This study provides comprehensive documentation of silk production in the pest moth Helicoverpa armigera from gland secretion to extrusion of silk thread. The structure of the silk glands, accessory structures and extrusion apparatus are reported. The general schema of the paired silk glands follows that found for Lepidoptera. Morphology of the duct, silk press, muscle attachments and spigot are presented as a three-dimensional reconstruction and the cuticular crescent-shaped profile of the silk press is demonstrated in both open and closed forms with attendant muscle blocks, allowing advances in our knowledge of how the silk press functions to regulate the extrusion of silk. Growth of the spigot across instars is documented showing a distinctive developmental pattern for this extrusion device. Its shape and structure are related to use and load-bearing activity.

The genus Hymenaea is characterized by a great diversity of secretory structures, but there are no reports of colleters yet. The objectives of this study are to report the occurrence and describe the origin and structure of colleters in Hymenaea stigonocarpa Mart. ex Hayne. Shoot apex samples were collected, fixed, and processed for light microscopy, scanning electron microscopy, and transmission electron microscopy as per usual methods. Colleters occur predominantly on the stipule's adaxial side. These structures are found at the base on a narrow strip, corresponding to the median vein up to half the length of the stipule. When present on the abaxial side, they are concentrated at the base and restricted to the margins. Colleters develop from the protoderm; they are elongate and club-shaped. Their body has no stratification; their surface cells differ from the inner cells only in position and presence of cuticle. Colleter cells have thin walls, dense cytoplasm, large nuclei, many mitochondria, rough endoplasmic reticulum, and abundant dictyosomes. Histochemical tests with Ruthenium red showed pectic compounds in the cytosol. In H. stigonocarpa, colleter arrangement is compatible with the hypothesis that they protect shoot apex. In this species, protection is reinforced by the sheath formed by the stipule pairs.

Morphological and functional alterations caused by chronic alcohol ingestion in testes and accessory sex organs have been studied both in man and in laboratory animals. The aim of the present study was to examine the possible occurrence of deleterious effects of chronic alcohol ingestion on the secretory epithelium of the ventral prostate of mice. Twenty-four adult male C57BL/6J mice were divided into three groups. The alcohol-treated group was allowed to drink only 6% (v/v) ethanol, the isocaloric group received a diet of water/sucrose with a calorie content equivalent to a 6% alcohol solution and the control group received water. Both groups were fed ad libitum with solid Purina rat chow. After 120 days, animals from each group were anesthetized with ethyl ether, weighed and processed for light and transmission electron microscopy. The results demonstrated reduction in the glandular epithelium cell height and disorganization of the Golgi complex. Moreover, abundant membrane-bound structures, most likely representing cytoplasmic material, were observed, as well as accumulation of dense bodies. Statistical analysis showed that bodyweight gain was similar for both groups. In conclusion, chronic alcohol ingestion has harmful effects on the secretory epithelium cells of the ventral lobe of the prostate of mice after 120 days of treatment. Diabetes mellitus can lead to reproductive disorders that in turn result in weakened fertility brought about by morphofunctional changes in the testes and accessory sex glands. However, doubts persist concerning the basic biology of the secretory epithelial cells and the stroma of the coagulating gland of diabetic mice. Thus, the objective of the present study was to analyze the histological and ultrastructural changes associated with stereology of the coagulating gland of mice with alloxan-induced diabetes, and of spontaneously diabetic mice. Sixteen mice of the C57BL/6J strain, and eight non-obese diabetic (NOD) mice were used. The animals were divided into three groups: 1) control (C), 2) alloxan diabetic (AD), and 3) NOD. Thirty days after the detection of diabetic status in group 2, all of the animals were killed and then perfused with Karnovsky's solution through the left cardiac ventricle. The coagulating gland was then removed and processed for morphometric study by light microscopy and electron microscopy. The results showed thickening of the stroma, atrophy of secretory epithelial cells, and disorganization of the organelles involved in the secretory process in both NOD and alloxan-induced mice. Thus, it may be concluded that the coagulating gland suffered drastic morphological changes, and consequently impaired glandular function, in the presence of diabetes mellitus type I in both NOD and AD mice. The structure and ultrastructure of immature to fully mature glandular dots in the leaf, floral organs and fruit, and their secretion components were described in Caesalpinia echinata Lam. (Leguminosae) for the first time. Data showed that glandular dots were groups of idioblasts with contents that reacted positively for both lipophilic and hydrophilic substances. Idioblasts originated from successive divisions of the ground meristem cells or mesophyll cells of an ovary of a fertilized flower. Following division, cells enlarged, the cytoplasm became denser and its content became full. No idioblasts were observed after fruit sclerification. Besides these mixed-content idioblasts, some cells in the sepals, petals and mesocarp were found to contain phenolic compounds, which probably represent a kind of constitutive defense mechanism, once the flowers and fruits become highly fitness-valued parts of the plant and can be commonly attacked. The contents of the idioblasts are released as the growth rate of the embryo increases, indicating that the plant probably diverts the precursors of secondary metabolites into the primary metabolism, at this critical time of embryo development.

A incidência de doenças renais crônicas está aumentando no mundo, e há uma grande necessidade de identificar as terapias capazes de deter ou reduzir a progressão da doença. Há crescente evidência clínica e experimental de que as estatinas poderiam desempenhar um papel terapêutico. Recentes estudos clínicos e experimentais têm mostrado que as estatinas têm "efeitos pleiotrópicos", além da modulação lipídica. Estudos têm avaliado os efeitos das estatinas sobre a progressão da doença renal crônica, mas os resultados são controversos. Estudos ultra-estruturais em humanos e em ratos demonstraram a presença de junções GAP dentro de todas as células do glomérulo e os podocitos demonstraram conter principalmente conexina-43 (Cx-43). O presente estudo tem como objetivo observar os efeitos da rosuvastatina na estrutura e ultra-estrutura renal e a expressão glomerular de Cx-43 em ratos normotensos (WKY) e em ratos espontaneamente hipertensos (SHR). O foco do estudo foi avaliar os efeitos pleiotrópicos da rosuvastatina em rins de animais hipertensos normocolesterolêmicos. Os ratos foram divididos aleatoriamente em quatro grupos: WKY-C: animais normotensos que não receberam rosuvastatina; WKY-ROS: animais normotensos que receberam rosuvastatina 20mg/kg/dia por gavagem orogástrica; SHR-C: animais hipertensos que não receberam rosuvastatina; SHR-ROS: animais hipertensos que receberam rosuvastatina, como descrito no grupo WKY-ROS. Os animais dos grupos SHR-C e SHR-ROS apresentaram níveis de pressão arterial maiores que os animais dos grupos WKY-C e WKY-Ros. A massa corporal dos grupos de animais não diferiram significativamente durante o experimento. Não houve diferença nos níveis sanguíneos de uréia, creatinina, ácido úrico e creatinafosfoquinase entre os animas dos grupos estudados. No entanto, houve um aumento da excreção de proteína de 24 horas nos animais do grupo SHR-C. Houve um aumento na área capsular nos animais do grupo SHR-C. Por microscopia eletrônica de transmissão observou-se que nos animais SHR-C a barreira de filtração glomerular, o diafragma de fenda e os podócitos estão alterados exibindo os vacúolos nos podócitos e pedicelos mais curtos e mais espessos. Por microscopia eletrônica de varredura, os animais SHR-C exibiram pedicelos mais afilados, curtos e tortuosos. Um aumento da imunofluorescência para Cx-43 foi observada em células epiteliais viscerais dos glomérulos dos animais do grupo WKY-ROS e nas células parietais e viscerais dos glomérulos dos animais do grupo SHR-ROS, se comparado com os grupos WKY-C e SHR-C. Em conclusão, podemos supor que o efeito pleiotrópico renal da rosuvastatina pode ser uma ferramenta terapêutica para melhorar a estrutura e conseqüentemente a função renal em indivíduos hipertensos.

Desordens do sistema renal podem ser as causas da hipertensão arterial, a qual pode, por sua vez, causar doenças renais. A pressão sanguínea elevada é muito comum também nas doenças crônicas dos rins, e é, além disso, um conhecido fator de risco para uma mais rápida progressão da falha renal. A incidência de doenças renais crônicas está aumentando no mundo, e há uma grande necessidade de identificar as terapias capazes de deter ou reduzir a progressão da doença. Há crescente evidência de que as estatinas poderiam desempenhar um papel terapêutico. Além disso, tem sido demonstrado que a atividade física melhora a função renal em pacientes. Estudos ultra-estruturais em humanos e em ratos demonstraram a presença de junções gap dentro de todas as células do glomérulo e os podócitos demonstraram conter principalmente conexina-43 (Cx-43). O presente estudo tem como objetivo observar os efeitos da rosuvastatina e da atividade física de baixa intensidade na estrutura e ultra-estrutura renal e na expressão glomerular de Cx-43 em ratos normotensos (WKY) e em ratos espontaneamente hipertensos (SHR). Os ratos foram divididos aleatoriamente em oito grupos: WKY-C: animais normotensos que não receberam rosuvastatina; WKY-ROS: animais normotensos que receberam rosuvastatina 20mg/kg/dia por gavagem orogástrica; SHR-C: animais hipertensos que não receberam rosuvastatina; SHR-ROS: animais hipertensos que receberam rosuvastatina, como descrito no grupo WKY-ROS; SED-WKY: animais normotensos sedentários; EX-WKY: animais normotensos exercitados; SED-SHR: animais hipertensos sedentários; e, EX-SHR: animais hipertensos exercitados. Os animais dos grupos SHR-C, SHR-ROS e SED-SHR apresentaram níveis de pressão arterial maiores que os animais dos grupos WKY-C, WKY-ROS, SED-WKY, EX-WKY e EX-SHR. The female prostate has aroused scientific interest because it is subjected to the same diseases compromising the male prostate during aging. The objective of this work was to characterize structurally, cytochemically, and ultrastructurally the tissue compartments of the normal adult female prostate of Meriones unguiculatus gerbils. The morphological analyses showed that the gerbil's female prostate is constituted of a cluster of glands and ducts inserted in a musculofibrous stroma. The alveolar epithelium is differentiated and consisted of basal proliferating cells, intermediary cells, and secretory cells. The secretory cells are the most numerous cell type and continuously secrete glycoproteins. The basal cells are the source of the secretory cells and they are then responsible for the alveolus renovation. The prostatic stroma is abundant and rich in elastic and collagen fibers, which are closely associated with smooth muscle cells and fibroblasts. The results showed that the gerbil's female prostate shows morphological and ultrastructural homology to the human female prostate (Skene's gland), and despite being a small organ, it is a mature and physiologically active gland. Chiroptera, the second largest mammalian order, presents different reproductive strategies and unique reproductive features. However, there are few reports regarding male reproductive accessory glands (RAGs) in Chiroptera. Thus, the aim of the present study was to characterise the RAGs of the exclusively neotropical bat Artibeus planirostris (Chiroptera: Phyllostomidae) macroscopically, microscopically and ultrastructurally. The RAGs were composed of a prostatic complex with two regions (ventral and dorsal) and paraurethral and bulbourethral glands, but no seminal vesicles. The ventral region had an undefined epithelium, with secretory and basal cells, and its secretions were periodic acid-Schiff (PAS) positive. The dorsal region received both deferens ducts, had a columnar pseudostratified epithelium with secretory and basal cells. There were two types of secretions from the dorsal region: one that was basophilic and another that was mixed PAS positive and PAS negative. The paraurethral glands were dispersed in the connective tissue of the urethra, whereas the bulbourethral glands were located in the penile root. Histological and ultrastructural data confirmed the prostatic nature of the ventral and dorsal regions and the holocrine nature of the ventral region, with the latter finding never having been described previously for the prostate gland. Our findings demonstrate the wide discrepancy of RAGs between A. planirostris and other mammals in terms of their composition, structure and morphology. © CSIRO 2013.
