Background: Podosphaera aphanis, the causal agent of strawberry powdery mildew causes significant economic loss worldwide. Methods: We used the diploid strawberry species Fragaria vesca as a model to study plant pathogen interactions. RNA-seq was employed to generate a transcriptome dataset from two accessions, F. vesca ssp. vesca Hawaii 4 (HW) and F. vesca f. semperflorens Yellow Wonder 5AF7 (YW) at 1 d (1 DAI) and 8 d (8 DAI) after infection. Results: Of the total reads identified about 999 million (92%) mapped to the F. vesca genome. These transcripts were derived from a total of 23,470 and 23,464 genes in HW and YW, respectively from the three time points (control, 1 and 8 DAI). Analysis identified 1,567, 1,846 and 1,145 up-regulated genes between control and 1 DAI, control and 8 DAI, and 1 and 8 DAI, respectively in HW. Similarly, 1,336, 1,619 and 968 genes were up-regulated in YW. Also 646, 1,098 and 624 down-regulated genes were identified in HW, while 571, 754 and 627 genes were down-regulated in YW between all three time points, respectively. Conclusion: Investigation of differentially expressed genes (log2 fold changes �5) between control and 1 DAI in both HW and YW identified a large number of genes related to secondary metabolism, signal transduction; transcriptional regulation and disease resistance were highly expressed. These included flavonoid 3´-monooxygenase, peroxidase 15, glucan endo-1,3-β-glucosidase 2, receptor-like kinases, transcription factors, germin-like proteins, F-box proteins, NB-ARC and NBS-LRR proteins. This is the first application of RNA-seq to any pathogen interaction in strawberry

Background and aims: To form nitrogen-fixing nodules on pea roots, Rhizobium leguminosarum biovar viciae must be competitive in the rhizosphere. Our aim was to identify genes important for rhizosphere fitness. Methods: Signature-tagged mutants were screened using microarrays to identify mutants reduced for growth in pea rhizospheres. Candidate mutants were assessed relative to controls for growth in minimal medium, growth in pea rhizospheres and for infection of peas in mixed inoculants. Mutated genes were identified by DNA sequencing and confirmed by transduction. Results: Of 5508 signature-tagged mutants, microarrays implicated 50 as having decreased rhizosphere fitness. Growth tests identified six mutants with rhizosphere-specific phenotypes. The mutation in one of the genes (araE) was in an arabinose catabolism operon and blocked growth on arabinose. The mutation in another gene (pcaM), encoding a predicted solute binding protein for protocatechuate and hydroxybenzoate uptake, decreased growth on protocatechuate. Both mutants were decreased for nodule infection competitiveness with mixed inoculants, but nodulated peas normally when inoculated alone. Other mutants with similar phenotypes had mutations predicted to affect secondary metabolism. Conclusions: Catabolism of arabinose and protocatechuate in the pea rhizosphere is important for competitiveness of R.l. viciae. Other genes predicted to be involved in secondary metabolism are also important.

Transcription factors of the plant-specific apetala2/ethylene response factor (AP2/ERF) family control plant secondary metabolism, often as part of signalling cascades induced by jasmonate (JA) or other elicitors. Here, we functionally characterized the JA-inducible tobacco (Nicotiana tabacum) AP2/ERF factor ORC1, one of the members of the NIC2-locus ERFs that control nicotine biosynthesis and a close homologue of ORCA3, a transcriptional activator of alkaloid biosynthesis in Catharanthus roseus. ORC1 positively regulated the transcription of several structural genes coding for the enzymes involved in nicotine biosynthesis. Accordingly, overexpression of ORC1 was sufficient to stimulate alkaloid biosynthesis in tobacco plants and tree tobacco (Nicotiana glauca) root cultures. In contrast to ORCA3 in C. roseus, which needs only the GCC motif in the promoters of the alkaloid synthesis genes to induce their expression, ORC1 required the presence of both GCC-motif and G-box elements in the promoters of the tobacco nicotine biosynthesis genes for maximum transactivation. Correspondingly, combined application with the JA-inducible Nicotiana basic helix–loop–helix (bHLH) factors that bind the G-box element in these promoters enhanced ORC1 action. Conversely, overaccumulation of JAZ repressor proteins that block bHLH activity reduced ORC1 functionality. Finally, the activity of both ORC1 and bHLH proteins was post-translationally upregulated by a JA-modulated phosphorylation cascade, in which a specific mitogen-activated protein kinase kinase, JA-factor stimulating MAPKK1 (JAM1), was identified. This study highlights the complexity of the molecular machinery involved in the regulation of tobacco alkaloid biosynthesis and provides mechanistic insights about its transcriptional regulators.

Oilseeds are a high-value natural resource, due to its use as a substitute for petroleum. However, the storage time can reduce seed viability and oil quality. Therefore, scientific efforts have been made to provide a increment of storage time, germination rates and plant establishment of high-value oilseeds. The seedling establishment depends of the plant pass over the functional transition stage, characterized by a metabolic change from heterotrophic condition to autotrophic one. The storage oil mobilization is performed by β-oxidation process and the glyoxylate cycle. Also, the functional transition involves acclimation to photosynthetic condition, which generally includes the participation of antioxidant system and the reactive oxygen species, the latter are produced in various reactions of primary and secondary metabolism. In the present study, Catalase was inhibited during the functional transition of sunflower and safflower, after were performed many analyzes to elucidate the effects caused on the SOD and APX antioxidant systems. Also, were checked the changes in expression pattern of the glyoxylate cycle enzymes markers, ICL and MLS. It was observed that after CAT inhibition, the SOD and APX antioxidant systems allow the seedling establishment. Besides, was verified that both oilseeds can be accelerate the reverse mobilization and the photosynthetic establishment when Catalase activity has dramatically decreased

The increased rate of sugarcane harvest without previous burn has provided a very favorable environment to the froghopper Mahanarva fimbriolota (Stal, 1854), with high Moisture and low temperature variation. Few works have Studied the response of sugarcane to this pest, so little is known about resistant cultivars. Plant phenolics are widely studied compounds because of their known antifierbivore effect. This research aims to determine if the attack of M.fimbriolata nymphs stimulates the acccumulation of total phenolics in sugarcane. The experiment was carried Out in greenhouse and arranged in completely randomized design, in a 3 x 2 x 4 factorial with three replications. Second instar nymphs of M. fimbriolota were infested at the following rates: control, 2-4 and 4-8 nymphs per pot (first-second infestations, respectively). Pots were covered with nylon net and monitored daily to isolate the effect of leaf Sucking adults. Leaf and root samples were collected and kept frozen in liquid nitrogen until analyses. Infested plants showed higher levels of phenolics in both root and leaf tissues. In roots, the cultivar SP80-1816 accumulated more phenolic compounds in response to the infestation of M. fimbriolata. on the other hand, higher levels were found in leaves and roots of control plants of SP86-42, which might be an indication of a non-preference mechanism. The increase of total phenolics in sugarcane infested with root-sucking froghopper nymphs does not seem to be useful to detect the resistance to this pest.

O bloqueio parcial das rotas onde atuam os herbicidas, com uso de baixas doses, pode ter implicações importantes, como a alteração do balanço de processos metabólicos nas plantas. Assim, foi conduzido no ano agrícola 2006/2007 um experimento em cana-soca de segundo corte na Fazenda Jurema, pertencente ao grupo COSAN, município de Barra Bonita-SP, para verificar os efeitos do glyphosate e do sulfumeturon-methyl, em subdoses, no comportamento fisiológico da cana-de-açúcar pelos níveis de clorofila e carotenoides. Os tratamentos constituíram-se da aplicação de dois herbicidas: sulfumeturon-methyl (Curavial 360 e.a. kg-1) e glyphosate (Roundup 480 i.a. kg-1), isolados e em misturas, em diferentes doses, e um tratamento controle, sem a aplicação dos herbicidas. As doses utilizadas foram: glyphosate 200 mL p.c. ha-1; glyphosate 400 mL p.c. ha -1; glyphosate 200 mL p.c. ha -1 + 10 g p.c. ha-1 de sulfumeturon-methyl; glyphosate 150 mL p.c. ha -1 + 12 g p.c. ha -1 de sulfumeturon-methyl; e sulfumeturon-methyl 20 g p.c. ha -1. O delineamento experimental utilizado foi o de blocos casualizados, com quatro repetições. As avaliações foram realizadas 15 e 30 dias após o plantio (DAP) e 30, 60, 90, 120, 150 e 180 dias após a colheita (DAC). As folhas foram cortadas padronizando-se o mesmo peso e área foliar. Para determinação do conteúdo de clorofila e carotenoides, amostras de 0,2 g de tecido foliar fresco foram preparadas e os extratos filtrados, sendo efetuadas leituras em espectrofotômetro (663 e 645 nm para clorofilas a e b, respectivamente). A aplicação de glyphosate e sulfumeturon-methyl nas maiores doses interferiu no conteúdo de carotenoides quando estes foram comparados com a testemunha. A maior dose de glyphosate diminuiu significativamente o conteúdo de clorofilas e carotenoides na cana-de-açúcar, porém esse resultado não se manteve quando a dose foi reduzida para 200 mL p.c. ha-1 . Os teores de clorofila foram inversamente proporcionais aos níveis Fe. A aplicação de sulfumeturon-methyl não interferiu nos teores de clorofila, no entanto os níveis de carotenoides se mostraram mais sensíveis e seus teores reduzidos. As alterações observadas nos níveis de clorofilas e carotenoides pela aplicação dos produtos podem afetar de maneira distinta o metabolismo da fotossíntese pela absorção e/ou conversão de energia.

O objetivo deste trabalho foi avaliar os teores de lignina e celulose em plantas de cana-de-açúcar após a aplicação de dois maturadores para a colheita. O experimento foi conduzido em uma área de cana-soca, cultivar SP 803280, no município de Igaraçu do Tietê/SP. O delineamento experimental utilizado foi o de blocos casualizados, com quatro repetições. Os tratamentos constituíram-se da aplicação de dois maturadores: sulfometuron-methyl (Curavial) e glyphosate (Roundup original). As doses utilizadas foram: glyphosate a 72 g e.a. ha-1; glyphosate a 144 g e.a. ha-1 ; glyphosate a 72 g e.a. ha-1 + sulfometuron methyl a 10 g p.c. ha-1; glyphosate a 108 g e.a. ha-1 + sulfometuron-methyl a 12 g p.c. ha-1; sulfometuron-methyl a 20 g p.c. ha-1; e a testemunha sem aplicação de maturadores. As análises de lignina e celulose foram realizadas pelo método lignina em detergente ácido modificado. O glyphosate e o sulfometuron-methyl alteraram os níveis de lignina no momento da colheita, e esse efeito foi observado também durante o crescimento da cana-de-açúcar (meses após a aplicação desses produtos). O glyphosate a 72 g e.a. ha-1 promoveu reduções nos teores de lignina, na colheita e durante o crescimento da cana-de-açúcar, quando comparados com os da testemunha, enquanto o sulfometuron-methyl isolado na menor dose (10 g ha-1) promoveu aumento nos teores desse biopolímero na soqueira da cana-de-açúcar.

Este trabalho teve como objetivo verificar se o horário de colheita da erva-cidreira brasileira [Lippia alba (Mill.) N. E. Br.], fenotipo carvona-limoneno, tem influência sobre a produção de massa foliar, rendimento e composição do óleo essencial. Foram avaliados cinco horários de colheita quando a cultura estava com 145 dias desde o transplante: 8:00, 10:00, 12:00, 14:00 e 16:00 h com cinco repetições, distribuídos em blocos casualizados. O experimento foi conduzido na Fazenda Experimental Lageado da FCA-UNESP/Botucatu no Setor de Horticultura do Departamento de Produção Vegetal. A colheita foi realizada a 15 cm da superfície do solo e o óleo essencial obtido através de hidrodestilação, em aparelho tipo Clevenger. Levou-se em consideração os fatores agronômicos e o rendimento obtido por hidrodestilação. O óleo essencial foi analisado em cromatógrafo gasoso acoplado a espectrômetro de massas (CG/EM). Os resultados foram submetidos à análise de variância (Teste F) e as médias comparadas pelo teste de Tukey. Não houve diferença estatística para produção de massa foliar, teor de óleo essencial e produtividade de óleo essencial em massa fresca e seca. No entanto, entre os compostos majoritários do óleo essencial das folhas, carvona e limoneno, a melhor produtividade de carvona foi obtida às 10:00 h, em matéria fresca (2,050 L ha-1) e em matéria seca (2,068 L ha-1), e para o limoneno às 16:00 h, em matéria fresca (1,068 L ha-1) e em matéria seca (1,060 L ha-1).

O yacon (Polymnia sonchifolia Poep. Endl.) é uma espécie da família Asteraceae que apresenta um complexo sistema subterrâneo. Suas raízes tuberosas e rizóforos contêm grandes quantidades de frutose e glicose livres, além de fruto-oligossacarídeos do tipo inulina como carboidrato de reserva. Vem despertando interesse principalmente por suas propriedades medicinais, sendo utilizado como auxiliar no tratamento contra diabetes e colesterol. Foi introduzido no Brasil por volta de 1989, porém somente em 1994 iniciaram-se os primeiros cultivos comerciais. Atualmente é cultivado na região de Capão Bonito (SP), a partir de rizóforos pesando de 60 a 80 g. Estes são plantados em canteiros de 0,30 - 0,40 m de altura por 1,0 m de base, em um espaçamento de 1,00 x 0,90 m. O pH do solo é ajustado para 6,0 e a fertilização básica é realizada com NPK + Zn, de acordo com análise e a recomendação utilizada para batata doce. Posteriormente são aplicados 40 kg.ha-1 de N em duas parcelas. A irrigação é feita por aspersão e a colheita realizada entre os 8 e 10 meses após o plantio, obtendo-se um rendimento médio de 80 t.ha-1 de raízes e 1 t.ha-1 de folhas desidratadas. Tanto as raízes como as folhas podem ser consumidas frescas ou desidratadas em estufas com ventilação forçada, à temperatura máxima de 50°C, para se evitar a degradação dos carboidratos de reserva e das substâncias do metabolismo secundário.

Endophytic fungi are a rich source of new and biologically active natural products. They colonize a relatively unexplored ecological habitat and their secondary metabolism is particularly active, presumably due to metabolic interactions with their hosts. In the course of our continuing investigations for new and bioactive compounds from endophytic fungi from brazilian flora Alibertia macrophylla, Caseria sylvestris, Ocotea corymbosa, Cassia spectabilis, Piper aduncum, Cryptocaria mandioccana, Xylopia aromatica and Palicourea marcgravii were investigated. Forty two natural products were isolated and their structures were established on the basis of comprehensive spectral analysis, mainly using 1D and 2D NMR experiments. The compounds were tested in their antifungal, antioxidant, anticholinesterasic and anticancer activities.

Elicitation with chitosan is a tool used to improve the responses of plant defense by inducing secondary metabolism routs. In addition, the adequate application of this elicitor on medicinal plants can promote the increase of major components in the composition of the oil. However, we lack information concerning which are the main physiological processes responsible for the changes in the composition of the oil. Thus, we aimed at evaluating the action of chitosan and determine an ideal concentration for optimizing the production of essential oil in Achillea millefolium L. and the changes in the physiological processes responsible for this increase. The research was conducted in greenhouse of the Plant Physiology sector of the Universidade Federal de Lavras (UFLA) with A. millefolium plants. The treatments consisted of control (water); acetic acid solvent (with no elicitor); and chitosan in the concentrations of 2, 4 and 6 g L -1 . The measurements of growth, gas exchange, enzyme activity of the antioxidant system and phenylalanine ammonia lyase (PAL), in addition to the production and composition of the essential oil. We verified that the application of chitosan promoted decrease in plant growth. However, the concentration of 4 g L -1 of chitosan induced an increase in the content and yield of the essential oil. In the oil, there was predominance of sesquiterpenic compounds, including the major compounds borneol, β-caryophyllene, β-cubebene, α-farnesene and chamazulene. The elicitation with chitosan at 4 g L -1 promoted an increase of the photosynthetic rate, activity of the antioxidant system and of PAL, however, this increase occurred in short-term, only in the first days after elicitation.
