Aiming to identify novel genetic variants and to confirm previously identified genetic variants associated with bone mineral density (BMD), we conducted a three-stage genome-wide association (GWA) meta-analysis in 27 061 study subjects. Stage 1 meta-analyzed seven GWA samples and 11 140 subjects for BMDs at the lumbar spine, hip and femoral neck, followed by a Stage 2 in silico replication of 33 SNPs in 9258 subjects, and by a Stage 3 de novo validation of three SNPs in 6663 subjects. Combining evidence from all the stages, we have identified two novel loci that have not been reported previously at the genome-wide significance (GWS; 5.0 × 10-8) level: 14q24.2 (rs227425, P-value 3.98 × 10-13, SMOC1) in the combined sample of males and females and 21q22.13 (rs170183, P-value 4.15 × 10-9, CLDN14) in the female-specific sample. The two newly identified SNPs were also significant in the GEnetic Factors for OSteoporosis consortium (GEFOS, n 5 32 960) summary results. We have also independently confirmed 13 previously reported loci at the GWS level: 1p36.12 (ZBTB40), 1p31.3 (GPR177), 4p16.3 (FGFRL1), 4q22.1 (MEPE), 5q14.3 (MEF2C), 6q25.1 (C6orf97, ESR1), 7q21.3 (FLJ42280, SHFM1), 7q31.31 (FAM3C, WNT16), 8q24.12 (TNFRSF11B), 11p15.3 (SOX6), 11q13.4 (LRP5), 13q14.11 (AKAP11) and 16q24 (FOXL1). Gene expression analysis in osteogenic cells implied potential functional association of the two candidate genes (SMOC1 and CLDN14) in bone metabolism. Our findings independently confirm previously identified biological pathways underlying bone metabolism and contribute to the discovery of novel pathways, thus providing valuable insights into the intervention and treatment of osteoporosis. © The Author 2013. Richard Lewontin proposed that the ability of a scientific field to create a narrative for public understanding garners it social relevance. This article applies Lewontin's conceptual framework of the functions of science (manipulatory and explanatory) to compare and explain the current differences in perceived societal relevance of genetics/genomics and proteomics. We provide three examples to illustrate the social relevance and strong cultural narrative of genetics/genomics for which no counterpart exists for proteomics. We argue that the major difference between genetics/genomics and proteomics is that genomics has a strong explanatory function, due to the strong cultural narrative of heredity. Based on qualitative interviews and observations of proteomics conferences, we suggest that the nature of proteins, lack of public understanding, and theoretical complexity exacerbates this difference for proteomics. Lewontin's framework suggests that social scientists may find that omics sciences affect social relations in different ways than past analyses of genetics.

Doctoral dissertation work in sociology examines how human heredity became a scientific, political and a personal issue in the 20th century Finland. The study focuses on the institutionalisation of rationales and technologies concerning heredity, in the context of Finnish medicine and health care. The analysis concentrates specifically on the introduction and development of prenatal screening within maternity care. The data comprises of medical articles, policy documents and committee reports, as well as popular guidebooks and health magazines. The study commences with an analysis on the early 20th century discussions on racial hygiene. It ends with an analysis on the choices given to pregnant mothers and families at present. Freedom to choose, considered by geneticists and many others as a guarantee of the ethicality of medical applications, is presented in this study as a historically, politically and scientifically constructed issue. New medical testing methods have generated new possibilities of governing life itself. However, they have also created new ethical problems. Leaning on recent historical data, the study illustrates how medical risk rationales on heredity have been asserted by the medical profession into Finnish health care. It also depicts medical professions ambivalence between maintaining the patients autonomy and utilizing for example prenatal testing according to health policy interests. Personalized risk is discussed as a result of the empirical analysis. It is indicated that increasing risk awareness amongst the public, as well as offering choices, have had unintended consequences. According to doctors, present day parents often want to control risks more than what is considered justified or acceptable. People s hopes to anticipate the health and normality of their future children have exceeded the limits offered by medicine. Individualization of the government of heredity is closely linked to a process that is termed as depolitization. The concept refers to disembedding of medical genetics from its social contexts. Prenatal screening is regarded to be based on individual choice facilitated by neutral medical knowledge. However, prenatal screening within maternity care also has its basis in health policy aims and economical calculations. Methodological basis of the study lies in Michel Foucault s writings on the history of thought, as well as in science and technology studies.

In order to investigate the modes of inheritance of serum immunoglobulin E (IgE) levels and atopic disease, serum IgE levels and data on allergic disease were obtained from 42 families ascertained through asthmatic children visiting an allergy clinic. Although the mean IgE levels were elevated (mean 637 U/ml), the prevalence of atopic disease in this population was surprisingly low. When the data were analyzed using complex segregation analysis, no major locus could be detected. Moreover, the polygenic heritability was unexpectedly small even though the correlation between serum IgE levels and the liability to atopic disease was around 0.4. Given this unusual set of findings, it is postulated that parasitic infections in this population have (in accordance with well-established results of parasitic disease) caused both elevated levels of serum IgE and a decreased prevalence of allergic disease with the possible masking of the various genetic components of serum IgE levels and atopic disease.

Starting from the early decades of the twentieth century, evolutionary biology began to acquire mathematical overtones. This took place via the development of a set of models in which the Darwinian picture of evolution was shown to be consistent with the laws of heredity discovered by Mendel. The models, which came to be elaborated over the years, define a field of study known as population genetics. Population genetics is generally looked upon as an essential component of modern evolutionary theory. This article deals with a famous dispute between J. B. S. Haldane, one of the founders of population genetics, and Ernst Mayr, a major contributor to the way we understand evolution. The philosophical undercurrents of the dispute remain relevant today. Mayr and Haldane agreed that genetics provided a broad explanatory framework for explaining how evolution took place but differed over the relevance of the mathematical models that sought to underpin that framework. The dispute began with a fundamental issue raised by Mayr in 1959: in terms of understanding evolution, did population genetics contribute anything beyond the obvious? Haldane's response came just before his death in 1964. It contained a spirited defense, not just of population genetics, but also of the motivations that lie behind mathematical modelling in biology. While the difference of opinion persisted and was not glossed over, the two continued to maintain cordial personal relations.

A gene is a unit of heredity in a living organism. It normally resides on a stretch of DNA that codes for a type of protein or for an RNA chain that has a function in the organism. All living things depend on genes, as they specify all proteins and functional RNA chains. Genes hold the information to build and maintain an organism’s cells and pass genetic traits to offspring. The gene has to be transferred to bacteria or eukaryotic cells for basic and applied molecular biology studies. Bacteria can uptake exogenous genetic material by three ways: conjugation, transduction and transformation. Genetic material is naturally transferred to bacteria in case of conjugation and transferred through bacteriophage in transduction. Transformation is the acquisition of exogenous genetic material through cell wall. The ability of bacteria of being transformed is called competency and those bacteria which have competency are competent cells. Divalent Calcium ions can make the bacteria competent and a heat shock can cause the bacteria to uptake DNA. But the heat shock method cannot be used for all the bacteria. In electroporation, a brief electric shock with an electric field of 10-20kV/cmmakes pores in the cell wall, facilitates the DNA to enter into the bacteria. Microprecipitates, microinjection, liposomes, and biological vectors are also used to transfer polar molecules like DNA into host cells.

Resumen: La crueldad, el prejuicio y los estereotipos eran algo común en la mirada que tenían los romanos de los demás pueblos. En el caso particular de los griegos, la visión que los romanos tenían de ellos era ambivalente: por un lado, admiraban a los escritores, a los filósofos, a los artistas, pero, por el otro, tenían en muy baja consideración al resto de los griegos. Marcaban una gran diferencia entre la Grecia clásica y su herencia y sus sometidos, los griegos contemporáneos. El Satyricon de Petronio, aunque no es una excepción dentro de la tradicional xenofobia contra los restantes pueblos, presenta con relación a los griegos una peculiaridad que lo distancia de la mayoría de los escritores latinos. La causa de esta singularidad quizá podría hallarse en el espíritu filohelénico de la corte de Nerón, de la que Petronio era el elegantiae arbiter.

对插入形心的Delaunay逐点插入算法,提出按单元可插度分组的双向链表组数据结构,避免了对最大可插度单元的搜索。采用了邻接单元搜索、双向链表存储、随机方向搜索、邻接旋转、几何量继承等技术,使算法的计算时间与生成单元数近似呈线性关系,时间复杂度达到O(N1.05),N为生成单元数。算例表明,在一台AMD Athlon3200+(主频2.0 GHz)PC上,该算法的四面体单元生成速度达50 000个/s以上。

A influência da dieta e da hereditariedade nas características dentofaciais foi avaliada através do exame de duas populações indígenas amazônicas divididas por um processo de fissão linear. Os indígenas que constituem a aldeia Arara-Iriri são descendentes de um único casal expulso da aldeia Arara-Laranjal. O crescimento da aldeia Iriri ocorreu pelo acasalamento de parentes próximos, ratificado por um alto coeficiente de consanguinidade (F=0,25, p<0,001). A epidemiologia da má oclusão e das características da face foi analisada nos indivíduos entre dois e 22 anos, das aldeias Iriri (n=46) e Laranjal (n=130). A biometria da dentição e da face foi obtida em 55 indígenas em dentição permanente sem perdas dentárias, através da fotogrametria facial e dos modelos de gesso. O desgaste dentário foi examinado em 126 indivíduos através da análise de regressão múltipla. Os resultados revelaram uma determinação significativa da idade no desgaste dos dentes (R2=87,6, p<0,0001), que se mostrou semelhante entre as aldeias (R2=0,027, p=0,0935). Por outro lado, diferenças marcantes foram observadas nas características dentofaciais. Revelou-se uma face mais vertical (dolicofacial) entre os índios Iriri e o predomínio do tipo braquifacial nos indígenas da aldeia original, corroborado pela fotogrametria. Uma face sagitalmente normal foi observada em 97,7% da aldeia Laranjal, enquanto faces convexas (26,1%, RR-16,96) e côncavas (15,2%, RR=19,78) eram mais prevalentes na aldeia Iriri (p<0,001). A biprotrusão, com consequente redução do ângulo nasolabial, era uma característica comum entre os Arara, porém com maior prevalência no grupo Iriri (RP=1,52, p=0,0002). A prevalência da má oclusão foi significativamente mais alta na aldeia Iriri (RP= 1,75, p=0,0007). A maioria da população da aldeia original (83,8%) apresentou uma relação normal entre os arcos dentários, contudo, na aldeia resultante (Iriri), 34,6% dos indivíduos era Classe III (RP=6,01, p<0,001) e 21,7% era Classe II (RP=2,02, p=0,05). Enquanto nenhum caso de apinhamento e de sobremordida foi observado na aldeia Iriri, a razão da prevalência era 2,64 vezes maior para a mordida aberta anterior (p=0,003), 2,83 vezes (p<0,001) para a mordida cruzada anterior, 3,93 (p=0,03) para a sobressaliência aumentada, e de 4,71 (p=0,02) para a mordida cruzada posterior. Observou-se uma alta prevalência das perdas dentárias, sem diferença entre as aldeias (RP=1,46, p=0,11). O exame dos modelos revelou uma tendência de incisivos maiores e pré-molares e caninos menores na aldeia Iriri, delineando uma semelhança na massa dentária total entre as aldeias, que, aliada a arcadas dentárias maiores, justificaram o menor índice de irregularidade dos incisivos entre esses indígenas. Esses resultados minimizam a influência do desgaste dentário, uma evidência direta de como um indivíduo se alimentou no passado, no desenvolvimento dentofacial e enfatizam o predomínio da hereditariedade, através da endogamia, na etiologia da variação anormal da oclusão dentária e da morfologia da face.

O estilo parental tem influência no desenvolvimento humano e pode ser definido como um padrão de comportamentos identificáveis nos pais quando se relacionam com seus filhos. A literatura tem utilizado alguns parâmetros para compreender os estilos parentais. Um deles se caracteriza pelo binômio: limites-afeto ou responsividade e exigência, delimitando três estilos parentais: Autoritário, Permissivo e Autoritativo. O estilo autoritário caracteriza-se por pais que dão muitos limites e pouco afeto. Estes pais procuram manter a ordem e o controle da família, devendo a obediência ser alcançada ainda que sob padrões punitivos. O estilo permissivo caracteriza-se por pais que dão pouco limite e muito afeto. Nesse caso, há uma crença naturalista segundo a qual a criança deve expressar livremente suas necessidades e aprender por si só. Estes pais não se vêem como responsáveis por ensinar ou modificar comportamentos infantis. O estilo autoritativo caracteriza-se por pais que dão muito limite e muito afeto. O cuidador apresenta condutas intermediárias entre as permissivas e autoritárias, estimulando verbalizações emocionais, permitindo que os filhos participem das decisões, estimulando argumentações, autonomia e disciplina. A empatia é considerada uma habilidade essencial para a manutenção dos laços sociais, que propicia a percepção da necessidade do outro e ao mesmo tempo favorece a sensação de ser compreendido. A assertividade é a habilidade de expressar e defender sua opinião com firmeza e segurança, sem, contudo desrespeitar o outro. Esta pesquisa tem como hipótese a idéia de que pais autoritativos seriam mais empáticos e assertivos do que pais autoritários. O objetivo deste estudo foi correlacionar os Estilos Parentais s Habilidades Sociais: Assertividade e Empatia. Participaram da pesquisa 64 pais, 21 homens e 43 mulheres. Os instrumentos utilizados foram: o Inventário de Habilidades Sociais (IHS-Del Prette), ao Inventario de Empatia (IE FALCONE) e ao Parental Autority Questionary (PAQ). A Análise dos dados foi feita através do SPSS. Foram efetuadas correlações r de Person entre os fatores do PAQ e os fatores do IHS e do IE. Os resultados apontaram para uma correlação positiva (r = 0,30 ; p < 0,05) entre o fator Enfrentamento e Auto-afirmação do IHS e o estilo Autoritativo. Uma relação negativa (r = -0,24 ; p < 0,05) foi observada entre este mesmo fator e o estilo Autoritário. Em relação a empatia, os fatores Tomada de Perspectiva (r = 0,33 ; p < 0,01), Sensibilidade Afetiva (r = 0,25 ; p < 0,05) e Autruismo (r = 0,25 ; p < 0,05) correlacionaram-se positivamente ao estilos Autoritativo, Em relação ao fator Autoritário, foi verificada uma correlação negativa ( r = -0,24 ; p < 0,05) com o fator Altruísmo do IE. A partir deste resultado, foram realizadas três análises de regressão linear múltipla (stepwise) considerando cada um dos fatores do PAQ como variáveis dependentes dos fatores do IHS e do IE. Os Fatores Enfrentamento e Auto-afirmação do IHS e o fator Autruísmo do IE junto explicam 15% da variância do Fator autoritativo. Os resultados apresentados são discutidos a luz da psicologia do desenvolvimento, psicologia social e neuropsicologia.

我国牡丹资源丰富、药用历史悠久,但长期以来,资源与药效关系不清严重阻碍着丹皮的产业化发展。建立一套综合评价体系,比较不同牡丹资源间的差异,科学评价、准确定位各地不同品种的利用价值,对稳定我国丹皮质量、可持续、集约化发展丹皮产业具有重要的意义。 本实验以我国丰富的药用牡丹资源为材料,从观赏和药用价值两方面首先探讨综合评价牡丹资源的体系。在观赏价值考察中重点比较了花型、花色、花期等性状;药用价值则从根的产量、重金属含量以及七种主要药效成分含量三方面来比较;重金属含量以原子分光光度法检测,药效成分则以高效液相色谱和质谱进行分离鉴定和定性、定量检测。利用建立的综合评价体系对野生种、各地药用品种进行了评价,对丹皮生产相关环节的问题进行了讨论,并初步探讨了药效成分的遗传规律。结果如下: 1、引种的野生牡丹药用价值不比栽培品种高,建议作为育种材料。 2、不同品种间药效成分含量存在显著差异,综合比较‘JSF’、‘CKL’、‘凤丹’适于药用栽培,‘赵粉’、‘朱砂垒’、‘首案红’等品种适宜观赏栽培,‘太平红’、‘JPH’等可花药兼用。 3、不同的土壤、气候环境对丹皮药效成分含量有明显的影响,丹皮中重金属含量主要与土壤环境关系密切。 4、根的粗细程度与药效成分含量关系不大,建议市场药材分级时充分考虑。 5、丹皮的药效成分含量与株龄、采收期、储藏时期关系密切;建议采收、应用根据各地的品种、气候条件及市场情况而定,在保证较高药效成分含量的前提下,尽量缩短生产、流通周期。 6、催花、嫁接对一些牡丹品种的药用价值影响不大,可合理开发利用。 7、药效成分的遗传有一定的规律,86%的子代丹皮酚含量高于父母本或亲本之一;丹皮酚原苷和没食子酰羟基芍药苷含量受父本影响较大,而芍药苷、苯甲酰芍药苷、丹皮酚新苷、没食子酰丹皮酚苷含量受母本影响较大。
