Monochamus beetles are the dispersing vectors of the nematode Bursaphelenchus xylophilus, the causative agent of pine wilt disease (PWD). PWD inflicts significant damages in Eurasian pine forests. Symbiotic microorganisms have a large influence in insect survival. The aim of this study was to characterize the bacterial community associated to PWD vectors in Europe and East Asia using a culture-independent approach. Twenty-three Monochamus galloprovincialiswere collected in Portugal (two different locations); twelve Monochamus alternatus were collected in Japan. DNA was extracted from the insects’ tracheas for 16S rDNA analysis through denaturing gradient gel electrophoresis and barcoded pyrosequencing. Enterobacteriales, Pseudomonadales, Vibrionales and Oceanospirilales were present in all samples. Enterobacteriaceae was represented by 52.2% of the total number of reads. Twenty-three OTUs were present in all locations. Significant differences existed between the microbiomes of the two insect species while for M. galloprovincialis there were no significant differences between samples from different Portuguese locations. This study presents a detailed description of the bacterial community colonizing the Monochamus insects’ tracheas. Several of the identified bacterial groups were described previously in association with pine trees and B. xylophilus, and their previously described functions suggest that they may play a relevant role in PWD.

The tribe Longidorini within the subfamily Longidorinae (Longidorus spp. and Paralongidorus spp.) and the subfamily Xiphineminae (Xiphinema spp.) are two large nematode groups with about 260 and 230 known species, respectively. They are globally two important groups of ectoparasitic nematodes considered to be major pests because of their activity as vectors of important plant nepovirus, with some species included in the list of quarantine pathogenic organisms in many European countries. Knowledge of the biodiversity and occurrence of this nematode group is a prerequisite for the establishment of sound management strategies and control measures. According to data collected from the databases (such as EPPO, FSTA, and Web of Science) and published in specialised literature, a total of 15 Longidorus, 1 Paralongidorus and 40 Xiphinema species have been recorded as present in Portugal. However, the taxonomic status of some species is controversial, and thus needs to be clarified. A comprehensive review for unravelling the biodiversity and occurrence of nematode species of the genus Longidorus, Paralongidorus and Xiphinema in Portugal is herein provided. This review includes an updated checklist of species with information on the localities, host plants and geographical distribution. Additionally, maps on the species distributions of Longidorinae and Xiphineminae nematodes present in Continental Portugal and the Portuguese Macaronesian islands are provided, as well as unpublished data on authors and comments on the current taxonomic status. Finally, new insights and directions for future research on Longidoridae in Portugal are presented.

The Columbia root-knot nematode (CRKN), Meloidogyne chitwoodi, is an EPPO A2 type quarantine pest since 1998. This nematode causes severe damage in economically important crops such as potato and tomato, making agricultural products unacceptable for the fresh market and food processing. Commonly used nematicidal synthetic chemicals are often environmentally unsafe. Essential oils (EOs) may constitute safer alternatives against RKN. EOs, isolated from 56 plant samples, were tested against CRKN hatching, in direct contact bioassays. Some of the most successful EOs were fractionated and the hydrocarbon molecules (HM) and oxygen-containing molecules (OCM) fractions tested separately. 24 EOs displayed very strong hatching inhibitions (≥90 %) at 2 µL mL−1 and were further tested at lower concentrations. Dysphaniaambrosioides, Filipendula ulmaria, Ruta graveolens, Satureja montana and Thymbra capitata EOs revealed the lowest EC50 values (<0.15 µL mL−1). The main compounds of these EOs, namely 2-undecanone, ascaridol, carvacrol, isoascaridol, methyl salicylate, p-cymene and/or γ-terpinene, were putatively considered responsible for CRKN hatching inhibition. S. montana and T. capitata OCM fractions showed hatching inhibitions higher than HM fractions. The comparison of EO and corresponding fractions EC50 values suggests interactions between OCM and HM fractions against CRKN hatching. These species EOs showed to be potential environmentally friendly CRKN hatching inhibitors; nonetheless, bioactivity should be considered globally, since its HM and OCM fractions may contribute, diversely, to the full anti-hatching activity.

The Pine Wood Nematode (PWN) Bursaphelenchus xylophilus is a severe forest pathogen in countries where it has been introduced and is considered a worldwide quarantine organism. In this study, protein markers for differentiating populations of this nematode were identified by studying differences among four selected Iberian and one American population. These populations were compared by quantitative proteomics (iTRAQ). From a total of 2860 proteins identified using the public database from the B. xylophilus genome project, 216 were unambiguous and significantly differentially regulated in the studied populations. Comparisons of their pairwise ratio were statistically treated and supported in order to convert them into discrete character states, suggesting that 141 proteins were not informative as population specific markers. Application of the Character Compatibility methodology on the remaining 75 proteins (belonging to families with different biological functions) excludes 27 which are incompatible among them. Considering only the compatible proteins, the method selects a subset of 30 specific unique protein markers which allowed the compared classification of the Iberian isolates. This approach makes it easier search for diagnostic tools and phylogenetic inference within species and populations of a pathogen exhibiting a high level of genetic diversity.

Yacon, Smallanthus sonchifolius, an Andean species. is a rich source of dictetíc oligofructans with low glucose content. proteins and phenolic compounds. These constituents have shown efficacy in the prevention of diet-related ehronic diseases, including gastroin-testinal disorders and diabetes |1,2|. Yacon is part of a research program at the National Center for Natural Products Research (NCNPR) and University of Mississippi Field Station to develop new alternative root crops for Mississippi while attempting to im-prove the diet of low incorne families. Yacon can be easily propa-gated by cultings. Virus and nematode infections have been re-ported on plants propagated by cuttings in Brazil. a country that hás adopted Yacon as specialty crop [3|. We have developed two culture systems. autotrophic and heterotrophic, to produce healthy plants. Herem we describe the presence of endophytic bactéria m micropropagated Yacon. In auxin free media, new roots were induced. Overa 15day period. the average root mduction per expiam was 5.45 to 8.75 under autotrophic and heterotrophic cul-tures, respectively. Root lenglh vaned between 3 and 60mrn. The presence of root hairs and lateral roots was noticed only in auto-trophic condilions. The root knot nematode (RKN), Meloidogyne incognita, is widespread worldwide and a major pathogen of several cultivated crops. The use of resistant genotypes is the most effective and environmentally sound way to manage RKN. In this study, we screened 16 selected sweet potato cultivars including Amanda, Bárbara, Beatriz, Beauregard, Brazlândia Branca, Brazlândia Rosada, Brazlândia Roxa, BRS Amélia, BRS Cuia, BRS Rubissol, Carolina Vitória, Duda, Júlia, Marcela, PA-26/2009, and Princesa obtained from Embrapa and Universidade Federal do Tocantins? germplasm bank. Studies were conducted under greenhouse and field conditions and the agronomic performance of the cultivars was evaluated in a nematode and soilborne insect-infested field. All 16 sweet potato cultivars tested were rated as resistant to this nematode both under greenhouse and field conditions with reproduction factors < 1. In the field infested with M. incognita, sweet potato cultivars Duda, BRS Amélia, Beauregard, Brazlândia Rosada, and Brazlândia Roxa stood out as superior cultivars, with average yield ranging from 26 to 47 tons per ha. Overall, most cultivars exhibited a fusiform to near fusiform root shape, a good characteristic for the market, and were moderately affected by insects (attack incidence 1 to 30%). As global demand for energy continues to rise, selecting new cultivars of sweet potatoes with increased resistance to nematode diseases and with high yield will be important for food security and biofuel production.

The aim of this study was to conduct a survey on nematodes associated with five cultivated fruit trees growing in the Agroforestry System (AFS) of the municipality of Oiapoque, Amapá, Brazil. Rhizosphere samples were collected from three points at the base of murici (Byrsonima sp.), soursop (Annona muricata), cupuaçu (Theobroma grandiflorum), passion fruit (Passiflora sp.) and peach palm (Bactris gasipaes) trees. Four species of phytonematodes were identified, namely Pratylenchus brachyurus, Helicotylenchus dihystera, Mesocriconema xenoplax and Rotylenchulus reniformis. The most prevalent and abundant species was R. reniformis, which was found in the rhizospheres of passion fruit, cupuaçu, soursop, and peach palm. The first record in Brazil of the nematode P. brachyurus, found in the roots of murici is reported here, and all of the nematode species identified here are the first records for fruit trees in the state of Amapá.

The continuous soybean-maize crop succession in the tropical region of Brazil has led to significant increases in the population size of root-knot (Meloidogyne incognita and M. javanica ) and root-lesion nematodes (Pratylenchus brachyurus), which make soils unsuitable for soybean cropping. A greenhouse study was conducted to identify sunflower genotypes adapted to the tropical region of Brazil and that are resistant to M. incognita, M. javanica and/or P. brachyurus . Two experiments for each nematode were conducted in a completely randomized design with six replicates. Gall index was calculated from visual scores (0?5) of gall intensity on roots for the root-knot nematode. Initial and final population density and reproduction factor were also measured for each nematode. Sunflower genotypes varied in resistance to the nematodes. Sunflower hybrids BRS 321 and BRS 323 were resistant to M. javanica and P. brachyurus and exhibited low gall index for M. incognita . The cultivars are good alternatives to using in the succession of soybean in nematode-infested areas of the tropical regions of Brazil. No sunflower genotype was identified as resistant to M. incognita and thus sunflower cropping is not indicated in areas infested with this nematode.
