astrocytes and progenitor cells but devoid of neuronal cell bodies. At this age, Western blots of ONs were positively stained by neurofilament and synapsin I specific antibodies. These bands increased in intensity during postnatal in situ development. In explant cultures, the glia cells reach a stage of functional differentiation and they maintain, together with undifferentiated cells, a complex histotypic organization. After 6 days in vitro, neurofilaments and synapsin I could not be detected on immunoblots, indicating that 1) axonal degeneration was completed, and 2) neuronal somata were absent at the time. Surprisingly, after about 4-5 weeks in culture, a new cell type appeared, which showed characteristics typical of neurons. After 406 days in vitro, neurofilaments and synapsin I were unequivocally detectable on Western blots. Furthermore, both immunocytochemical staining and light and electron microscopic examinations corroborated the presence of this earlier-observed cell type. These in vitro results clearly show the high developmental plasticity of ON progenitor cells, even late in development. The existence of a common neuron-glia precursor, which never gives rise to neurons in situ, is suggested.

Management of chronic pain is a real challenge, and current treatments focusing on blocking neurotransmission in the pain pathway have only resulted in limited success. Activation of glia cells has been widely implicated in neuroinflammation in the central nervous system, leading to neruodegeneration in many disease conditions such as Alzheimer's and multiple sclerosis. The inflammatory mediators released by activated glial cells, such as tumor necrosis factor-α and interleukin-1β can not only cause neurodegeneration in these disease conditions, but also cause abnormal pain by acting on spinal cord dorsal horn neurons in injury conditions. Pain can also be potentiated by growth factors such as BDNF and bFGF that are produced by glia to protect neurons. Thus, glia cells can powerfully control pain when they are activated to produce various pain mediators. We will review accumulating evidence supporting an important role of microglia cells in the spinal cord for pain control under injury conditions (e.g. nerve injury). We will also discuss possible signaling mechanisms in particular MAP kinase pathways that are critical for glia control of pain. Investigating signaling mechanisms in microglia may lead to more effective management of devastating chronic pain.

Evidências crescentes na literatura têm sugerido papel importante para os fatores ambientais, como a exposição a pesticidas, na patogênese da doença de Parkinson. Em animais experimentais, a exposição à rotenona, um pesticida e piscicida de uso comum, induz características de parkinsonismo através da inibição do complexo I mitocondrial. O objetivo deste estudo foi investigar a morte de neurônios induzida por rotenona utilizando culturas primárias mistas neurônio/glia derivadas de hipocampo e de mesencéfalo ventral de ratos, bem como o papel do Ca2+ na neste modelo experimental e a utilização de extrato aquoso de folhas de mogno com substâncias com alto poder antioxidante. A perda neuronal foi analisada com ensaios colorimétricos (MTT e LDH). Nossos resultados mostraram significativa redução na viabilidade celular após exposição à rotenona de maneira dependente de concentração, mas não dependente de tempo. Foi observada igual e elevada suscetibilidade em culturas mistas neurônio/glia derivadas de hipocampo e de mesencéfalo ventral ao agente neurotóxico. Em termos mecanicísticos, nossos resultados mostraram um papel discreto desempenhado pelo Ca2+ mitocondrial na neurodegeneração induzida por rotenona. Além disso, neste paradigma utilizado, verificamos que o extrato aquoso de folhas de mogno não promoveu proteção contra a toxicidade da rotenona, na concentração testada; ainda, promoveu efeito sinérgico em associação com rotenona. Verificou-se ainda que a rotenona, bem como o extrato de mogno promoveu indução de morte celular tanto por necrose quanto por apoptose, nas concentrações utilizadas. Os resultados deste estudo devem avançar nosso conhecimento sobre o mecanismo de ação de fatores ambientais na patogênese da doença de Parkinson.

Die Bildung, Aufrechterhaltung und die Funktionalität von Oligodendrozyten, den myelinisierenden Zellen des ZNS, bedarf einer präzisen Regulation von Ereignissen wie Migration, Proliferation und Differenzierung. Die Src-Kinasen spielen in vielen Signalkaskaden eine zentrale Rolle. In murinen Oligodendrozyten werden die beiden Src-Kinasen Fyn und Lyn exprimiert, diese Arbeit konzentriert sich auf die Analyse der Lyn Kinase. Es konnte die Expression von Lyn in Oligodendrozyten und Myelin in vitro und in vivo bestätigen werden, sowie die Lokalisation innerhalb und außerhalb von rafts. Je älter die Mäuse, desto weniger Lyn wird in Myelin exprimiert und desto stärker ist das verbleibende Lyn in den rafts lokalisiert. Die Aktivität von Lyn ist im Myelin 12-Tage alter Mäuse am höchsten. Synchronisation des Zellzyklus in Oli-neu Zellen zeigte eine zyklische Expression von Lyn. Transfektion aktiver und inaktiver Konstrukte machte deutlich, dass eine Lyn Aktivität die Zellausläufer Bildung, sowie die Differenzierung der Zelle, hemmt. Es konnte eine Assoziation der GluRB/C Untereinheit des AMPA Rezeptors mit Lyn nachgewiesen werden. Stimulierung des AMPA-Rezeptors mit Glutamat führte zu einer Aktivierung der Lyn Kinase und daraus resultierend, zu einer Inhibierung der Zelldifferenzierung. Welche biologische Relevanz diese Ergebnisse haben könnten, wurde in drei Hypothesen festgehalten. Zum einen könnte die Lyn Kinase den zeitlichen Übergang von Proliferation und Differenzierung in Oligodendrozyten regulieren und zwar sowohl in perinatalen oligodendroglialen Vorläuferzellen, als auch, unter besonderen Umständen, in adulten oligodendroglialen Vorläuferzellen. Des Weiteren könnte die Lyn Kinase auch eine Rolle in der Neuron-Glia-Synapsenbildung spielen. Weitere Versuche, insbesondere in vivo, müssen folgen, um diese Hypothesen zu bestätigen oder zu widerlegen.

Im ersten Teil dieser Doktorarbeit beabsichtigte meine Arbeit, die funktionelle Beteiligung des CB1 Rezeptors, einer Hauptkomponente des neuronalen Endocannabinoid-Systems (ECS), an der Ausbildung von verschiedenen Verhaltensphänotypen mit Hilfe von konditionalen Mausmutanten, denen der CB1 Rezeptor auf verschiedenen neuronalen Unterpopulationen fehlt, aufzuschlüsseln und zu untersuchen. Verschiedene Verhaltensmodelle wurden hierzu getestet. Dabei lag der Fokus dieser Arbeit auf der CB1f/f;D1-Cre Mauslinie, welche der CB1 Rezeptor auf den D1 Rezeptor exprimierenden Neuronen des Striatums fehlt. Ich konnte zeigen, dass der Verlust des CB1 Rezeptors auf diesen Neuronen keinen Einfluss auf basale neurologische Funktionen, Gewicht, Bewegung, Exploration, Sozialverhalten, Angst und Stressbewältigung der Tiere hat, jedoch eine Beteiligung an der Entwicklung von Suchtverhalten gegeben ist. Bei Betrachtung des Kokain-induzierten Suchtverhaltens zeigten die konditionalen Mausmutanten eine reduzierte Suchtanfälligkeit sowohl im Vergleich zu Tieren mit einem totalen CB1 Rezeptor Verlust in allen Körperzellen, als auch zu genetisch unveränderten Kontrollmäusen beider Linien.rnDes Weiteren zeigen die Ergebnisse dieser Studie eine große, aber gegensätzliche Beteiligung des ECS bei der Regulation von Exploration in Abhängigkeit des Verlustes des CB1 Rezeptors auf GABAergen Neuronen des Vorderhirns und kortikalen glutamatergen Neuronen, jedoch nicht auf striatalen Neuronen alleine. Zusätzlich war ich in der Lage, die Wichtigkeit des genetischen Hintergrunds von Mauslinien nicht nur auf die Ausbildung von spezifischen Verhaltensphänotypen, sondern auch auf die Genexpression zu zeigen.rnIn dem zweiten Teil dieser Arbeit, in dem ich mich auf die Funktion von Gliazellen konzentrierte, wurden ebenfalls Mausmutanten in verschiedenen Verhaltensmodellen getestet. Ein genetisches Auslöschen des NG2 Glykoproteins in Gliazellen sorgt in den Knock-out Mäusen für ein schlechteres Hörvermögen und ein reduziertes Depressionsverhalten im Vergleich zu ihren Wildtyp-Kontrollmäusen. Interessanterweise zeigten diese Tiere auch eine reduzierte Empfänglichkeit bei chemisch induzierten epileptischen Krämpfen, was eine Rolle des NG2 Glykoproteins bei der Kontrolle der glutamatergen Homöostase vorschlägt, die wahrscheinlich durch Strukturänderungen der Neuron-Glia-Synapse verursacht wird. rn

Typical neuroleptic drugs elicit their antipsychotic effects mainly by acting as antagonists at dopamine D2 receptors. Much of this activity is thought to occur in the cerebral cortex, where D2 receptors are found largely in inhibitory GABAergic neurons. Here we confirm this localization at the electron microscopic level, but additionally show that a subset of cortical interneurons with low or undetectable expression of D2 receptor isoforms are surrounded by astrocytic processes that strongly express D2 receptors. Ligand binding of isolated astrocyte preparations indicate that cortical astroglia account for approximately one-third of the total D2 receptor binding sites in the cortex, a proportion that we found conserved among rodent, monkey, and human tissues. Further, we show that the D2 receptor-specific agonist, quinpirole, can induce Ca2+ elevation in isolated cortical astrocytes in a pharmacologically reversible manner, thus implicating this receptor in the signaling mechanisms by which astrocytes communicate with each other as well as with neurons. The discovery of D2 receptors in astrocytes with a selective anatomical relationship to interneurons represents a neuron/glia substrate for cortical dopamine action in the adult cerebral cortex and a previously unrecognized site of action for antipsychotic drugs with affinities at the D2 receptor.

Recently we have shown that growth hormone (GH) inhibits neuronal differentiation and that this process is blocked by suppressor of cytokine signalling-2 (SOCS2). Here we examine several cortical and subcortical neuronal populations in GH hyper-responsive SOCS2 null (-/-) mice and GH non-responsive GH receptor null (GHR-/-) mice. While SOCS2-/- mice showed a 30% decrease in density of NeuN positive neurons in cortex compared to wildtype, GHR-/- mice showed a 25% increase even though brain size was decreased. Interneuron sub-populations were variably affected, with a slight decrease in cortical parvalbumin expressing interneurons in SOCS2-/- mice and an increase in cortical calbindin and calretinin and striatal cholinergic neuron density in GHR-/- mice. Analysis of glial cell numbers in cresyl violet or glial fibrillary acidic protein (GFAP) stained sections of cortex showed that the neuron: glia ratio was increased in GHR-/- mice and decreased in SOCS2-/- mice. The astrocytes in GHR-/- mice appeared smaller, while they were larger in SOCS2-/- mice. Neuronal soma size also varied in the different genotypes, with smaller striatal cholinergic neurons in GHR-/- mice. While the size of layer 5 pyramidal neurons was not significantly different from wildtype, SOCS2-/- neurons were larger than GHR-/- neurons.
