First published April 25, 2012; doi:10.1152/jn.00057.2012.-The involvement of glutamatergic neurotransmission in the rostral ventrolateral medulla/Botzinger/pre-Botzinger complexes (RVLM/BotC/pre-BotC) on the respiratory modulation of sympathoexcitatory response to peripheral chemoreflex activation (chemoreflex) was evaluated in the working heart-brain stem preparation of juvenile rats. We identified different types of baro- and chemosensitive presympathetic and respiratory neurons intermingled within the RVLM/BotC/pre-BotC. Bilateral microinjections of kynurenic acid (KYN) into the rostral aspect of RVLM (RVLM/BotC) produced an additional increase in frequency of the phrenic nerve (PN: 0.38 +/- 0.02 vs. 1 +/- 0.08 Hz; P < 0.05; n = 18) and hypoglossal (HN) inspiratory response (41 +/- 2 vs. 82 +/- 2%; P < 0.05; n = 8), but decreased postinspiratory (35 +/- 3 vs. 12 +/- 2%; P < 0.05) and late-expiratory (24 +/- 4 vs. 2 +/- 1%; P < 0.05; n = 5) abdominal (AbN) responses to chemoreflex. Likewise, expiratory vagal (cVN; 67 +/- 6 vs. 40 +/- 2%; P < 0.05; n = 5) and expiratory component of sympathoexcitatory (77 +/- 8 vs. 26 +/- 5%; P < 0.05; n = 18) responses to chemoreflex were reduced after KYN microinjections into RVLM/BotC. KYN microinjected into the caudal aspect of the RVLM (RVLM/pre-BotC; n = 16) abolished inspiratory responses [PN (n = 16) and HN (n = 6)], and no changes in magnitude of sympathoexcitatory (n = 16) and expiratory (AbN and cVN; n = 10) responses to chemoreflex, producing similar and phase-locked vagal, abdominal, and sympathetic responses. We conclude that in relation to chemoreflex activation 1) ionotropic glutamate receptors in RVLM/BotC and RVLM/pre-BtC are pivotal to expiratory and inspiratory responses, respectively; and 2) activation of ionotropic glutamate receptors in RVLM/BotC is essential to the coupling of active expiration and sympathoexcitatory response.

Type II DNA topoisomerases catalyse DNA double-strand cleavage, passage and re-ligation to effect topological changes. There is considerable interest in elucidating topoisomerase II roles, particularly as these proteins are targets for anti-cancer drugs. Here we uncover a role for topoisomerase IIa in RNA polymerase I-directed ribosomal RNA gene transcription, which drives cell growth and proliferation and is upregulated in cancer cells. Our data suggest that topoisomerase IIa is a component of the initiation-competent RNA polymerase Iß complex and interacts directly with RNA polymerase I-associated transcription factor RRN3, which targets the polymerase to promoter-bound SL1 in pre-initiation complex formation. In cells, activation of rDNA transcription is reduced by inhibition or depletion of topoisomerase II, and this is accompanied by reduced transient double-strand DNA cleavage in the rDNA-promoter region and reduced pre-initiation complex formation. We propose that topoisomerase IIa functions in RNA polymerase I transcription to produce topological changes at the rDNA promoter that facilitate efficient de novo pre-initiation complex formation.

Central chemoreception is the mechanism by which the brain regulates breathing in response to changes in tissue CO2/H+. Abrainstemregion called the retrotrapezoid nucleus (RTN) contains a population of CO2/H+-sensitive neurons that appears to function as an important chemoreceptor. Evidence also indicates that CO2-evoked ATP release from RTN astrocytes modulates activity of CO2/H+-sensitive neurons; however, the extent to which purinergic signalling contributes to chemoreception by RTN neurons is not clear and the mechanism(s) underlying CO2/H+-evoked ATP release is not fully elucidated. The goals of this study are to determine the extent to which ATP contributes to RTN chemoreception both in vivo and in vitro, andwhether purinergic drive to chemoreceptors relies on extracellularCa(2+) or gap junction hemichannels. We also examine the possible contribution of P2Y1 receptors expressed in theRTNto the purinergic drive to breathe. We showthat purinergic signalling contributes, in part, to the CO2/H+ sensitivity of RTN neurons. In vivo, phrenic nerve recordings of respiratory activity in adult rats show that bilateral injections of pyridoxal-phosphate-6-azophenyl-2',4'-disulfonate (PPADS, a P2 receptor blocker) decreased the ventilatory response to CO2 by 30%. In vitro, loose-patch recordings from RTN neurons show that P2 receptor blockers decreased responsiveness to both 10% and 15% CO2 also by 30%. In the slice, the contribution of purinergic signalling to RTN chemoreception did not increase with temperature (22-35 degrees C) and was retained in low extracellular Ca2+ medium. Conversely, the gap junction blockers carbenoxolone and cobalt decreased neuronal CO2/H+ sensitivity by an amount similar to P2 receptor antagonists. Inhibition of the P2Y1 receptor in the RTN had no effect on CO2 responsivness in vitro or in vivo; thus, the identity of P2 receptors underlying the purinergic component of RTN chemoreception remains unknown. These results support the possibility that CO2/H+-evoked ATP release is mediated by a mechanism involving gap junction hemichannels.

Aim: It has been suggested that the medullary raphe (MR) plays a key role in the physiological responses to hypoxia. As opioid mu-receptors have been found in the MR, we studied the putative role of opioid mu-receptors in the rostral MR (rMR) region on ventilation in normal and 7% hypoxic conditions. Methods: We measured pulmonary ventilation ((V) over dotE) and the body temperatures (Tb) of male Wistar rats before and after the selective opioid l-receptor antagonist CTAP ( d-Phe-Cys-Tyr-d-Trp-Arg-Thr-Pen-Thr-NH2, cyclic, 0.1 mu g per 0.1 mu L) was microinjected into the rMR during normoxia or after 60 min of hypoxia. Results: The animals treated with intra-rMR CTAP exhibited an attenuation of the ventilatory response to hypoxia ( 430 +/- 86 mL kg) 1 min) 1) compared with the control group ( 790 +/- 82 mL kg) 1 min) 1) ( P < 0.05). No differences in the Tb were observed between groups during hypoxia. Conclusion: These data suggest that opioids acting on l-receptors in the rMR exert an excitatory modulation of hyperventilation induced by hypoxia.

In eukaryotes, pre-rRNA processing depends on a large number of nonribosomal trans-acting factors that form intriguingly organized complexes. One of the early stages of pre-rRNA processing includes formation of the two intermediate complexes pre-40S and pre-60S, which then form the mature ribosome subunits. Each of these complexes contains specific pre-rRNAs, ribosomal proteins and processing factors. The yeast nucleolar protein Nop53p has previously been identified in the pre-60S complex and shown to affect pre-rRNA processing by directly binding to 5.8S rRNA, and to interact with Nop17p and Nip7p, which are also involved in this process. Here we show that Nop53p binds 5.8S rRNA co-transcriptionally through its N-terminal region, and that this protein portion can also partially complement growth of the conditional mutant strain Delta nop53/GAL:NOP53. Nop53p interacts with Rrp6p and activates the exosome in vitro. These results indicate that Nop53p may recruit the exosome to 7S pre-rRNA for processing. Consistent with this observation and similar to the observed in exosome mutants, depletion of Nop53p leads to accumulation of polyadenylated pre-rRNAs.

We have analysed the extent of base-pairing interactions between spacer sequences of histone pre-mRNA and U7 snRNA present in the trans-acting U7 snRNP and their importance for histone RNA 3' end processing in vitro. For the efficiently processed mouse H4-12 gene, a computer analysis revealed that additional base pairs could be formed with U7 RNA outside of the previously recognised spacer element (stem II). One complementarity (stem III) is located more 3' and involves nucleotides from the very 5' end of U7 RNA. The other, more 5' located complementarity (stem I) involves nucleotides of the Sm binding site of U7 RNA, a part known to interact with snRNP structural proteins. These potential stem structures are separated from each other by short internal loops of unpaired nucleotides. Mutational analyses of the pre-mRNA indicate that stems II and III are equally important for interaction with the U7 snRNP and for processing, whereas mutations in stem I have moderate effects on processing efficiency, but do not impair complex formation with the U7 snRNP. Thus nucleotides near the processing site may be important for processing, but do not contribute to the assembly of an active complex by forming a stem I structure. The importance of stem III was confirmed by the ability of a complementary mutation in U7 RNA to suppress a stem III mutation in a complementation assay using Xenopus laevis oocytes. The main role of the factor(s) binding to the upstream hairpin loop is to stabilise the U7-pre-mRNA complex. This was shown by either stabilising (by mutation) or destabilising (by increased temperature) the U7-pre-mRNA base-pairing under conditions where hairpin factor binding was either allowed or prevented (by mutation or competition). The hairpin dependence of processing was found to be inversely related to the strength of the U7-pre-mRNA interaction.

An early stage in thymocyte development, after rearrangement of the β chain genes of the T cell receptor (TCR), involves expression of the pre-TCR complex and accompanying differentiation of CD4−CD8− double negative (DN) cells to CD4+CD8+ double positive (DP) cells. The ZAP-70 and Syk tyrosine kinases each contain two N-terminal SH2 domains that bind phosphorylated motifs in antigen receptor subunits and are implicated in pre-T receptor signaling. However, mice deficient in either ZAP-70 or Syk have no defect in the formation of DP thymocytes. Here we show that, in mice lacking both Syk and ZAP-70, DN thymocytes undergo β chain gene rearrangement but fail to initiate clonal expansion and are incapable of differentiating into DP cells after expression of the pre-TCR. These data suggest that the ZAP-70 and Syk tyrosine kinases have crucial but overlapping functions in signaling from the pre-TCR and hence in early thymocyte development.

Computational epigenetics is a new area of research focused on exploring how DNA methylation patterns affect transcription factor binding that affect gene expression patterns. The aim of this study was to produce a new protocol for the detection of DNA methylation patterns using computational analysis which can be further confirmed by bisulfite PCR with serial pyrosequencing. The upstream regulatory element and pre-initiation complex relative to CpG islets within the methylenetetrahydrofolate reductase gene were determined via computational analysis and online databases. The 1,104 bp long CpG island located near to or at the alternative promoter site of methylenetetrahydrofolate reductase gene was identified. The CpG plot indicated that CpG islets A and B, within the island, contained 62 and 75 % GC content CpG ratios of 0.70 and 0.80–0.95, respectively. Further exploration of the CpG islets A and B indicates that the transcription start sites were GGC which were absent from the TATA boxes. In addition, although six PROSITE motifs were identified in CpG B, no motifs were detected in CpG A. A number of cis-regulatory elements were found in different regions within the CpGs A and B. Transcription factors were predicted to bind to CpGs A and B with varying affinities depending on the DNA methylation status. In addition, transcription factor binding may influence the expression patterns of the methylenetetrahydrofolate reductase gene by recruiting chromatin condensation inducing factors. These results have significant implications for the understanding of the architecture of transcription factor binding at CpG islets as well as DNA methylation patterns that affect chromatin structure.

Pore forming toxins are being classified in the protein community based on their ability of forming pores in living cell membranes. Some initial study has apparently pointed out the crystallographic pathway rather can be viewed as a structural as well as morphological changes of proteins in terms of self assembly before and during the pore formation process in surfactant medium. Being a water soluble compound, it changes its conformation and originates some pre-pore complex, which later partially goes inside the cell membrane causing a pore. The physical mechanism for this whole process is still unknown. In this study we have tried to understand these types of biological processes from physical point of view by using supported lipid bilayer as a model system.

三重基序蛋白TRIM5α(Tripartite motif protein 5 alpha)是哺乳动物细胞中一种重要的限制因子,广泛分布于各种哺乳动物细胞中。人类TRIM5α mRNA 广泛表达于人类各个组织中,并且I 型干扰素IFN-α/β/γ 均能与TRIM5α 基因启动子的ISRE 元件结合,上调TRIM5α mRNA 的表达。恒河猴(Macaca mulatta)TRIM5α 是恒河猴体内重要的限制因子。目前对恒河猴尤其是中国恒河猴TRIM5α 的组织分布以及在受到外界刺激时TRIM5α mRNA 表达量的变化研究还未见报道。本论文通过从中国恒河猴各组织中提取总RNA,以β-actin 基因作为内参照,通过逆转录PCR 检测各组织中TRIM5α mRNA 的表达。我们选择用HIV-GFP-VSVG 感染、用佛波脂(Phorbol myfismte acetate, PMA)+离子霉素(Ionomycin, Ion),CD28 抗体+CD49d 抗体分别共刺激恒河猴PBMC,研究不同刺激对中国恒河猴TRIM5α mRNA 表达量的影响。研究发现:TRIM5α mRNA 广泛表达于恒河猴各组织中,在免疫系统和泌尿生殖系统各组织,如腹淋巴结、睾丸和附睾中表达量最高,而在神经系统各组织如大脑、脊髓中表达量比较少,在其他各组织中未见明显的表达差异。此外HIV-GFP-VSVG 感染、PMA+ Ion 与CD28 抗体+CD49d 抗体分别共刺激PBMC 均能促进PBMC TRIM5α mRNA 表达量的上调。 TRIM5α 作为恒河猴体内的最主要的限制HIV-1 感染的限制因子,除了可能通过促进HIV-1 的脱壳和阻止整合前复合物PIC(pre-integration complex)入核,恒河猴TRIM5α 还能限制HIV-1 病毒颗粒的产生。在这个过程中B30.2 结构域是非必需的,而B-box2 和Coiled-Coil 结构域起着决定性的作用。因为鹰猴(Aotes trivirgatus)TRIMCyp(omTRIMCyp) 蛋白和北平顶猴(Macaca leouina) TRIMCyp(npmTRIMCyp)蛋白的B-box2 和Coiled-Coil 结构域与恒河猴TRIM5α 的B-box2 和Coiled-Coil 具有很高的同源性,我们希望了解鹰猴TRIMCyp 蛋白和北平顶猴TRIMCyp 蛋白对HIV-1 病毒颗粒的产生是否有限制作用。本论文主要通过将质粒pNL4.3 分别与质粒pLPCX 、pLPCX-npmTRIMCyp-HA 、 pLPCX-omTRIMCyp-HA和pLPCX-rhTRIM5α-HA共转染293T细胞,通过western blot 检测细胞内Gag 蛋白和TRIM5 蛋白的表达情况,研究omTRIMCyp 蛋白和 npmTRIMCyp 蛋白对HIV-1 病毒颗粒产生的限制作用。结果表明:北平顶猴 TRIMCyp 蛋白、鹰猴TRIMCyp 蛋白都能不同程度的促进HIV-1 病毒Gag 蛋白的降解。

Os estudos de maquinabilidade de biomateriais e outros materiais aplicados na área médica são extensos. Todavia, muitos destes estudos recorrem a modelos de geometria regular e operações elementares de maquinagem. Relativamente a estas, os estudos académicos atualmente disponíveis mostram que a tecnologia preferencial é o torneamento, opção que se fundamenta na simplicidade de análise (corte ortogonal). Saliente-se ainda que, neste contexto, a liga de titânio Ti-6Al-4V constitui o biomaterial mais utilizado. Numa perspetiva complementar, refira-se que as publicações científicas evidenciam que a informação disponível sobre a fresagem Ti-6Al-4V não é muito extensa e a do Co-28Cr-6Mo é quase inexistente. A presente dissertação enquadra-se neste domínio e representa mais uma contribuição para o estudo da maquinabilidade das ligas de Titânio e de crómio-cobalto. A aplicação de operações de maquinagem complexas, através do recurso a programas informáticos de fabrico assistido por computador (CAM), em geometrias complexas, como é o caso das próteses femorais anatómicas, e o estudo comparativo da maquinabilidade das ligas Co-28Cr-6Mo e Ti-6Al-4V, constituem os objetivos fundamentais deste trabalho de doutoramento. Neste trabalho aborda-se a problemática da maquinabilidade das ligas metálicas usadas nos implantes ortopédicos, nomeadamente as ligas de titânio, de crómiocobalto e os aços Inoxidáveis. Efetua-se ainda um estudo da maquinagem de uma prótese femoral com uma forma geométrica complexa, onde as operações de corte foram geradas recorrendo às tecnologias de fabrico assistido por computador (CAD/CAM). Posteriormente, procedeu-se ao estudo da maquinabilidade das duas ligas usadas neste trabalho, dando uma atenção particular à determinação das forças de corte para diferentes velocidades de corte. Para além da monitorização da evolução da força de corte, o desgaste das ferramentas, a dureza e a rugosidade foram avaliadas, em função da velocidade de corte imposta. Por fim, com base nas estratégias de maquinagem adotadas, analisa-se a maquinabilidade e selecionam-se os parâmetros de corte mais favoráveis para as ligas de Titânio e Crómio-cobalto. Os resultados obtidos mostram que a liga de crómio-cobalto induz maior valor de força de corte do que a liga de titânio. Observa-se um aumento progressivo das forças de corte quando a velocidade de corte aumenta, até atingir o valor máximo para a velocidade de corte de 80m/min, após a qual, a força de corte tende a diminuir. Apesar do fabricante das ferramentas recomendar a velocidade de corte de 50 m/min para ambos os materiais, conclui-se que a velocidade de corte de 65 m/min induz o mesmo desgaste na ferramenta de corte no caso da liga de titânio, e menor desgaste no caso da liga de crómio-cobalto.

Les toxines de l’anthrax font partie de la famille des toxines A-B dans laquelle la moitié B se fixe à la membrane de la cellule permettant par la suite la translocation de la moitié A. Dans le cas de l’anthrax, la moitié B est représentée par le Protective Antigen (PA) et la moitié A par les deux protéines Edema Factor (EF) et Lethal Factor (LF). Après le recrutement par les récepteurs cellulaires (CMG2 et TEM8), PA s’organise en heptamère. Il peut fixer jusqu'à 3 ligands (EF et LF) avant d'être endocyté. Les modèles actuels de PA suggèrent que la baisse de pH à l’intérieur des endosomes permet un changement de conformation de la forme pré-pore vers la forme pore et que les ligands EF et LF passeraient au travers le pore pour entrer dans le cytoplasme. Cependant, le diamètre du pore est environ dix fois inférieur à celui des ligands (10 Å contre 100 Å). Un processus de folding/unfolding a été proposé mais demeure controversé. Afin d'identifier le processus de passage des facteurs EF et LF dans le cytoplasme, nous avons déterminé par cryo-microscopie électronique combinée avec l’analyse d’image les structures tridimensionnelles des complexes formés par PA et LF aux étapes prépore et pore. Par la suite, une étude complémentaire par dynamique moléculaire nous a permis de modéliser à haute résolution les différentes interactions qui ont lieu au sein du complexe. La structure 3D du complexe prépore combiné à 3 LF a été déterminée à une résolution de 14 Å. Nous avons aussi calculé une structure préliminaire du complexe pore également combiné à 3 LF Celles-ci n’ont jamais été résolues auparavant et leur connaissance permet d’envisager l’étude en profondeur du mécanisme infectieux de l’Anthrax in vivo.
