Utilizou-se o músculo Iliotibialis lateralis da sobrecoxa da ema (Rhea americana) para a realização das análises de estabilidade da carne sob armazenamento em refrigeração e congelamento. Na avaliação da carga microbiológica da carne foram realizadas análises de Coliformes Totais, Coliformes Fecais (Escherichia coli) e psicrotróficos. No período de zero a 12 dias de refrigeração determinou-se o pH da carne e a degradação protéica através da análise de Nitrogênio Volátil Total (NVT). Durante o congelamento (período de 60 dias) avaliou-se o pH e a oxidação lipídica pela análise do número de TBA da carne armazenada. As amostras sob refrigeração e congelamento mostraram-se adequadas ao consumo, não apresentando contaminação microbiológica durante todo o período de estocagem. Os valores de Nitrogênio Volátil Total (NVT) da carne sob refrigeração tiveram uma evolução insignificante (4%) entre o tempo zero e o 12º dia de estocagem. Durante o congelamento ocorreu um aumento significativo do número de TBA nas amostras, com variações de 1,5-2,5 mg de SRATB/1.000 g amostra. Os valores de pH da carne de ema armazenada sob refrigeração e congelamento mantiveram-se praticamente estáveis em torno de 5,8.

The free form of the iron ion is one of the strongest oxidizing agents in the cellular environment. The effect of iron at different concentrations (0, 1, 5, 10, 50, and 100 µM Fe3+) on the normal human red blood cell (RBC) antioxidant system was evaluated in vitro by measuring total (GSH) and oxidized (GSSG) glutathione levels, and superoxide dismutase (SOD), catalase, glutathione peroxidase (GSH-Px) and reductase (GSH-Rd) activities. Membrane lipid peroxidation was assessed by measuring thiobarbituric acid reactive substance (TBARS). The RBC were incubated with colloidal iron hydroxide and phosphate-buffered saline, pH 7.45, at 37oC, for 60 min. For each assay, the results for the control group were: a) GSH = 3.52 ± 0.27 µM/g Hb; b) GSSG = 0.17 ± 0.03 µM/g Hb; c) GSH-Px = 19.60 ± 1.96 IU/g Hb; d) GSH-Rd = 3.13 ± 0.17 IU/g Hb; e) catalase = 394.9 ± 22.8 IU/g Hb; f) SOD = 5981 ± 375 IU/g Hb. The addition of 1 to 100 µM Fe3+ had no effect on the parameters analyzed. No change in TBARS levels was detected at any of the iron concentrations studied. Oxidative stress, measured by GSH kinetics over time, occurs when the RBC are incubated with colloidal iron hydroxide at concentrations higher than 10 µM of Fe3+. Overall, these results show that the intact human RBC is prone to oxidative stress when exposed to Fe3+ and that the RBC has a potent antioxidant system that can minimize the potential damage caused by acute exposure to a colloidal iron hydroxide in vitro.

O presente trabalho teve por objetivo a produção de farinha de carne a partir das vísceras do jacaré do pantanal (Caiman crocodilus yacare) e o estudo das propriedades físico-químicas, bioquímicas e bacteriológicas da farinha elaborada. Essas características que servem de parâmetro de controle de qualidade, são normalmente exigidas na comercialização de farinhas de carne elaboradas a partir de vísceras e outros materiais de descarte para animais de consumo doméstico. Os resultados obtidos do pH, acidez solúvel, composição percentual dos componentes, dos minerais Ca e P, digestibilidade em pepsina, aspecto microbiológico (presença de salmonela) e estabilidade avaliada pelo nº de TBARS da farinha mostram ser viável o uso das vísceras do jacaré do pantanal como fonte alternativa de nutrientes para incorporação nas formulações de ração animal .

OBJETIVO: Avaliar a reprodutibilidade de dois modelos experimentais de isquemia e reperfusão cerebral. MÉTODOS: 60 ratos foram distribuídos, aleatoriamente, em três grupos experimentais, com 20 animais cada: I - pinçamento temporário de artéria carótida esquerda; II - cauterização prévia das artérias vertebrais e pinçamento temporário da artéria carótida esquerda; simulado - sem isquemia nem reperfusão. Todos os animais tiveram oclusão definitiva de artéria carótida direita e os três grupos foram subdivididos em dois períodos de reperfusão: A - 60 minutos e B - 120 minutos. Os parâmetros verificados foram: medidas de pressão arterial média sistêmica e fluxo sangüíneo carotídeo; medida de malondialdeído cerebral através do teste TBARS e avaliação histológica do hemisfério cerebral submetido à isquemia e reperfusão. Foi feito também um estudo complementar com angiografia cerebral em 5 animais adicionais. RESULTADOS: Não houve diferenças significativas nas dosagens de malondialdeído cerebral e na freqüência e gravidade das alterações histológicas cerebrais entre os três grupos. Nos grupos GI e GII, a PAM foi significantemente maior no período de isquemia. O fluxo sangüíneo entre os períodos pré e pós-pinçamento aumentou nos grupos IA e IIB, diminuiu no grupo IB e no grupo IIA manteve-se inalterado. As angiografias do estudo complementar mostraram aporte sangüíneo para cérebro através de circulação colateral. CONCLUSÃO: Os modelos de isquemia e reperfusão estudados não demonstraram alterações consistentes de marcadores de lesão cerebral, seja quanto à produção de lipoperóxidos ou de lesões histológicas.

In this study, the influence of the addition of antioxidants in vivo on the fatty acid composition of the flesh of a freshwater fish known as pacu (Piaractus mesopotamicus) is verified. Four groups (one being the control group) of juvenile pacu were cultured on isocaloric and isoproteic diets. The lipid source was soybean oil and diets were added with either 100 ppm of alpha-tocopheryl acetate, or 100 ppm of BHT or 1.4 g of rosemary extract (Herbalox(R))/kg diet. The fatty acid composition of the lipids of the different groups was determined before and after irradiation at 2 and 3 kGy, respectively, for the evaluation of the protective effects of the different antioxidants. Similarly, thiobarbituric acid reactive substances (TBARS) were determined from irradiated and nonirradiated samples. The results showed that the use of antioxidants altered the fatty acid composition of the fillets. TEARS and irradiation confirmed their important role in protecting against lipid oxidation. Among all the antioxidants used, tocopherol was the most efficient, as shown by the highest percentage of polyunsaturated fatty acids (PUFA), by the lowest values of TEARS and by the analyses of the individual fatty acid levels at different irradiation doses. Salting have been a very utilized fish conservation method, however only in the last years the basic mechanism involved in salting fish has been understood. The objectives of this study were determine the addition in brine salt of rosemary leaves, rosemary extracts and tocopherol, and your action in the followed parameters: water activity (Aw), moisture, ash, salt content and TBARS. The results showed that the addition of antioxidants was difficulted the salt absorption, however didn't have differences between rosemary or tocopherol use. In the salting time of 3 hours the values of Aw and salt levels, was respectively: 0.77±0.01 and 14.42±1.69. for control treatment; 0.85±0.02 and 9.09±1.39for rosemary filtrate; 0.78±0.03 and 10.63±0.69 rosemary without filtrating and 0.85±0.02 and 11.96±1.78 tocopherol, showed that didn't grow indigenous bacterias. Lipid oxidation was evaluated by TBARS and the results showed the oxidative effect of salt and the pro oxidant effects of alls antioxidants used in brine salting.

Protein malnutrition leads to functional impairment in several organs, which is not fully restored with nutritional recovery. Little is known about the role of oxidative stress in the genesis of these alterations. This study was designed to assess the sensitivity of blood oxidative stress biomarkers to a dietary protein restriction. Male Wistar rats were divided into two groups, according to the diet fed from weaning (21 days) to 60 day old: normal protein (17% protein) and low protein (6% protein). Serum protein, albumin, free fatty acid and liver glycogen and lipids were evaluated to assess the nutritional status. Blood glutathione reductase (GR) and catalase (CAT) activities, plasma total sulfhydryl groups concentration (TSG) as well as plasma thiobarbituric acid reactive substances (TBARs) and reactive carbonyl derivatives (RCD) were measured as biomarkers of the antioxidant system and oxidative damage, respectively. The glucose metabolism in soleus muscle was also evaluated as an index of stress severity imposed to muscular mass by protein malnutrition. No difference was observed in muscle glucose metabolism or plasma RCD concentration between both groups. However, our results showed that the low protein group had higher plasma TBARs (62%) concentration and lower TSG (44%) concentration than control group, indicating increased reactive oxygen species production in low protein group. The enhancement of erythrocyte GR (29%) and CAT (28%) activities in this group also suggest an adaptation to the stress generated by the protein deficiency. Taken together, the results presented here show that the biomarkers used were able to reflect the oxidative stress level induced by this specific protein deficient diet.

Quercetin, a typical bioflavonoid ubiquitously present in fruits and vegetables, is considered to be helpful for human health. Cisplatin (cDDP) is one of the most active cytotoxic agents in the treatment of a wide range of solid tumors. The aim of this study was to investigate the possible effect of quercetin, a bioflavonoid with antioxidant potential, on cisplatin-induced nophrotoxicity and lipid peroxidation in rats. Gavage administrations of water, propylene glycol and quercetin (50 mg/kg) were made 24 and 1 h before saline or cDDP (5 mg/kg) ip injections and were repeated daily for 2, 5 or 20 subsequent days. Rats were killed 2, 5 and 20 days after ip injections, and blood and urine samples were collected to determine plasma creatinine, urine volume and osmolality. The kidneys were removed to determine the levels of thiobarbituric acid-reactive substances (TBARS) and for histological studies. Cisplatin increased lipid peroxidation, urine volume and plasma creatinine levels and decreased urine osmolality. Treatment with quercetin attenuated these alterations. These results demonstrate the role of oxidative stress and suggest a protective effect of quercetin on cisplatin-induced nephrotoxicity in adult Wistar rats.
