This investigation has resulted in the chemical identification and isolation of the egg-laying hormone from Aplysia californica, Aplysia vaccaria, and Aplysia dactylomela. The hormone, which was originally identified as the Bag Cell-Specific protein (BCS protein) on polyacrylamide gels, is a polypeptide of molecular weight ≈ 6000, which is localized in the neurosecretory bag cells of the parietovisceral ganglion and the surrounding connective tissue sheath which contains the bag cell axons. All three species produce a hormone of similar molecular weight, but varying electrophoretic mobility as determined on polyacrylamide gels. As tested, the hormone is completely cross-reactive among the three species.

Although the bag cells of sexually immature animals contain the active hormone, sexual maturation of the animal results in a 10-fold increase in the BCS protein content of these neurons.

A seasonal variation in the BCS protein content was also observed, with 150 times more hormone contained in the bag cells of Aplysia californica in August than in January. This correlates well with the variation in the animals' ability to lay eggs throughout the year (Strumwasser et al., 1969). There are some indications that the receptivity of the animal to the available hormone also fluctuates during the year, being lower in winter than in swmner. The seasonal rhythm of the other species, Aplysia vaccaria and Aplysia dactylomela, has not been investigated.

A polyacrylamide gel electrophoresis analysis of water-soluble proteins in Aplysia californica revealed several other nerve-specific proteins. One of these is also located in the bag cell somas and stains turquoise with Amido Schwarz. The function of this protein has not been investigated.

Chronic excessive alcohol intoxications evoke cumulative damage to tissues and organs. We examined prefrontal cortex (Brodmann's area (BA) 9) from 20 human alcoholics and 20 age, gender, and postmortem delay matched control subjects. H & E staining and light microscopy of prefrontal cortex tissue revealed a reduction in the levels of cytoskeleton surrounding the nuclei of cortical and subcortical neurons, and a disruption of subcortical neuron patterning in alcoholic subjects. BA 9 tissue homogenisation and one dimensional polyacrylamide gel electrophoresis (PAGE) proteomics of cytosolic proteins identified dramatic reductions in the protein levels of spectrin beta II, and alpha- and beta-tubulins in alcoholics, and these were validated and quantitated by Western blotting. We detected a significant increase in a-tubulin acetylation in alcoholics, a non-significant increase in isoaspartate protein damage, but a significant increase in protein isoaspartyl methyltransferase protein levels, the enzyme that triggers isoaspartate damage repair in vivo. There was also a significant reduction in proteasome activity in alcoholics. One dimensional PAGE of membrane-enriched fractions detected a reduction in beta-spectrin protein levels, and a significant increase in transmembranous alpha 3 (catalytic) subunit of the Na+, K+-ATPase in alcoholic subjects. However, control subjects retained stable oligomeric forms of a-subunit that were diminished in alcoholics. In alcoholics, significant loss of cytosolic alpha-and beta-tubulins were also seen in caudate nucleus, hippocampus and cerebellum, but to different levels, indicative of brain regional susceptibility to alcohol-related damage. Collectively, these protein changes provide a molecular basis for some of the neuronal and behavioural abnormalities attributed to alcoholics

Diabetes mellitus e doenças periodontais são altamente prevalentes na população mundial. Doenças periodontais (DPs) compreendem um grupo de condições crônicas inflamatórias induzidas por microorganismos que levam à inflamação gengival, à destruição tecidual periodontal e à perda óssea alveolar. Diabetes mellitus (DM) é o termo utilizado para descrever um grupo de desordens metabólicas associadas à intolerância à glicose e ao metabolismo inadequado de carboidratos. Uma vez que DPs poderiam agir de forma similar a outros estados infecciosos sistêmicos, aumentando a severidade do diabetes, uma possível relação entre ambas tem sido considerada em todo o mundo. Polimorfismos genéticos de um único nucleotídeo (SNPs) têm sido estudados em diversas doenças. Nas periodontites, acredita-se que possam estar envolvidos na exacerbação da resposta inflamatória frente ao desafio bacteriano, modificando a susceptibilidade do hospedeiro. Neste estudo, a prevalência de periodontite foi avaliada em portadores de diabetes mellitus tipo I. Posteriormente, o SNP localizado na região promotora do gene TNFA (-1031T>C) foi analisado e sua importância para a doença periodontal destrutiva foi avaliada. O grupo teste foi constituído por diabéticos tipo I (DGT, n=113) enquanto o grupo controle por indivíduos não diabéticos (ND, n=73). Para as análises dos polimorfismos genéticos, um subgrupo foi retirado do grupo teste (DG, n=58) e comparado ao grupo ND. Os seguintes parâmetros clínicos e demográficos foram avaliados: percentual de sítios com profundidade de bolsa  6,0 mm (%PBS6,0 mm); índice gengival (IG); perda óssea radiográfica (POR); fumo; duração do diabetes ; idade; índice de massa corpórea (IMC), n de internações e n de dentes presentes. Amostras de sangue e/ou esfregaço bucal foram colhidas de 58 pacientes do grupo teste e de 73 controles. Após a extração do DNA genômico e amplificação da região genômica de interesse por PCR (Polymerase Chain Reaction), o polimorfismo TNFA 1031T>C foi analisado por BbsI RFLP (Restriction Fragment Length Polymorphism). A análise dos produtos de digestão foi feita por eletroforese em gel de poliacrilamida 8%. A análise estatística das freqüências alélica e genotípica juntamente com os dados clínicos e epidemiológicos entre os 2 grupos foi feita através do teste do Mann-Whitney e do Qui-quadrado. Os grupos de estudo obedecem ao princípio de Hardy-Weinberg. No grupo ND, as seguintes freqüências genotípicas foram encontradas: 78,1% (T/T); 20,5% (T/C) e 1,4% (C/C) enquanto no grupo D foram: 42,4%(T/T); 37,3% (T/C) e 20,3% (C/C). A frequência do alelo T no grupo diabético (D) foi de 0,610 ao passo que no grupo ND foi de 0,883. Não foi possível encontrar uma relação entre o polimorfismo -1031 T>C do gene TNFA e a presença de periodontite em diabéticos tipo I. Entretanto, o polimorfismo estudado se mostrou significativamente relacionado (p<0,0001 e OR= 4.85 95%IC 2,271-10,338) à presença do diabetes tipo I.

Enterococcus faecium tem se destacado no cenário das infecções hospitalares, particularmente, amostras portadoras de características de multirresistência. Apesar de serem responsabilizados como agentes etiológicos em diferentes quadros clínicos, fatores associados a patogênese das infecções ainda não estão esclarecidos. Entretanto, sabe-se que a capacidade de formação de biofilmes pode ser responsabilizada como um dos atributos capazes de promover o papel patogênico desses microrganismos. Assim sendo, este estudo se propôs a investigar a capacidade de formação de biofilmes por duas amostras de E. faecium: SS-1274 (derivada da amostra tipo da espécie) e CL-6729 (multirresistente e pertencente a um complexo clonal globalmente disperso). As amostras foram caracterizadas fenotipica e genotipicamente para confirmação da identificação, determinação da susceptibilidade a 17 antimicrobianos, caracterização da concentração mínima inibitória para ampicilina, gentamicina e vancomicina, determinação do genótipo vanA e detecção de genes associados a expressão dos genes asa1, cylA, esp, gelE e hyl. A análise quantitativa da formação por 24h e 72h dos biofilmes foi realizada por metodologia do cristal violeta, em placas de microtitulação de poliestireno. Foram construídas curvas de formação pela avaliação da DO570nm das preparações coradas por cristal violeta em períodos de tempo de 2h a 74h. A influência de subCMIs de ampicilina, gentamicina e vancomicina na formação de biofilmes de E. faecium foi também caracterizada em ensaios de quantificação da biomassa. A presença de DNA e proteínas foi avaliada em ensaios de destacamento e por fluorimetria. A arquitetura, distribuição espacial e reação aos fluorocromos Syto9 e SYPRO Ruby Protein foram evidenciadas por microscopia confocal de varredura a laser. Análises proteômicas através da avaliação por SDS-PAGE e por ESI-Q-TOF também foram empregadas para avaliação de biofilmes versus crescimento planctônico. Nossos resultados demonstraram que a amostra CL-6729 apresentou uma maior biomassa nos tempos analisados. Apesar da menor quantidade de biomassa da amostra SS-1274, o perfil da curva de formação foi semelhante a CL-6729. As análises da matriz por ensaios quantitativos e microscopia confocal revelaram que proteína parece ser um importante constituinte para ambas as amostras, entretanto biofilmes formados na presença de da CMI e durante 24h, parecem sofrer alterações na constituição. Os resultados relativos às espectrometrias de massa sugerem que células de biofilmes de E. faecium podem também estar metabolicamente ativas, devido a identificação de um considerável número de proteínas relacionadas ao metabolismo e divisão celular, similarmente às células planctônicas. No entanto, investigações complementares são necessárias para quantificar as diferenças relacionadas a sua expressão. Foi observado que ambas as amostras independente das variações fenotípicas e genotípicas são capazes de formar biofilmes maduros exibindo constituição e arquitetura similares. Entretanto, nossos resultados sugeriram que cada amostra responde as diferentes situações de acordo com seus determinantes de virulência e resistência.

植物与昆虫的互作关系是个长期进化的过程,虫害给农业生产带来巨大损失。本研究以甘蓝型油菜(Brassica napus)为例,研究了不同环境条件和遗传背景下外源基因的表达与效用,同时利用蛋白质组技术,研究了虫害损伤模拟条件下植物可能存在的内源抗性机制。甘蓝型油菜中转入了人工合成的Bt(Bacillus thuringiensis)杀虫基因,能使植物产生抗虫蛋白抵御虫害。我们在湖北湖南两个实验点进行了大田实验,按植株生长发育的4个不同时期从转基因植株的叶片上采样,研究抗虫蛋白在植物体内的表达动态。植株顶部第三片展开叶的Bt毒蛋白浓度在结荚期前随植物生长而不断增加,而在结荚期出现或增或减的现象。采样叶片的可溶性总蛋白浓度含量一直呈增加的趋势,直到结荚以后出现含量的明显降低。同时,收集了转基因油菜与湘油15号在田间自然杂交形成的杂交后代种子用于栽培,用GFP仪检测杂交后代的绿色荧光蛋白(green fluorescent protein),并用聚合酶链式反应(polymerase chain reaction, PCR)检测并确认带有转基因的杂交植株。为了检测带有转基因的杂交后代油菜中Bt毒蛋白的杀虫效率,用对Bt毒蛋白敏感的试虫品系——初孵棉铃虫幼虫(Helicoverpa armigera)进行杀虫活性检测实验。结果表明,携带Bt基因的杂交湘油及其转基因亲本对试虫的体重增长量均产生了负面影响,可以推断在调查取样的植株生长发育阶段,转基因杂交后代与其转基因亲本植株的杀虫效率没有显著差异。转基因植物及其杂交后代中抗虫蛋白的持续表达及田间带有转基因的自播植物的出现会使害虫产生耐受抗性的潜在可能性增加。 相对于人为增加的抗虫基因,植物在长期对抗昆虫的过程中也进化形成了自我防御机制,能够产生特异的抗性蛋白来应对昆虫的取食。本研究用机械损伤模拟害虫取食,对比了油菜受到物理损伤前后可溶性总蛋白的含量变化并试图通过蛋白质组学技术来检测可能发生变化的蛋白质。Bradford定量测定发现,同一植株同一叶片损伤前后可溶性总蛋白含量差异显著,损伤后蛋白表达量显著增高。蛋白质组双向凝胶电泳及其差异分析显示,损伤前后有8个蛋白质点发生明显的上调或下调。选择其中2个差异蛋白点经过MALDI-TOF质谱鉴定,它们分别是Rubisco小亚基前体以及果糖-1,6-二磷酸醛缩酶和粪卟啉-3-氧化酶的混合物,这些蛋白质在其他植物的抗逆研究中也有报道,它们可能在油菜叶片应答机械损伤过程中对维持植物的生理功能也有重要作用。

A specific blood coagulation factor X activator was purified from the venom of Ophiophagus hannah by gel filtration and two steps of FPLC Mono-Q column ion-exchange chromatography. It showed a single protein band both in sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and alkaline polyacrylamide gel electrophoresis. The mol. wt was estimated to be 62,000 in non-reducing conditions and 64,500 in reducing conditions by SDS-PAGE. The isoelectric point was found to be pH 5.6. The enzyme had weak amidolytic activities toward CBS 65-25, but it showed no activities on S-2266, S-2302, thrombin substrate S-2238, plasmin substrate S-2251 or factor Xa substrate S-2222. It had no arginine esterase activity toward substrate benzoylarginine ethylester (BAEE). The enzyme activated factor X in vitro and the effect was absolutely Ca2+ dependent, with a Hill coefficient of 6.83. It could not activate prothrombin nor had any effect on fibrinogen and thus appeared to act specifically on factor X. The procoagulant activity of the enzyme was almost completely inhibited by serine protease inhibitors like PMSF, TPCK and soybean trypsin inhibitor; partially inhibited by L-cysteine. Metal chelator EDTA did not inhibit its procoagulant activity. These results suggest that the factor X activator from O. hannah venom is a serine protease.

Stejnulxin, a novel snake C-type lectin-like protein with potent platelet activating activity, was purified and characterized from Trimeresurus stejnegeri venom. Under non-reducing conditions, it migrated on a SDS-polyacrylamide gel with an apparent molecular mass of 120 kDa. On reduction, it separated into three polypeptide subunits with apparent molecular masses of 16 kDa (alpha), 20 kDa (beta(1)) and 22 kDa (beta(2)), respectively. The complete amino acid sequences of its subunits were deduced from cloned cDNAs. The N-terminal sequencing and cDNA cloning indicated that beta(1) and beta(2) subunits of stejnulxin have identical amino acid sequences and each contains two N-glycosylation sites. Accordingly, the molecular mass difference between 1 and 2 is caused by glycosylation heterogenity. The subunit amino acid sequences of stejnulxin are similar to those of convulxin, with sequence identities of 52.6% and 66.4% for the U. and beta, respectively. Stejnulxin induced human platelet aggregation in a dose-dependent manner. Antibodies against UNA inhibited the aggregation response to stejnulxin, indicating that activation of alpha(IIb)beta(3) and binding of fibrinogen are involved in stejnulxin-induced platelet aggregation. Antibodies against GPIbalpha or alpha(2)beta(1) as well as echicetin or rhodocetin had no significant effect on stejnulxin-induced platelet aggregation. However, platelet activation induced by stejnulxin was blocked by anti-GPVI antibodies. In addition, stejnulxin induced a tyrosine phosphorylation profile in platelets that resembled that produced by convulxin. Biotinylated stejnulxin bound specifically to platelet membrane GPVI.

A blood coagulation factor IX-binding protein (TSV-FIX-BP) was isolated from the snake venom of Trimeresurus stejnegeri. On SDS-polyacrylamide gel electrophoresis, TSV-FIX-BP showed a single band with an apparent molecular weight of 23,000 under non-reducing conditions. and two distinct bands with apparent molecular weights of 14,800 and 14,000 under reducing conditions. cDNA clones containing the coding sequences of TSV-FIX-BP were isolated and sequenced to determine the structure of the precusors of TSV-FIX-BP subunits. The deduced amino acid sequences of two subunits of TSV-FIX-BP were confirmed by N-terminal protein sequencing and trypsin-digested peptide mass fingerprinting. TSV-FIX-BP was a nonenzymatic C-type lectin-like anti-coagulant. The anti-coagulant activity of TSV-FIX-BP was mainly caused by its dose dependent interaction with blood coagulation factor IX but not with blood coagulation factor X. (C) 2003 Elsevier Science Ltd. A platelet glycoprotein Ib-binding protein, termed TSV-GPIb-BP, was isolated from the venom of Trimeresurus stejnegeri. On SDS-polyacrylamide gel electrophoresis, TSV-GPIb-BP showed a single band with an apparent molecular weight of 28,000 and two distinct bands with apparent molecular weights of 16,000 and 15,000 under non-reducing and reducing conditions, respectively. cDNA clones containing the coding sequences for both TSV-GPIb-BP subunits were isolated and sequenced. The deduced amino acid sequences of TSV-GPIb-BP subunits were confirmed by N-terminal protein sequencing and trypsin-digested peptide mass fingerprinting. Interestingly, the a subunit of TSV-GPIb-BP is identical to that of alboaggregin-B, and the sequence identity of their beta subunits is 94.3%. TSV-GPIb-BP inhibited ristocetin-induced human platelet agglutination in platelet-rich plasma under lower dosages (<5 mug/ml). On the other hand, it directly aggregated washed human platelets in the absence of additional Ca2+ or any other cofactors under higher dosages (>5 mug/ml). This platelet aggregation activity was dose-dependently inhibited by specific GPIbalpha antibodies, but not by those antibodies against platelet GPIa, GPIIa, GPIIb and GPIIIa. A phospholipase A(2) (PLA(2)) called jerdoxin, was isolated from Trimeresurus jerdonni snake venom and partially characterized. The protein was purified by three chromatographic steps. SDS-polyacrylamide gel electrophoresis in the presence or absence of dithiothreitol showed that it had a molecular mass of 15 kDa. Jerdoxin had an enzymatic activity of 39.4 mumol/min/mg towards egg yolk phosphatidyl choline (PC). It induced edema in the footpads of mice. In addition, jerdoxin exhibited indirect hemolytic activity. About 97% hemolysis was observed when 2 mug/ml enzyme was incubated for 90 min in the presence of PC and Ca2+. No detectable hemolysis was noticed when PC was not added. Ca2+ was necessary for jerdoxin to exert its hemolytic activity, since only 52% hemolysis was seen when Ca2+ was absent in the reaction mixture. Furthermore, jerdoxin inhibited ADP induced rabbit platelet aggregation and the inhibition was dose dependent with an IC50 of 1.0 muM. The complete amino acid sequence of jerdoxin deduced from cDNA sequence shared high homology with other snake venom PLA(2)s, especially the D49 PLA(2)s. Also, the residues concerned to Ca2+ binding were conserved. This is the first report of cDNA sequence of T jerdonii venom PLA(2). (C) 2002 Elsevier Science Ltd. All rights reserved.

An L-amino acid oxidase (TM-LAO) from the venom of Hunan Trimeresurus mucrosquamatus was purified to homogenicity by three steps including DEAE Sephadex A-50 ion-exchange chromatography, Sephadex G-75 gel filtration and Resourse Q ion-exchange chromatography. TM-LAO is composed of two identical subunits with a molecular weight of 55 kD by SDS-polyacrylamide gel electrophoresis. The molecular weight was different with that of LAO purified from the same species distributed in Taiwan that was 70 kD. The 24 N-terminal ammo acid sequence of TM-LAO is ADNKNPLEECFRETNYEEFLEIAR, which shares high similarity with other Viperid snake venom LAOs and has moderate similarity with Elapid snake venom LAOs. Further studies found that TM-LAO inhibited the growth of E. colt, S. aurues and B. dysenteriae. TM-LAO also showed cytotoxicity and platelet aggregation activity. All the biological activities were eliminated by catalase, a H2O2 scavenger. It shows that these biological effects are possibly due to the formation of H2O2 produced by TM-LAO.

Electrophoretic patterns of the muscle myogens of prawns and some common Indian food fishes of marine and fresh water origin were obtained on polyacrylamide gel. It was observed that the characteristic band pattern which was species specific and was not altered by storage of fish in ice could be employed as a means of identification of fish species.

The stress response, at the molecular level, of the soft corals Dendronephthya klunzingeri and Heteroxenia sp., hard corals Acropora hyacinthus and A. valenciennesi, an ascidian Symplegma sp. and sponges Latruncula cortica and Callyspongia crassa to germanium oxide (GeO sub(2)) was evaluated. Evaluation was carried out using bioindicators. such as the level of expression of each of the heat shock proteins (HSPs) and the silicatein enzyme in response to the compound. However, the expression was measured by SDS Polyacrylamide Gel Electrophoresis (SDS PAGE) and western blotting. The harmful concentration of GeO sub(2) that produced noticeable molecular changes in the studied samples during the first 6-24 hours was 6 μg/ml. The two studied soft corals as well as the ascidian responded to the harmful concentration of germanium oxide by expressing the heat-shock protein 90 (hsp90), while the two hard corals responded by expressing hsp70, C. crassa by decreasing the level of silicatein enzyme and sponge L. cortica produced no change by any of the used biomarkers, The soft coral Heteroxenia sp. was found to be sensitive to mechanical stress during the experiment and it was more sensitive to 6 μg/ml of GeO sub(2) than the other soft coral D. klunzingeri. The two studied hard corals were sensitive to mechanical stress during the experiment, but A. hyacinth us showed higher sensitivity than A. valenciennesi. However, these 2 corals displayed reverse response to GeO sub(2). Primitive evidences were found in the SDS PAGE to distinguish the tissue of the soft coral from that of the hard coral on the molecular level; the soft coral showed two prominent protein bands (45 and 50 kDa) while the two prominent protein bands for hard corals were 31 and 116 kDa.
