Subterranean mammals spend their lives in dark, unventilated environments that are rich in carbon dioxide and ammonia and low in oxygen. Many of these animals are also long-lived and exhibit reduced aging-associated diseases, such as neurodegenerative disorders and cancer. We sequenced the genome of the Damaraland mole rat (DMR, Fukomys damarensis) and improved the genome assembly of the naked mole rat (NMR, Heterocephalus glaber). Comparative genome analyses, along with the transcriptomes of related subterranean rodents, revealed candidate molecular adaptations for subterranean life and longevity, including a divergent insulin peptide, expression of oxygen-carrying globins in the brain, prevention of high CO2-induced pain perception, and enhanced ammonia detoxification. Juxtaposition of the genomes of DMR and other more conventional animals with the genome of NMR revealed several truly exceptional NMR features: unusual thermogenesis, an aberrant melatonin system, pain insensitivity, and unique processing of 28S rRNA. Together, these genomes and transcriptomes extend our understanding of subterranean adaptations, stress resistance, and longevity.

Genome maintenance (GM) is an essential defense system against aging and cancer, as both are characterized by increased genome instability. Here, we compared the copy number variation and mutation rate of 518 GM-associated genes in the naked mole rat (NMR), mouse, and human genomes. GM genes appeared to be strongly conserved, with copy number variation in only four genes. Interestingly, we found NMR to have a higher copy number of CEBPG, a regulator of DNA repair, and TINF2, a protector of telomere integrity. NMR, as well as human, was also found to have a lower rate of germline nucleotide substitution than the mouse. Together, the data suggest that the long-lived NMR, as well as human, has more robust GM than mouse and identifies new targets for the analysis of the exceptional longevity of the NMR.

Foram realizados ensaios de adensamento, SIC e CRS, em amostras retiradas de um depósito argiloso muito mole, na Baixada de Jacarepaguá, 15 anos após a execução de um aterro. As amostras foram retiradas do mesmo local onde foram obtidas as amostras da primeira campanha, por ocasião do projeto. Os ensaios CRS, realizados com diferentes velocidades de deformação, são comparados aos resultados dos ensaios SIC da campanha atual de investigação. Os parâmetros geotécnicos da camada de argila muito mole, 15 anos após a construção do aterro, são comparados aos parâmetros da camada original. O aumento das tensões de sobreadensamento e redução do OCR são obtidos da interpretação dos ensaios atuais. A grandeza do recalque foi inferida a partir da nova estratigrafia, através da espessura atual da camada na região investigada, pela variação do índice de vazios e pela variação do teor médio de umidade. Os recalques previstos originalmente, incluindo a parcela de compressão secundária, são comparados aos recalques inferidos e medidos através de placa de recalque. As principais conclusões da pesquisa sugerem que a qualidade dos corpos de prova da primeira campanha foram superiores aos atuais, apesar dos cuidados com a amostragem, transporte das amostras e preparação dos corpos de prova no laboratório na segunda campanha de ensaios. Atribuiu-se esta ocorrência ao processo construtivo, que impôs movimentação excessiva ao maciço argiloso, interferindo com suas características de maior uniformidade em seu processo de deposição natural. Os ensaios de adensamento com diferentes velocidades de carregamento apresentaram comportamento similar, com variação da posição relativa das curvas e x σv, com ensaios mais rápidos exibindo maiores índices de vazios. As curvas do índice de vazios versus tensão efetiva ilustram, de forma acentuada, a redução significativa do índice de vazios da segunda campanha em relação ao solo natural, antes do lançamento do aterro. Os recalques previstos e os obtidos, seja pela instrumentação, seja pelos demais registros, indicam valores bastante próximos, em face da variabilidade da estratigrafia e dos parâmetros geotécnicos inerentes à natureza dos depósitos sedimentares.

A argila encontrada na Barra da Tijuca, Rio de Janeiro, é conhecida por ser muito mole e de peso específico bastante baixo. Passados 17 anos após a execução de um aterro em um dos depósitos argilosos da região, foram realizados ensaios de piezocone e SPT para aferir a espessura atual da camada mole. Tendo-se as espessuras originais da camada, determinadas a partir de ensaios de palheta e SPT na época da realização da obra, foi possível,assim, determinar o recalque que realmente ocorreu. O recalque primário foi calculado através dos parâmetros de compressibilidade previamente conhecidos daquele material. Foi aplicada a correção devido à submersão do aterro e, para isso, foi desenvolvida neste trabalho uma nova metodologia de cálculo para este efeito, considerando a submersão do aterro de forma gradual em todo o seu desenvolvimento matemático. Através da diferença entre o recalque total verificado nos ensaios e o recalque primário calculado, determinou-se a magnitude do recalque secundário. Com isso, estimou-se o OCR secundário do local. Este parâmetro surge da teoria que considera que os recalques secundários também ocorrem devido à variação das tensões efetivas, mas, nesse caso, horizontais. As principais conclusões desta pesquisa apontam para um valor de OCR secundário maior do que aquele já conhecido da argila de Sarapuí, que serviu de base para a referida teoria. Além disso, a proposta de cálculo do efeito da submersão se mostrou fácil e rápida de ser aplicada e forneceu resultados próximos daqueles obtidos pelas metodologias usuais.

Cross-species painting (fluorescence in situ hybridization) with 23 human (Homo sapiens (HSA)) chromosome-specific painting probes (HSA 1-22 and the X) was used to delimit regions of homology on the chromosomes of the golden mole (Ghrysochloris asiaticus) and elephant-shrew (Elephantulus rupestris). A cladistic interpretation of our data provides evidence of two unique associations, HSA 1/19p and 5/21/3, that support Afrotheria. The recognition of HSA 5/3/21 expands on the 3/21 synteny originally designated as an ancestral state for all eutherians. We have identified one adjacent segment combination (HSA2/8p/4) that is supportive of Afroinsectiphillia (aardvark, golden mole, elephant-shrew). Two segmental combinations (HSA 10q/17 and HSA 3/20) unite the aardvark and elephant-shrews as sister taxa. The finding that segmental syntenies in evolutionarily distant taxa can improve phylogenetic resolution suggests that they may be useful for testing sequence-based phylogenies of the early eutherian mammals. They may even suggest clades that sequence trees are not recovering with any consistency and thus encourage the search for additional rare genomic changes among afrotheres.

AlInGaN quaternary epilayers with varying In mole fraction were investigated using triple-axis x-ray diffraction and photoluminescence measurements. The indium compositional fluctuation is enhanced with increasing In mole fraction, whereas the mosaicity of the AlInGaN epilayers is determined through the GaN template quality. Based on the analysis of the temperature dependence of the PL peak position, it is found that the localization effect strengthens with increasing In mole fraction due to the larger fluctuations of the In distribution. Increasing the influence of the localized state results in increasing the emission intensity and FWHM with the In content.

We have studied the optical and structural properties of InAs/GaAs QDs covered by InxGa1-xAs (0 less than or equal to x less than or equal to 0.3) layer using transmission electron microscopy, photoluminescence (PL) spectra and atomic force microscopy. We find that the strain reduces in the growth direction of InAs islands covered by InGaAs instead of GaAs layer. Significant redshift of PL peak energy and narrowing of PL linewidth are observed for the InAs QDs covered by 3 nm thick InGaAs layer. In addition, atomic force microscopy measurements indicate that the InGaAs islands will nucleate on top of InAs quantum dots, when 3 nm In0.3Ga0.7As overgrowth layer is deposited. This result can well explain the PL intensify degradation and linewidth increment of quantum dots with a higher In-mole-fraction InGaAs layer. The energy gap change of InAs QDs covered by InGaAs may be explained in terms of reducing strain, suppressing compositional mixing and increasing island height. (C) 2000 Elsevier Science B.V. Based on the n(x, lambda), the calculation of the reflection spectrum for vertical cavity surface emitting lasers shows that the deviation of the central wavelength caused by the change of layer thickness is much more than that caused by the change of AlAs mole fractions. Therefore the control of the MBE growth rate is very important.

Different reproductive strategies of males and females may lead to the evolution of differences in their energetic costs of reproduction, overall energetic requirements and physiological performances. Sexual dimorphism is often associated with costly behaviours (e.g. large males might have a competitive advantage in fighting, which is energetically expensive). However, few studies of mammals have directly compared the energy costs of reproductive activities between sexes. We compared the daily energy expenditure (DEE) and resting metabolic rate (RMR) of males and females of two species of mole-rat, Bathyergus janetta and Georychus capensis (the former is sexually dimorphic in body size and the latter is not) during a period of intense digging when males seek females. We hypothesized that large body size might be indicative of greater digging or fighting capabilities, and hence greater mass-independent DEE values in males of the sexually dimorphic species. In contrast to this prediction, although absolute values of DEE were greater in B. janetta males, mass-independent values were not. No differences were apparent between sexes in G. capensis. By comparison, although RMR values were greater in B. janetta than G. capensis, no differences were apparent between the sexes for either species. The energy cost of dimorphism is most likely to be the cost of maintenance of a large body size, and not the cost of behaviours performed when an individual is large.

Small rodents with a large surf ace-area-to-volume ratio and a high thermal conductance are likely to experience conditions where they have to expend large: amounts of energy in order to maintain a constant body temperature at low ambient temperatures. The survival of small rodents is thus dependent on their ability to reduce heat loss and increase heat production at low ambient temperatures. Two such animals are the social subterranean rodents Cryptomys damarensis (the Damaraland mole-rat) and Cryptomys hottentotus natalensis (the Natal mole-rat). This study examined the energy savings associated with huddling as a behavioural thermoregulatory mechanism to conserve energy in both these species. Individual oxygen consumption (VO2) was measured in groups ranging in size from one to 15 huddling animals for both species at ambient temperatures of 14, 18, 22, 26 and 30 degrees C.
