The protective effect of various Salmonella vaccines regimens against an experimental Salmonella Gallinarum challenge (SGNalr strain at 12 wk of age) was evaluated in two experiments. In Experiment 1 commercial brown layers were vaccinated according to one of the following programs: (i) two doses of a SE bacterin (Layermune SE; group 1); (ii) a first dose of a live SG9R vaccine (Cevac SG9R) followed by a SE bacterin (Layermune SE; group 2); (iii) one dose of each of two different multivalent inactivated vaccines containing SE cells (Corymune 4 & Corymune 7; group 3) or (iv) not vaccinated (group 4). In Experiment 2, broiler breeders were given the same vaccination treatments except for the group vaccinated with the multivalent vaccines. Overall, in both experiments, all vaccination schemes were effective in reducing mortality after challenge with a SG field strain. Primary vaccination with an initial dose of a live SG9R vaccine followed some weeks later by a dose of an inactivated SE bacterin was the most effective (p<0.05) vaccination program against mortality induced by field SG experimental challenge in both experiments. In conclusion, Salmonella vaccination programs containing SE bacterins alone or in combination with a live SG9R vaccine are effective in preventing mortality induced by infection of field SG. Nevertheless, it is important to emphasize that any vaccination program against any Salmonella serotype will only be effective if it is part of a sound and comprehensive biosecurity program designed for Salmonella control in poultry farms.

Carbohydrates and cultures of faecal microflora were administered to newly hatched chicks to prevent infection with Salmonella typhimurium Salmonella enteritidis, Salmonella agona and Salmonella infantis. Birds were killed 72 hours after challenge and the number of viable Salmonella organisms in their caecal contents estimated. Carbohydrates did not promote efficient control of infection with the Salmonella serotypes tested whereas cultures of faecal microflora completely prevented infection with all serotypes.

Salmonella sp. é um dos principais microrganismos causadores de surtos de enfermidades transmitidas por alimentos associados ao consumo de ovos e de alimentos formulados com este ingrediente. Ovos desidratados são largamente utilizados pelas indústrias de alimentos, por oferecer maior praticidade e maior padronização em relação ao produto \"in natura\". Apesar do processo tecnológico de desidratação do ovo incluir uma etapa de pasteurização, existe um risco de haver microrganismos sobreviventes, já que a pasteurização é feita em temperatura branda. Além disso, a pasteurização pode destruir os fatores intrínsecos antimicrobianos presentes na clara, possibilitando a multiplicação de microrganismos que sobreviveram ao processo de pasteurização ou que contaminaram o produto após a pasteurização. O controle da Aa do produto desidratado e o tempo de armazenamento são, portanto, fatores fundamentais para o controle da multiplicação de microrganismos indesejáveis. Nesse estudo, avaliou-se a cinética de multiplicação de Salmonella experimentalmente adicionada a ovo em pó Aa ajustada para 0,4, 0,6, 0,8 e 0,9, durante o armazenamento em quatro temperaturas: 8°C, 15°C, 25°C e 35°C. Os resultados indicaram que S. Enteritidis é capaz de sobreviver por longo tempo (pelo menos 56 dias) em ovo em pó com Aa próximo de 0,4 quando armazenado a 8°C, 15° e a 25°C. Essa sobrevivência é menor (até 28 dias) quando o armazenamento é feito a 35°C. No ovo em pó com Aa em tomo de 0,6 ou 0,8, S. Enteritidis sobrevive por menos tempo do que no produto com Aa de cerca de 0,4, independentemente da temperatura de armazenamento. No produto com Aa de cerca de 0,9, há grande multiplicação de S. Enteritidis quando o armazenamento é feito a 15°C, 25°C ou 35°C. Nesse produto, o armazenamento a 8°C impede a multiplicação do patógeno. Verificou-se também que Salmonella Radar, resistente a diversos antibióticos, apresentou o mesmo comportamento que S. Enteritidis nas amostras de ovo estudadas.

Shrimp are an important commodity in the international fisheries trade and there is an indication of an increase in worldwide consumption of this crustacean. Salmonella and Listeria have been isolated from shrimps and shrimp products on a regular basis since the 1980s. The continued reporting of the presence of these pathogens in fresh and frozen shrimps, and even in the lightly preserved and ready-to-eat products, indicates that the existing practices used by the manufacturers or processors are insufficient to eliminate these pathogens. This paper reviews the information available on Salmonella and Listeria in shrimp and makes recommendations on control options and avenues for future research in order to improve shrimp safety and quality.

Campylobacter jejuni is a zoonotic bacterial pathogen of worldwide importance. It is estimated that 460,000 human infections occur in the United Kingdom per annum and these involve acute enteritis and may be complicated by severe systemic sequelae. Such infections are frequently associated with the consumption of contaminated poultry meat and strategies to control C. jejuni in poultry are expected to limit pathogen entry into the food chain and the incidence of human disease. Toward this aim, a total of 840 Light Sussex chickens were used to evaluate a Salmonella enterica serovar Typhimurium ΔaroA vaccine expressing the C. jejuni amino acid binding protein CjaA as a plasmid-borne fusion to the C-terminus of fragment C of tetanus toxin. Chickens were given the vaccine at 1-day-old and two weeks later by oral gavage, then challenged after a further two weeks with C. jejuni. Across six biological replicates, statistically significant reductions in caecal C. jejuni of c. 1.4 log10 colony-forming units/g were observed at three and four weeks post-challenge relative to age-matched unvaccinated birds. Protection was associated with the induction of CjaA-specific serum IgY and biliary IgA. Protection was not observed using a vaccine strain containing the empty plasmid. Vaccination with recombinant CjaA subcutaneously at the same intervals significantly reduced the caecal load of C. jejuni at three and four weeks post-challenge. Taken together these data imply that responses directed against CjaA, rather than competitive or cross-protective effects mediated by the carrier, confer protection. The impact of varying parameters on the efficacy of the S. Typhimurium ΔaroA vaccine expressing TetC-CjaA was also tested. Delaying the age at primary vaccination had little impact on protection or humoral responses to CjaA. The use of the parent strain as carrier or changing the attenuating mutation of the carrier to ΔspaS or ΔssaU enhanced the protective effect, consistent with increased invasion and persistence of the vaccine strains relative to the ΔaroA mutant. Expression in the ΔaroA strain of a TetC fusion to Peb1A, but not TetC fusions to GlnH or ChuA, elicited protection against intestinal colonisation by C. jejuni that was comparable to that observed with the TetC-CjaA fusion. Our data are rendered highly relevant by use of the target host in large numbers and support the potential of CjaA- and Peb1A-based vaccines for control of C. jejuni in poultry. © 2009 Elsevier Ltd. The authors previously reported increased expression of the Salmonella enterica serovar Typhi (S. typhi) rfaH gene when the bacterial cells reach stationary phase. In this study, using a lacZ fusion to the rfaH promoter region, they demonstrate that growth-dependent regulation of rfaH expression occurs at the level of transcription initiation. It was also observed that production of the lipopolysaccharide (LPS) O-antigen by S. typhi Ty2 correlated with the differential expression of rfaH during bacterial growth. This was probably due to the increased cellular levels of RfaH, since expression of the distal gene in the O-antigen gene cluster of S. typhi Ty2, wbaP, was also increased during stationary growth, as demonstrated by RT-PCR analysis. Examination of the sequences upstream of the rfaH coding region revealed homologies to potential binding sites for the RcsB/RcsA dimer of the RcsC/YopJ/RcsB phosphorelay regulatory system and for the RpoN alternative sigma factor. The expression of the rfaH gene in rpoN and rcsB mutants of S. typhi Ty2 was measured. The results indicate that inactivation of rpoN, but not of rcsB, suppresses the growth-phase-dependent induction of rfaH expression. Furthermore, production of beta-galactosidase mediated by the rfaH-lacZ fusion increased approximately fourfold when bacteria were grown in a nitrogen-limited medium. Nitrogen limitation was also shown to increase the expression of the O-antigen by the wild-type S. typhi Ty2, as demonstrated by a similar electrophoretic profile to that observed during the stationary phase of growth in rich media. It is therefore concluded that the relationship between LPS production and nitrogen limitation parallels the pattern of rfaH regulation under the control of RpoN and is consistent with the idea that RpoN modulates LPS formation via its effect on rfaH gene expression during bacterial growth.

Biocides play an essential role in limiting the spread of infectious disease. The food industry is dependent on these agents, and their increasing use is a matter for concern. Specifically, the emergence of bacteria demonstrating increased tolerance to biocides, coupled with the potential for the development of a phenotype of cross-resistance to clinically important antimicrobial compounds, needs to be assessed. In this study, we investigated the tolerance of a collection of susceptible and multidrug-resistant (MDR) Salmonella enterica strains to a panel of seven commercially available food-grade biocide formulations. We explored their abilities to adapt to these formulations and their active biocidal agents, i.e., triclosan, chlorhexidine, hydrogen peroxide, and benzalkonium chloride, after sequential rounds of in vitro selection. Finally, cross-tolerance of different categories of biocidal formulations, their active agents, and the potential for coselection of resistance to clinically important antibiotics were investigated. Six of seven food-grade biocide formulations were bactericidal at their recommended working concentrations. All showed a reduced activity against both surface-dried and biofilm cultures. A stable phenotype of tolerance to biocide formulations could not be selected. Upon exposure of Salmonella strains to an active biocidal compound, a high-level of tolerance was selected for a number of Salmonella serotypes. No cross-tolerance to the different biocidal agents or food-grade biocide formulations was observed. Most tolerant isolates displayed changes in their patterns of susceptibility to antimicrobial compounds. Food industry biocides are effective against planktonic Salmonella. When exposed to sublethal concentrations of individual active biocidal agents, tolerant isolates may emerge. This emergence was associated with changes in antimicrobial susceptibilities.

Chilled breasts of chicken were inoculated with Salmonella infantis or Pseudomonas aeruginosa and then given one of the following treatments: (i) exposure to gaseous ozone (>2000 ppm for up to 30 min); (ii) storage under 70% CO2:30% N-2; and (iii) exposure to gaseous ozone (>2000 ppm for 15 min) followed by storage under 70% CO2:30% N-2; all storage at 7degreesC. Gaseous ozone reduced the counts of samnonellae by 97(Y,, and pseudomonads by 95%, but indigenous coliforms were unaffected. Under the modified atmosphere, the cell count of S. infantis was reduced by 72% following initial exposure and then stabilised, coliforms grew, but Ps. aeruginosa behaved like S. infantis-initial reduction (58%) followed by stability. Exposure to gaseous ozone followed by gas packaging allowed survival of S. infantis, Ps. aeruginosa and coliforms over 9 days at 7degreesC, but there was no evidence of any sensory deterioration. It is proposed that the latter treatment could, in a modified form perhaps, be used to reduce the contamination of chicken carcasses with salmonellae and improve their shelf-life. Experiments were carried out on the antibacterial effects of a commercial formic acid-propionic acid mixture (Bio-add(TM)) against different Salmonella serptj pes. The preparation exerted a strong antibacterial effect on S. typhimurium strain F98 in artificially contaminated feed. After 28 days storage, the bactericidal effect was still considerable. When chickens were reared on feed that had been treated with Bio-add(TM) and artificially contaminated with different serotypes, S. enteritidis, S. typhimurium and S. agona were not isolated from the caecal contents, but S. infantis was. No organisms of this strain were isolated when a lower feed-contamination rate of bacteria was used.

Freshly hatched chickens show a very high susceptibility to Salmonella infections and control measures are therefore frequently focused on the period shortly after hatching. Experimental investigations using one strain against itself, differentiated by different antibiotic resistance markers, have shown that colonisation with Salmonella prevents the establishment of subsequently inoculated challenge organisms in the chicken gut. The inhibition effect lasts for several days and is detectable even when a challenge dose of 10(8) organisms is used. It is independent of the breed of bird. Chickens colonised with Salmonella shed a subsequently inoculated challenge strain with significant lower numbers for several weeks than do non colonised control birds. The phenomenon is strain specific but not serovarspecific as has been shown in investigations using different strains of the same and other serovars for colonisation and challenge. The phenomenon shows a large variability between strains. Using other Enterobacteriaceae strains comparable inhibition against Salmonella was not observed.One important topic for further investigation is the capability of Salmonella live Vaccines given orally to establish a protection effect, based on the inhibition phenomenon in the first few days of live, developing into a long-lasting immunity when the birds reach immunological maturity.
