Knowledge of the temporal and spatial characteristics of chokka squid (Loligo vulgaris reynaudii) biology in South African waters is limited, so the possibility of there being a geographically fragmented stock was examined by investigating the distribution of maturity patterns for the species, covering all known spawning areas and using both historical and recent data. Gonadosomatic indices (GSI) varied between year-round consistency and apparent seasonal peaks in both summer and winter; there was no clear spatial pattern. Monthly percentage maturity provided further evidence for two peak reproductive periods each year, although mature squid were present throughout. Sex ratios demonstrated great variability between different areas and life history stages. Male-biased sex ratios were only apparent on the inshore spawning grounds and ranged between 1.118:1 and 4.267:1. Size at sexual maturity was also seasonal, squid maturing smaller in winter/spring than in summer/autumn. Also, squid in the east matured smaller than squid in the west. Although the results from the present study do not provide conclusive evidence of distinct geographic populations, squid likely spawn over a significantly larger area of the Agulhas Bank than previously estimated, and squid on the west coast of South Africa may return to spawn on the western portion of the Agulhas Bank. It remains likely, however, that the east and west coast populations are a single stock and that migration of juveniles to the west coast and their subsequent return as sub-adults is an integral but non-essential and variable part of the life history.

Table beet production in the Lockyer Valley of south-eastern Queensland is known to be adversely affected by soilborne root disease from infection by Pythium spp. However, little is known regarding the species or genotypes that are the causal agents of both pre- and post-emergence damping off. Based on RFLP analysis with HhaI, HinfI and MboI of the PCR amplified ITS region DNA from soil and diseased plant samples, the majority of 130 Pythium isolates could be grouped into three genotypes, designated LVP A, LVP B and LVP C. These groups comprised 43, 41 and 7% of all isolates, respectively. Deoxyribonucleic acid sequence analysis of the ITS region indicated that LVP A was a strain of Pythium aphanidermatum, with greater than 99% similarity to the corresponding P. aphanidermatum sequences from the publicly accessible databases. The DNA sequences from LVP B and LVP C were most closely related to P. ultimum and P. dissotocum, respectively. Lower frequencies of other distinct isolates with unique RFLP patterns were also obtained with high levels of similarity (>97%) to P. heterothallicum, P. periplocum and genotypes of P. ultimum other than LVP B. Inoculation trials of 1- and 4-week-old beet seedlings indicated that compared with isolates of the LVP B genotype, a higher frequency of LVP A isolates caused disease. Isolates with the LVP A, LVP B and LVP C genotypes were highly sensitive to the fungicide Ridomil MZ, which suppressed radial growth on V8 agar between approximately four and thirty fold at 5 μg/mL metalaxyl and 40 μg/mL mancozeb, a concentration far lower than the recommended field application rate.

The response of soybean (Glycine max) and dry bean (Phaseolus vulgaris) to feeding by Helicoverpa armigera during the pod-fill stage was studied in irrigated field cages over three seasons to determine the relationship between larval density and yield loss, and to develop economic injury levels. H. armigera intensity was calculated in Helicoverpa injury equivalent (HIE) units, where 1 HIE was the consumption of one larva from the start of the infestation period to pupation. In the dry bean experiment, yield loss occurred at a rate 6.00 ± 1.29 g/HIE while the rates of loss in the three soybean experiments were 4.39 ± 0.96 g/HIE, 3.70 ± 1.21 g/HIE and 2.12 ± 0.71 g/HIE. These three slopes were not statistically different (P > 0.05) and the pooled estimate of the rate of yield loss was 3.21 ± 0.55 g/HIE. The first soybean experiment also showed a split-line form of damage curve with a rate of yield loss of 26.27 ± 2.92 g/HIE beyond 8.0 HIE and a rapid decline to zero yield. In dry bean, H. armigera feeding reduced total and undamaged pod numbers by 4.10 ± 1.18 pods/HIE and 12.88 ± 1.57 pods/HIE respectively, while undamaged seed numbers were reduced by 35.64 ± 7.25 seeds/HIE. In soybean, total pod numbers were not affected by H. armigera infestation (out to 8.23 HIE in Experiment 1) but seed numbers (in Experiments 1 and 2) and the number of seeds/pod (in all experiments) were adversely affected. Seed size increased with increases in H. armigera density in two of the three soybean experiments, indicating plant compensatory responses to H. armigera feeding. Analysis of canopy pod profiles indicated that loss of pods occurred from the top of the plant downwards, but with an increase in pod numbers close to the ground at higher pest densities as the plant attempted to compensate for damage. Based on these results, the economic injury levels for H. armigera on dry bean and soybean are approximately 0.74 HIE and 2.31 HIE/m2, respectively (0.67 and 2.1 HIE/row-m for 91 cm rows).

The relative amounts of chloroplast tRNAs(Leu), tRNA(Glu), tRNA(Phe), tRNAs(Thr), and tRNA(Tyr) and of chloroplastic and cytoplasmic aminoacyl-tRNA synthetases were compared in green leaves, yellowing senescing leaves, and N(6)-benzyladenine-treated senescing leaves from bean (Phaseolus vulgaris, var Contender). Aminoacylation of the tRNAs using Escherichia coli aminoacyl-tRNA synthetases indicated that in senescing leaves the relative amount of chloroplast tRNA(Phe) was significantly lower than in green leaves. Senescing leaves treated with N(6)-benzyladenine contained higher levels of this tRNA than untreated senescing leaves. No significant change in the relative amounts of chloroplast tRNAs(Leu), tRNAs(Thr), and tRNA(Tyr) was detected in green, yellow senescing, or N(6)-benzyladine-treated senescing leaves. Relative levels of chloroplast tRNAs were also estimated by hybridization of tRNAs to DNA blots of gene specific probes. These experiments confirmed the results obtained by aminoacylation and revealed in addition that the relative level of chloroplast tRNA(Glu) is higher in senescing leaves than in green leaves. Transcription run-on assays indicated that these changes in tRNA levels are likely to be due to a differential rate of degradation rather than to a differential rate of transcription of the tRNA genes. Chloroplastic and cytoplasmic leucyl-, phenylalanyl-, and tyrosyl-tRNA synthetase activities were greatly reduced in senescing leaves as compared to green leaves, whereas N(6)-benzyladenine-treated senescing leaves contained higher enzyme activities than untreated senescing leaves. These results suggest that during senescence, as well as during senescence-retardation by cytokinins, changes in enzyme activities, such as aminoacyl-tRNA synthetases, rather than reduced levels of tRNAs, affect the translational capacity of chloroplasts.

Protein-energy malnutrition and mineral deficiencies are two of the three forms of nutritional deficiencies that affect most developing countries due to inadequate access to food and diets based on a sole crop. Common bean (Phaseolus vulgaris L.) is the staple crop of Nicaragua and it has the potential to improve the nutritional status of the poorest group of the nation. Its high content of both protein and nonhaem iron provides many nutrients, but inhibitors also may prevent absorption of iron and zinc by the human consumer. A proper production chain must be followed to ensure the best grain quality for the consumer. To achieve food security, both production and high nutritional content must be maintained. Four nationally important accessions of common bean, with different harvesting dates, were selected to be submitted to two treatments: to evaluate the impact of storage conditions on the end quality of the grain. The duration of the study was six months with sampling every six weeks, and the two treatments were controlled one stored at 40°C and 75 RH %, and the other was stored in in-situ conditions. Proximate and mineral composition was evaluated as well as tannin, phytate and bioavailability. Significant differences among different accessions were found, being the most significant in protein, Fe and Zn content, tannins and phytate. Protein values ranged from 21-23%. Iron content was 61-81 mg/kg but only 3-4% was bioavailable. Zinc content was 21-25 mg/kg and 10-12% was bioavailable. The concentration of phytate ranged from 8.6-9.6 mg/g while tannin values ranged within 37.7-43.8 mg/g. Storage at high temperatures was demonstrated to have an impact on certain nutritional compounds and proved detrimental to final grain quality. Soluble sugar content and tannin content decreased after six months in both storage conditions, IDF decreased in the in-situ and SDF in the stress. The iron content and bioavailability in INTA Biofortificado were not as outstanding as expected, so experiments should be conducted to compare its iron uptake and delivery with other cultivars.

Indole butyric acid (IBA) initiates roots in the hypocotyl tissue of Phaseolus vulgaris (French bean). The response is dependent on the concentration of IBA and the duration of exposure to the hormone. IBA enhances the rate of total protein synthesis in ca 30 min after exposure of the hypocotyl segments to the hormone. There is no detectable change in total or poly(A)-containing RNA synthesis in this period although significant increases are seen 2 hr after hormone pre-treatment. The early IBA-mediated increase in protein synthesis (30 min) is not sensitive to Actinomycin D but the antibiotic blocks the increase manifested 2 hr after hormone pre-treatment. Inhibition of early protein synthesis by cycloheximide depresses and delays root initiation. Cytosol prepared from IBA-treated hypocotyl tissue stimulates protein synthesis in vitro to a greater extent than that of the control.

Objective: In this study, we report the role of miRNAs involved under nitrogen starvation from widely grown vegetable crop, French bean. In recent years, a great deal of attention has been paid to the elucidation of miRNAs involved in low nitrate stress. Methods: To identify miRNAs expressed under stress, cDNA libraries were analyzed. Results: We reported the nine potential miRNAs with 67 targets involved in nutrient transporters and other stress specific genes. Among the miRNA sequences obtained 6 sequences belong to miR172 family, one with miR169. RT-PCR analysis of expression of miR172 family was induced upon low nitrate stress while miR169 family was repressed. In addition, Pvu-SN7b and Pvu-miR16 may be new members of miRNA172 and miR169 families, respectively. Conclusion: The targets of Pvu-SN7b were major protein kinases, one among which is the Protein Kinase CK2. CK2 Kinase is found to involve in transcription-directed signaling, gene control and cell-cycle regulation. Other targets of Pvu-SN7b were involved in DNA-dependent transcription regulation, photo-periodism, calcium-mediated signaling. Pvu-miR16 targets Thymidine kinase, the key enzyme of deoxy-nucleotide synthesis. The cleavage of these targets affects cell proliferation there by affecting nodule formation. Pvu-miR8 inhibits translation of its target protein Pre-protein translocase, a membrane-bound protein transporter involved in trans-membrane protein transportation. Together these results denote the response and role of miRNAs to nitrate-limiting conditions in French bean.

El experimento fue realizado con el objetivo de evaluar el efecto de aplicaciones de fungicidas (Benomyl + Mancozeb) durante diferentes etapas de desarrollo del cultivo de frijol, sobre la calidad final de la semilla producida. Se utilizó la variedad mejorada (DOR-364) y los tratamientos fueron 4 frecuencias de aplicación (0,1,2,3,4) a partir de la etapa V3 (20 días después de la siembra) hasta la etapa R8 (55 días después de la siembra), más un testigo sin aplicación. El experimento se desarrolló en época de postrera Octubre 1993, en la región 1V, finca experimental "La Compañía" en el departamento de Carazo. En el campo se registró la severidad de Mustia hilachosa (Thanatephorua cucumeris) en 2 etapas 40 y 55 días después de la siembra, observándose niveles de infección que van desde 7.5 hasta 27% para el testigo y niveles inferiores para loe tratamientos que van desde 3.5 hasta 9.5% para la mayor frecuencia de aplicación. Al realizar el análisis de calidad de la semilla cosechada se observó que los tratamientos no tuvieron ningún efecto diferencial en relación a su germinación, velocidad de emergencia y componentes de rendimiento. Sin embargo se pudo apreciar en el análisis de estado sanitario mayor porcentaje de infección generada por hongos en el testigo que en los tratamientos aplicados.

Este trabajo experimental fue llevado a cabo con el objetivo de evaluar crecimiento, desarrollo y rendimiento del cultivo del café (Coffea arábica L.) sembrando frijol (Phasheolus vulgaris L.) en las calles de café, al mismo tiempo determinar el efecto que tengan las malezas de dicho cultivo sobre el café, y que el frijol sirva de cobertura viva para el café y coma fuente de ingreso económico y alimenticio para el agricultor. El trabajo fue realizado en el Centro Experimental de café del Pacifico (C.E.C.P.) jardín Botánico (Masatepe) durante las dos épocas de siembra para el frijol, y la cosecha anual de café (Dic-Ene) de 1992. Este ensayo se estableció con las variedades. Rev-81 para frijol y catuai amarillo para el café, este tiene una edad de 5 años. El frijol fue sembrado en las calles de café de manera tradicional con el propósito de aprovechar esa área disponible. No hubo diferencia estadística significativa en cuanto al crecimiento, desarrollo y rendimiento del café, por lo que se deduce que el frijol (P. vulgaris) no ejerce efecto negativo alguno sobre el café, lo cual indica que el cultivo en asocio no perjudica la cosecha del café por lo que se recomienda llevar a cabo esta práctica en los primeros alias de edad del café ya que no perjudica el espaciamiento de siembra.

Este estudio fue realizado en la estación experimental "La Compañía", ubicada en el municipio de San Marcos, departamento de Carazo (IV región de Nicaragua). Los objetivos que se evaluaron fueron: (1) determinar cuál de los tres sistemas de labranza (Cero, mínima y convencional) es económicamente más rentable dentro de cada uno de los cultivos (frijol y maíz) y épocas (primera y postrera). (2) determinar cuál de los sistemas de labranza es más rentable dado el efecto de la rotación (maíz-frijol y frijol-frijol) • El experimento fue establecido en la época de primera (mayo-junio) y la de postrera (septiembre-diciembre). El experimento se organizó en un diseño de Bloque Completo al Azar (BCA) en arreglo de parcelas divididas con tres tratamientos y cuatro repeticiones. Se tomaron datos sobre los costos agrícolas de cada uno de los tratamientos y sus respectivos rendimientos. Se analizaron mediante el análisis de presupuestos parciales incluyendo el análisis de retorno mínimo y sensibilidad de los precios del producto final. El análisis económico mostró que en la época de primera en el cultivo de maíz y en la época de postrera bajo cualquier de los dos cultivos la labranza mínima resultó ser el mejor sistema. Mientras tanto, en la época de primera en el cultivo de frijol el tratamiento recomendado fue la labranza cero.

En el Centro Experimental de Café del Pacífico Central Jardín Botánico, municipio de Masatepe, departamento de Masaya" se realizó en los meses de Junio a Diciembre 1991, el presente trabajo con el objetivo de determinar el efecto del frijol común Phaseolus vulgaris L. como cultivo intercalado sobre el crecimiento y rendimiento del café coffea arabica L. y las malezas. Se usó un diseño de bloques completos al azar (B.C.A.) con cuatro tratamientos y cuatro repeticiones: Frijol en primera (Junio - Agosto), frijol en postrera (Septiembre - Noviembre) frijol en primera más postrera (Junio - Agosto, Septiembre - Noviembre) y sin frijol (testigo) dando como respuesta que el frijol común como cobertura en las calles de café tuvo influencia sobre la abundancia y dominancia de las malezas, y con respecto al cafeto, los resultados obtenidos en los tratamientos en estudio mostraron que no existe diferencias significativas estadísticamente en la influencia del frijol común sobre todas las variables estudiadas en el cafeto. El cultivo intercalado del frijol común no influyó en la producción del café; ejerció control sobre las malezas y presentó rendimiento adicional en la misma área donde se encuentra el cultivo del café.

En el ciclo de postrera del año 1992 fueron sembradas ocho parcelas para producción de semilla de cuatro lotes propiedad de igual número de pequeños productores de la comarca Pio XII, Masatepe. Cuatro parcelas fueron sometidas al manejo tradicional de los productores participantes (Manejo I), y las otras cuatro sometidas a un manejo diferente con el fin de producir semillas de calidad (Manejo II). A los lotes de semillas obtenidos se les hizo Prueba Analítica, Prueba de Humedad, Determinación de Daño Mecánico, Peso de 1000 Semillas, Pruebas de Germinación, Vigor y Materia Seca de Plántulas y, Pruebas de Sanidad. Ambos manejos fueron comparados mediante los resultados de estas pruebas, los que indicaron que la semilla de frijol producidad por los productores participantes fue de menor calidad que la producida bajo el Manejo II; en el primer caso, los porcentajes de Daños Mecánico y de infección por hongos fueron mayores. Los porcentajes de germinación y vigo resultaron menores en el mismo caso. En el Manejo II resultó superior el peso de las semillas respecto al contenido de materia seca de las plántulas, los lotes A y C del manejo II resultaron con valores menores. Mediante la Prueba de Humedad se encontró que la semilla manejada por los productores posee alto contenido de agua al momento de almacenarla, lo que hace que ésta se deteriore más fácilmente. Con los resultados obtenidos en el presente trabajo se puede decir que es posible producir semilla de calidad por parte de los pequeños productores y, dado el interés mostrado por éstos, se recomiendad dar continuidad a este tipo de trabajos.
