A Resolução CONAMA N 430/2011 exige a utilização de dois bioensaios (dois níveis tróficos) para avaliação ecotoxicológica de efluentes, mas a seleção ao acaso de bioensaios pode permitir lançamentos tóxicos. A sensibilidade dos bioindicadores irá depender da substância tóxica avaliada. Assim, baterias de bioensaios sensíveis devem ser estabelecidas às classes de contaminantes. Na literatura não há estudos que indiquem uma bateria de bioensaios ecotoxicológicos sensíveis para avaliação de efluentes contendo principalmente metais. Esse trabalho teve como objetivo selecionar uma bateria de bioensaios ecotoxicológicos que conjuntamente detectem toxicidade ao maior número de metais isolados e em misturas e que sejam realizados no menor tempo indicado pelas normas de padronização. Foram avaliadas as sensibilidades de seis bioensaios, incluindo três níveis tróficos (produtores, algas: Pseudokirchneriella subcapitata e Chlorella vulgaris; consumidores primários, cladóceros: Daphnia similis e Ceriodaphnia dubia; consumidores secundários, peixes: Poecilia reticulata e Danio rerio), a 10 espécies metálicas individuais (Ag+, Cd2+, Cu+, Cu2+, Cr3+, Cr6+, Pb2+, Ni2+, Zn2+ e Hg2+) e a efluentes reais (siderúrgicos) e simulados em laboratoriais (baseado nos limites máximos permitidos para descarte). Os bioensaios com peixes foram os menos sensíveis, D. rerio não detectou toxicidade em nenhum dos efluentes testados. P. subcapitata foi um bom bioindicador de toxicidade de Cr3+ e D. similis foi o organismo mais sensível a Hg2+. O uso combinado do bioensaio crônico de 72h com C. vulgaris e do bioensaio agudo de 48h com C. dubia garantiu a detecção das menores concentrações dos metais tanto individualmente quanto em efluentes reais e simulados. Apesar de P. subcapitata ser um bom bioindicador da toxicidade de Cr3+, a interação dos metais em misturas tornou C. vulgaris igualmente sensível. Da mesma forma, apesar de D. similis ter sido mais sensível ao Hg2+, o efeito da toxicidade dos efluentes com maiores teores de Hg2+ foi detectado por C. dubia

Neste trabalho foi avaliada a qualidade das águas da Bacia do Rio Morto, localizado na Baixada de Jacarepaguá Rio de Janeiro, com base em análise físicoquímicas e ensaios ecotoxicológicos agudos com Danio rerio, Daphnia similis e Aliivibrio fischeri e ensaios ecotoxicológicos crônicos referentes à reprodução com Daphnia similis. Foram comparadas as sensibilidades dos organismos-teste, que pertencem a níveis tróficos diferentes, nos quatro pontos selecionados para coleta de amostras de água no Rio Morto e seus principais tributários: Rio Branco, Rio Sacarrão e canal do Morro do Bruno. Além disso, foi implementado no laboratório o método de ensaio crônico com o microcrustáceo Daphnia similis. As amostras, em sua maioria, apresentaram parâmetros físico-químicos dentro dos limites permitidos pela legislação nacional para a classe de águas doces em que a Bacia estudada está inserida. Não foram observados efeitos agudos nos organismos-teste, não sendo possível o cálculo da CE50 ou CL50, por conseqüência, o FT ficou fixado em 1. No teste agudo para Aliivibrio fischeri, para algumas amostras, foi constatado efeito Hormesis. O mesmo foi verificado em algumas amostras submetidas aos testes crônicos com Daphnia similis.

Estudos em animais experimentais evidenciaram associações significativas entre esquistossomose mansoni e hipercolesterolemia. Estudos in vitro e in vivo já demonstraram que o colesterol é essencial para Schistosoma mansoni, embora este não tenha capacidade de sintetizá-lo. A captação é realizada a partir do ambiente (cultivo ou hospedeiro) através do tegumento. O colesterol está envolvido nos mecanismos de evasão do helminto contra a resposta imunológica, além de poder participar na modulação da sinalização celular e reprodução, estimulando os órgãos reprodutores dos helmintos adultos como observado na fase aguda da infecção experimental. Este trabalho tem como objetivo avaliar se o mesmo fenômeno ocorre na fase crônica. Os helmintos foram recuperados de dez camundongos submetidos à dieta hiperlipídica ou padrão (controle) foram corados pelo carmin cloridrico e montados, individualmente, em lâmina histológica com bálsamo do Canadá. A preparação foi analisada por microscopia de campo claro nos seguintes caracteres: tegumento e o sistema reprodutor nos vermes machos (lobos testiculares, vesícula seminal, lobos testiculares supranumerários e canal ginecóforo) e, nas fêmeas (ovário, oótipo, útero, ovo, glândulas vitelínicas e espermateca). Posteriormente, algumas lâminas foram separadas para visualização pela microscopia confocal dos órgãos do sistema reprodutores acima descritos. Apesar de ter sido observado uma maior quantidade de espermatozoides, uma maior quantidade de oócitos sendo liberados no grupo da dieta, não houve diferença estatística significativa (p>0,05) entre os grupos analisados. Houve um aumento na oogênese como observado na fase aguda. Dessa forma, o colesterol pode estar relacionado com a estimulação na atividade dos órgãos reprodutores dos helmintos adultos na fase crônica da infecção.

Karenia brevis is the dominant toxic red tide algal species in the Gulf of Mexico. It produces potent neurotoxins (brevetoxins [PbTxs]), which negatively impact human and animal health, local economies, and ecosystem function. Field measurements have shown that cellular brevetoxin contents vary from 1–68 pg/cell but the source of this variability is uncertain. Increases in cellular toxicity caused by nutrient-limitation and inter-strain differences have been observed in many algal species. This study examined the effect of P-limitation of growth rate on cellular toxin concentrations in five Karenia brevis strains from different geographic locations. Phosphorous was selected because of evidence for regional P-limitation of algal growth in the Gulf of Mexico. Depending on the isolate, P-limited cells had 2.3- to 7.3-fold higher PbTx per cell than P-replete cells. The percent of cellular carbon associated with brevetoxins (%C-PbTx) was ~ 0.7 to 2.1% in P-replete cells, but increased to 1.6–5% under P-limitation. Because PbTxs are potent anti-grazing compounds, this increased investment in PbTxs should enhance cellular survival during periods of nutrient-limited growth. The %C-PbTx was inversely related to the specific growth rate in both the nutrient-replete and P-limited cultures of all strains. This inverse relationship is consistent with an evolutionary tradeoff between carbon investment in PbTxs and other grazing defenses, and C investment in growth and reproduction. In aquatic environments where nutrient supply and grazing pressure often vary on different temporal and spatial scales, this tradeoff would be selectively advantageous as it would result in increased net population growth rates. The variation in PbTx/cell values observed in this study can account for the range of values observed in the field, including the highest values, which are not observed under N-limitation. These results suggest P-limitation is an important factor regulating cellular toxicity and adverse impacts during at least some K. brevis blooms.

This report contains a chemical and biological characterization of sediments from the St. Thomas East End Reserves (STEER) in St. Thomas, U.S. Virgin Islands (USVI). The STEER Management Plan (published in 2011) identified chemical contaminants and habitat loss as high or very high threats and called for a characterization of chemical contaminants as well as an assessment of their effects on natural resources. The baseline information contained in this report on chemical contaminants, toxicity and benthic infaunal community composition can be used to assess current conditions, as well as the efficacy of future restoration activities. In this phase of the project, 185 chemical contaminants, including a number of organic (e.g., hydrocarbons and pesticides) and inorganic (e.g., metals) compounds, were analyzed from 24 sites in the STEER. Sediments were also analyzed using a series of toxicity bioassays, including amphipod mortality, sea urchin fertilization impairment, and the cytochrome P450 Human Reporter Gene System (HRGS), along with a characterization of the benthic infaunal community. Higher levels of chemical contaminants were found in Mangrove Lagoon and Benner Bay in the western portion of the study area than in the eastern area. The concentrations of polychlorinated biphenyls (PCBs), DDT (dichlorodiphenyltrichloroethane), chlordane, zinc, copper, lead and mercury were above a NOAA sediment quality guideline at one or more sites, indicating impacts may be present in more sensitive species or life stages in the benthic environment. Copper at one site in Benner Bay, however, was above a NOAA guideline indicating that effects on benthic organisms were likely. The antifoulant boat hull ingredient tributyltin, or TBT, was found at the third highest concentration in the history of NOAA’s National Status and Trends (NS&T) Program, which monitors the Nation’s coastal and estuarine waters for chemical contaminants and bioeffects. Unfortunately, there do not appear to be any established sediment quality guidelines for TBT. Results of the bioassays indicated significant sediment toxicity in Mangrove Lagoon and Benner Bay using multiple tests. The benthic infaunal communities in Mangrove Lagoon and Benner Bay appeared severely diminished.

Ciguatoxins (CTX) are polyether neurotoxins that target voltage-gated sodium channels and are responsible for ciguatera, the most common fish-borne food poisoning in humans. This study characterizes the global transcriptional response of mouse liver to a symptomatic dose (0.26 ng/g) of the highly potent Pacific ciguatoxin-1 (P-CTX-1). At 1 h post-exposure 2.4% of features on a 44K whole genome array were differentially expressed (p ≤ 0.0001), increasing to 5.2% at 4 h and decreasing to 1.4% by 24 h post-CTX exposure. Data were filtered (|fold change| ≥ 1.5 and p ≤ 0.0001 in at least one time point) and a trend set of 1550 genes were used for further analysis. Early gene expression was likely influenced prominently by an acute 4°C decline in core body temperature by 1 h, which resolved by 8 h following exposure. An initial downregulation of 32 different solute carriers, many involved in sodium transport, was observed. Differential gene expression in pathways involving eicosanoid biosynthesis and cholesterol homeostasis was also noted. Cytochrome P450s (Cyps) were of particular interest due to their role in xenobiotic metabolism. Twenty-seven genes, mostly members of Cyp2 and Cyp4 families, showed significant changes in expression. Many Cyps underwent an initial downregulation at 1 h but were quickly and strongly upregulated at 4 and 24 h post-exposure. In addition to Cyps, increases in several glutathione S-transferases were observed, an indication that both phase I and phase II metabolic reactions are involved in the hepatic response to CTX in mice.

California sea lions have been a repeated subject of investigation for early life toxicity, which has been documented to occur with increasing frequency from late February through mid-May in association with organochlorine (PCB and DDT) poisoning and infectious disease in the 1970's and domoic acid poisoning in the last decade. The mass early life mortality events result from the concentrated breeding grounds and synchronization of reproduction over a 28 day post partum estrus cycle and 11 month in utero phase. This physiological synchronization is triggered by a decreasing photoperiod of 11.48 h/day that occurs approximately 90 days after conception at the major California breeding grounds. The photoperiod trigger activates implantation of embryos to proceed with development for the next 242 days until birth. Embryonic diapause is a selectable trait thought to optimize timing for food utilization and male migratory patterns; yet from the toxicological perspective presented here also serves to synchronize developmental toxicity of pulsed environmental events such as domoic acid poisoning. Research studies in laboratory animals have defined age-dependent neurotoxic effects during development and windows of susceptibility to domoic acid exposure. This review will evaluate experimental domoic acid neurotoxicity in developing rodents and, aided by comparative allometric projections, will analyze potential prenatal toxicity and exposure susceptibility in the California sea lion. This analysis should provide a useful tool to forecast fetal toxicity and understand the impact of fetal toxicity on adult disease of the California sea lion.

Phenyl-mercuric lactate is included in the pulp processing reagents by some paper mills to eliminate slime formation in the pulp. Small quantities of this chemical are added to the wet pulp in the beaters, particularly for the bactericidal action against Aerobacter aerogenes. Subsequently the mecurial is carried away in the wash waters. However, as the highly poisonous nature of many compounds of mercury is well known, questions have been raised concerning the pollution hazards created by phenyl-mercuric lactate in streams receiving effluents from mills using this substance.

A bioassay study was conducted using three organic pesticides to determine their comparative toxicity to fingerlings of Hypophthalmichthys molitrix. There was wide variation in the toxicity of different pesticides with 24 hr LC sub(50) values ranging from 0.000403 to 0.294 mg/l. Endosulfan appeared to be the most toxic, whereas BHC was the least.

Bioassay were carried out on 48h cultured nauplii of brine shrimp Artemia by exposing them to seven trace elements viz. copper (Cu), zinc (Zn), cadmium (Cd), nickel (Ni), lead (Pb), iron (Fe) and manganese (Mn). Synergistic effects of all these elements and additive effects of Cu and Zn, Cd and Pb, and Ni and Fe were also investigated. Comparatively, the degree of toxicity for compound bioassays was higher than individual simple tests. The values were averaged and the expected median lethal concentration [LC sub(50)] of tested heavy metals was obtained by probit analysis on the basis of cumulative numbers of dead organisms after 24 and 48h. The order of toxicity of the metals to Artemia was Pb>Cd>Cu>Ni>Zn>Fe>Mn. Potency ratios of the seven metals were also calculated. The 24 and 48h variations obtained in LC sub(50) values were significantly different and relative implications of these are discussed.

Some biochemical variations during ovarian vitellogenic growth in hill-stream teleost Garra mullya due to sublethal concentration of cadmium has been discussed. Total protein, cholesterol and glycogen in ovary and liver along with gonadosomatic index (GSI) and hepatosomatic index (HSI) in Cd-treated fish exhibited significant decrease while liver glycogen remained unaltered.

The acute toxic effect of the toxicant sumithion (50% E.C.) on mortality rate (after 24, 48, 72, and 96 h), total RBC count and haemoglobin content (after 48 and 72 h) on Heteropneustes fossilis was investigated at four concentrations (9.7, 10.7 and 11.1 ppm). The sumithion treated fishes showed lower RBC and Hb levels than the untreated ones. A gradual decrease in the total RBC counts and Hb contents was recorded with increasing concentration of toxicant after 72 h but the blood showed fluctuating values after 48 h of treatment.

The comparative toxicity was evaluated using four detergents, viz, linear alkylbenzene sulfonate (LAS), branched alkylbenzene sulfonate (BAS), sodium sulfonate (SS) and alfa-olefin sulfonate (AOS) on an estuarine fish, Ambassis commersonii, abundant in Kali estuarine system. Standard toxicity bioassay method was followed as per APHA (1980). AOS concentration in "Mega" soap was determined by the standard MBAS method described in APHA (1980). The results indicate that LAS was the most toxic detergent to fish relative to the other types and the other of toxicity was LAS > AOS > SS > BAS. A significant correlation was seen between observed and calculated mortalities for all the detergents. Some behavioural responses of fishes to all detergents were also observed.

The toxicity of methyl amine was studied by finding out its LC 50 values for Catla catla fingerlings. On the basis of LC 50 values, the harmless concentration of methyl amine was found to be 12.8 ppm. This indicates that methyl amine is fairly toxic to C. catla fingerlings and needs care for its disposal in aquatic environment.

Three new acylic diterpenoids belonging to the class of phytol series have been isolated. They were obtained from the ethyl acetate soluble fractions of four siphonaceous green seaweeds, Bryopsis pennata Lamour., Caulerpa taxifolia (Vahl) C. Ag., Codium decorticatum (Woodw.) Howe and Valoniopsis pachynema (Mart.) Børg., collected from Karachi coast of Pakistan. Structures of these compounds were elucidated with the help of spectroscopic methods and confirmed by comparison with the known compounds. Even the known compounds are being reported for the first time from a green algal source. All the compounds were found to display a strong toxicity at all the three concentrations tested in the brine shrimp bioassay.
